    Case studies of six CBFM-2 water bodies

    All-trans retinoic acid suppresses exocrine differentiation and branching morphogenesis in the embryonic pancreas

    Recent evidence has shown that retinoic acid (RA) signalling is required for early pancreatic development in zebrafish and frog but its role in later development in mammals is less clear cut. In the present study, we determined the effects of RA on the differentiation of the mouse embryonic pancreas. Addition of all-trans retinoic acid (atRA) to embryonic pancreatic cultures induced a number of changes. Branching morphogenesis and exocrine differentiation were suppressed and there was premature formation of endocrine cell clusters (although the total area of β cells was not different in control and atRA-treated buds). We investigated the mechanism of these changes and found that the premature formation of β cells was associated with the early expression of high-level Pdx1 in the endocrine cell clusters. In contrast, the suppressive effect of RA on exocrine differentiation may be due to a combination of two mechanisms (i) up-regulation of the extracellular matrix component laminin and (ii) enhancement of apoptosis. We also demonstrate that addition of fibroblast growth factor (FGF)-10 is able to partially prevent apoptosis and rescue exocrine differentiation and branching morphogenesis in atRA-treated cultures but not in mice lacking the FGF receptor 2-IIIb, suggesting the effects of FGF-10 are mediated through this receptor

    AD51B in Familial Breast Cancer

    Common variation on 14q24.1, close to RAD51B, has been associated with breast cancer: rs999737 and rs2588809 with the risk of female breast cancer and rs1314913 with the risk of male breast cancer. The aim of this study was to investigate the role of RAD51B variants in breast cancer predisposition, particularly in the context of familial breast cancer in Finland. We sequenced the coding region of RAD51B in 168 Finnish breast cancer patients from the Helsinki region for identification of possible recurrent founder mutations. In addition, we studied the known rs999737, rs2588809, and rs1314913 SNPs and RAD51B haplotypes in 44,791 breast cancer cases and 43,583 controls from 40 studies participating in the Breast Cancer Association Consortium (BCAC) that were genotyped on a custom chip (iCOGS). We identified one putatively pathogenic missense mutation c.541C>T among the Finnish cancer patients and subsequently genotyped the mutation in additional breast cancer cases (n = 5259) and population controls (n = 3586) from Finland and Belarus. No significant association with breast cancer risk was seen in the meta-analysis of the Finnish datasets or in the large BCAC dataset. The association with previously identified risk variants rs999737, rs2588809, and rs1314913 was replicated among all breast cancer cases and also among familial cases in the BCAC dataset. The most significant association was observed for the haplotype carrying the risk-alleles of all the three SNPs both among all cases (odds ratio (OR): 1.15, 95% confidence interval (CI): 1.11–1.19, P = 8.88 x 10−16) and among familial cases (OR: 1.24, 95% CI: 1.16–1.32, P = 6.19 x 10−11), compared to the haplotype with the respective protective alleles. Our results suggest that loss-of-function mutations in RAD51B are rare, but common variation at the RAD51B region is significantly associated with familial breast cancer risk

    Value chain analysis of the aquaculture feed sector in Egypt

    This study was carried out to evaluate the value chain performance of the aquaculture feed sector in Egypt, in terms of value addition, employment and profitability. The strengths and weaknesses of each link of the value chain were assessed and appropriate upgrading, management and development strategies were suggested. Quantitative data were collected for each link in the value-chain through structured questionnaires that were drafted and distributed to the key players in the sector; 25 fish feed mills and 34 fish farms covering different geographical and production regions. The results indicated that the Egyptian aquafeed value-chain is relatively simple; including only four main stakeholder groups. These are feed input suppliers, aquafeed producers, aquafeed marketers and traders and fish farmers. Between 50 and 99% of feed ingredients used in aquafeed production in Egypt are imported. About 90% of Egyptian aquafeeds are produced by the private sector in the form of conventionally pressed, pelleted feeds (80–85%) and extruded feeds (15–20%). About 85% of those producers sell their feeds directly to farmers with payment either in cash or on credit, while the remaining 15% sell through intermediaries such as traders. State-owned mills produced only 10% of total commercial fish feed production in 2012, exclusively in the form of pressed, pelleted feeds. Employment generation in private sector feed mills was 29.2 full-time equivalent (FTE) jobs per mill, with an average of 3.9 jobs per 1000 tonnes of feed produced. Employment generation in state-owned mills was much higher; with an average of 90.3 FTE per mill. Males represented 90% of the full-time employment in the state-owned mills and 96.6% in the private sector. Feed costs represent 75–90% of the total operating costs of the fish farms. The major factors impacting on the performance of the value-chain relate to inputs, to feed production, to fish farmers and to marketing and financial services. The study recommends actions to mitigate these issues including the local production of more feed raw materials, strengthening quality control and inspection, providing training for feed mills, better organization of fish farmers and improving the legal and policy environment
