    Isolation and characterization of Faecalibacterium prausnitzii from calves and piglets.

    The goal of our study was to isolate and characterize Faecalibacterium prausnitzii from fecal samples of healthy calves and piglets, in order to develop a novel probiotic for livestock animals. We identified 203 isolates of Faecalibacterium sp., which were clustered in 40 genetically distinct groups. One representative isolate from each cluster was selected for further characterization. The concentrations of the short chain fatty acids (SCFA) acetate, butyrate, propionate and isobutyrate in the culture media were measured by gas chromatography. We observed reduction in the concentration of acetate followed by concomitant increase in the concentration of butyrate, suggesting that the isolates were consuming acetate present in the media and producing butyrate. Butyrate production correlated positively with bacterial growth. Since butyrate has many benefits to the colonic epithelial cells, the selection of strains that produce higher amounts of butyrate is extremely important for the development of this potential probiotic. The effect of pH and concentration of bile salts on bacterial growth was also evaluated in order to mimic the conditions encountered by F. prausnitzii in vivo. The optimal pH for growth ranged between 5.5 and 6.7, while most isolates were inhibited by of the lowest concentration of bile salts tested (0.1%). Antimicrobial resistance profile showed that most isolates of Faecalibacterium sp. were resistant against ciprofloxacin and sulfamethoxazole-trimethoprim. More than 50% of the isolates were resistant to tetracycline, amikacin, cefepime and cefoxitin. A total of 19 different combinations of multidrug resistance were observed among the isolates. Our results provide new insights into the cultural and physiological characteristics of Faecalibacterium prausnitzii illustrating large variability in short chain fatty acid production, in vitro growth, sensitivity to bile salts, and antibiotic resistance and suggesting that future probiotic candidates should be carefully studied before elected for in vivo studies

    Both cetaceans in the Brazilian Amazon show sustained, profound population declines over two decades

    <div><p>Obligate river dolphins occur only in the rivers of Asia and South America, where they are increasingly subject to damaging pressures such as habitat degradation, food competition and entanglement in fishing gear as human populations expand. The Amazon basin hosts two, very different, dolphins—the boto or Amazon river dolphin (<i>Inia geoffrensis</i>) and the smaller tucuxi (<i>Sotalia fluviatilis</i>). Both species have wide geographical ranges and were once considered to be relatively abundant. Their IUCN Red List conservation status of Data Deficient (DD), due to limited information on threats, ecology, population numbers and trends, did not initially cause alarm. However, the development of dolphin hunting to provide fish bait at around the beginning of this millennium broadly coincided with the onset of a widespread perception that numbers of both species were in decline. Consequently, the need for population trend data to inform conservation advice and measures became urgent. This paper presents a 22-year time series of standardised surveys for both dolphins within the Mamirauá Reserve, Amazonas State, Brazil. Analysis of these data show that both species are in steep decline, with their populations halving every 10 years (botos) and 9 years (tucuxis) at current rates. These results are consistent with published, independent information on survival rates of botos in this area, which demonstrated a substantial drop in annual survival, commencing at around the year 2000. Mamirauá is a protected area, and is subject to fewer environmental pressures than elsewhere in the region, so there is no reason to suspect that the decline in dolphins within the Reserve is more pronounced than outside it. If South America's freshwater cetaceans are to avoid following their Asian counterparts on the path to a perilous conservation status, effective conservation measures are required immediately. Enforcement of existing fishery laws would greatly assist in achieving this.</p></div

    Characterization of ZSM-5 modified with niobium pentoxide: the study of thiophene adsorption

