    Rhizobia contribute to salinity tolerance in common beans (Phaseolus vulgaris L.)

    Rhizobia are soil bacteria that induce nodule formation on leguminous plants. In the nodules, they reduce dinitrogen to ammonium that can be utilized by plants. Besides nitrogen fixation, rhizobia have other symbiotic functions in plants including phosphorus and iron mobilization and protection of the plants against various abiotic stresses including salinity. Worldwide, about 20% of cultivable and 33% of irrigation land is saline, and it is estimated that around 50% of the arable land will be saline by 2050. Salinity inhibits plant growth and development, results in senescence, and ultimately plant death. The purpose of this study was to investigate how rhizobia, isolated from Kenyan soils, relieve common beans from salinity stress. The yield loss of common bean plants, which were either not inoculated or inoculated with the commercial R. tropici rhizobia CIAT899 was reduced by 73% when the plants were exposed to 300 mM NaCl, while only 60% yield loss was observed after inoculation with a novel indigenous isolate from Kenyan soil, named S3. Expression profiles showed that genes involved in the transport of mineral ions (such as K+, Ca2+, Fe3+, PO43−, and NO3−) to the host plant, and for the synthesis and transport of osmotolerance molecules (soluble carbohydrates, amino acids, and nucleotides) are highly expressed in S3 bacteroids during salt stress than in the controls. Furthermore, genes for the synthesis and transport of glutathione and γ-aminobutyric acid were upregulated in salt-stressed and S3-inocculated common bean plants.     Overexpression of the aphid-induced serine protease inhibitor <i>CI2c </i>gene in barley affects the generalist green peach aphid, not the specialist bird cherry-oat aphid

    <div><p>Aphids are serious pests in crop plants. In an effort to identify plant genes controlling resistance against aphids, we have here studied a protease inhibitor, CI2c in barley (<i>Hordeum vulgare</i> L.). The <i>CI2c</i> gene was earlier shown to be upregulated by herbivory of the bird cherry-oat aphid <i>(Rhopalosiphum padi</i> L.<i>)</i> in barley genotypes with moderate resistance against this aphid, but not in susceptible lines. We hypothesized that CI2c contributes to the resistance. To test this idea, cDNA encoding <i>CI2c</i> was overexpressed in barley and bioassays were carried out with <i>R</i>. <i>padi</i>. For comparison, tests were carried out with the green peach aphid (<i>Myzus persicae</i> Sulzer), for which barley is a poor host. The performance of <i>R</i>. <i>padi</i> was not different on the <i>CI2c</i>-overexpressing lines in comparison to controls in test monitoring behavior and fecundity. <i>M</i>. <i>persicae</i> preference was affected as shown in the choice test, this species moved away from control plants, but remained on the <i>CI2c</i>-overexpressing lines. <i>R</i>. <i>padi</i>-induced responses related to defense were repressed in the overexpressing lines as compared to in control plants or the moderately resistant genotypes. A putative susceptibility gene, coding for a β-1,3-glucanase was more strongly induced by aphids in one of the <i>CI2c</i>-overexpressing lines. The results indicate that the CI2c inhibitor in overexpressing lines affects aphid-induced responses by suppressing defense. This is of little consequence to the specialist <i>R</i>.<i>padi</i>, but causes lower non-host resistance towards the generalist <i>M</i>. <i>persicae</i> in barley.</p></div

    This article reviews the present status of the technology and instrumentation for the joint ESA/NASA gravitational wave detector LISA. It briefly describes the measurement principle and the mission architecture including the resulting sensitivity before focussing on a description of the main payload items, such as the interferomtric measurement system, comprising the optical system with the optical bench and the telescope, the laser system, and the phase measurement system; and the disturbance reduction system with the inertial sensor, the charge control system, and the micropropulsion system.     Adipose Tissue in Persons With HIV Is Enriched for CD4+ T Effector Memory and T Effector Memory RA+ Cells, Which Show Higher CD69 Expression and CD57, CX3CR1, GPR56 Co-expression With Increasing Glucose Intolerance

