Kompozit yama olarak cam/epoksiden üretilmiş farklı kalınlık ve fiber takviye açılarında yamalar kullanılmış olup, merkezinde yükleme doğrultusuna göre farklı açılarda konumlanmış çatlağa sahip levha ise 2024-T3 Alüminyum alaşımıdır. Yorulma yükü etkisi altında levhada çatlak ucu gerilme şiddet faktörleri, çatlak ilerleme doğrultusu ve levha ömrü sayısal olarak hesaplanmıştır. Çözümde sonlu elemanlar paket programı ANSYS,12.1 kullanılmıştır ve tamir edilmiş levha üç boyutlu olarak modellenmiştir. Gerilme şiddet faktörü; Yer değiştirme Extrapolasyon (YE) metodu ile hesaplanmıştır. Kompozit yama özelliklerinin, çatlak doğrultularının ve yapıştırıcı kalınlıklarının değişiminin çatlak ilerlemesi ve ömür üzerindeki etkisi araştırılmıştır. Elde edilen sonuçlar grafikler halinde sunulmuştur. Anahtar Kelimeler: Kompozit yama, yorulma, çatlak ilerlemesi, sonlu elemanlar metodu, gerilme şiddet faktörü.In this study, fatigue behavior of single-side repaired aluminum plates is investigated, numerically. Composite patch obtained from glass/epoxy laminate which are manufactured for different thickness and fiber orientation angles are used. Aluminum Alloy plates having centered crack whose axes are selected at different angles according to the load direction is 2023-T3. Crack tip stress intensity factor, crack growth direction and plate life are solved under fatigue load condition. In the numerical solution, ANSYS, 12.1 finite element package program is used and the repaired plate is modeled as the three dimensional. Stress intensity factor is calculated by using Displacement Extrapolation (DE) method. Effect of variation of composite patch properties, crack directions and adhesive thickness on crack growth and life is examined. Obtained results are presented in the form of graphs.     Exercise training and detraining process affects plasma adiponectin level in healthy and spontaneously hypertensive rats

    BACKGROUND: Adiponectin levels with long-term swimming exercise have been never investigated in spontaneously hypertensive rats (SHR). OBJECTIVE: This study was aimed to investigate the effects of exercise and detraining process on the adiponectin plasma levels of spontaneously hypertensive rats (SHR) and healthy Wistar-Kyoto rats (WKY). MATERIAL AND METHODS: The rats in the exercise groups were swimming for 10 weeks, 5 days/week, one hour in a day. The detraining rats were left to be sedentary in their cages for 5 weeks after 10 weeks of exercise period. RESULTS: The plasma adiponectin levels decreased in E and SHRE groups compared to the SC and the SHR groups, respectively. In addition, blood pressure was decreased in the exercise groups vs their controls. The adiponectin level was not found to be significantly different in ED and SHRED groups compared to their controls. The blood pressure did not differ between SDC and ED groups, although in the SHRED group it was found to be lower than in SHRSD group rats. CONCLUSION: The results of this study showed that exercise reduced plasma levels of adiponectin in healthy and spontaneously hypertensive rats. However, this difference disappeared at the end of the training processes. Our results suggest, that changes in plasma adiponectin levels are not responsible for changes in blood pressure

    An epigenomic approach to understanding the mechanism of nucleosome retention in mouse spermatozoa

    In mammals fusion of sperm and oocyte gives rise to a totipotent embryo. Origin of totipotency of the early embryo is highly debated: whether it is achieved by inheritance of the epigenetic states of the gametes or by reprogramming of such parental epigenetic marks in the embryo. Oocyte and sperm differ in their potential to transmit epigenetic information. The oocyte is full of maternal transcripts, proteins and its DNA is packed into nucleosomes while a spermatozoon is in highly compact structure and the majority of its histones are exchanged by protamines. It has been determined that in mature sperm histone-to-protamine exchange is not complete, still around 10% and 1% of histones are retained in human and mouse sperm, respectively. During the initial period of my PhD, we and others showed that in human sperm retained histones are not randomly distributed in the genome but to some extent are enriched at loci important for developmental and signaling pathways. We obtained similar findings in mouse sperm at single loci. Nevertheless, genome-wide localization of nucleosomes in mouse sperm and the main principles defining specific nucleosome retention were not known. In my project, the major aim was to determine the logic of nucleosome retention by using mouse sperm as the model system. In addition, I investigated transcript dynamics during late spermatogenesis to identify characteristics of the transcriptomes in maturing germ cells and spermatozoa. By taking a genome-wide approach we have identified that combinatorial effects of sequence composition, histone variants, histone modifications and gene expression uniquely package sperm DNA. Importantly, H3.3 constitutes the main histone H3 variant retained in mature sperm and localizes to CpG islands. The majority of the genomic regions containing H3.3 are marked by H3K4me3. H3.3 retention in sperm reflects high nucleosome turnover in round spermatids. Canonical histone H3 variants H3.1 and H3.2 are present in low amounts in mature sperm and their retention pattern mostly shows the history from non-replicating round spermatids. GC-rich genomic regions marked by H3K27me3 retain H3.1/H3.2, likely related to low nucleosome turnover in round spermatids. Investigating transcript dynamics during later stages of spermatogenesis showed that overall transcript levels towards sperm development are static. Nevertheless, our data relating changes in gene expression to changes in chromatin states highly suggest for ongoing transcriptional activity during differentiation of spermatids into sperm. Overall, we identified that histone modification states of retained nucleosomes and spermatozoal RNA pool highly relate to early embryonic gene expression, which argues that sperm carries critical information to the early embryo

