    Evaluación de sistemas de producción y alimentación bovina (Quindío-Colombia).

    Se realizó una encuesta en 50 explotaciones ganaderas en el departamento de Quindio (Colombia) cubriendo aspectos: físicos (altura m.s.n.m.), socio-económicos (precio de leche, uso de mano de obra), manejo de forrajes (area de pastos mejorados), manejo animal (sistemas de crianza de terneros), alimentación (uso de concentrados, pasto de corte), parámetros de productividad (producción por vaca, carga animal). Los objetivos básicos del trabajo, fueron describir y caracterizar los sistemas de producción y evaluar técnicamente las formas de alimentación tradicional utilizadas. Para el primer objetivo se emplearon las técnicas para análisis multivariante de datos: análisis factorial y análisis de conglomerados ((cluster), de la cual se obtuvo la tipificación de los productores en 4 grupos de fincas con caractersticas productivas y socio-agroenonómicas diferentes. Para cada grupo se obtuvo un modelo esquemático del sistema de producción respectivo. La evaluación alimentaria se realizó con modelos sencillos para el cálculo de raciones en vacas en producción, vacas secas, novillas y terneros, empleando en el modelo dietas típicas utilizadas en los diferentes sistemas de producción. Al analizar las dietas se encontraton tendencias dentro de la muestra en estudio tales como: deficiencias de energía en el aporte nutricional de las dietas para las vacas en producción, bajos niveles de producción de leche/vaca y bajas ganancias de peso para animales en crecimiento. También se encontró en la zona de estudio costosos sistemas de crianza para terneros, manejo irracional del componente forrajero: praderas - pasto de corte y utilización ineficiente de subproductos y recursos no convencionales de alta disponibilidad para la alimentación de bovinos.;A partir del análisis integrado de los aspectos estudiados, se proponen recomendaciones prácticas dirigidas al asistente técnico e instituciones tendientes a solucionar los problemas encontrados y con el criterio de reducir costos de producción y aumentar productividad/ha en las finca

    Analysis of the common genetic component of large-vessel vasculitides through a meta-Immunochip strategy.

    Giant cell arteritis (GCA) and Takayasu’s arteritis (TAK) are major forms of large-vessel vasculitis (LVV) that share clinical features. To evaluate their genetic similarities, we analysed Immunochip genotyping data from 1,434 LVV patients and 3,814 unaffected controls. Genetic pleiotropy was also estimated. The HLA region harboured the main disease-specific associations. GCA was mostly associated with class II genes (HLA-DRB1/HLA-DQA1) whereas TAK was mostly associated with class I genes (HLA-B/MICA). Both the statistical significance and effect size of the HLA signals were considerably reduced in the cross-disease meta-analysis in comparison with the analysis of GCA and TAK separately. Consequently, no significant genetic correlation between these two diseases was observed when HLA variants were tested. Outside the HLA region, only one polymorphism located nearby the IL12B gene surpassed the study-wide significance threshold in the meta-analysis of the discovery datasets (rs755374, P = 7.54E-07; ORGCA = 1.19, ORTAK = 1.50). This marker was confirmed as novel GCA risk factor using four additional cohorts (PGCA = 5.52E-04, ORGCA = 1.16). Taken together, our results provide evidence of strong genetic differences between GCA and TAK in the HLA. Outside this region, common susceptibility factors were suggested, especially within the IL12B locus

    Role of PTPN22 and CSK gene polymorphisms as predictors of susceptibility and clinical heterogeneity in patients with Henoch-Schönlein purpura (IgA vasculitis)

    INTRODUCTION: To determine whether the PTPN22 (protein tyrosine phosphatase nonreceptor 22)/CSK (c-src tyrosine kinase) pathway is implicated in the susceptibility and clinical heterogeneity of Henoch-Schönlein purpura (HSP) in the largest series of Caucasian HSP patients ever assessed for genetic studies. METHODS: A set of 329 Spanish patients diagnosed with HSP fulfilling the American College of Rheumatology and the Michel et al. classification criteria and 515 sex and ethnically matched controls were recruited in this study. Two well-known CSK (CSK rs34933034 and CSK rs1378942) and two functional PTPN22 (PTPN22 rs2476601 (R620W) and PTPN22 rs33996649 (R263Q)) polymorphisms, previously associated with autoimmunity, were genotyped with TaqMan single nucleotide polymorphism (SNP) genotyping assays. RESULTS: No significant differences in the genotype and allele frequencies between HSP patients and controls were observed when the CSK rs34933034, CSK rs1378942, PTPN22 rs2476601 (R620W) and PTPN22 rs33996649 (R263Q) polymorphisms were analyzed independently. In keeping with this observation, no significant differences were found when we assessed these polymorphisms combined conforming haplotypes. In addition, there were no differences in the allele or genotype frequencies when HSP patients were stratified according the age at disease onset, sex, presence of arthralgia/arthritis, nephritis or gastrointestinal manifestations. CONCLUSIONS: Our results do not support association between PTPN22/CSK and HSP