    Chronic T cell activation and accelerated immune senescence are hallmarks of HIV infection, which may contribute to the increased risk of cardiometabolic diseases in people living with HIV (PLWH). T lymphocytes play a central role in modulating adipose tissue inflammation and, by extension, adipocyte energy storage and release. Here, we assessed the CD4+ and CD8+ T cell profiles in the subcutaneous adipose tissue (SAT) and blood of non-diabetic (n = 9; fasting blood glucose [FBG] &lt; 100 mg/dL), pre-diabetic (n = 8; FBG = 100–125 mg/dL) and diabetic (n = 9; FBG ≥ 126 mg/dL) PLWH, in addition to non- and pre-diabetic, HIV-negative controls (n = 8). SAT was collected by liposuction and T cells were extracted by collagenase digestion. The proportion of naïve (TNai) CD45RO−CCR7+, effector memory (TEM) CD45RO+CCR7−, central memory (TCM) CD45RO+CCR7+, and effector memory revertant RA+(TEMRA) CD45RO−CCR7− CD4+ and CD8+ T cells were measured by flow cytometry. CD4+ and CD8+ TEM and TEMRA were significantly enriched in SAT of PLWH compared to blood. The proportions of SAT CD4+ and CD8+ memory subsets were similar across metabolic status categories in the PLWH, but CD4+ T cell expression of the CD69 early-activation and tissue residence marker, particularly on TEM cells, increased with progressive glucose intolerance. Use of t-distributed Stochastic Neighbor Embedding (t-SNE) identified a separate group of predominantly CD69lo TEM and TEMRA cells co-expressing CD57, CX3CR1, and GPR56, which were significantly greater in diabetics compared to non-diabetics. Expression of the CX3CR1 and GPR56 markers indicate these TEM and TEMRA cells may have anti-viral specificity. Compared to HIV-negative controls, SAT from PLWH had an increased CD8:CD4 ratio, but the distribution of CD4+ and CD8+ memory subsets was similar irrespective of HIV status. Finally, whole adipose tissue from PLWH had significantly higher expression of TLR2, TLR8, and multiple chemokines potentially relevant to immune cell homing compared to HIV-negative controls with similar glucose tolerance

    Antiinflammatory Therapy with Canakinumab for Atherosclerotic Disease

    Background: Experimental and clinical data suggest that reducing inflammation without affecting lipid levels may reduce the risk of cardiovascular disease. Yet, the inflammatory hypothesis of atherothrombosis has remained unproved. Methods: We conducted a randomized, double-blind trial of canakinumab, a therapeutic monoclonal antibody targeting interleukin-1β, involving 10,061 patients with previous myocardial infarction and a high-sensitivity C-reactive protein level of 2 mg or more per liter. The trial compared three doses of canakinumab (50 mg, 150 mg, and 300 mg, administered subcutaneously every 3 months) with placebo. The primary efficacy end point was nonfatal myocardial infarction, nonfatal stroke, or cardiovascular death. RESULTS: At 48 months, the median reduction from baseline in the high-sensitivity C-reactive protein level was 26 percentage points greater in the group that received the 50-mg dose of canakinumab, 37 percentage points greater in the 150-mg group, and 41 percentage points greater in the 300-mg group than in the placebo group. Canakinumab did not reduce lipid levels from baseline. At a median follow-up of 3.7 years, the incidence rate for the primary end point was 4.50 events per 100 person-years in the placebo group, 4.11 events per 100 person-years in the 50-mg group, 3.86 events per 100 person-years in the 150-mg group, and 3.90 events per 100 person-years in the 300-mg group. The hazard ratios as compared with placebo were as follows: in the 50-mg group, 0.93 (95% confidence interval [CI], 0.80 to 1.07; P = 0.30); in the 150-mg group, 0.85 (95% CI, 0.74 to 0.98; P = 0.021); and in the 300-mg group, 0.86 (95% CI, 0.75 to 0.99; P = 0.031). The 150-mg dose, but not the other doses, met the prespecified multiplicity-adjusted threshold for statistical significance for the primary end point and the secondary end point that additionally included hospitalization for unstable angina that led to urgent revascularization (hazard ratio vs. placebo, 0.83; 95% CI, 0.73 to 0.95; P = 0.005). Canakinumab was associated with a higher incidence of fatal infection than was placebo. There was no significant difference in all-cause mortality (hazard ratio for all canakinumab doses vs. placebo, 0.94; 95% CI, 0.83 to 1.06; P = 0.31). Conclusions: Antiinflammatory therapy targeting the interleukin-1β innate immunity pathway with canakinumab at a dose of 150 mg every 3 months led to a significantly lower rate of recurrent cardiovascular events than placebo, independent of lipid-level lowering.     Feeding of Triportheus angulatus (Spix & Agassiz, 1829) in the Camaleão lake, Manaus, Amazonas state, Brazil

