    The Interactions between Insulin and Androgens in Progression to Castrate-Resistant Prostate Cancer

    An association between the metabolic syndrome and reduced testosterone levels has been identified, and a specific inverse relationship between insulin and testosterone levels suggests that an important metabolic crosstalk exists between these two hormonal axes; however, the mechanisms by which insulin and androgens may be reciprocally regulated are not well described. Androgen-dependant gene pathways regulate the growth and maintenance of both normal and malignant prostate tissue, and androgen-deprivation therapy (ADT) in patients exploits this dependence when used to treat recurrent and metastatic prostate cancer resulting in tumour regression. A major systemic side effect of ADT includes induction of key features of the metabolic syndrome and the consistent feature of hyperinsulinaemia. Recent studies have specifically identified a correlation between elevated insulin and high-grade PCa and more rapid progression to castrate resistant disease. This paper examines the relationship between insulin and androgens in the context of prostate cancer progression. Prostate cancer patients present a promising cohort for the exploration of insulin stabilising agents as adjunct treatments for hormone deprivation or enhancers of chemosensitivity for treatment of advanced prostate cancer

    Swaddling infants has been known to provide them comfort when transitioning from the womb to the world. There has been an ongoing debate of whether or not this intervention is as helpful as we have known it to be in the past. We have performed a systematic literature review to answer the question: In infants, what are the effects of swaddling on sleeping practices and comfort? We studied peer-reviewed articles and have gathered information related to answering our research question. Some research has shown that swaddling is no longer in use due to the risks it presents with. We developed a practice protocol to address the effect that swaddling has on infants. The impact of this research is to determine if swaddling is beneficial regarding the sleeping practices and comfort of infants

    Depression of the ULF geomagnetic pulsation related to ionospheric irregularities

    We consider a depression in intensity of ULF magnetic pulsations, which is observed on the ground surface due to appearance of the irregularities in the ionosphere. It is supposed that oblique Alfven waves in the ULF frequency range are downgoing from the magnetosphere and the horizontal irregularities of ionospheric conductivity are created by upgoing atmospheric gravity waves from seismic source. Unlike the companion paper by Molchanov et al. (2003), we used a simple model of the ionospheric layer but took into consideration the lateral inhomogeneity of the perturbation region in the ionosphere. It is shown that ULF intensity could be essentially decreased for frequencies f = 0.001-0.1 Hz at nighttime but the change is negligible at daytime in coincidence with observational results

    Increasing Colorectal Cancer Testing: Translating Physician Interventions Into Population-Based Practice

    Colorectal cancer (CRC) screening in the Medicare population remains low despite Medicare coverage. We describe a population-based effort to increase CRC testing of Medicare enrollees in two States through promotion and distribution of office-based tools to primary care physicians and gastroenterologists. Small increases in colonoscopy test use by primary care physicians were observed, but the differences were not statistically significant. Results in one State were stronger than the other, and two components of the intervention appeared more promising than others. Use of CRC tests can be increased, but additional approaches are needed

    Permeability Prediction in Tight Carbonate Rocks using Capillary Pressure Measurements

    The prediction of permeability in tight carbonate reservoirs presents ever more of a challenge in the hydrocarbon industry today. It is the aim of this paper to ascertain which models have the capacity to predict permeability reliably in tight carbonates, and to develop a new one, if required. This paper presents (i) the results of laboratory Klinkenberg-corrected pulse decay measurements of carbonates with permeabilities in the range 65 nD to 0.7 mD, (ii) use of the data to assess the performance of 16 permeability prediction models, (iii) the development of an improved prediction model for tight carbonate rocks, and (iv) its validation using an independent data set. Initial measurements including porosity, permeability and mercury injection capillary pressure measurements (MICP) were carried out on a suite of samples of Kometan limestone from the Kurdistan region of Iraq. The prediction performance of sixteen different percolation-type and Poiseuille-type permeability prediction models were analysed with the measured data. Analysis of the eight best models is included in this paper and the analysis of the remainder is provided in supplementary material. Some of the models were developed especially for tight gas sands, while many were not. Critically, none were developed for tight gas carbonates. Predictably then, the best prediction was obtained from the generic model and the RGPZ models (R2 = 0.923, 0.920 and 0.915, respectively), with other models performing extremely badly. In an attempt to provide a better model for use with tight carbonates, we have developed a new model based on the RGPZ theoretical model by adding an empirical scaling parameter to account for the relationship between grain size and pore throat size in carbonates. The generic model, the 28 new RGPZ Carbonate model and the two original RGPZ models have been tested against independent data from a suite of 42 samples of tight Solnhofen carbonates. All four models performed very creditably with the generic and the new RGPZ Carbonate models performing well (R2 = 0.840 and 0.799, respectively). It is clear from this study that the blind application of conventional permeability prediction techniques to carbonates, and particularly to tight carbonates, will lead to gross errors and that the development of new methods that are specific to tight carbonates is unavoidable

