    IPSC differentiation into ependymal progenitors to treat ventricular damage during hydrocephalus

    Introduction: During both obstructive congenital hydrocephalus and post-hemorrhagic hydrocephalus additional pathological events are intimately associated with their ethiology: a) a detrimental inflammatory response; b) severe damage of the underlying periventricular nervous tissue, including white matter, and c). Therapeutic approaches have been directed to overcome a) and b), however recovery of damaged neuroepithelium/ependyma is, in our present, an important therapeutic gap. Methods: Human and mouse induced pluripotent stem cells (iPSC) have been artificially differented into ependymal progenitors. Intracerebroventricular (ICV) injections of iPCS are performed ex vivo and in vivo in the damaged ventricular wall. Their integration and differentiation has been studied by immunohistochemistry and histopathological analysis. Results: Mice and human ependymal progenitors are able to integrate and differentiate into ependyma in damaged ventricular wall. Stage of ependymal differentiation by the time of the injection defined different degrees of integration.     Globalización, funcionalidad económica y estructura urbana en Guadalajara, 1990-2000

    Este trabajo tiene como propósito analizar las transformaciones que experimentaron la funcionalidad económica y la estructura urbana de la zona conurbada de Guadalajara (ZCG) en la última década del siglo XX, en el marco del proceso de globalización. Partimos del supuesto de que la integración de la economía de esta metrópoli a dicho fenómeno contribuye a que asuma las funciones de plataforma de exportación de la industria globalizada y de centro de distribución regional de mercancías producidas en otros países1. Buscamos hacer una aportación distinta a la “nueva geografía urbana” (Sassen 1999), en la que la ciudad global se ha convertido en el epicentro y escenario protagónico de la globalización. En este trabajo el objeto de estudio no es la ciudad global, sino una de ellas que ocupan una posición periférica y tienen una cobertura subnacional y local, como ocurre con la ZCG.     Traffic Crimes and risky driving: The role of personality and driving anger

    The current study aims to examine the influence of personality traits (alternative Zuckerman model) and driving anger in the explanation of risky driving style in individuals convicted for road safety offences (N = 245), using as a basis an adaptation of the context-mediated model. This is a transversal, descriptive study designed to be implemented by means of surveys, in which took part 245 men convicted of road safety offences from five prisons in Galicia (a region in northwestern Spain) took part. The average age of the participants was 38.73 years (Sx-9.61), with a range between 18 and 64 years. All participants had three or more years of driving experience. Our data shows that the Impulsive-Sensation Seeking (Imp-SS) personality trait had a direct and positive effect on dangerous driving, while the Activity (Act) trait had a direct but negative effect. The Aggression-Hostility (Agg-Host) trait, in turn, influenced the risky driving style, but not directly, but by raising driving anger levels, so it acted as a powerful mediator between the Aggression-Hostility (Agg-Hos) trait and the risky driving style. In general, our research partially replicates and expands previous findings regarding the model used, the aggression-hostility personality trait (Agg-Host) was placed in the distal context, driving anger in the proximal context, while age and personality traits Activity (Act) and Impulsive-Sensation Seeking (Imp-SS) were direct predictors. The results of this study may have practical implications for the detection and rehabilitation of offenders and penalties for road safety offences.Universidade de Vigo/CISU

    Genetic distance as an alternative to physical distance for definition of gene units in association studies

    Background: Some association studies, as the implemented in VEGAS, ALIGATOR, i-GSEA4GWAS, GSA-SNP and other software tools, use genes as the unit of analysis. These genes include the coding sequence plus flanking sequences. Polymorphisms in the flanking sequences are of interest because they involve cis-regulatory elements or they inform on untyped genetic variants trough linkage disequilibrium. Gene extensions have customarily been defined as ±50 Kb. This approach is not fully satisfactory because genetic relationships between neighbouring sequences are a function of genetic distances, which are only poorly replaced by physical distances. Results: Standardized recombination rates (SRR) from the deCODE recombination map were used as units of genetic distances. We searched for a SRR producing flanking sequences near the ±50 Kb offset that has been common in previous studies. A SRR≥2 was selected because it led to gene extensions with median length=45.3 Kb and the simplicity of an integer value. As expected, boundaries of the genes defined with the ±50 Kb and with the SRR≥2 rules were rarely concordant. The impact of these differences was illustrated with the interpretation of top association signals from two large studies including many hits and their detailed analysis based in different criteria. The definition based in genetic distance was more concordant with the results of these studies than the based in physical distance. In the analysis of 18 top disease associated loci form the first study, the SRR≥2 genes led to a fully concordant interpretation in 17 loci; the ±50 Kb genes only in 6. Interpretation of the 43 putative functional genes of the second study based in the SRR≥2 definition only missed 4 of the genes, whereas the based in the ±50 Kb definition missed 10 genes. Conclusions: A gene definition based on genetic distance led to results more concordant with expert detailed analyses than the commonly used based in physical distance. The genome coordinates for each gene are provided to maintain a simple use of the new definitions

