    Challenges to seismic rehabilitation decision process in New Zealand: a focus of decision environment

    Implementing seismic rehabilitation requires a substantial investment for substandard building owners. Seismic retrofitting can significantly reduce earthquake damages to the built envi­ronment and thus decrease the risk posing to the public and the community. However, many countries with active seismic zones, including New Zealand, experience slow progress of seismic retrofit. This paper examines the decision environment which has significant impact on stakeholders’ behaviours, to identify challenges in seismic rehabilitation decision-making. A qualitative approach was adopted with semi-structured interviews. A selection of building owners, government officials, and practical professio­nals involved in seismic retrofitting decision-making were interviewed. Major challenges identified by the interview results include various options, diverse considerations, assorted stakeholders, conflicting multiple objectives, and unaided decision making process. The inconsistency in expectation of whether building owners have sufficient aid in decision-making process offers plausible explanation regarding the key impediment to successful seismic rehabilitation decisions. A decision-making model is thus a necessity to assist building owners making an informed decision

    Optic Nerve Head Gene Transcription Sequelae to a Single Elevated IOP Exposure Provides Insights Into Known Responses to Chronically Elevated IOP

    Purpose: To clarify the optic nerve head (ONH) gene expression responses associated with a single, axon-damaging exposure to elevated IOP in relation to the composite cellular events previously identified in models of chronically elevated IOP. Methods: Anesthetized rats were exposed unilaterally to an 8-hour pulse-train controlled elevation of IOP (PT-CEI) at 60 mm Hg, while others received normotensive CEI at 20 mm Hg. ONH RNA was harvested at 0 hours and 1, 2, 3, 7, and 10 days after either CEI and from naïve animals. RNA sequencing was performed to analyze ONH gene expression. DAVID Bioinformatics tools were used to identify significant functional annotation clusters. Gene function was compared between PT-CEI and two models of chronic ocular hypertension from the literature. Results: The number of significantly changed genes peaked immediately (n = 1354) after PT-CEI (0 hours). This was followed by a lull (point) at 1 and 2 days after PT-CEI. Gene activity increased again at 3 days (136 genes) and persisted at 7 (78 genes) and 10 (339 genes) days. Significant gene functional categories included an immediate upregulation of Defense Response at 0 hours, followed by upregulation in Cell Cycle, a reduction in Axonal-related genes at 3 to 10 days, and upregulation of Immune Response–related genes at 10 days following PT-CEI. The most commonly upregulated gene expression across our PT-CEI study and two chronic models of ocular hypertension were cell cycle related. Conclusions: The PT-CEI model places in sequence ONH gene expression responses previously reported in models with chronically elevated IOP and may provide insights into their role in optic nerve damage

    Acupuncture for treatment of erectile dysfunction : a systematic review and meta-analysis

    Purpose: To assess the effectiveness and safety of acupuncture for erectile dysfunction (ED). Materials and Methods: We searched six major English and Chinese databases included randomized controlled trials (RCTs) testing acupuncture alone or in combination for ED. Dichotomous data were presented as risk ratio (RR) and continuous data were presented as mean difference (MD) both with 95% confidence interval (CI). The Revman (v.5.3) was used for data analyses. Quality of evidence across studies was assessed by the online GRADEpro tool. Results: We identified 22 RCTs, fourteen of them involving psychogenic ED. Most of the included RCTs had high or unclear risk of bias. There was no difference between electro-acupuncture and sham acupuncture with electrical stimulation on the rate of satisfaction and self-assessment (RR, 1.50; 95% CI, 0.71-3.16; 1 trial). Acupuncture combined with tadalafil appeared to have better effect on increasing cure rate (RR, 1.31; 95% CI, 1.00-1.71; 2 trials), and International Index of Erectile Function-5 scores (MD, 5.38; 95% CI, 4.46-6.29; 2 trials). When acupuncture plus herbal medicine compared with herbal medicine alone, the combination therapy showed significant better improvement in erectile function (RR, 1.68; 95% CI, 1.31-2.15; 7 trials). Only two trials reported facial red and dizziness cases, and needle sticking and pruritus cases in acupuncture group. Conclusions: Low quality evidence shows beneficial effect of acupuncture as adjunctive treatment for people mainly with psychogenic ED. Safety of acupuncture was insufficiently reported. The findings should be confirmed in large, rigorously designed and well-reported trials