    Son yirmi yıl içerisinde taşınmaz sektöründe yeni yaklaşımlar kullanılmaya başlanmıştır. Bu yaklaşımlar toprağın hala temel iki yatırım aracından birisi olduğunu göstermektedir. Günümüz yatırım araçları arasında her ne kadar hisse senetleri, devlet tahvilleri vb. olsa da insanoğlu toprak ve altından asla vazgeçmemiştir. Taşınmaz yatırım kararlarında; sermaye, proje ve yer seçimi esaslarına dayanan taşınmaz geliştirme ve taşınmaz değerleme birbirini tamamlayan aşamalardır. Bu çalışmada, taşınmaz geliştirmenin olmazsa olmaz üç bileşeninden arsa ve sermayenin olduğu projenin hangisi olması gerektiği konusunda karar vericilere alternatifler ve bu alternatiflerin gelecekteki getirileri incelenmiştir. Çalışmada ele alınan alanı Tapu Kadastro Genel Müdürlüğü (TKGM) Oran yerleşkesi olup kullanılan yöntem en etkin ve en verimli kullanım analizidir (EVKA). Oran yerleşkesinin bulunduğu bölgeye ilişkin veriler toplanmış, düzenlenmiş ve analiz çalışmalarında kullanılmıştır. Bölgenin ihtiyaçlarına göre elde edilen sonuçlar, alternatifler ve öneriler paylaşılmıştır. Aynı zamanda gelişen teknolojilere bağlı olarak gerçek zamanlı bir EVKA yapılabilmesi için PROPTECH yatırımcılarına yönelik önerilere de yer verilmiştir.In the last twenty years, new approaches have been used in the real estate sector. These approaches show that the land is still one of the two main investment tools. Although today's investment instruments, stocks, government bonds, etc. though mankind has never given up on land and gold. In immovable investment decisions; Real estate development and real estate valuation, which are based on capital, project and location selection, are complementary stages. In this study, alternatives to the decision makers and the future returns of these alternatives on the land that has land and capital, which are the indispensable components of real estate development, are examined. The area covered in the study is TKGM ratio campus, and the method used is the most effective and most efficient use analysis (EVKA). Data on the region where the rate campus is located was collected, organized and used in analysis studies. The results, alternatives and suggestions obtained according to the needs of the region were shared. At the same time, suggestions for PROPTECH investors are included in order to make a real-time EVKA depending on the developing technologies

    Hanein-1, a novel conserved eukaryotic protein ubiquitously expressed in human tissues

The protein is highly conserved among eukaryotes and bears no domain similarities with any known proteins. In databases, it is described as coiled coil containing-124 hypothetical protein and predicted to encode a 223 amino acid-protein. In this project, our aim was to characterize this highly conserved protein by using several bioinformatics and biochemical methodologies. Sequence similarity search analysis showed that it had around 75% identity in mammals, 50% identity with insects and nematodes. Expressional analysis revealed that HANEIN-1 was expressed in all tissues ubiquitously with a remarkable expression status in skeletal muscle. Beside providing information about expression status of HANEIN-1, northern blotting showed that HANEIN-1 transcript size was approximately 1000 bp. Regarding protein level expression, western blotting revealed that HANEIN-1 encoded a 33 kDa protein and protein stability was affected in a different way upon labeling with Flag epitope at N-ter and C-ter. Yeast double hybrid screening performed in our laboratory showed that HANEIN-1 interacted with RASGEF1B, which was a guanine nucleotide exchange factor not fully characterized. Expressional analysis displayed that RASGEF1B expression profile inversely correlated with HANEIN-1. Finally, serine-scanning mutagenesis analysis showed that site-directed mutagenesis of serine at position 194 significantly affected the stability of the protein.Erkek, SerapM.S

