In vitro and in vivo expression of foreign genes by transmissible gastroenteritis coronavirus-derived minigenomes

A helper-dependent expression system based on transmissible gastroenteritis coronavirus (TGEV) has been developed using a minigenome of 3·9 kb (M39). Expression of the reporter gene {beta}-glucuronidase (GUS) (2–8 µg per 106 cells) and the porcine respiratory and reproductive syndrome virus (PRRSV) ORF5 (1–2 µg per 106 cells) has been shown using a TGEV-derived minigenome. GUS expression levels increased about eightfold with the m.o.i. and were maintained for more than eight passages in cell culture. Nevertheless, instability of the GUS and ORF5 subgenomic mRNAs was observed from passages five and four, respectively. About a quarter of the cells in culture expressing the helper virus also produced the reporter gene as determined by studying GUS mRNA production by in situ hybridization or immunodetection to visualize the protein synthesized. Expression of GUS was detected in the lungs, but not in the gut, of swine immunized with the virus vector. Around a quarter of lung cells showing replication of the helper virus were also positive for the reporter gene. Interestingly, strong humoral immune responses to both GUS and PRRSV ORF5 were induced in swine with this virus vector. The large cloning capacity and the tissue specificity of the TGEV-derived minigenomes suggest that these virus vectors are very promising for vaccine development

Effects of adenotonsillectomy on plasma inflammatory biomarkers in obese children with obstructive sleep apnea: A community-based study.

BackgroundObesity and obstructive sleep apnea syndrome (OSA) are highly prevalent and frequently overlapping conditions in children that lead to systemic inflammation, the latter being implicated in the various end-organ morbidities associated with these conditions.AimTo examine the effects of adenotonsillectomy (T&A) on plasma levels of inflammatory markers in obese children with polysomnographically diagnosed OSA who were prospectively recruited from the community.MethodsObese children prospectively diagnosed with OSA, underwent T&A and a second overnight polysomnogram (PSG) after surgery. Plasma fasting morning samples obtained after each of the two PSGs were assayed for multiple inflammatory and metabolic markers including interleukin (IL)-6, IL-18, plasminogen activator inhibitor-1 (PAI-1), monocyte chemoattractant protein-1 (MCP-1), matrix metalloproteinase-9 (MMP-9), adiponectin, apelin C, leptin and osteocrin.ResultsOut of 122 potential candidates, 100 obese children with OSA completed the study with only one-third exhibiting normalization of their PSG after T&A (that is, apnea-hypopnea index (AHI) ≤1/hour total sleep time). However, overall significant decreases in MCP-1, PAI-1, MMP-9, IL-18 and IL-6, and increases in adropin and osteocrin plasma concentrations occurred after T&A. Several of the T&A-responsive biomarkers exhibited excellent sensitivity and moderate specificity to predict residual OSA (that is, AHI⩾5/hTST).ConclusionsA defined subset of systemic inflammatory and metabolic biomarkers is reversibly altered in the context of OSA among community-based obese children, further reinforcing the concept on the interactive pro-inflammatory effects of sleep disorders such as OSA and obesity contributing to downstream end-organ morbidities

An experience of educational technology reported from other times and spaces.

En este artículo narramos nuestra experiencia con alumnas y alumnos de Tecnología Educativa en el curso 2005-2006 de la Facultad de Pedagogía de la Universidad de Barcelona. Se describe el modo en el que confluyen y divergen los tiempos subjetivos, de docentes y estudiantes, los tiempos del grupo clase, los tiempos sociales y de cómo una asignatura como Tecnología educativa nos ayuda a cuestionar, desmontar y proponer alternativas a los tiempos objetivados desde el plan de estudios para permitir la entrada de otros tiempos y espacios.In this article we narrate our experience with students of Educational Technology of the Faculty of Pedagogy of the University of Barcelona in the course 2005-2006. There is described the way in which the subjective times of teachers and students come together and diverge, the times of the group class, the social times and of how a subject as Educational Technology helps us to question, to dismantle and to propose alternatives to the times from the curriculum to allow the entry of other times and spaces

