    Assessment of the protein interaction between coagulation factor XII and corn trypsin inhibitor by molecular docking and biochemical validation

    Background: Corn trypsin inhibitor (CTI) has selectivity for serine proteases coagulation factor XII (FXII) and trypsin. CTI is in widespread use as a reagent that specifically inhibits the intrinsic pathway of blood coagulation but not the extrinsic pathway. Objectives: To investigate the molecular basis of FXII inhibition by CTI. Methods: We performed molecular docking of CTI, using its known crystal structure, with a model of the activated FXII (FXIIa) protease domain. The interaction model was verified using a panel of recombinant CTI variants tested for their ability to inhibit FXIIa enzymatic activity using a substrate cleavage assay. Results: The docking predicted that (i) the CTI central inhibitory loop P1 Arg34 side chain forms a salt bridge to the FXIIa S1 pocket Asp189 side chain (ii) residue Trp22 from the CTI helix α1 interacts with the FXIIa S3 pocket (iii) Arg43 from CTI helix α2 forms a salt bridge to FXIIa H1 pocket Asp60A. CTI amino acid substitution R34A negated all inhibitory activity whereas variants G32W, L35A, W22A or R42A-R43A reduced activity by a large degree of 108, 41, 158 and 100-fold respectively, with R27A, W37A, W39A, R42A having no effect. Synthetic peptides spanning CTI residues 20-44 had inhibitory activity 3-4000-fold less than full-length CTI. Conclusions: The data confirm the validity of a canonical model of the FXIIa-CTI interaction with helix α1 (Trp22), central inhibitory loop (Arg34) and helix α2 (Arg43) of CTI required for effective binding by contacting the S1, S3 and H1 pockets of FXIIa, respectively

    Immunological characterization of chromogranins A and B and secretogranin II in the bovine pancreatic islet

    Antisera against chromogranin A and B and secretogranin II were used for analysing the bovine pancreas by immunoblotting and immunohistochemistry. All three antigens were found in extracts of fetal pancreas by one dimensional immunoblotting. A comparison with the soluble proteins of chromaffin granules revealed that in adrenal medulla and in pancreas antigens which migrated identically in electrophoresis were present. In immunohistochemistry, chromogranin A was found in all pancreatic endocrine cell types with the exception of most pancreatic polypeptide-(PP-) producing cells. For chromogranin B, only a faint immunostaining was obtained. For secretorgranin II, A-and B-cells were faintly positive, whereas the majority of PP-cells exhibited a strong immunostaining for this antigen. These results establish that chromogranins A and B and secretogranin II are present in the endocrine pancreas, but that they exhibit a distinct cellular localization

    Comparison of two combinations of opioid and non-opioid analgesics for acute periradicular abscess : a randomized clinical trial

    Acute periradicular abscess is a condition characterized by the formation and propagation of pus in the periapical tissues and generally associated with debilitating pain. Objective: The aim of this study was to compare the overall analgesic effectiveness of two combinations of opioid and non-opioid analgesics for acute periradicular abscess. Material and Methods: This study included 26 patients who sought emergency care in a Brazilian dental school. The patients were randomly divided into two groups: Co/Ac - oral prescription of codeine (30 mg) plus acetaminophen (500 mg), every 4h, for 3 days or Tr/Ac - oral prescription of tramadol hydrochloride (37.5 mg) plus acetaminophen (500 mg) on the same schedule. Two factors were evaluated: (1) pain scores recorded by the patients in a pain diary 6, 12, 24, 48, and 72 h after treatment, using the Visual Analogue Scale; and (2) the occurrence of adverse effects. Results: In both groups, there was a reduction in pain scores over time. For the Co/Ac group, there was a significant reduction in the scores 12, 24,48, and 72 hours after treatment (P0.05), i.e.,both treatments were effective in controlling pain caused by APA; however, the combination of Tr/Ac caused more adverse reactions as two patients had to stop using the medication. Conclusion: This study suggests that, considering both analgesic efficacy and safety, the combination of codeine and acetaminophen is more effective to control moderate to severe pain from acute periradicular abscesses