    ZSM-5 adsorbents impregnated with Nb2O5 were applied in the sulfur removal in the form of thiophene, refractory substance of difficult removal of liquid fuels. For this purpose, a model fuel containing iso-octane contaminated with thiophene in concentrations of 877.5 to 1155 ppmw was prepared. The samples were characterized by X-ray diffractometry (XRD), Fourier transform infrared (FTIR) and Fourier transform-Raman (FT-Raman) spectroscopies for confirmation of the adsorbents, being prioritized the adsorption study with that containing 5 wt.% of niobium pentoxide, because it showed a greater capacity for removal of thiophene. The best results of adsorption were achieved at 353 K, a longer time to reach equilibrium was observed. Under these conditions, the best kinetic fitting was achieved using the equation of pseudo-second order, demonstrating the domain of the phenomenon of chemisorption. While under lower temperatures, the diffusion model presented a better approximation of the experimental results. Also, the increasing of temperature did enhance spontaneous processes.Adsorventes ZSM-5 impregnados com Nb2O5 foram aplicados na remoção de enxofre sob a forma de tiofeno, substância refratária de difícil remoção dos combustíveis líquidos. Para tanto, foi preparado um combustível modelo contendo iso-octano contaminado com tiofeno nas concentrações de 877,5 a 1155 ppmw. As amostras foram caracterizadas por difratometria de raios X (XRD), espectroscopias no infravermelho (FTIR) e Raman (FT-Raman) por transformada de Fourier para confirmação dos adsorventes obtidos, sendo priorizado o estudo de adsorção com aquele contendo 5% de pentóxido de nióbio, que apresentou uma maior capacidade de remoção de tiofeno. Os melhores resultados de adsorção foram alcançados a 353 K, sendo observado um maior tempo para atingir o equilíbrio. Sob essas condições, o melhor ajuste cinético foi alcançado utilizando a equação de pseudo-segunda ordem, evidenciando o domínio do fenômeno de quimissorção. Por outro lado, sob menores temperaturas, o modelo de difusão apresentou uma melhor aproximação dos resultados experimentais. O aumento da temperatura também favoreceu os processos espontâneos

    Efficacy of pilates in functional body composition : a systematic review

    The aim of this review was to collect and systematize results of studies from the last 5 years concerning the influence of the Pilates method (PM) on functional body composition (FBC). Methods: The criteria of Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) were used in this review. We conducted research in three scientific databases: (i) Web of Science Core Collection, (ii) SCOPUS, and (iii) search directory of the library catalog of the Faculty of Sport and Physical Education of the University of Coimbra—EBSCO Discovery Services. We found 334 articles, covering the period between 1 January 2017 and 31 December 2021. After the selection process, we found 33 eligible articles. Results: The main results seem to point to a tendency to get a better body weight and body fat percentage (BFP), hip circumference (HC), waist, and skinfolds from the chest, abdomen, triceps, and supra-iliac in mostly female samples. Available evidence indicates that, in body composition (BC) and FBC, Pilates practice tends to be effective in reducing obesity as a multifactorial condition. Conclusions: It is concluded that there is a marked trend in the benefits of Pilates in FBC, which is in line with other systematic reviews at the BC level (which includes body weight (BW) and body mass index (BMI)) and in the reduction of the percentage of fat mass (FM). It is also concluded that there is a vast lack of studies on the male population, preventing further scientific development in this area. The limitations of this systematic review can be overcome with studies that bring together multidisciplinary aspects of FBC, better designed and methodologically more robust, which will allow more reliable analyses for the implementation of the Pilates method in terms of FBC. In addition, further studies with a male sample or mixed samples (men vs. women) could confirm the trend of no gender differences in the benefits of Pilates practice.info:eu-repo/semantics/publishedVersio

    The use of transvaginal synthetic mesh for anterior vaginal wall prolapse repair: a randomized controlled trial

    The aim of the study was to compare the efficacy and safety of transvaginal trocar-guided polypropylene mesh insertion with traditional colporrhaphy for treatment of anterior vaginal wall prolapse.This is a randomized controlled trial in which women with advanced anterior vaginal wall prolapse, at least stage II with Ba a parts per thousand yenaEuro parts per thousand+1 cm according to the Pelvic Organ Prolapse Quantification (POP-Q) classification, were randomly assigned to have either anterior colporrhaphy (n = 39) or repair using trocar-guided transvaginal mesh (n = 40). the primary outcome was objective cure rate of the anterior compartment (point Ba) assessed at the 12-month follow-up visit, with stages 0 and I defined as anatomical success. Secondary outcomes included quantification of other vaginal compartments (POP-Q points), comparison of quality of life by the prolapse quality of life (P-QOL) questionnaire, and complication rate between the groups after 1 year. Study power was fixed as 80 % with 5 % cutoff point (p 0.05).Trocar-guided transvaginal synthetic mesh for advanced anterior POP repair is associated with a higher anatomical success rate for the anterior compartment compared with traditional colporrhaphy. Quality of life equally improved after both techniques.
