    Territorio y Paisaje Cultural en México. Análisis y reconstrucción histórica para su conservación patrimonial

    Esta comunicación tiene por objetivo presentar algunas reflexiones acerca de la problemática actual que afecta a los paisajes culturales de Michoacán, tanto a nivel urbe como a escala territorial. La historia de este patrimonio cultural, ha estado marcada por las acciones que las diferentes culturas que se han apropiado de estos espacios han ejercido sobre el contexto natural. Partiendo de que el paisaje cultural está conformado por formas físicas y formas culturales determinadas por el tiempo, el análisis de los paisajes y los asentamientos humanos permite realizar reconstrucciones históricas, que aporten datos sobre el pasado, den explicaciones del presente y permitan tomar decisiones a futuro que permitan su conservación.

    El presente diplomado de profundización y acompañamiento Psicosocial en escenarios de violencia está orientado al análisis de las distintas realidades que vivencian los individuos y los territorios que han sido víctimas del conflicto armado en Colombia, reflexión que suscita a partir de la composición de subjetividades que se encuentran inmersas en los diferentes escenarios de violencia expuestos de acuerdo a las circunstancias e incidencias que a lo largo de cada historia se visibiliza en el impacto negativo que la violencia desencadena a nivel social, cultural, emocional, económico y político en el horizonte social colombiano. Al existir una identificación de eventos traumáticos, nuestro objetivo está enfocado en el abordaje y profundización de las disímiles herramientas psicosociales que puedan brindar una adecuada atención he intervención de afrontamiento en crisis tanto a nivel individual como colectivo. Así mismo, al abordar las diversas narrativas y relatos expuestos a lo largo del proceso en el diplomado, se encontró que para las víctimas se hace indispensable una intervención psicosocial con responsabilidad donde se logre identificar distintos elementos que permitan subsanar las secuelas que dejo la violencia y los simbólicos subjetivos. Es por ello que desde los diferentes enfoques psicosociales se propone un puñado de estrategias que permitan transformar todas las experiencias traumáticas vividas por causa de la violencia, donde se encuentra en este documento como ejemplo el caso Pandurí, donde las estrategias planteadas están encaminadas a la transformación de los diferentes eventos violentos que vivieron las víctimas.The present Diploma of deepening and Psychosocial accompaniment in violence scenarios is oriented to the analysis of the different realities experienced by the individuals and territories that have been victims of the armed conflict in Colombia, reflection than arises from the composition of subjectivities that are immersed in the different scenarios of violence exposed according to the circumstances and incidents that throughout each story are visible in the negative impact that violence triggers at the social, cultural, emotional, economic and political level on the Colombian social horizon. As there is an identification of traumatic events, our objective is focused on the approach and deepening of the different psychosocial tools that can provide adequate attention and intervention in crisis coping both individually and collectively. Likewise, when addressing the various narratives and stories exposed throughout the process at the diploma course, it was found that a psychosocial intervention with responsibility is indispensable for the victims, where it is possible to identify different elements that can correct the aftermath left by the violence and the subjective symbolics. That is why from the different psychosocial approaches a handful of strategies are proposed that allow transforming all the traumatic experiences lived due to violence, where the Pandurí case is found in this document as an example, where the strategies proposed are aimed at transformation of the different violent events experienced by the victims

