"2nd World Congress of Health Research: Viseu – Portugal, 7-8 October 2014"The European project HEPCOM (Promoting Healthy Eating and Physical Activity in Local Communities) supported by the EU’s Health Programme (2008-2013) is the structural backbone to consider the role of health literacy in health promotion. The aim of HEPCOM is to increase the quality and level of local community and school interventions all over Europe on promoting healthy eating and physical activity among children and young people. For this a platform has been constructed to promote the actual use and up scaling of the existing high quality tools, making them easy accessible and usable to incorporate in the daily work of policy makers and professional practitioners. The target audience includes policy makers and professional practitioners that directly benefit from the project, as they will gain knowledge and inspiration from the learning platform (launched recently for experimental testing, September 2014), which can assist them to improve their daily work with health promotion. Therefore it will also have an impact in children and young people and their families. The 21 HEPCOM partners, coming from 16 EU countries, are closely collaborating with local communities. In Portugal we are working with three Municipalities: Braga, Esposende and Guarda. In the sequence of the HEPCOM project, the Portuguese version of the European Health Literacy Scale “HLS-EU-PT: www. http://literacia-saude.info/” will be applied to primary school teachers and pupils of the above three municipalities in order to find out any putative differences in health literacy. Specific solutions will be designed and applied in order to solve critical health literacy deficiencies found in these municipal communities.CIEC – Research Centre on Child Studies, UM (FCT R&D 317)HEPCOM “Promoting healthy eating and physical activity in local communities” European Commission Health Programme Contract nº 2012120

Safeguarding and revitalizing this cultural heritage is on the agenda of all entities with responsibility for this heritage. Taking as reference the analysis of Oeiras and having as background, the concern expressed in the preservation of buildings that illustrate the past of the territory and constituting this Quintas de Recreio a legacy to conserve, emerges the question that will be the guiding thread of this research: In what way is it possible to correct existing pathologies in the Paço Real de Caxias, through a process of sustainable rehabilitation in order to recover the memory and the identity of this place? This work aims to problematize the concept of Sustainable Rehabilitation applied to this Quintas de Recreio architectural typology, as a strategy for the appropriation of space according to sustainability principles and, thus, to encourage the practice of an architecture that uses (re) constructive techniques of lower environmental impact, preserving and valuing the heritage. The methodology chosen is the case study as a qualitative research strategy, suggesting the recovery of the Paço Real de Caxias with sustainable strategies to be applied in the rehabilitation of the architectural heritage with historical and cultural value. It is assumed as a guiding principle the recovery of this building to respond to a need of the parish of Caxias at the level of a social equipment, which was identified through the crossing of several sources of information. This study, which is structured in the concept of sustainability and in a perspective of conservation of the identity and cultural memory of the architectural work, is a contribution to a strategy to be adopted in a process of sustainable rehabilitation for the architectural typology of Quintas de Recreio, in the Royal Palace of Caxias.N/

Meiotic wave adds extra asymmetry to the development of female chicken gonads

    Get PDF
Development of female gonads in the chicken is asymmetric. This asymmetry affects gene expression, morphology, and germ cell development; consequently only the left ovary develops into a functional organ, whereas the right ovary remains vestigial. In males, on the other hand, both gonads develop into functional testes. Here, we revisited the development of asymmetric traits in female (and male) chicken gonads between Hamburger Hamilton stage 16 (HH16) and hatching. At HH16, primordial germ cells migrated preferentially to the left gonad, accumulating in the left coelomic hinge between the gut mesentery and developing gonad in both males and females. Using the meiotic markers SYCP3 and phosphorylated H2AFX, we identified a previously undescribed, pronounced asymmetryc meiotic progression in the germ cells located in the central, lateral, and extreme cortical regions of the left female gonad from HH38 until hatching. Moreover, we observed that-in contrast to the current view-medullary germ cells are not apoptotic, but remain arrested in pre-leptotene until hatching. In addition to the systematic analysis of the asymmetric distribution of germ cells in female chicken gonads, we propose an updated model suggesting that the localization of germ cells-in the left or right gonad; in the cortex or medulla of the left gonad; and in the central part or the extremities of the left cortex-has direct consequences for their development and participation in adult reproduction

Identification of Brucella sp. isolated in Brazil from 1976 to 2013 by Bruce-Ladder PCR

    Get PDF
Background: Brucella sp. are the causative agents of brucellosis, an infectious disease that affects various species of animals and can be transmitted to humans through direct contact with infected animals, indirectly by the ingestion of rawmilk products, and during the handling of strains or infected material in the laboratory. Being a zoonosis, the detection of Brucella species in animals is essential for the prevention of the disease in humans and to perform a good program of control in infected herds. This study aimed at identifying Brucella field strains isolated from 1976 to 2013 in Brazil, using the modified Bruce-Ladder method, to evaluate the performance of this technique. Materials, Methods & Results: Eighty-three strains of Brucella sp. were included in the study, i.e. 21 reference strains (nine B. abortus, one B. canis, four B. melitensis, two B. ovis and five B. suis) and 62 field strains (six B. canis, one B. suis and 55 B. abortus). For the identification of the genus and/or species of Brucella, biochemical and physiological tests, including MacConkey-agar growth, glucose fermentation, haemolysis, catalase, oxidase and urease tests, nitrate reduction, citrate utilization, H2 S production and CO2 requirement, were performed. Genomic DNA was extracted from pure cultures through heat-lysis of bacterial cultures and the genus was confirmed by a genus-specific PCR (bcsp31 target gene), before performing the modified Bruce-Ladder PCR for the confirmation of the Brucella species. No problems of specificity were observed with the Bruce-Ladder PCR. However, the 1,682 bp fragment was not systematically amplified, even after several modifications such as the concentration of mix components, annealing temperatures and time. Therefore, an individual PCR using primers specific to this fragment was needed for complete identification of some strains. Also, only one kind of Polymerase gave the best results. All Brucella reference strains and negative controls gave the expected results. All field strains previously identified as B. abortus, B. canis and B. suis by biochemical and physiological tests were confirmed by the modified Bruce-Ladder PCR. All isolated Brucella abortus presented a Bruce-Ladder PCR profile expected for field strains, excluding the vaccine strains. Discussion: The modified Bruce-Ladder PCR identified properly all Brucella species (reference and field strains) and proved to be a reliable technique, thus facilitating the identification of the species in the laboratory, reducing the manipulation of these bacteria and the associated danger. Albeit the difficulties of amplification of one fragment for some strains, when using the multiplex technique, this method is fast and without risks after inactivation of the strains. Most studies on animal brucellosis in Brazil were only based on serological tests without identification of the pathogen; while the knowledge of the particular species and/or biovars that occur in Brazil, as well as their distribution, is important to monitor the spread of Brucella among sensitive species and among farms. Our results showed also that B. abortus is still the predominant species isolated in cattle in Brazil. The knowledge of the species that occur in Brazil can help to identify the source of infection and the measures of control to be applied, while it is also very important to trace the dispersion of strains among farms
