    In-plane optical response of Bi2Sr2CuO6

    We report on infrared reflectivity measurements of the abab-plane response of superconducting Bi2_2Sr2_2CuO6_6 single crystals. The frequency dependent conductivity has a maximum near 700 cm1^{-1} at room temperature, which shifts to lower frequency and merges with a Drude-peak below 100 K. We attribute the unusual behaviour of the mid-infrared conductivity to low frequency transitions between electronic bands of mainly BiO character near the M\overline{M} point. The linear temperature dependence of the low-frequency resistivity can be followed down to approximately 40 K where it saturates.Comment: Revtex, 4 pages, 4 postscript figures, Phys. Rev. B, in pres

    Systematics of c-axis Phonons in the Thallium and Bismuth Based Cuprate Superconductors

    We present grazing incidence reflectivity measurements in the far infrared region at temperatures above and below Tc for a series of thallium (Tl2Ba2CuO6, Tl2Ba2CaCu2O8) and bismuth (Bi2Sr2CuO6, Bi2Sr2CaCu2O8, and Bi(2-x)Pb(x)Sr2CaCu2O8) based cuprate superconductors. From the spectra, which are dominated by the c-axis phonons, longitudinal frequencies (LO) are directly obtained. The reflectivity curves are well fitted by a series of Lorentz oscillators. In this way the transverse (TO) phonon frequencies were accurately determined. On the basis of the comparative study of the Bi and Tl based cuprates with different number of CuO2 layers per unit cell, we suggest modifications of the assignment of the main oxygen modes. We compare the LO frequencies in Bi2Sr2CaCu2O8 and Tl2Ba2Ca2Cu3O10 obtained from intrinsic Josephson junction characteristics with our measurements, and explain the discrepancy in LO frequencies obtained by the two different methods.Comment: 8 pages Revtex, 6 eps figures, 3 tables, to appear in Phys.     Genotyping by sequencing in almond : SNP discovery, linkage mapping, and marker sesign

    In crop plant genetics, linkage maps provide the basis for the mapping of loci that affect important traits and for the selection of markers to be applied in crop improvement. In outcrossing species such as almond (Prunus dulcis Mill. D. A. Webb), application of a double pseudotestcross mapping approach to the F progeny of a biparental cross leads to the construction of a linkage map for each parent. Here, we report on the application of genotyping by sequencing to discover and map single nucleotide polymorphisms in the almond cultivars "Nonpareil" and "Lauranne." Allele-specific marker assays were developed for 309 tag pairs. Application of these assays to 231 Nonpareil × Lauranne F progeny provided robust linkage maps for each parent. Analysis of phenotypic data for shell hardness demonstrated the utility of these maps for quantitative trait locus mapping. Comparison of these maps to the peach genome assembly confirmed high synteny and collinearity between the peach and almond genomes. The marker assays were applied to progeny from several other Nonpareil crosses, providing the basis for a composite linkage map of Nonpareil. Applications of the assays to a panel of almond clones and a panel of rootstocks used for almond production demonstrated the broad applicability of the markers and provide subsets of markers that could be used to discriminate among accessions. The sequence-based linkage maps and single nucleotide polymorphism assays presented here could be useful resources for the genetic analysis and genetic improvement of almond

    Scanning tunneling spectroscopy of high-temperature superconductors

    Tunneling spectroscopy played a central role in the experimental verification of the microscopic theory of superconductivity in the classical superconductors. Initial attempts to apply the same approach to high-temperature superconductors were hampered by various problems related to the complexity of these materials. The use of scanning tunneling microscopy/spectroscopy (STM/STS) on these compounds allowed to overcome the main difficulties. This success motivated a rapidly growing scientific community to apply this technique to high-temperature superconductors. This paper reviews the experimental highlights obtained over the last decade. We first recall the crucial efforts to gain control over the technique and to obtain reproducible results. We then discuss how the STM/STS technique has contributed to the study of some of the most unusual and remarkable properties of high-temperature superconductors: the unusual large gap values and the absence of scaling with the critical temperature; the pseudogap and its relation to superconductivity; the unprecedented small size of the vortex cores and its influence on vortex matter; the unexpected electronic properties of the vortex cores; the combination of atomic resolution and spectroscopy leading to the observation of periodic local density of states modulations in the superconducting and pseudogap states, and in the vortex cores.Comment: To appear in RMP; 65 pages, 62 figure