    Periodical flooding cause strong modifications in Central Amazonian floodplain. Periodical rising and decreasing of the water level influences abiotic and biotic factors of the aquatic environments. During the flooding season, there is a high feed availability, in contrast with dry season in which feed availability is at the lowest level. Limnological parameters also change seasonally. Oxygen concentrations vary both daily and seasonally, sometimes even showing hypoxic conditions. Despite these alterations, floodplain lakes are inhabited by several fish species, having the most diverse and abundant fish fauna among the Amazonian environments. This study focused on Triportheus angulatus feeding in temporal distribution in order to understand the influences of hydrological changes on individual size, diet composition and feeding even in low oxygen concentrations. In rising waters season, small individuals are predominant while the decreasing and the dry seasons there are mostly larger ones. There is a seasonal change on diet composition and this is related to feed type that is more available: during the rising, fishes eat mainly insects and zooplankton; in the flood season and drying season, seeds and fruits are predominant, and at the dry season they eat mainly insects. Feed ingestion was higher in the rising waters and mostly in the flood, and it was lower in the drying season. Feeding activity was not related with oxygen concentrations.As inundações periódicas na Amazônia Central causam profundas modificações no meio ambiente. A alternância dos períodos de cheias e secas tem influência nos fatores bióticos e abióticos do meio aquático. Na enchente e na cheia é alta a oferta de alimentos, ampliando o espectro alimentar que é restrito na seca. As variáveis limnológicas também sofrem modificações. As concentrações de oxigênio têm variações sazonais e diárias, às vezes com períodos de hipoxia. Apesar dessas alterações, os lagos de várzea são habitados por muitas espécies de peixes e estão entre os ambientes de maior abundância e riqueza de peixes na Amazônia. A distribuição temporal e a alimentação de Triportheus angulatus (Spix & Agassiz, 1829) foi estudada em um lago de várzea da Amazônia Central para entender a influência das modificações hídricas sobre o tamanho dos indivíduos, a composição da dieta e a ingestão dos alimentos mesmo em condições de baixas concentrações de oxigênio. Na enchente predominam os indivíduos menores, enquanto que na cheia, vazante e seca os maiores. Houve mudança sazonal na composição da dieta que foi relacionada com a disponibilidade de alimentos no ambientena enchente os peixes ingerem principalmente insetos e zooplâncton; na cheia e vazante frutos e sementes, e na seca insetos. O consumo de alimento foi alto na enchente e cheia, decrescendo na vazante e atingindo as menores quantidades na seca. A atividade alimentar, no período estudado, não foi influenciada pelas baixas concentrações de oxigênio existentes no lago

    Composition, diversity and structure of vascular epiphytes in two contrasting Central Amazonian floodplain ecosystems

    Research focusing on assemblages of vascular epiphytes in the Amazon are scarce. This is especially true for Amazonian floodplain forests, for which only two previous studies have been published. We compared composition, richness and structure of epiphyte assemblages in white-water and black-water floodplains (várzea and igapó) in Central Amazonia in order to close knowledge gaps concerning the distribution and richness of epiphytes. We established sixteen 25x25 m plots in each forest type, and counted and identified all species of vascular epiphytes occurring on trees with a diameter at breast height (DBH) ≥10 cm. We observed a clear distinction in epiphytic species composition (r2=0.83, p=0.001) and diversity (t=3.24, P=0.003) between the two environments, with 61.5 % of species being restricted to várzea, 22.9 % restricted to igapó and only 15.6 % common to both ecosystems. The floodplains were also structurally different for the most abundant species and those with the highest Epiphytic Importance Value (IVe). The diversity of trees did not influence the epiphyte diversity in either ecosystem. The forests were found to differ in the composition, diversity and structure of their epiphytic assemblages, which must be taken into account when designing conservation action plans for these ecosystems and for their vascular epiphytes