Microscopic Model for Granular Stratification and Segregation

We study segregation and stratification of mixtures of grains differing in size, shape and material properties poured in two-dimensional silos using a microscopic lattice model for surface flows of grains. The model incorporates the dissipation of energy in collisions between rolling and static grains and an energy barrier describing the geometrical asperities of the grains. We study the phase diagram of the different morphologies predicted by the model as a function of the two parameters. Doce años después.... recuperamos el saber acumulado y elaboramos una nueva propuesta docente para los estudiantes de Tecnología educativa de la Universidad de Barcelona

En este artículo se relatan los cambios introducidos en nuestra propuesta docente de la asignatura de Tecnología Educativa en la Universidad de Barcelona y se intenta mostrar el trabajo, el conocimiento y el saber acumulado en todos estos años. El texto se divide en dos bloques. El primero, describe la propuesta actual y el recorrido realizado desde el curso 1993/94. Es una síntesis – con las limitaciones que conlleva – donde se destaca el sentido y algunos cambios significativos para las profesoras y profesores de la asignatura, en su intento de alejarse de una práctica docente de tradición reproductora, que la mayoría de las alumnas y los alumnos habían experimentado en propuestas educativas anteriores. El segundo, recoge algunas de sus voces y nos muestra como se han apropiado y han significado el trabajo realizado durante este curso 2004/05. La valoración de los alumnos y alumnas, es un mosaico de testimonios que nos permite comprender o, al menos, intuir el sentido que ha podido tener para ellas y ellos esta vivencia. Algunos de éstos son reveladores en cuanto confirman o desmienten tópicos o creencias, asumidas por el profesorado, y por mostrar elementos sutiles que quizás durante las sesiones de clase pasaron desapercibidos.In this article we describe the changes introduced in our educational proposal of the subject of Educational Technology in the University of Barcelona and we show the work and the knowledge accumulated in every year. The text is divided in two blocks. The first one, describes the current proposal and the journey carried out from the course 1993/94. It is a synthesis, where he/she stands out the sense and some significant changes for the teachers of the subject, in their intent of moving away from an educational practice of reproductional tradition that most of the students had experienced in previous educational proposals. The second, it picks up some of their voices and it shows us like they have appropriated and they have meant the work carried out during this course 2004/05. The valuation of the students, it is a mosaic of testimonies that allows us to understand this experience

Diseño de maqueta accesible para el yacimiento arqueológico de Tiermes (Soria): La topografía aterrazada del frente sur y el acueducto romano

Este proyecto estudia la representación y recreación de una parte del paisaje arqueológico y el acueducto romano de Tiermes, en Soria. Se trata de un estudio minucioso de la topografía del lugar en el que se representa esta estructura excavada en la misma roca. Concretamente, se trata del ramal sur, donde el recorrido del agua presenta partes vistas y subterráneas en la roca arenisca. El hecho de que el acueducto se mimetice con el terreno, apareciendo y ocultándose dentro del mismo, determina el tipo de maqueta elegida, facilita la comprensión de la zona a los usuarios y les otorga una perspectiva del paisaje más amplia. Diseño y construcción de un sistema de monitorización portátil para la caracterización térmica de equipos evaporativos

El uso de sistemas de climatización juega un papel fundamental en la vida cotidiana de las personas originado por la calidad de vida y los niveles de confort de la sociedad actual. Según la Guía Práctica. Consumo eficiente y responsable de IDAE (Instituto para la Diversificación y Ahorro de la Energía), el consumo en los hogares es aproximadamente un 30% del consumo total de energía final en España, y este se reparte en un 18% en las viviendas [1]. A esto debemos sumar que de ese 18%, los usos térmicos (calefacción, refrigeración y ACS) suponen el 69% del consumo. Un porcentaje tan alto de consumo, y por tanto de emisiones de dióxido de carbono al ambiente, lleva a una búsqueda constante de mejora de la eficiencia de estos sistemas. Una decisión clave que condiciona la eficiencia de estos equipos es el sistema de condensación. Las alternativas actuales son sistemas condensados por agua y condensados por aire. Cada una de ellas tiene unas ventajas y unas desventajas con respecto a la otra