    Proteolytic cleavage of p53 mutants in response to mismatched DNA

    Interaction of p53 with mismatched DNA induces proteolytic cleavage with release of a 35-kDa protein fragment from the p53–DNA complexes. The 35-kDa cleavage product is activated for specific biochemical function(s) and may play a role in the cellular response to DNA damage (Molinari et al (1996) Oncogene13: 2077–2086; Okorokov et al (1997) EMBO J16: 6008–6017). In the present study we have asked if mutants of p53 retain the ability to undergo similar proteolytic cleavage, and compared sequence-specific ‘DNA contact’ with ‘structural’ mutants commonly found in human cancer. In addition, a series of phosphorylation site mutants were generated to investigate the possible effects of phosphorylation/dephosphorylation on the proteolytic cleavage of p53. All mutants tested bound to a mismatched DNA target in vitro. Moreover, studies in vitro and in vivo indicate that p53 mutants with intact conformational structure (as determined by immunoreactivity with PAb246 and PAb1620) retain the ability to undergo proteolytic cleavage similar, if not identical, to the wild-type p53 protein. Our results suggest that the capacity for p53 to bind mismatched DNA is independent of structural conformation of the central core domain. Proteolytic cleavage, however, is crucially dependent upon a wild-type conformation of the protein. © 1999 Cancer Research Campaig

    Ligand engagement of Toll-like receptors regulates their expression in cortical microglia and astrocytes

    BACKGROUND: Toll-like receptor (TLR) activation on microglia and astrocytes are key elements in neuroinflammation which accompanies a number of neurological disorders. While TLR activation on glia is well-established to up-regulate pro-inflammatory mediator expression, much less is known about how ligand engagement of one TLR may affect expression of other TLRs on microglia and astrocytes. METHODS: In the present study, we evaluated the effects of agonists for TLR2 (zymosan), TLR3 (polyinosinic-polycytidylic acid (poly(I:C)), a synthetic analogue of double-stranded RNA) and TLR4 (lipopolysaccaride (LPS)) in influencing expression of their cognate receptor as well as that of the other TLRs in cultures of rat cortical purified microglia (>99.5 %) and nominally microglia-free astrocytes. Elimination of residual microglia (a common contaminant of astrocyte cultures) was achieved by incubation with the lysosomotropic agent L-leucyl-L-leucine methyl ester (L-LME). RESULTS: Flow cytometric analysis confirmed the purity (essentially 100 %) of the obtained microglia, and up to 5 % microglia contamination of astrocytes. L-LME treatment effectively removed microglia from the latter (real-time polymerase chain reaction). The three TLR ligands robustly up-regulated gene expression for pro-inflammatory markers (interleukin-1 and interleukin-6, tumor necrosis factor) in microglia and enriched, but not purified, astrocytes, confirming cellular functionality. LPS, zymosan and poly(I:C) all down-regulated TLR4 messenger RNA (mRNA) and up-regulated TLR2 mRNA at 6 and 24 h. In spite of their inability to elaborate pro-inflammatory mediator output, the nominally microglia-free astrocytes (>99 % purity) also showed similar behaviours to those of microglia, as well as changes in TLR3 gene expression. LPS interaction with TLR4 activates downstream mitogen-activated protein kinase and nuclear factor-κB signalling pathways and subsequently causes inflammatory mediator production. The effects of LPS on TLR2 mRNA in both cell populations were antagonized by a nuclear factor-κB inhibitor. CONCLUSIONS: TLR2 and TLR4 activation in particular, in concert with microglia and astrocytes, comprise key elements in the initiation and maintenance of neuropathic pain. The finding that both homologous (zymosan) and heterologous (LPS, poly(I:C)) TLR ligands are capable of regulating TLR2 gene expression, in particular, may have important implications in understanding the relative contributions of different TLRs in neurological disorders associated with neuroinflammation

    Risk Factors Associated with Adverse Fetal Outcomes in Pregnancies Affected by Coronavirus Disease 2019 (COVID-19): A Secondary Analysis of the WAPM study on COVID-19

