    Experimental Study of Tapping Wear Mechanisms on Nodular Cast Iron

    AbstractTapping by cutting is one of the most common operations in manufacturing. This multi-teeth tool, known as a tap, cuts the thread in a hole when the piece has a high added value. The thread quality is ensured when the tap is new or slightly worn, yet when tap wear is high; the quality of profiles exceeds tolerance limits and therefore a defect occurs in the manufacturing line.The aim of this paper is to study the tap wear of titanium nitride coated taps measured on nodular cast iron. The level of tap wear is determined by optical images and the wear mechanics are classified by scanning images and energy dispersion spectroscopy analysis. The results highlight that the critical part in measured taps is between the last chamfer and the first cylinder teeth and, consequently, the thread profile is under-sized. Beside adhesive wear, coating removal and chipping are the main wear aspects during tapping operations

    Optimization of fourteen microsatellite loci in a Mediterranean demosponge subjected to population decimation, Ircinia fasciculata

    The recovery potential of decimated populations of sponges will largely hinge on their populations' size retrieval and their connectivity with conspecifics in unaffected locations. Here, we report on the development of microsatellite markers for estimation of the population connectivity and bottleneck and inbreeding signals in a Mediterranean sponge suffering from disease outbreaks, Ircinia fasciculata. From the 220,876 sequences obtained by genomic pyrosequencing, we isolated 14 polymorphic microsatellite loci and assessed the allelic variation of loci in 24 individuals from 2 populations in the Northwestern Mediterranean. The allele number per locus ranged from 3 to 11, observed heterozygosity from 0.68 to 0.73, and expected heterozygosity from 0.667 to 0.68. No significant linkage disequilibrium between pairs of loci was detected. The 14 markers developed here will be valuable tools for conservation strategies across the distributional range of this species allowing the detection of populations with large genetic diversity loss and high levels of inbreeding

    The synthesis and characterization of composites of oligomeric chitosan with propolis extract which allow the incorporation of a third component (silver nanoparticles) are reported, together with their application in aqueous or hydroalcoholic solutions with a view to the formation of adhesive substances or nanofilms for the protection of vineyards against harmful xylophagous fungi. The antimicrobial properties of the association of the two biological products or those resulting from the incorporation of silver nanoparticles (NPs) are studied and discussed. The efficacy of the chitosan oligomers/propolis/silver NPs ternary system is assessed in vitro for Diplodia fungi. A preliminary study on the convenience of replacing propolis with gentisic acid is also presented

    The Neutralino Sector of the Next-to-Minimal Supersymmetric Standard Model

    The Next-to-Minimal Supersymmetric Standard Model (NMSSM) includes a Higgs iso-singlet superfield in addition to the two Higgs doublet superfields of the minimal extension. If the Higgs fields remain weakly coupled up to the GUT scale, as naturally motivated by the concept of supersymmetry, the mixing between singlet and doublet fields is small and can be treated perturbatively. The mass spectrum and mixing matrix of the neutralino sector can be analyzed analytically and the structure of this 5-state system is under good theoretical control. We also determine decay modes and production channels in sfermion cascade decays to these particles at the LHC and pair production in e+e- colliders.Comment: 27 pages, 8 figure

    WadD, a New Brucella Lipopolysaccharide Core Glycosyltransferase Identified by Genomic Search and Phenotypic Characterization

