    Cellulose hydrolysis-hydrogenolysis to ethyleneglycol and propyleneglycol over Ru and heteropolyacid catalysts

    Еthylene and propylene glycols (EG and PG) are widely used in industry to produce cooling systems and other valuable chemical products. But PG is non-toxic, therefore it is used in industries where EG can not be used: pharmaceutical, food, etc. This polyols produced by "one-pot" method, which is one of the promising and effective methods for producing alcohols from cellulose under harsh conditions. The purpose of this study was to determine the optimal composition of the solid bifunctional catalyst and the conditions of its preparation for the hydrolysis-hydrogenolysis of cellulose. Catalists are Ru-HPA/ZrO[2], RuHPA/Nb[2]O[5] and Ru/CsHPK. As a result of the study, the most promising catalyst system is 1%Ru/Cs[3.5]H[0.5]SiW[12]O[40]. In the presence of 1%Ru/CsHPA, the yield of 25% EG and 11% PG was detected (EG and PG selectivity is 60 and 27%). The activity of the catalysts was studied in the presence of Ca(OH)[2]

    A Crystal Structure of the Bifunctional Antibiotic Simocyclinone D8, Bound to DNA Gyrase

    Simocyclinones are bifunctional antibiotics that inhibit bacterial DNA gyrase by preventing DNA binding to the enzyme. We report the crystal structure of the complex formed between the N-terminal domain of the Escherichia coli gyrase A subunit and simocyclinone D8, revealing two binding pockets that separately accommodate the aminocoumarin and polyketide moieties of the antibiotic. These are close to, but distinct from, the quinolone-binding site, consistent with our observations that several mutations in this region confer resistance to both agents. Biochemical studies show that the individual moieties of simocyclinone D8 are comparatively weak inhibitors of gyrase relative to the parent compound, but their combination generates a more potent inhibitor. Our results should facilitate the design of drug molecules that target these unexploited binding pockets

    Interaction of anticancer reduced Schiff base coumarin derivatives with human serum albumin investigated by fluorescence quenching and molecular modeling

    The specific binding of five reduced Schiff base derived 7-amino-coumarin compounds with antitumor activity to human serum albumin, the principal binding protein of blood, was studied by fluorescence spectroscopy. Their conditional binding constants were computed and the reversible binding at the Sudlow’s site I was found to be strong (KD ~ 0.03-2.09 M). Based on the data albumin can provide a depot for the compounds and is responsible for their biodistribution and transport processes. The experimental data is complemented by protein– ligand docking calculations for two representatives which support the observations. The proton dissociation constants of the compounds were also determined by UV-Vis spectrophotometric and fluorometric titrations to obtain the actual charges and distribution of the species in the various protonation states at physiological pH

    Ökologische Erfolgskontrolle Dachbegrünungen Europaallee Zürich

    Die Blauflügelige Sandschrecke gehört zu den gefährdetsten Heuschreckenarten der Schweiz. Ihre Lebensräume sind infolge der veränderten Landnutzung stark eingeschränkt. Die Dachbegrünungen der Bauten der Europaallee in Zürich sind im Sinne des ökologischen Ausgleichs eigens für die Blauflügelige Sandschrecke als Zielart konzipiert worden. Diese Arbeit untersucht die Population der Blauflügelige Sandschrecke auf den Dachflächen der Europaallee und vergleicht die Ergebnisse mit den bestehenden Erhebungen, um Erkenntnisse über die Populationsdynamik zu erlangen. Weiter wird untersucht, ob sich die Art entlang der Gleise auf weitere Flachdächer ausbreitet. 11 Dachflächen werden zur Erfassung des Vorkommens von Larven und Adulten besucht und bei besiedelten Dächern zur Schätzung der ungefähren Populationsgrössen kommt die Fang-Wiederfang-Methode zum Einsatz. Die Blauflügelige Sandschrecke konnte wie bereits in den Vorjahren auf den Dächern der Baufelder A und C nachgewiesen werden, was das Konzept der begrünten Dachflächen für den ökologischen Ausgleich stützt. Die Population auf dem Baufeld C konnte als stabil eingestuft werden. Auf dem benachbarten Baufeld A wurden überraschend wenig adulte Tiere gefunden, wofür verschiedene Erklärungsansätze vorhanden sind. Das Vorkommen der Pionierart konnte auf einem Dach (Letzibach C, Mitte Ost) entlang der Gleise bestätigt werden

    В данной работе рассмотрена возможность использования барий-стронциевого карбонатита при изготовлении сварочных флюсов на основе шлака производства силикомарганца, а так же на основе ковшевых электросталеплавильных шлаков, образованных при производстве рельсовых марок стали.     Inhibitor of differentiation 4 (Id4) is a potential tumor suppressor in prostate cancer

    Abstract Background Inhibitor of differentiation 4 (Id4), a member of the Id gene family is also a dominant negative regulator of basic helix loop helix (bHLH) transcription factors. Some of the functions of Id4 appear to be unique as compared to its other family members Id1, Id2 and Id3. Loss of Id4 gene expression in many cancers in association with promoter hypermethylation has led to the proposal that Id4 may act as a tumor suppressor. In this study we provide functional evidence that Id4 indeed acts as a tumor suppressor and is part of a cancer associated epigenetic re-programming. Methods Data mining was used to demonstrate Id4 expression in prostate cancer. Methylation specific polymerase chain reaction (MSP) analysis was performed to understand molecular mechanisms associated with Id4 expression in prostate cancer cell lines. The effect of ectopic Id4 expression in DU145 cells was determined by cell cycle analysis (3H thymidine incorporation and FACS), expression of androgen receptor, p53 and cyclin dependent kinase inhibitors p27 and p21 by a combination of RT-PCR, real time-PCR, western blot and immuno-cytochemical analysis. Results Id4 expression was down-regulated in prostate cancer. Id4 expression was also down-regulated in prostate cancer line DU145 due to promoter hyper-methylation. Ectopic Id4 expression in DU145 prostate cancer cell line led to increased apoptosis and decreased cell proliferation due in part by an S-phase arrest. In addition to S-phase arrest, ectopic Id4 expression in PC3 cells also resulted in prolonged G2/M phase. At the molecular level these changes were associated with increased androgen receptor (AR), p21, p27 and p53 expression in DU145 cells. Conclusion The results suggest that Id4 acts directly as a tumor suppressor by influencing a hierarchy of cellular processes at multiple levels that leads to a decreased cell proliferation and change in morphology that is possibly mediated through induction of previously silenced tumor suppressors.</p