    Contamination level and spatial distribution of heavy metals in water and sediments of El Guájaro Reservoir, Colombia

    Heavy metals have become a subject of special concern worldwide, mainly due to high persistence in the environment, toxicity, biogeochemical recycling and ecological risk. Therefore, the objective of this investigation was to analyze the spatial–temporal distribution of heavy metals in water and sediments to determine the environmental status of El Guájaro Reservoir, where such studies have not been developed. Two measurement campaigns (dry and wet period) were carried out and eight sampling stations were selected. A comparison of water and sediment quality parameters with existing national and international regulations was done. Also, heavy metal distribution maps were generated, and the geoaccumulation index was calculated to identify sources and sediments contamination level. Based on the obtained results, agriculture and mining activities are the main causes of the reservoir contamination. This metals levels could be a potential risk for the aquatic life and the populations that are supplied from this water body

    The Hoopoe's Uropygial Gland Hosts a Bacterial Community Influenced by the Living Conditions of the Bird

    Molecular methods have revealed that symbiotic systems involving bacteria are mostly based on whole bacterial communities. Bacterial diversity in hoopoe uropygial gland secretion is known to be mainly composed of certain strains of enterococci, but this conclusion is based solely on culture-dependent techniques. This study, by using culture-independent techniques (based on the 16S rDNA and the ribosomal intergenic spacer region) shows that the bacterial community in the uropygial gland secretion is more complex than previously thought and its composition is affected by the living conditions of the bird. Besides the known enterococci, the uropygial gland hosts other facultative anaerobic species and several obligated anaerobic species (mostly clostridia). The bacterial assemblage of this community was largely invariable among study individuals, although differences were detected between captive and wild female hoopoes, with some strains showing significantly higher prevalence in wild birds.     Es importante mencionar que este proyecto corresponde al seguimiento de los ciclos anteriores, Primavera y Verano 2022. Enunciando el plan de continuidad que se tuvo, se avanzó con la creación de un taller de capacitación para la profesionalización de las experiencias, llevando a la existencia la estructura y contenido que se presentará con el objetivo de que durante la siguiente etapa (Primavera 2023) se pueda llevar a cabo. Durante el actual semestre, se realizaron diversos productos, levantamientos de información y entregas que derivaron en la creación del “Taller de Profesionalización de Experiencias Turísticas para la Ruta de la Raicilla.” Los objetivos de este producto son crear interés entre los raicilleros y taberneros a través de la información y el conocimiento del potencial turístico que tiene la raicilla no solo como un objeto de evento, sino como un producto turístico. Además, se crearon productos para redes sociales que sirvieran como guía para aquellas tabernas que busquen impulsar sus marcas y productos a través de campañas de comunicación en estos medios, el objetivo de estos productos fue unificar la manera en que las marcas de raicilla se comunican con sus potenciales clientes y el público interesado.ITESO, A.C

    The direct effect of fibroblast growth factor 23 on vascular smooth muscle cell phenotype and function

    [Background] In chronic kidney disease (CKD) patients, increased levels of fibroblast growth factor 23 (FGF23) are associated with cardiovascular mortality. The relationship between FGF23 and heart hypertrophy has been documented, however, it is not known whether FGF23 has an effect on vasculature. Vascular smooth muscle cells VSMCs may exhibit different phenotypes; our hypothesis is that FGF23 favours a switch from a contractile to synthetic phenotype that may cause vascular dysfunction. Our objective was to determine whether FGF23 may directly control a change in VSMC phenotype.[Methods] This study includes in vitro, in vivo and ex vivo experiments and evaluation of patients with CKD stages 2–3 studying a relationship between FGF23 and vascular dysfunction.[Results] In vitro studies show that high levels of FGF23, by acting on its specific receptor FGFR1 and Erk1/2, causes a change in the phenotype of VSMCs from contractile to synthetic. This change is mediated by a downregulation of miR-221/222, which augments the expression of MAP3K2 and PAK1. miR-221/222 transfections recovered the contractile phenotype of VSMCs. Infusion of recombinant FGF23 to rats increased vascular wall thickness, with VSMCs showing a synthetic phenotype with a reduction of miR-221 expression. Ex-vivo studies on aortic rings demonstrate also that high FGF23 increases arterial stiffening. In CKD 2–3 patients, elevation of FGF23 was associated with increased pulse wave velocity and reduced plasma levels of miR-221/222.[Conclusion] In VSMCs, high levels of FGF23, through the downregulation of miR-221/222, causes a change to a synthetic phenotype.