    To evaluate the strength of association between maternal and pregnancy characteristics and the risk of adverse perinatal outcomes in pregnancies with laboratory confirmed COVID-19. Secondary analysis of a multinational, cohort study on all consecutive pregnant women with laboratory-confirmed COVID-19 from February 1, 2020 to April 30, 2020 from 73 centers from 22 different countries. A confirmed case of COVID-19 was defined as a positive result on real-time reverse-transcriptase-polymerase-chain-reaction (RT-PCR) assay of nasal and pharyngeal swab specimens. The primary outcome was a composite adverse fetal outcome, defined as the presence of either abortion (pregnancy loss before 22 weeks of gestations), stillbirth (intrauterine fetal death after 22 weeks of gestation), neonatal death (death of a live-born infant within the first 28 days of life), and perinatal death (either stillbirth or neonatal death). Logistic regression analysis was performed to evaluate parameters independently associated with the primary outcome. Logistic regression was reported as odds ratio (OR) with 95% confidence interval (CI). Mean gestational age at diagnosis was 30.6\ub19.5 weeks, with 8.0% of women being diagnosed in the first, 22.2% in the second and 69.8% in the third trimester of pregnancy. There were six miscarriage (2.3%), six intrauterine device (IUD) (2.3) and 5 (2.0%) neonatal deaths, with an overall rate of perinatal death of 4.2% (11/265), thus resulting into 17 cases experiencing and 226 not experiencing composite adverse fetal outcome. Neither stillbirths nor neonatal deaths had congenital anomalies found at antenatal or postnatal evaluation. Furthermore, none of the cases experiencing IUD had signs of impending demise at arterial or venous Doppler. Neonatal deaths were all considered as prematurity-related adverse events. Of the 250 live-born neonates, one (0.4%) was found positive at RT-PCR pharyngeal swabs performed after delivery. The mother was tested positive during the third trimester of pregnancy. The newborn was asymptomatic and had negative RT-PCR test after 14 days of life. At logistic regression analysis, gestational age at diagnosis (OR: 0.85, 95% CI 0.8-0.9 per week increase; p<0.001), birthweight (OR: 1.17, 95% CI 1.09-1.12.7 per 100 g decrease; p=0.012) and maternal ventilatory support, including either need for oxygen or CPAP (OR: 4.12, 95% CI 2.3-7.9; p=0.001) were independently associated with composite adverse fetal outcome. Early gestational age at infection, maternal ventilatory supports and low birthweight are the main determinants of adverse perinatal outcomes in fetuses with maternal COVID-19 infection. Conversely, the risk of vertical transmission seems negligible

    The goal of the study is to reveal the species of Ixodid ticks in Eastern Siberia and Mongolia, having epidemiological value and pathogens that transmit to humans via their bites. The tasks is to determine ecologo-epidemiologial characteristics of the main vectors and genetic characteristics of the agents of tick-borne infections. Characterization of the materials. There are materials of the study of more than 200 000 Ixodid ticks of 4 species and their rate of infection by different pathogens with zooparasitological, epidemiological, virological, microbiological, molecular-biological standard and modified to the goals and tasks of the study. Most abundance and dangerous species is Ixodes persulcatus P.Sch. ticks, that is widespread in region investigated. The agents of known vector-borne infections in Eastern Siberia and Mongolia are tick-borne encephalitis virus, Borrelia garinii, Borrelia afze- lii, Rickettsia sibirica, R. sp. DnS14 group.C целью выявления видов иксодовых клещей, имеющих эпидемическое значение на территории Восточной Сибири и Мон- голии, и возбудителей инфекций, передающихся человеку через их укусы, были исследованы эколого-эпидемиологические и ге- нетические характеристики основных переносчиков клещевых инфекций и связанных с ними возбудителей болезней человека. В статье приведены материалы изучения более 200 тыс. экземпляров иксодовых клещей четырех видов, исследован- ных при помощи общепринятых зоопаразитологических, эпидемиологических, вирусологических, микробиологических, молекулярно-биологических и модифицированных применительно к целям и задачам исследования методов. Наиболее массовым и опасным видом является таежный клещ Ixodes persulcatus P. Sch., широко распространенный в регионе. Эпидемиологическое значение имеют также клещи видов Dermacentor nuttalli Ol., Dermacentor silvarum Ol., Hae- maphysalis concinna Koch. Возбудителями трансмиссивных инфекций в Восточной Сибири и Монголии являются вирус КЭ, боррелии, риккетсии, возможно, эрлихии и другие патогены