    The use of corn gluten meal as a source of colour material in broilers

    In this study was carried out to investigate the influence of corn gluten meal on broiler skin colour, shank colour and criteria related to production. In the experiment, 400 broiler chicks (Arbor Acres) were used as the animal material. Chicks were divided into five groups, 80 chicks in each. First group as a control group consumed ration that contained 60% yellow corn as the source of the carotenoid. The other groups consumed rations containing 2.5%, 5.0%, 7.5% and 10% corn gluten meal respectively. Broilers were fed starter rations at the first two weeks. After then, they were fed finisher rations at the four weeks. The experiment was finished at the end of six weeks of age. In this results, The groups feeding rations with containing 10% corn gluten meal showed higher Roche colour Fan value and higher B.C.E value than the other groups, significantly.Arastırmada, mısır gluten ununun broyler deri rengi, incik rengi ve verimle ilgili kriterler üzerine etkilerini incelemek amacıyla yapılmıstır. Denemede,hayvan materyali olarak 400 adet etlik civcivi (Arbor acres) kullanılmıstır. Civcivler her birinde 80 adet hayvan olacak sekilde 5 gruba ayrılmıstır. Kontrol grubu olan 1. Grup, karotenoid kaynagı olarak %60 sarı mısır içeren rasyonu tüketmistir. Diger gruplar sırasıyla, %2.5, %5.0, %7.5 ve %10 mısır gluten unu içeren rasyonları tüketmislerdir. Bütün gruplar ilk 2 hafta baslatma rasyonu, daha sonra ki 4 hafta ise bitirme rasyonu tüketmislerdir. Deneme 6 haftada bitirilmistir. Arastırma sonucunda, %10mısır gluteni tüketen gruplar istatistiksel olarak daha yüksek Roche renk yelpazesi ve B.C.E. degeri göstermislerdir

    Contribution of heme oxygenase 2 to blood pressure regulation in response to swimming exercise and detraining in spontaneously hypertensive rats

    Background: We aimed to determine the effects of exercise followed by detraining on systolic blood pressure (SBP), heme oxygenase 2 (HO-2) expression, and carboxyhemoglobin (COHb) concentration in spontaneously hypertensive rats (SHR) to explain the role of carbon monoxide (CO) in this process. Material/Methods: Animals were randomized into exercised and detrained groups. Corresponding sedentary rats were grouped as Time 1–2. Swimming of 60 min/5 days/week for 10 weeks was applied. Detraining rats discontinued training for an additional 5 weeks. Gene and protein expressions were determined by real-time PCR and immunohistochemistry. Results: Aorta HO-2 histological scores (HSCORE) of hypertensive rats were lower, while SBP was higher. Swimming caused enhancement of HO-2 immunostaining in aorta endothelium and adventitia of SHR. Exercise induced elevation of blood COHb index in SHR. Synchronous BP lowering effect of exercise was observed. HO-2 mRNA expression, HSCORE, and blood COHb index were unaltered during detraining, while SBP was still low in SHR. Conclusions: CO synthesized by HO-2 at least partly plays a role in SBP regulation in the SHR-and BP-lowering effect of exercise. Regular exercise with short-term pauses may be advised to both hypertensives and individuals who are at risk. © Med Sci Monit

    Vatandaş Gözünden E-Sağlık Hizmetlerinin Değerlendirilmesi: Farkındalık, Kullanım ve Memnuniyet Düzeyleri

    Çalışma, e-Devletin önemli ayaklarından biri olan e-sağlık hizmetlerinin kullanıcılar açısından incelenmesi amacıyla yapılmıştır. Katılımcıların e-sağlık hizmetleri farkındalık, kullanım ve memnuniyet düzeyleri ölçülmüştür. Memnuniyet düzeyleri ile sosyo-ekonomik ve demografik yapı arasında anlamlı ve pozitif bir ilişki olup olmadığı incelenmiştir. Yöntem olarak alan araştırması tekniği, veri toplama aracı olarak ise online ve yüz yüze anket tekniği kullanılmıştır. Araştırma sonucunda eğitim düzeyi ile e-sağlık hizmetleri memnuniyet düzeyi arasında anlamlı bir sonuç ortaya çıkmıştır. E-sağlık hizmetlerinin çalışma hızından memnuniyetin, talep edilen hizmetlerin taleplerini randevu beklemeden karşılama düzeyinin yüksek olduğu gözlemlendi. Katılımcıların çoklu ifadelerinin değerlendirildiği e-nabız kullanım sonuçlarına göre en çok kullanılan hizmetin ‘Test Sonucu Öğrenme’ hizmeti olduğu anlaşıldı. Katılımcıların en sık kullandıkları e-sağlık hizmetleri arasında “Hayat Eve Sığar Mobil uygulaması” birinci, “MHRS Mobil uygulaması” ikinci sırada yer aldı. En az kullanılan mobil uygulamalar arasında yerel bir uygulama olan Konya Şehir Hastanesi Mobil uygulaması yer aldı. Çalışma bulgularının kamu politika yapıcılarına e-sağlık hizmetlerinin geliştirilmesinde kullanabilecekleri önemli bilgiler sağlayacağı değerlendirilmektedir