    Brucellosis, an infectious disease caused by Brucella, is one of the most extended bacterial zoonosis in the world and an important cause of economic losses and human suffering. The lipopolysaccharide (LPS) of Brucella plays a major role in virulence as it impairs normal recognition by the innate immune system and delays the immune response. The LPS core is a branched structure involved in resistance to complement and polycationic peptides, and mutants in glycosyltransferases required for the synthesis of the lateral branch not linked to the O-polysaccharide (O-PS) are attenuated and have been proposed as vaccine candidates. For this reason, the complete understanding of the genes involved in the synthesis of this LPS section is of particular interest. The chemical structure of the Brucella LPS core suggests that, in addition to the already identified WadB and WadC glycosyltransferases, others could be implicated in the synthesis of this lateral branch. To clarify this point, we identified and constructed mutants in 11 ORFs encoding putative glycosyltransferases in B. abortus. Four of these ORFs, regulated by the virulence regulator MucR (involved in LPS synthesis) or the ByrR/ByrS system (implicated in the synthesis of surface components), were not required for the synthesis of a complete LPS neither for virulence or interaction with polycationic peptides and/or complement. Among the other seven ORFs, six seemed not to be required for the synthesis of the core LPS since the corresponding mutants kept the O-PS and reacted as the wild type with polyclonal sera. Interestingly, mutant in ORF BAB1_0953 (renamed wadD) lost reactivity against antibodies that recognize the core section while kept the O-PS. This suggests that WadD is a new glycosyltransferase adding one or more sugars to the core lateral branch. WadD mutants were more sensitive than the parental strain to components of the innate immune system and played a role in chronic stages of infection. These results corroborate and extend previous work indicating that the Brucella LPS core is a branched structure that constitutes a steric impairment preventing the elements of the innate immune system to fight against Brucella

    Genome-wide association study in an admixed case series reveals IL12A as a new candidate in Behçet Disease

    Introduction: The etiology of Behçet's disease (BD) is unknown, but widely considered an excessive T-cell mediated inflammatory response in a genetically susceptible host. Recent genomewide association studies (GWAS) have shown limited number of novel loci-associations. The rarity and unequal distribution of the disease prevalence amongst different ethnic backgrounds have hampered the use of GWAS in cohorts of mixed ethnicity and sufficient sample size. However, novel statistical approaches have now enabled GWAS in admixed cohorts. Methods: We ran a GWAS on 336 BD cases and 5,843 controls. The cases consisted of Western Europeans, Middle Eastern and Turkish individuals. Participants from the Generation R study, a multiethnic birth cohort in Rotterdam, The Netherlands were used as controls. All samples were genotyped and data was combined. Linear regression models were corrected for population stratification using Genomic Principal Components and Linear Mixed Modelling. Meta-analysis was performed on selected results previously published. Results: We identified SNPs associated at genome-wide significant level mapping to the 6p21.33 (HLA) region. In addition to this known signal two potential novel associations on chromosomes 6 and 18 were identified, yet with low minor allele frequencies. Extended metaanalysis reveal a GWS association with the IL12A variant rs17810546 on chromosome 3. Discussion: We demonstrate that new statistical techniques enable GWAS analyses in a limited sized cohort of mixed ethnicity. After implementation, we confirmed the central role of the HLA region in the disease and identified new regions of interest.     On exact solutions for quintessential (inflationary) cosmological models with exponential potentials

    We first study dark energy models with a minimally-coupled scalar field and exponential potentials, admitting exact solutions for the cosmological equations: actually, it turns out that for this class of potentials the Einstein field equations exhibit alternative Lagrangians, and are completely integrable and separable (i.e. it is possible to integrate the system analytically, at least by quadratures). We analyze such solutions, especially discussing when they are compatible with a late time quintessential expansion of the universe. As a further issue, we discuss how such quintessential scalar fields can be connected to the inflationary phase, building up, for this class of potentials, a quintessential inflationary scenario: actually, it turns out that the transition from inflation toward late-time exponential quintessential tail admits a kination period, which is an indispensable ingredient of this kind of theoretical models.     Glucocorticoids for acute urticaria: study protocol for a double-blind non-inferiority randomised controlled trial

