Chinese Communist Materials at the Bureau of Investigation Archives, Taiwan
During the long years of civil strife in China the Nationalist authorities amassed extensive materials on their Communist adversaries. Now stored in government institutions on Taiwan, these materials are an excellent source for the study of the Chinese Communist movement. Among them is the Bureau of Investigation Collection (BIC), which holds over 300,000 volumes of primary documents on the Chinese Communist movement. The purpose of Chinese Communist Materials is, without any attempt at comprehensive listing of the Bureau’s holdings, to give scholars a representative description of the collection, to point out its implications for research, and suggest new areas for research at the Bureau in the fields of political science and history [1, 4]
Self-control exertion and glucose supplementation prior to endurance performance
Objectives:
Completion of a task requiring self-control may negatively impact on subsequent self-regulatory efforts. This study explored a) whether this effect occurs during a well-practiced endurance task, b) the potential for glucose supplementation to moderate this effect, and c) whether this effect differed over time.
Method:
Fourteen trained cyclists completed four simulated 16 km time trials on an electromagnetically braked cycle ergometer. Prior to each time trial, participants completed a congruent Stroop task or an incongruent Stroop task that required self-control. They also received either a glucose-based drink or placebo. Participants’ performance time and heart rate were recorded throughout the time trials.
Results:
Multilevel growth curve analysis revealed a significant three-way interaction between self-control, glucose, and time (b = -0.91; p = 0.02). When participants did not exert self-control (congruent Stroop) or consume glucose (placebo drink) they were slowest during the early stages of the time trial but quickest over the full distance. No differences were found in heart rate across the four conditions.
Conclusions:
Findings suggest that pacing may explain why self-control exertion interferes with endurance performance. Moreover, the debate revolving around depletion of self-control must consider that any observed effects may be dependent on the timing of performance inspection
Finishing the euchromatic sequence of the human genome
The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

Therapeutic targeting of prenatal pontine ID1 signaling in diffuse midline glioma.
BACKGROUND: Diffuse midline gliomas (DMG) are highly invasive brain tumors with rare survival beyond two years past diagnosis and limited understanding of the mechanism behind tumor invasion. Previous reports demonstrate upregulation of the protein ID1 with H3K27M and ACVR1 mutations in DMG, but this has not been confirmed in human tumors or therapeutically targeted. METHODS: Whole exome, RNA, and ChIP-sequencing was performed on the ID1 locus in DMG tissue. Scratch-assay migration and transwell invasion assays of cultured cells were performed following shRNA-mediated ID1-knockdown. In vitro and in vivo genetic and pharmacologic [cannabidiol (CBD)] inhibition of ID1 on DMG tumor growth was assessed. Patient-reported CBD dosing information was collected. RESULTS: Increased ID1 expression in human DMG and in utero electroporation (IUE) murine tumors is associated with H3K27M mutation and brainstem location. ChIP-sequencing indicates ID1 regulatory regions are epigenetically active in human H3K27M-DMG tumors and prenatal pontine cells. Higher ID1-expressing astrocyte-like DMG cells share a transcriptional program with oligo/astrocyte-precursor cells (OAPCs) from the developing human brain and demonstrate upregulation of the migration regulatory protein SPARCL1. Genetic and pharmacologic (CBD) suppression of ID1 decreases tumor cell invasion/migration and tumor growth in H3.3/H3.1K27M PPK-IUE and human DIPGXIIIP* in vivo models of pHGG. The effect of CBD on cell proliferation appears to be non-ID1 mediated. Finally, we collected patient-reported CBD treatment data, finding that a clinical trial to standardize dosing may be beneficial. CONCLUSIONS: H3K27M-mediated re-activation of ID1 in DMG results in a SPARCL1+ migratory transcriptional program that is therapeutically targetable with CBD