    Restoration of CFTR function in patients with cystic fibrosis carrying the F508del-CFTR mutation

    <div><p>Restoration of BECN1/Beclin 1-dependent autophagy and depletion of SQSTM1/p62 by genetic manipulation or autophagy-stimulatory proteostasis regulators, such as cystamine, have positive effects on mouse models of human cystic fibrosis (CF). These measures rescue the functional expression of the most frequent pathogenic CFTR mutant, F508del, at the respiratory epithelial surface and reduce lung inflammation in <i>Cftr<sup>F508del</sup></i> homozygous mice. Cysteamine, the reduced form of cystamine, is an FDA-approved drug. Here, we report that oral treatment with cysteamine greatly reduces the mortality rate and improves the phenotype of newborn mice bearing the <i>F508del-CFTR</i> mutation. Cysteamine was also able to increase the plasma membrane expression of the F508del-CFTR protein in nasal epithelial cells from <i>F508del</i> homozygous CF patients, and these effects persisted for 24 h after cysteamine withdrawal. Importantly, this cysteamine effect after washout was further sustained by the sequential administration of epigallocatechin gallate (EGCG), a green tea flavonoid, both <i>in vivo</i>, in mice, and <i>in vitro</i>, in primary epithelial cells from CF patients. In a pilot clinical trial involving 10 <i>F508del-CFTR</i> homozygous CF patients, the combination of cysteamine and EGCG restored BECN1, reduced SQSTM1 levels and improved CFTR function from nasal epithelial cells <i>in vivo</i>, correlating with a decrease of chloride concentrations in sweat, as well as with a reduction of the abundance of <i>TNF/TNF-alpha (tumor necrosis factor)</i> and <i>CXCL8</i> (<i>chemokine [C-X-C motif] ligand 8</i>) transcripts in nasal brushing and TNF and CXCL8 protein levels in the sputum. Altogether, these results suggest that optimal schedules of cysteamine plus EGCG might be used for the treatment of CF caused by the <i>F508del-CFTR</i> mutation.</p></div

    Genomes of Fasciola hepatica from the Americas reveal colonization with Neorickettsia Endobacteria related to the agents of Potomac Horse and Human Sennetsu Fevers

    Food borne trematodes (FBTs) are an assemblage of platyhelminth parasites transmitted through the food chain, four of which are recognized as neglected tropical diseases (NTDs). Fascioliasis stands out among the other NTDs due to its broad and significant impact on both human and animal health, as Fasciola sp., are also considered major pathogens of domesticated ruminants. Here we present a reference genome sequence of the common liver fluke, Fasciola hepatica isolated from sheep, complementing previously reported isolate from cattle. A total of 14,642 genes were predicted from the 1.14 GB genome of the liver fluke. Comparative genomics indicated that F. hepatica Oregon and related food-borne trematodes are metabolically less constrained than schistosomes and cestodes, taking advantage of the richer millieux offered by the hepatobiliary organs. Protease families differentially expanded between diverse trematodes may facilitate migration and survival within the heterogeneous environments and niches within the mammalian host. Surprisingly, the sequencing of Oregon and Uruguay F. hepatica isolates led to the first discovery of an endobacteria in this species. Two contigs from the F. hepatica Oregon assembly were joined to complete the 859,205 bp genome of a novel Neorickettsia endobacterium (nFh) closely related to the etiological agents of human Sennetsu and Potomac horse fevers. Immunohistochemical studies targeting a Neorickettsia surface protein found nFh in specific organs and tissues of the adult trematode including the female reproductive tract, eggs, the Mehlis' gland, seminal vesicle, and oral suckers, suggesting putative routes for fluke-to-fluke and fluke-to-host transmission. The genomes of F. hepatica and nFh will serve as a resource for further exploration of the biology of F. hepatica, and specifically its newly discovered transkingdom interaction with nFh and the impact of both species on disease in ruminants and humans

    The VAST Survey - I. Companions and the unexpected X-ray detection of B6-A7 stars

