    Modular detergents tailor the purification and structural analysis of membrane proteins including G-protein coupled receptors

    Detergents enable the purification of membrane proteins and are indispensable reagents instructural biology. Even though a large variety of detergents have been developed in the lastcentury, the challenge remains to identify guidelines that allowfine-tuning of detergents forindividual applications in membrane protein research. Addressing this challenge, here weintroduce the family of oligoglycerol detergents (OGDs). Native mass spectrometry (MS)reveals that the modular OGD architecture offers the ability to control protein purificationand to preserve interactions with native membrane lipids during purification. In addition to abroad range of bacterial membrane proteins, OGDs also enable the purification and analysisof a functional G-protein coupled receptor (GPCR). Moreover, given the modular design ofthese detergents, we anticipatefine-tuning of their properties for specific applications instructural biology. Seen from a broader perspective, this represents a significant advance forthe investigation of membrane proteins and their interactions with lipids

    International audienceAnthropogenic fires affected the temperate deciduous forests of Central Europe over millennia. Biomass burning releases carbon to the atmosphere and produces charcoal, which potentially contributes to the stable soil carbon pools and is an important archive of environmental history. The fate of charcoal in soils of temperate deciduous forests, i.e. the processes of charcoal incorporation and transportation, and the effects on soil organic matter are still not clear. In a long-term experimental burning site, we investigated the effects of slash-and-burn and determined soil organic carbon, charcoal carbon and nitrogen concentrations and the soil lightness of colour (L*) in the topmost soil material (0?1, 1?2.5 and 2.5?5 cm depths) before, immediately after the fire and one year after burning. The main results are that (i) only few charcoal particles from the forest floor were incorporated into the soil matrix by soil mixing animals. In 0?1 cm and during one year, the charcoal C concentrations increased only by 0.4 g kg?1 and the proportion of charcoal C to SOC concentrations increased from 2.8 to 3.4%; (ii) the SOC concentrations did not show any significant differences; (iii) soil lightness significantly decreased in the topmost soil layer and correlated with the concentrations of charcoal C (r=-0.87**) and SOC (r=?0.94**) in samples 0?5 cm. We concluded that the soil colour depends on the proportion of aromatic charcoal carbon in total organic matter and that Holocene burning could have influenced soil charcoal concentrations and soil colour

    Evaluation of the expansion of Mantis religiosa (L.) in Poland based on a questionnaire survey

    Mantis religiosa (L.) is the only species of praying mantis in Poland. The main habitat where its occurrence is permanent and confirmed is the Sandomierska Basin (SE Poland). Numerous reports suggest a significant dispersion of these insects in Poland. Therefore, we decided to use and collect information to update the knowledge about the spread of the praying mantis in Poland. With the use of Google Maps, we created a map on which respondents were asked to select the location where they encountered a M. religiosa specimen in Poland in recent years (2013–2016). In total, we obtained 159 locations for the European mantis. These findings show the significant spread of this species in all directions from its main habitat in Sandomierska Basin. However, there is a need for more studies on this topic, especially to confirm the existence of reproductive populations in the provided locations and to confirm the existence of two different M. religiosa lineages in Poland

    Franchise Business Development Model: Theoretical Considerations

    Franchising is not a popular business development form in Lithuania. Only 0.02% of all Lithuanian companies use franchising for business development, while in most of the developed countries franchise is recognized as a convenient business expansion form. So, what factors determine such differences in the use of franchising? Analysis of related literature showed, that mainly researchers analyse some aspects of franchising, however there is no systematized analysis which covers all the main franchise development elements. Therefore, the aim of this article is to systematize franchise business development phases, main elements and factors, influencing this process and to propose a theoretical franchise system development model. Thus, theoretical considerations about the main phases and elements of franchise system development as well as advantages and disadvantages of the system are analysed in the article. These theoretical considerations are generalized in theoretical franchise system development model, which shows, that development of franchise business consists of particular steps, starting from favourable conditions for the system to emerge; intention and ability of franchisor to form franchise based on his business; the establishment and initial development of franchise system; and its further functioning. The article does not test the model and even does not discuss the operationalization of it. Operationalization of the model as well as empirical evidence is presented by the authors in the forthcoming articles

    The splicing landscape is globally reprogrammed during male meiosis

    Meiosis requires conserved transcriptional changes, but it is not known whether there is a corresponding set of RNA splicing switches. Here, we used RNAseq of mouse testis to identify changes associated with the progression from mitotic spermatogonia to meiotic spermatocytes. We identified ∼150 splicing switches, most of which affect conserved protein-coding exons. The expression of many key splicing regulators changed in the course of meiosis, including downregulation of polypyrimidine tract binding protein (PTBP1) and heterogeneous nuclear RNP A1, and upregulation of nPTB, Tra2β, muscleblind, CELF proteins, Sam68 and T-STAR. The sequences near the regulated exons were significantly enriched in target sites for PTB, Tra2β and STAR proteins. Reporter minigene experiments investigating representative exons in transfected cells showed that PTB binding sites were critical for splicing of a cassette exon in the Ralgps2 mRNA and a shift in alternative 5′ splice site usage in the Bptf mRNA. We speculate that nPTB might functionally replace PTBP1 during meiosis for some target exons, with changes in the expression of other splicing factors helping to establish meiotic splicing patterns. Our data suggest that there are substantial changes in the determinants and patterns of alternative splicing in the mitotic-to-meiotic transition of the germ cell cycle

    Changes in the Red Sea overturning circulation during Marine Isotope Stage 3

    The oceanography of the Red Sea is controlled by the restricted exchange of water masses with the Indian Ocean and by high evaporation rates due to the arid climate of the surrounding land areas. In the northern Red Sea, the formation of oxygen-rich subsurface water ventilates the deeper parts of the basin, but little is known about the variability in this process in the past. The stable oxygen and carbon isotope records of epibenthic foraminifera from a sediment core of the central Red Sea and comparison with existing isotope records allow for the reconstruction of changes in the Red Sea overturning circulation (ROC) during Marine Isotope Stage 3. The isotope records imply millennial-scale variations in the ROC, in phase with the climate variability in the high northern latitudes. This suggests an immediate response of dense-water formation to the regional climate and hydrology of the northern Red Sea. Deep-water formation was intensified under the influence of cold and hyper-arid conditions during Heinrich stadials and was diminished during Dansgaard–Oeschger interstadials. While these changes are reflected in both stable oxygen and carbon isotope records, the latter data also exhibit changes in phase with the African–Indian monsoon system. The decoupling of the stable carbon and oxygen isotope records at the summer monsoon maximum centered around 55–60 ka may be associated with an increased inflow of nutrient-rich intermediate waters from the Arabian Sea to the central Red Sea. This process fueled local surface water productivity, resulting in enhanced remineralization of sinking organic matter and release of 12C at intermediate water depths.</p
