    The soil organic carbon: Clay ratio in North Devon, UK: Implications for marketing soil carbon as an asset class

    Building up stocks of agricultural soil organic carbon (SOC) can improve soil conditions as well as contribute to climate change mitigation. As a metric, the ratio of SOC to clay offers a better predictor of soil condition than SOC alone, potentially providing a benchmark for ecosystem service payments. We determined SOC:clay ratios for 50 fields in the North Devon UNESCO World Biosphere Reserve using 30 cm soil cores (divided into 0-10 cm and 10-30 cm depth samples), with soil bulk density, soil moisture and land-use history recorded for each field. All the arable soils exceeded the minimum desirable SOC:clay ratio threshold, and the ley grassland soils generally exceeded it but were inconsistent at 10-30 cm. Land use was the primary factor driving SOC:clay ratios at 0-10 cm, with permanent pasture fields having the highest ratios followed by ley grass and then arable fields. Approximately half of the fields sampled had potential for building up SOC stock at 10-30 cm. However, at this depth, the effect of land use is significantly reduced. Within-field variability in SOC and clay was low (coefficient of variation was similar to 10%) at both 0-10 cm and 10-30 cm, suggesting that SOC:clay ratios precisely characterized the fields. Due to the high SOC:clay ratios found, we conclude that there is limited opportunity to market additional carbon sequestration as an asset class in the North Devon Biosphere or similar areas. Instead, preserving existing SOC stocks would be a more suitable ecosystem service payment basis

    For some female athletes, a risk exists for the development of one or more of the medical disorders that comprise the female athlete triad: the related conditions of disordered eating, amenorrhea, and osteoporosis. These disorders may lead to significant morbidity and a high rate of mortality. The female college athlete, driven to excel in her sport and pressured to fit a thin athletic image, is at risk for the development of premature osteoporosis. College counselors and advisors can be instrumental in the prevention and treatment of these problems in female col­lege athletes

    Current advisory interventions for grazing ruminant farming cannot close exceedance of modern background sediment loss – Assessment using an instrumented farm platform and modelled scaling out

    Water quality impairment by elevated sediment loss is a pervasive problem for global water resources. Sediment management targets identify exceedance or the sediment loss ‘gap’ requiring mitigation. In the UK, palaeo-limnological reconstruction of sediment loss during the 100–150 years pre-dating the post-World War II intensification of agriculture, has identified management targets (0.20−0.35 t ha−1 yr−1) representing ‘modern background sediment delivery to rivers’. To assess exceedance on land for grazing ruminant farming, an integrated approach combined new mechanistic evidence from a heavily-instrumented experimental farm platform and a scaling out framework of modelled commercial grazing ruminant farms in similar environmental settings. Monitoring (2012–2016) on the instrumented farm platform returned sediment loss ranges of 0.11−0.14 t ha−1 yr−1 and 0.21−0.25 t ha−1 yr-1 on permanent pasture, compared with between 0.19−0.23 t ha−1 yr-1 and 0.43−0.50 t ha−1 yr−1and 0.10−0.13 t ha−1 yr−1and 0.25−0.30 t ha−1 yr-1 on pasture with scheduled plough and reseeds. Excess sediment loss existed on all three farm platform treatments but was more extensive on the two treatments with scheduled plough and reseeds. Excessive sediment loss from land used by grazing ruminant farming more strategically across England, was estimated to be up to >0.2 t ha−1 yr−1. Modelled scenarios of alternative farming futures, based on either increased uptake of interventions typically recommended by visual farm audits, or interventions selected using new mechanistic understanding for sediment loss from the instrumented farm platform, returned minimum sediment loss reductions. On the farm platform these were 2.1 % (up to 0.007 t ha−1 yr−1) and 5.1 % (up to 0.018 t ha−1 yr-1). More strategically, these were up to 2.8 % (0.014 t ha−1 yr−1) and 4.1 % (0.023 t ha−1 yr−1). Conventional on-farm measures will therefore not fully mitigate the sediment loss gap, meaning that more severe land cover change is required

    Novel approaches to investigating spatial variability in channel bank total phosphorus at the catchment scale