    NEID Reveals that The Young Warm Neptune TOI-2076 b Has a Low Obliquity

    TOI-2076 b is a sub-Neptune-sized planet (R=2.39±0.10RR=2.39 \pm 0.10 \mathrm{R_\oplus}) that transits a young (204±50MYr204 \pm 50 \mathrm{MYr}) bright (V=9.2V = 9.2) K-dwarf hosting a system of three transiting planets. Using spectroscopic observations with the NEID spectrograph on the WIYN 3.5 m Telescope, we model the Rossiter-McLaughlin effect of TOI-2076 b, and derive a sky-projected obliquity of λ=315+16\lambda=-3_{-15}^{+16\:\circ}. Using the size of the star (R=0.775±0.015RR=0.775 \pm0.015 \mathrm{R_\odot}), and the stellar rotation period (Prot=7.27±0.23P_{\mathrm{rot}}=7.27\pm0.23 days), we estimate a true obliquity of ψ=189+10\psi=18_{-9}^{+10\:\circ} (ψ<34\psi < 34^\circ at 95% confidence), demonstrating that TOI-2076 b is on a well-aligned orbit. Simultaneous diffuser-assisted photometry from the 3.5 m Telescope at Apache Point Observatory rules out flares during the transit. TOI-2076 b joins a small but growing sample of young planets in compact multi-planet systems with well-aligned orbits, and is the fourth planet with an age 300\lesssim 300 Myr in a multi-transiting system with an obliquity measurement. The low obliquity of TOI-2076 b and the presence of transit timing variations in the system suggest the TOI-2076 system likely formed via convergent disk migration in an initially well-aligned disk.Comment: Submitted to ApJL, 13 pages, 4 figures, 3 table

    Functional Analysis of Human Hematopoietic Stem Cell Gene Expression Using Zebrafish

    Although several reports have characterized the hematopoietic stem cell (HSC) transcriptome, the roles of HSC-specific genes in hematopoiesis remain elusive. To identify candidate regulators of HSC fate decisions, we compared the transcriptome of human umbilical cord blood and bone marrow CD34(+)CD33(−)CD38(−)Rho(lo)c-kit(+) cells, enriched for hematopoietic stem/progenitor cells with CD34(+)CD33(−)CD38(−)Rho(hi) cells, enriched in committed progenitors. We identified 277 differentially expressed transcripts conserved in these ontogenically distinct cell sources. We next performed a morpholino antisense oligonucleotide (MO)-based functional screen in zebrafish to determine the hematopoietic function of 61 genes that had no previously known function in HSC biology and for which a likely zebrafish ortholog could be identified. MO knock down of 14/61 (23%) of the differentially expressed transcripts resulted in hematopoietic defects in developing zebrafish embryos, as demonstrated by altered levels of circulating blood cells at 30 and 48 h postfertilization and subsequently confirmed by quantitative RT-PCR for erythroid-specific hbae1 and myeloid-specific lcp1 transcripts. Recapitulating the knockdown phenotype using a second MO of independent sequence, absence of the phenotype using a mismatched MO sequence, and rescue of the phenotype by cDNA-based overexpression of the targeted transcript for zebrafish spry4 confirmed the specificity of MO targeting in this system. Further characterization of the spry4-deficient zebrafish embryos demonstrated that hematopoietic defects were not due to more widespread defects in the mesodermal development, and therefore represented primary defects in HSC specification, proliferation, and/or differentiation. Overall, this high-throughput screen for the functional validation of differentially expressed genes using a zebrafish model of hematopoiesis represents a major step toward obtaining meaningful information from global gene profiling of HSCs

    Careers in context: An international study of career goals as mesostructure between societies' career-related human potential and proactive career behaviour

    Careers exist in a societal context that offers both constraints and opportunities for career actors. Whereas most studies focus on proximal individual and/or organisational-level variables, we provide insights into how career goals and behaviours are understood and embedded in the more distal societal context. More specifically, we operationalise societal context using the career-related human potential composite and aim to understand if and why career goals and behaviours vary between countries. Drawing on a model of career structuration and using multilevel mediation modelling, we draw on a survey of 17,986 employees from 27 countries, covering nine of GLOBE's 10 cultural clusters, and national statistical data to examine the relationship between societal context (macrostructure building the career-opportunity structure) and actors' career goals (career mesostructure) and career behaviour (actions). We show that societal context in terms of societies' career-related human potential composite is negatively associated with the importance given to financial achievements as a specific career mesostructure in a society that is positively related to individuals' proactive career behaviour. Our career mesostructure fully mediates the relationship between societal context and individuals' proactive career behaviour. In this way, we expand career theory's scope beyond occupation- and organisation-related factors

    Integrated genomic characterization of pancreatic ductal adenocarcinoma

    We performed integrated genomic, transcriptomic, and proteomic profiling of 150 pancreatic ductal adenocarcinoma (PDAC) specimens, including samples with characteristic low neoplastic cellularity. Deep whole-exome sequencing revealed recurrent somatic mutations in KRAS, TP53, CDKN2A, SMAD4, RNF43, ARID1A, TGFβR2, GNAS, RREB1, and PBRM1. KRAS wild-type tumors harbored alterations in other oncogenic drivers, including GNAS, BRAF, CTNNB1, and additional RAS pathway genes. A subset of tumors harbored multiple KRAS mutations, with some showing evidence of biallelic mutations. Protein profiling identified a favorable prognosis subset with low epithelial-mesenchymal transition and high MTOR pathway scores. Associations of non-coding RNAs with tumor-specific mRNA subtypes were also identified. Our integrated multi-platform analysis reveals a complex molecular landscape of PDAC and provides a roadmap for precision medicine