    The association of urinary cadmium with sex steroid hormone concentrations in a general population sample of US adult men

    <p>Abstract</p> <p>Background</p> <p>Studies investigating the association of cadmium and sex steroid hormones in men have been inconsistent, but previous studies were relatively small.</p> <p>Methods</p> <p>In a nationally representative sample of 1,262 men participating in the morning examination session of phase I (1998–1991) of the third National Health and Nutrition Examination Survey, creatinine corrected urinary cadmium and serum concentrations of sex steroid hormones were measured following a standardized protocol.</p> <p>Results</p> <p>After adjustment for age and race-ethnicity, higher cadmium levels were associated with higher levels of total testosterone, total estradiol, sex hormone-binding globulin, estimated free testosterone, and estimated free estradiol (each p-trend < 0.05). After additionally adjusting for smoking status and serum cotinine, none of the hormones maintained an association with urinary cadmium (each p-trend > 0.05).</p> <p>Conclusion</p> <p>Urinary cadmium levels were not associated with sex steroid hormone concentrations in a large nationally representative sample of US men.</p

    The long non-coding RNA GHSROS reprograms prostate cancer cell lines toward a more aggressive phenotype

    It is now appreciated that long non-coding RNAs (lncRNAs) are important players in orchestrating cancer progression. In this study we characterized GHSROS, a human lncRNA gene on the opposite DNA strand (antisense) to the ghrelin receptor gene, in prostate cancer. The lncRNA was upregulated by prostate tumors from different clinical datasets. Transcriptome data revealed that GHSROS alters the expression of cancer-associated genes. Functional analyses in vitro showed that GHSROS mediates tumor growth, migration and survival, and resistance to the cytotoxic drug docetaxel. Increased cellular proliferation of GHSROS-overexpressing PC3, DU145, and LNCaP prostate cancer cell lines in vitro was recapitulated in a subcutaneous xenograft model. Conversely, in vitro antisense oligonucleotide inhibition of the lncRNA reciprocally regulated cell growth and migration, and gene expression. Notably, GHSROS modulates the expression of PPP2R2C, the loss of which may drive androgen receptor pathway-independent prostate tumor progression in a subset of prostate cancers. Collectively, our findings suggest that GHSROS can reprogram prostate cancer cells toward a more aggressive phenotype and that this lncRNA may represent a potential therapeutic target

    Biomarkers of folate status in NHANES: a roundtable summary123456

    A roundtable to discuss the measurement of folate status biomarkers in NHANES took place in July 2010. NHANES has measured serum folate since 1974 and red blood cell (RBC) folate since 1978 with the use of several different measurement procedures. Data on serum 5-methyltetrahydrofolate (5MTHF) and folic acid (FA) concentrations in persons aged ≥60 y are available in NHANES 1999–2002. The roundtable reviewed data that showed that folate concentrations from the Bio-Rad Quantaphase II procedure (Bio-Rad Laboratories, Hercules, CA; used in NHANES 1991–1994 and NHANES 1999–2006) were, on average, 29% lower for serum and 45% lower for RBC than were those from the microbiological assay (MA), which was used in NHANES 2007–2010. Roundtable experts agreed that these differences required a data adjustment for time-trend analyses. The roundtable reviewed the possible use of an isotope-dilution liquid chromatography–tandem mass spectrometry (LC-MS/MS) measurement procedure for future NHANES and agreed that the close agreement between the MA and LC-MS/MS results for serum folate supported conversion to the LC-MS/MS procedure. However, for RBC folate, the MA gave 25% higher concentrations than did the LC-MS/MS procedure. The roundtable agreed that the use of the LC-MS/MS procedure to measure RBC folate is premature at this time. The roundtable reviewed the reference materials available or under development at the National Institute of Standards and Technology and recognized the challenges related to, and the scientific need for, these materials. They noted the need for a commutability study for the available reference materials for serum 5MTHF and FA