    Phosphorus (P) is often a limiting nutrient that leads to the eutrophication of aquatic systems. While dissolved P forms are the most bioavailable, the form, mobility, transport and fate of P are directly related to its association with fine-grained riverine sediment. Therefore, to implement successful P catchment management strategies it is important to understand the relative contribution of different sediment sources to P loads across the river continuum. While agricultural topsoil and, to a lesser extent, riverbed sediment are important sources of sediment-associated P, channel banks have been shown to be an important sediment source in some catchments. However, comparatively little is known about the P concentration and corresponding spatial variability in channel bank sediment and the associated implications for catchment management. The present study examines the spatial variability of P associated with channel bank profiles within a series of three nested catchments using both non-spatial and spatial statistical methods, where for the latter, a novel spatial approach was used to estimate the spatial averages and variances of P in channel bank sediment along the stream network. Channel bank P concentrations were compared to factors such as catchment scale, stream order, land use, bank exposure and location along the stream network. Concentrations of TP ranged between 129.6 and 1206.9 mg P kg⁻¹ of which the water extractable P (WEP) content ranged from 0.01 to 0.12%. Stream order was found to influence TP concentrations, while land use and catchment scale provided only a moderate influence. This suggested that focussing channel bank sampling strategies at the largest catchment scale would capture key drivers of TP variability provided stream order is sufficiently represented. Whether the bank was had limited vegetation and was exposed and potentially eroding had a slight influence on TP variability in second-order stream banks in the larger of the two nested catchments. However, the slightly lower TP concentrations measured at these sites indicated that banks that are actually eroding may be contributing less TP than the total channel bank TP values measured across the catchments as a whole. The results of an explicitly spatial analysis demonstrated that local channel bank TP averages and TP variances vary along the stream network. Specifically, the most accurate spatial predictor of TP was local TP means with the use of ‘crow flies’ rather than stream network distances. Local TP variances were used to provide optimal designs for future channel bank TP sampling campaigns, given available resources. Throughout, both standard and outlier-resistant statistical analyses were applied to improve interpretation of the study findings

    Correlates of unintended pregnancy among currently pregnant married women in Nepal

    <p>Abstract</p> <p>Background</p> <p>Women living in every country, irrespective of its development status, have been facing the problem of unintended pregnancy. Unintended pregnancy is an important public health issue in both developing and developed countries because of its negative association with the social and health outcomes for both mothers and children. This study aims to determine the prevalence and the factors influencing unintended pregnancy among currently pregnant married women in Nepal.</p> <p>Methods</p> <p>This paper reports on data drawn from Nepal Demographic and Health Survey (NDHS) which is a nationally representative survey. The analysis is restricted to currently pregnant married women at the time of survey. Association between unintended pregnancy and the explanatory variables was assessed in bivariate analysis using Chi-square tests. Logistic regression was used to assess the net effect of several independent variables on unintended pregnancy.</p> <p>Results</p> <p>More than two-fifth of the currently pregnant women (41%) reported that their current pregnancy was unintended. The results indicate that age of women, age at first marriage, ideal number of children, religion, exposure to radio and knowledge of family planning methods were key predictors of unintended pregnancy. Experience of unintended pregnancy augments with women's age (odds ratio, 1.11). Similarly, increase in the women's age at first marriage reduces the likelihood of unintended pregnancy (odds ratio, 0.93). Those who were exposed to the radio were less likely (odds ratio, 0.63) to have unintended pregnancy compared to those who were not. Furthermore, those women who had higher level of knowledge about family planning methods were less likely to experience unintended pregnancy (odds ratio, 0.60) compared to those having lower level of knowledge.</p> <p>Conclusion</p> <p>One of the important factors contributing to high level of maternal and infant mortality is unintended pregnancy. Programs should aim to reduce unintended pregnancy by focusing on all these identified factors so that infant and maternal mortality and morbidity as well as the need for abortion are decreased and the overall well-being of the family is maintained and enhanced.</p

    SoTL Lab: Undergraduate student-faculty collaborative research in teaching and learning in CSD

    The University of Wisconsin-Eau Claire Communication Sciences and Disorders SoTL Lab was designed to provide hands-on research experiences to undergraduate students on a large scale. Student reflections on experiences within the SoTL Lab identify the value of collaboration, development of confidence, and exposure to the entire research process as key outcomes. These experiences foster development of research skills and may lead students to consider academic careers

    A randomized controlled trial of methotrexate for patients with generalized myasthenia gravis

    OBJECTIVE: To determine the steroid-sparing effect of methotrexate (MTX) in patients with symptomatic generalized myasthenia gravis (MG). METHODS: We performed a 12-month multicenter, randomized, double-blind, placebo-controlled trial of MTX 20 mg orally every week vs placebo in 50 acetylcholine receptor antibody-positive patients with MG between April 2009 and August 2014. The primary outcome measure was the prednisone area under the dose-time curve (AUDTC) from months 4 to 12. Secondary outcome measures included 12-month changes of the Quantitative Myasthenia Gravis Score, the Myasthenia Gravis Composite Score, Manual Muscle Testing, the Myasthenia Gravis Quality of Life, and the Myasthenia Gravis Activities of Daily Living. RESULTS: Fifty-eight patients were screened and 50 enrolled. MTX did not reduce the month 4-12 prednisone AUDTC when compared to placebo (difference MTX - placebo: -488.0 mg, 95% confidence interval -2,443.4 to 1,467.3, p = 0.26); however, the average daily prednisone dose decreased in both groups. MTX did not improve secondary measures of MG compared to placebo over 12 months. Eight participants withdrew during the course of the study (1 MTX, 7 placebo). There were no serious MTX-related adverse events. The most common adverse event was nonspecific pain (19%). CONCLUSIONS: We found no steroid-sparing benefit of MTX in MG over 12 months of treatment, despite being well-tolerated. This study demonstrates the challenges of conducting clinical trials in MG, including difficulties with recruitment, participants improving on prednisone alone, and the need for a better understanding of outcome measure variability for future clinical trials. CLASSIFICATION OF EVIDENCE: This study provides Class I evidence that for patients with generalized MG MTX does not significantly reduce the prednisone AUDTC over 12 months of therapy