    Actividades desarrolladas durante el período: Desde mediados de año se ha instalado un Taller Continuo que funciona los días viernes destinado a la puesta en común de información y la discusión de la marcha general del proyecto. El taller, se ha constituido en caja de resonancia de los avances y dificultades que se presentan y como el espacio donde se distribuyen tareas y responsabilices, se analizan las actividades y los aportes personales o de los integrantes de cada grupo responsable de alguna línea de investigación y se incorporan ideas y señalamientos. En este marco, se ha programado también un Ciclo de Lecturas teóricas y metodológicas que colaboran en la construcción de un lenguaje y una mirada común, necesaria para armonizar la labor de investigadores y becarios de postgrado con trayectorias disciplinares y experiencias muy disímiles; En la investigación sobre la historia económica de Misiones se ha avanzado en los siguientes puntos: Construcción de una periodización basada en ciclos económicos. Se trabaja en el reconocimiento y exploración de repositorios y fuentes escritas existentes a nivel local. exhaustivo relevamiento de las tesis de grado y postgrado, tanto en historia como en Antropología, existentes en las bibliotecas de la UNaM, referidas a temas empresariales y o al funcionamiento de las elites del poder. Se ha tomado contacto y accedido a ámbitos empresariales como las cámaras de la construcción, que en Misiones son 5, de PyMES y la Delegación Provincial de la Cámara Argentina de la Construcción que reúne a grandes empresas; Un grupo de investigadores y auxiliares del ESOHE está investigando los procesos de transformación del sistema educativo provincial (incluyendo tanto la componente pública como la privada), entendiendo que el sistema escolar es una de las instancias donde se socializa ideológicamente a la población y se promueven “sentidos de la realidad”; Entre las tareas en ejecución cabe mencionar: Lectura y fichaje de materia bibliográfico destinado a la actualización reajuste del encuadre teórico-metodológico. Identificación y caracterización de diferentes actores: funcionarios gubernamentales, empresas constructoras, e inmobiliarias, medios de comunicación, asociaciones civiles y otras organizaciones, grupos de interés y afectados por las obras que intervienen en la producción, disputa, apropiación y uso del espacio urbano en las ciudades de Encarnación y Posadas, Garupá y Candelaria. Elección y contacto de informantes calificados. Relevamiento de políticas y/o programas de intervención urbana a ambos lados de la frontera y sus procesos resultantes. Geo-referenciamiento de áreas de revalorización urbana, comerciales, de relegación y desplazamiento, así como de asentamientos precarios y zonas residenciales para sectores de alto poder adquisitivo. El trabajo realizado en esta primera etapa ha permitido avanzar en: Revisión bibliográfica; Redefinición del marco teórico de la investigación; Análisis de algunas relaciones concretas establecidas entre el estado y el sector civil organizado, en particular en el campo de las actividades económicas del tercer sector y en el marco de la implementación de planes y programas públicos; Análisis de las tensiones entre prácticas políticas y económicas que se desatan en el proceso de generación y funcionamiento de los emprendimientos productivos auto-gestionados y cooperativas de trabajo

    Thrombosis in vasculitis: from pathogenesis to treatment

    In recent years, the relationship between inflammation and thrombosis has been deeply investigated and it is now clear that immune and coagulation systems are functionally interconnected. Inflammation-induced thrombosis is by now considered a feature not only of autoimmune rheumatic diseases, but also of systemic vasculitides such as Behçet’s syndrome, ANCA-associated vasculitis or giant cells arteritis, especially during active disease. These findings have important consequences in terms of management and treatment. Indeed, Behçet’syndrome requires immunosuppressive agents for vascular involvement rather than anticoagulation or antiplatelet therapy, and it is conceivable that also in ANCA-associated vasculitis or large vessel-vasculitis an aggressive anti-inflammatory treatment during active disease could reduce the risk of thrombotic events in early stages. In this review we discuss thrombosis in vasculitides, especially in Behçet’s syndrome, ANCA-associated vasculitis and large-vessel vasculitis, and provide pathogenetic and clinical clues for the different specialists involved in the care of these patients

    Identification of novel risk loci, causal insights, and heritable risk for Parkinson's disease: a meta-analysis of genome-wide association studies