    With an adaptive optics imaging survey of 148 B6-A7 stars, we have tested the hypothesis that unresolved lower-mass companions are the source of the unexpected X-ray detections of stars in this spectral type range. The sample is composed of 63 stars detected in X-rays within the ROSAT All-Sky Survey and 85 stars that form a control sample; both subsets have the same restricted distribution of spectral type, age, X-ray sensitivity and separation coverage. A total of 68 companion candidates are resolved with separations ranging from 0.3" to 26.2", with 23 new detections. The multiple star frequency of the X-ray sample based on companions resolved within the ROSAT error ellipse is found to be 43 (+6,-6)%. The corresponding control sample multiple star frequency is three times lower at 12 (+4,-3)% -- a difference of 31\pm7%.     Human-environmental interactions and seismic activity in a Late Bronze to Early Iron Age settlement center in the southeastern Caucasus

    Long-term human-environmental interactions in naturally fragile drylands are a focus of geomorphological and geoarchaeological research. Furthermore, many dryland societies were also affected by seismic activity. The semi-arid Shiraki Plain in the tectonically active southeastern Caucasus is currently covered by steppe and largely devoid of settlements. However, numerous Late Bronze to Early Iron Age city-type settlements suggest early state formation between ca. 3.2-2.5 ka that abruptly ended after that time. A paleolake was postulated for the lowest plain, and nearby pollen records suggest forest clearcutting of the upper altitudes under a more humid climate during the Late Bronze/Early Iron Ages. Furthermore, also an impact of earthquakes on regional Early Iron Age settlements was suggested. However, regional paleoenvironmental changes and paleoseismicity were not systematically studied so far. We combined geomorphological, sedimentological, chronological and paleoecological data with hydrological modelling to reconstruct regional Holocene paleoenvironmental changes, to identify natural and human causes and to study possible seismic events during the Late Bronze/Early Iron Ages. Our results show a balanced to negative Early to Mid-Holocene water balance probably caused by forested upper slopes. Hence, no lake but a pellic Vertisol developed in the lowest plain. Following, Late Bronze/Early Iron Age forest clear-cutting caused lake formation and the deposition of lacustrine sediments derived from soil erosion. Subsequently, regional aridification caused slow lake desiccation. Remains of freshwater fishes indicate that the lake potentially offered valuable ecosystem services for regional prehistoric societies even during the desiccation period. Finally, colluvial coverage of the lake sediments during the last centuries could have been linked with hydrological extremes during the Little Ice Age. Our study demonstrates that the Holocene hydrological balance of the Shiraki Plain was and is situated near a major hydrological threshold, making the landscape very sensitive to small-scale human or natural influences with severe consequences for local societies. Furthermore, seismites in the studied sediments do not indicate an influence of earthquakes on the main and late phases of Late Bronze/Early Iron Age settlement. Altogether, our study underlines the high value of multi-disciplinary approaches to investigate human-environmental interactions and paleoseismicity in drylands on millennial to centennial time scales

    Variable strength of forest stand attributes and weather conditions on the questing activity of Ixodes ricinus ticks over years in managed forests

    Given the ever-increasing human impact through land use and climate change on the environment, we crucially need to achieve a better understanding of those factors that influence the questing activity of ixodid ticks, a major disease-transmitting vector in temperate forests. We investigated variation in the relative questing nymph densities of Ixodes ricinus in differently managed forest types for three years (2008–2010) in SW Germany by drag sampling. We used a hierarchical Bayesian modeling approach to examine the relative effects of habitat and weather and to consider possible nested structures of habitat and climate forces. The questing activity of nymphs was considerably larger in young forest successional stages of thicket compared with pole wood and timber stages. Questing nymph density increased markedly with milder winter temperatures. Generally, the relative strength of the various environmental forces on questing nymph density differed across years. In particular, winter temperature had a negative effect on tick activity across sites in 2008 in contrast to the overall effect of temperature across years. Our results suggest that forest management practices have important impacts on questing nymph density. Variable weather conditions, however, might override the effects of forest management practices on the fluctuations and dynamics of tick populations and activity over years, in particular, the preceding winter temperatures. Therefore, robust predictions and the detection of possible interactions and nested structures of habitat and climate forces can only be quantified through the collection of long-term data. Such data are particularly important with regard to future scenarios of forest management and climate warming