    Psychological Profile and Consumption of Healthcare Resources in Actively Employed People, Pre-Retirees, and Retirees

    Research on personality variables and consumption of health services in actively employed people or retirees has been extensive, but the group of pre-retirees has hardly been studied. This work attempts to determine the mental health and use of health resources of the three groups, hypothesizing that, as the group of pre-retirees does not receive the social stigma of work exclusion, it will present better mental health. A sample of 1332 Spanish participants aged between 51 and 69 years was randomly chosen, and various personality tests were applied, and consumption of healthcare resources was determined. Pairwise analysis of the three work situations using binary logistic regressions showed that pre-retirees present better mental health, although there were no differences in the consumption of healthcare resources. The implications of this study for the creation of health promotion policies targeting older people, depending specifically on their employment status, are discussedS

    El presente artículo analiza la influencia del deporte en la reinserción social de la población reclusa, centrándose en los resultados obtenidos a través de un estudio realizado por la Universidad Pablo de Olavide (UPO) mediante una ayuda concedida por el Consejo Superior de Deportes (CSD) (Ref. 38/UPB20/10). Para su desarrollo, se llevó a cabo un conjunto de entrevistas individuales en profundidad a informantes cualificados y microrrelatos de vida entre reclusos de los centros penitenciarios de Andalucía. Los resultados confirman la importancia del deporte en la rehabilitación social de la población reclusa, así como su influencia sobre la salud física, psíquica y emocional.This paper is an analysis of the influence of sports on the social integration of inmates in Andalusian prisons, focusing on results obtained from a study carried on by the University Pablo de Olavide, through a grant awarded by the Consejo Superior de Deportes (High Council of Sports). It was made through a series of in-depth individual interviews with qualified informants and microstories of inmates in such prisons. The results confirm the importance of sports in socialization and social rehabilitation of inmates, as well as its influence on the physical health, psychic and emotional changes in attitudes and values for the social integration of this population

    Approaching In Vivo Models of Pneumococcus–Host Interaction: Insights into Surface Proteins, Capsule Production, and Extracellular Vesicles

    Infections caused by the Gram-positive bacterium Streptococcus pneumoniae have become a major health problem worldwide because of their high morbidity and mortality rates, especially in developing countries. This microorganism colonizes the human upper respiratory tract and becomes pathogenic under certain circumstances, which are not well known. In the interaction with the host, bacterial surface structures and proteins play major roles. To gain knowledge into gradual changes and adaptive mechanisms that this pathogen undergoes from when it enters the host, we mimicked several in vivo situations representing interaction with epithelial and macrophage cells, as well as a condition of presence in blood. Then, we analyzed, in four pneumococcal strains, two major surface structures, the capsule and extracellular vesicles produced by the pneumococci, as well as surface proteins by proteomics, using the “shaving” approach, followed by LC-MS/MS. We found important differences in both surface ultrastructures and proteins among the culture conditions and strains used. Thus, this work provides insights into physiological adaptations of the pneumococcus when it interacts with the host, which may be useful for the design of strategies to combat infections caused by this pathogen

    Inter-rater and intra-rater reliability of the extended TUG test in elderly participants

    Background: To analyse the reliability, variance and execution time of the Extended Timed Up and Go (Extended TUG) test in three age groups of elderly participants (G1: 55–64 years; G2: 65–74 years; G3: 75–85 years). Methods: An analytical cross-sectional study of 114 recruited participants (63 women) of average age 70.17 (± 7.3) years was undertaken. Each participant performed the Extended TUG three consecutive times, with a rest break between tests of 120 s. Both the intragroup and intergroup reliability of the measurements in the Extended TUG were analysed. Results: The reliability of the Extended TUG test is excellent for the first and second decades but drops down to good for the third decade. Specifically, intragroup reliability ranged from 0.784 for G3 to 0.977 for G1 (G2 = 0.858). Intergroup reliability, compared with intragroup reliability, was slightly lower, ranging between 0.779 for G3 and 0.972 for G1 (G2 = 0.853). Conclusion: The reliability of the Extended TUG test progressively decreases with increasing age, being excellent for the younger age groups and good for the oldest age group