    Background Genome-wide association studies (GWAS) in Parkinson's disease have increased the scope of biological knowledge about the disease over the past decade. We aimed to use the largest aggregate of GWAS data to identify novel risk loci and gain further insight into the causes of Parkinson's disease. Methods We did a meta-analysis of 17 datasets from Parkinson's disease GWAS available from European ancestry samples to nominate novel loci for disease risk. These datasets incorporated all available data. We then used these data to estimate heritable risk and develop predictive models of this heritability. We also used large gene expression and methylation resources to examine possible functional consequences as well as tissue, cell type, and biological pathway enrichments for the identified risk factors. Additionally, we examined shared genetic risk between Parkinson's disease and other phenotypes of interest via genetic correlations followed by Mendelian randomisation. Findings Between Oct 1, 2017, and Aug 9, 2018, we analysed 7·8 million single nucleotide polymorphisms in 37 688 cases, 18 618 UK Biobank proxy-cases (ie, individuals who do not have Parkinson's disease but have a first degree relative that does), and 1·4 million controls. We identified 90 independent genome-wide significant risk signals across 78 genomic regions, including 38 novel independent risk signals in 37 loci. These 90 variants explained 16–36% of the heritable risk of Parkinson's disease depending on prevalence. Integrating methylation and expression data within a Mendelian randomisation framework identified putatively associated genes at 70 risk signals underlying GWAS loci for follow-up functional studies. Tissue-specific expression enrichment analyses suggested Parkinson's disease loci were heavily brain-enriched, with specific neuronal cell types being implicated from single cell data. We found significant genetic correlations with brain volumes (false discovery rate-adjusted p=0·0035 for intracranial volume, p=0·024 for putamen volume), smoking status (p=0·024), and educational attainment (p=0·038). Mendelian randomisation between cognitive performance and Parkinson's disease risk showed a robust association (p=8·00 × 10−7). Interpretation These data provide the most comprehensive survey of genetic risk within Parkinson's disease to date, to the best of our knowledge, by revealing many additional Parkinson's disease risk loci, providing a biological context for these risk factors, and showing that a considerable genetic component of this disease remains unidentified. These associations derived from European ancestry datasets will need to be followed-up with more diverse data.     Regulatory sites for splicing in human basal ganglia are enriched for disease-relevant information

    Genome-wide association studies have generated an increasing number of common genetic variants associated with neurological and psychiatric disease risk. An improved understanding of the genetic control of gene expression in human brain is vital considering this is the likely modus operandum for many causal variants. However, human brain sampling complexities limit the explanatory power of brain-related expression quantitative trait loci (eQTL) and allele-specific expression (ASE) signals. We address this, using paired genomic and transcriptomic data from putamen and substantia nigra from 117 human brains, interrogating regulation at different RNA processing stages and uncovering novel transcripts. We identify disease-relevant regulatory loci, find that splicing eQTLs are enriched for regulatory information of neuron-specific genes, that ASEs provide cell-specific regulatory information with evidence for cellular specificity, and that incomplete annotation of the brain transcriptome limits interpretation of risk loci for neuropsychiatric disease. We first perform a large genetic association study by merging all available case-control datasets and by-proxy study results (discovery n = 409,435 and validation size n = 58,190). Here, we add six variants associated with Alzheimer's disease risk (near APP, CHRNE, PRKD3/NDUFAF7, PLCG2 and two exonic variants in the SHARPIN gene). Assessment of the polygenic risk score and stratifying by APOE reveal a 4 to 5.5 years difference in median age at onset of Alzheimer's disease patients in APOE ɛ4 carriers. Because of this study, the underlying mechanisms of APP can be studied to refine the amyloid cascade and the polygenic risk score provides a tool to select individuals at high risk of Alzheimer's disease

    New insights into the genetic etiology of Alzheimer's disease and related dementias

    Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele

    Clonal chromosomal mosaicism and loss of chromosome Y in elderly men increase vulnerability for SARS-CoV-2

    The pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2, COVID-19) had an estimated overall case fatality ratio of 1.38% (pre-vaccination), being 53% higher in males and increasing exponentially with age. Among 9578 individuals diagnosed with COVID-19 in the SCOURGE study, we found 133 cases (1.42%) with detectable clonal mosaicism for chromosome alterations (mCA) and 226 males (5.08%) with acquired loss of chromosome Y (LOY). Individuals with clonal mosaic events (mCA and/or LOY) showed a 54% increase in the risk of COVID-19 lethality. LOY is associated with transcriptomic biomarkers of immune dysfunction, pro-coagulation activity and cardiovascular risk. Interferon-induced genes involved in the initial immune response to SARS-CoV-2 are also down-regulated in LOY. Thus, mCA and LOY underlie at least part of the sex-biased severity and mortality of COVID-19 in aging patients. Given its potential therapeutic and prognostic relevance, evaluation of clonal mosaicism should be implemented as biomarker of COVID-19 severity in elderly people.