    Sediment source fingerprinting: benchmarking recent outputs, remaining challenges and emerging themes

    Abstract: Purpose: This review of sediment source fingerprinting assesses the current state-of-the-art, remaining challenges and emerging themes. It combines inputs from international scientists either with track records in the approach or with expertise relevant to progressing the science. Methods: Web of Science and Google Scholar were used to review published papers spanning the period 2013–2019, inclusive, to confirm publication trends in quantities of papers by study area country and the types of tracers used. The most recent (2018–2019, inclusive) papers were also benchmarked using a methodological decision-tree published in 2017. Scope: Areas requiring further research and international consensus on methodological detail are reviewed, and these comprise spatial variability in tracers and corresponding sampling implications for end-members, temporal variability in tracers and sampling implications for end-members and target sediment, tracer conservation and knowledge-based pre-selection, the physico-chemical basis for source discrimination and dissemination of fingerprinting results to stakeholders. Emerging themes are also discussed: novel tracers, concentration-dependence for biomarkers, combining sediment fingerprinting and age-dating, applications to sediment-bound pollutants, incorporation of supportive spatial information to augment discrimination and modelling, aeolian sediment source fingerprinting, integration with process-based models and development of open-access software tools for data processing. Conclusions: The popularity of sediment source fingerprinting continues on an upward trend globally, but with this growth comes issues surrounding lack of standardisation and procedural diversity. Nonetheless, the last 2 years have also evidenced growing uptake of critical requirements for robust applications and this review is intended to signpost investigators, both old and new, towards these benchmarks and remaining research challenges for, and emerging options for different applications of, the fingerprinting approach

    SMI of Bcl-2 TW-37 is active across a spectrum of B-cell tumors irrespective of their proliferative and differentiation status

    The Bcl-2 family of proteins is critical to the life and death of malignant B-lymphocytes. Interfering with their activity using small-molecule inhibitors (SMI) is being explored as a new therapeutic strategy for treating B-cell tumors. We evaluated the efficacy of TW-37, a non-peptidic SMI of Bcl-2 against a range spectrum of human B-cell lines, fresh patient samples and animal xenograft models. Multiple cytochemical and molecular approaches such as acridine orange/ethidium bromide assay for apoptosis, co-immunoprecipitation of complexes and western blot analysis, caspase luminescent activity assay and apoptotic DNA fragmentation assay were used to demonstrate the effect of TW-37 on different B-cell lines, patient derived samples, as well as in animal xenograft models. Nanomolar concentrations of TW-37 were able to induce apoptosis in both fresh samples and established cell lines with IC50 in most cases of 165–320 nM. Apoptosis was independent of proliferative status or pathological classification of B-cell tumor. TW-37 was able to block Bim-Bcl-XL and Bim-Mcl-1 heterodimerization and induced apoptosis via activation of caspases -9, -3, PARP and DNA fragmentation. TW-37 administered to tumor-bearing SCID mice led to significant tumor growth inhibition (T/C), tumor growth delay (T-C) and Log10kill, when used at its maximum tolerated dose (40 mg/kg × 3 days) via tail vein. TW-37 failed to induce changes in the Bcl-2 proteins levels suggesting that assessment of baseline Bcl-2 family proteins can be used to predict response to the drug. These findings indicate activity of TW-37 across the spectrum of human B-cell tumors and support the concept of targeting the Bcl-2 system as a therapeutic strategy regardless of the stage of B-cell differentiation

    A review of source tracking techniques for fine sediment within a catchment

    Excessive transport of fine sediment, and its associated pollutants, can cause detrimental impacts in aquatic environments. It is therefore important to perform accurate sediment source apportionment to identify hot spots of soil erosion. Various tracers have been adopted, often in combination, to identify sediment source type and its spatial origin; these include fallout radionuclides, geochemical tracers, mineral magnetic properties and bulk and compound-specific stable isotopes. In this review, the applicability of these techniques to particular settings and their advantages and limitations are reviewed. By synthesizing existing approaches, that make use of multiple tracers in combination with measured changes of channel geomorphological attributes, an integrated analysis of tracer profiles in deposited sediments in lakes and reservoirs can be made. Through a multi-scale approach for fine sediment tracking, temporal changes in soil erosion and sediment load can be reconstructed and the consequences of changing catchment practices evaluated. We recommend that long-term, as well as short-term, monitoring of riverine fine sediment and corresponding surface and subsurface sources at nested sites within a catchment are essential. Such monitoring will inform the development and validation of models for predicting dynamics of fine sediment transport as a function of hydro-climatic and geomorphological controls. We highlight that the need for monitoring is particularly important for hilly catchments with complex and changing land use. We recommend that research should be prioritized for sloping farmland-dominated catchments