    INTRODUCTION: This study protocol describes a trial designed to investigate whether antihistamine alone in patients with acute urticaria does not increase the 7-day Urticaria Activity Score (UAS7) in comparison with an association of antihistamine and glucocorticoids and reduces short-term relapses and chronic-induced urticaria. METHODS AND ANALYSIS: This is a prospective, double-blind, parallel-group, multicentre non-inferiority randomised controlled trial. Two-hundred and forty patients with acute urticaria admitted to emergency department will be randomised in a 1:1 ratio to receive levocetirizine or an association of levocetirizine and prednisone. Randomisation will be stratified by centre. The primary outcome will be the UAS7 at day 7. The secondary outcomes will encompass recurrence of hives and/or itch at day 7; occurrence of spontaneous hives or itch for >6 weeks; patients with angioedema at day 7, and 2, 6, 12 and 24 weeks; new emergency visits for acute urticaria recurrences at days 7 and 14, and 3 months; Dermatology Life Quality Index at days 7 and 14, and 3 and 6 months; and Chronic Urticaria Quality of Life Questionnaire at 6 weeks. ETHICS AND DISSEMINATION: The protocol has been approved by the and will be carried out in accordance with the Declaration of Helsinki and Good Clinical Practice guidelines. A steering committee will oversee the progress of the study. Findings will be disseminated through national and international scientific conferences and publication in peer-reviewed journals.     Nas últimas décadas, qualidade do solo tem se tornado um tópico importante na ciência do solo. Embora esforços consideráveis tenham sido dedicados com o intuito de definir "qualidade do solo", ainda não há um conceito amplamente aceito pela comunidade cientifica. A seleção de índices qualitativos para definir qualidade do solo é uma tarefa extremamente difícil, e diversas propriedades químicas, físicas e biológicas tem sido sugeridas como potenciais indicadores. A matéria orgânica do solo está associada com processos químicos, físicos e biológicos no solo, e, portanto, é considerada um dos melhores indicadores de qualidade do solo. O manejo do solo pode influenciar significativamente a dinâmica do carbono orgânico e o ciclo de N, P, e S. Entretanto, mudanças na concentração total da matéria organica em resposta ao manejo pode ser dificil de ser detectada devido à variabilidade natural do solo. Quando comparada com a matéria orgânica total do solo, a fração mais prontamente disponível, como o carbono orgânico dissolvido (COD), é mais sensível às mudanças no manejo do solo a curto e médio prazo e, portanto, pode ser utilizada como indicador fundamental de qualidade do solo ou das alterações das condições naturais. Embora a fração dissolvida represente apenas uma pequena porção da matéria orgânica total do solo, o COD é móvel no solo e constitui uma importante fonte de C para os microorganismos, podendo facilmente refletir os efeitos de diferentes sistemas de manejo. Inúmeros métodos são utilizados para caracterizar o COD, mas os processos que influenciam sua mineralização e a disponibilidade dos elementos associado com a matéria orgânica (N, P, e S) ainda não são completamente entendidos. Pesquisas futuras devem buscar entender os processos que governam a dinâmica de nutrientes e do COD e como os mesmos afetam a qualidade do solo.Soil quality has become an important issue in soil science. Considerable attempts have been made to define soil quality, but a general concept has not yet been accepted by the scientific community. The selection of quantitative indices for soil quality is extremely difficult, and a considerable number of chemical, physical, and biochemical properties have been suggested as potential indicators of soil quality. Because soil organic matter (SOM) can be associated with different soil chemical, physical and biological processes, it has been widely considered as one of the best soil quality indicator. Land use can significantly influence dynamics of organic carbon and N, P, and S cycle. However, changes in total soil organic carbon (SOC) contents in response to land use may be difficult to detect because of the natural soil variability. In the short to medium term, biological properties and readily decomposable fractions of SOC, such as dissolved organic carbon (DOC), are much more sensitive to soil management than is SOM as a whole, and can be used as a key indicator of soil natural functions. Despite the fact that labile C accounts for a small portion of the total organic matter in the soils, DOC is the most mobile and important C-source for microorganisms, and can easily reflect the effects of land use on soil quality. Although several methods are used to characterize DOC, the factors influencing mineralization and bioavailability of elements associated with organic matter (N, P, and S) remains unclear. Future research should focus on the processes that govern DOC and nutrient dynamics and how they affect soil quality

    Teaching of Energy Issues: A debate proposal for a GLobal Reorientation

    The growing awareness of serious difficulties in the learning of energy issues has produced a great deal of research, most of which is focused on specific conceptual aspects. In our opinion, the difficulties pointed out in the literature are interrelated and connected to other aspects (conceptual as well as procedural and axiological), which are not sufficiently taken into account in previous research. This paper aims to carry out a global analysis in order to avoid the more limited approaches that deal only with individual aspects. From this global analysis we have outlined 24 propositions that are put forward for debate to lay the foundations for a profound reorientation of the teaching of energy topics in upper high school courses, in order to facilitate a better scientific understanding of these topics, avoid many students' misconceptions and enhance awareness of the current situation of planetary emergency
