    Online sobriety communities for women's problematic alcohol use: A mini review of existing qualitative and quantitative research

    The increase in women's drinking is one of the most prominent trends in alcohol consumption in the UK in recent history, possibly exacerbated by COVID-19 lockdown measures. Higher rates of drinking are associated with substantial economic, health, and social costs. However, women are less likely to seek treatment for Alcohol Use Disorder (AUD) than men and have less successful treatment outcomes from traditional treatment paths, such as 12-step programs and in-patient care. Female heavy drinkers may also experience particular forms of gendered stigma that affect their experiences of addiction and recovery and their desire or ability to access these more "traditional" services. This review provides an overview of existing qualitative and quantitative research regarding online sobriety communities that are predominantly utilised by women, such as non-12-step alcohol online support groups (AOSGs) and temporary abstinence initiatives (TAIs). This is a small-but expanding-body of literature emerging as "sober curiosity" and "mindful drinking" are trending in Western contexts such as the UK, particularly amongst young women who do not identify with traditional, binary recovery language such as "alcoholic" and "addict." This review highlights the gaps in research and concludes that further research regarding these new treatment pathways, and women's experiences when utilising them, must be conducted to provide more evidence-based options for women who want to address problematic drinking. Public health bodies could also learn more effective strategies from these innovative solutions to reduce alcohol consumption generally. [Abstract copyright: Copyright © 2021 Davey.

    Sober Women’s Feminist Resistance to Alcohol Marketing and Cultural Representations of Women’s Drinking Practices

    Alcohol is marketed to women as a glamorous and empowering reward for juggling the demands of work and family life. This essay explores the ways in which women who do not drink reject the feminization of alcohol and drinking practices and frame this rejection within discourses of feminist resistance. This essay draws on data collected as part of a mixed-method ethnographic research project that investigates women’s use of, and participation in, online sobriety communities. Findings suggest that women who lead or utilize online sobriety communities have considerable awareness of the feminized marketing of alcohol, and some express strong ideological opposition to it. The marketing of alcohol is positioned as a predatory force that takes advantage of women’s exhaustion as mothers and perpetuates the double standards associated with women’s drinking. Sobriety may prompt a feminist awakening regarding the connections between the feminization of alcohol and women’s inequality within society and, in turn, disrupt women’s identification with post-feminist cultural representations of women’s drinking practices. Through the public identification and critique of these marketing practices, women critically engage with feminism while raising consciousness and building a community of sober women

    The cationic region of Rhes mediates its interactions with specific Gβ subunits

    Ras homologue enriched in striatum (Rhes) is a small monomeric G protein which functions in a variety of cellular processes, including attenuation of G protein-coupled receptor (GPCR)signalling. There have been many studies into the effects of Rhes, but there is no molecular information about how Rhes might bring about these effects. Rhes shares striking sequence homology to AGS1 (activator of G protein signalling 1) and we considered whether the two proteins function in similar ways. AGS1 binds to the Gβ1 subunit of heterotrimeric G proteins and we have used yeast two-hybrid studies to show that Rhes binds selectively to Gβ1, Gβ2 and Gβ3 subunits. Binding to the Gβ subunits involves the cationic regions of AGS1 and Rhes, and we used Rhes-AGS1 chimeras to show that their different cationic regions determine the Gβ-specificity of the interactions. Possible implications of this interaction for the activity of Rhes are discussed

    Patterns of Consumption at the UK’s First “Alcohol-Free Off-Licence”: Who Engaged with No- and Low-Alcohol Drinks and Why?

    No- and low-alcohol beverages are currently experiencing high sales growth in the UK, but academic research regarding the production, regulation, marketing and consumption of these drinks remains limited. This article presents research findings from ethnographic customer observations and semi-structured staff interviews at Club Soda’s temporary “alcohol-free off-licence” in London – the UK’s first shop that sold exclusively no- and low-alcohol drinks. I analyse the demographics of who came to the off-licence, and how and why they engaged with no- and low-alcohol drinks. Findings suggest that relatively equal numbers of non-drinkers and current drinkers were customers of the off-licence, but there were differences across gender, ethnicity, and age groups. No- and low-alcohol drinks supported customers’ alcohol-free lifestyles or attempts to drink mindfully. However, whether customers were former drinkers, lifelong abstainers or current drinkers influenced what products they sought from the off-licence and shaped how these products were used in (non-)drinking practices. This paper also highlights the role of the off-licence as a stigma-free, sober space where customers could try no- and low-alcohol products, prior to purchase, and find connection with others

    Writing the risk of cancer: Cancer risk in public policy

    In this paper we examine how cancer risk is written in cancer policy documents from the English speaking OECD nations. We offer an audit of the multiple ways in which cancer risk is conceptualised and presented in health policy and professional contexts with the long term aim of comparing this with lay conceptualisations. Our study sampled cancer policy documents produced by six nations, the World Health Organization and the International Union for Cancer Control since 2000 and analysed them iteratively through questions and codes. Whilst the documents contained a comprehensive range of concepts and locations for cancer risk, our analysis found two predominant representations: firstly, well established metaphors that depict cancer as uniformly dreadful and life threatening; and secondly, through a concentration on five behavioural risk factors (tobacco smoking, drinking alcohol, inadequate nutrition, sun exposure and physical inactivity) and one bodily state (overweight). We discuss the implications of this dual focus and of other tensions within ideas about cancer risk that we identified for risk communication

    Differential effects of RGS proteins on Gαq and Gα11 activity

    Heterotrimeric G proteins play a pivotal role in GPCR signalling; they link receptors to intracellular effectors and their inactivation by RGS proteins is a key factor in resetting the pathway following stimulation. The precise GPCR:G protein:RGS combination determines the nature and duration of the response. Investigating the activity of particular combinations is difficult in cells which contain multiples of each component. We have therefore utilised a previously characterised yeast system to express mammalian proteins in isolation. Human Gαq and Gα11 spontaneously activated the yeast pheromone-response pathway by a mechanism which required the formation of Gα-GTP. This provided an assay for the specific activity of human RGS proteins. RGS1, RGS2, RGS3 and RGS4 inhibited the spontaneous activity of both Gαq and Gα11 but, in contrast, RGS5 and RGS16 were much less effective against Gα11 than Gαq. Interestingly, RGS2 and RGS3 were able to inhibit signalling from the constitutively active Gαq QL/Gα11 QL mutants, confirming the GAP-independent activity of these RGS proteins. To determine if the RGS-Gα specificity was maintained under conditions of GPCR stimulation, minor modifications to the C-terminus of Gαq/Gα11 enabled coupling to an endogenous receptor. RGS2 and RGS3 were effective inhibitors of both Gα subunits even at high levels of receptor stimulation, emphasising their GAP-independent activity. At low levels of stimulation RGS5 and RGS16 retained their differential Gα activity, further highlighting that RGS proteins can discriminate between two very closely related Gα subunits

    Dual positive and negative regulation of GPCR signaling by GTP hydrolysis

    G protein-coupled receptors (GPCRs) regulate a variety of intracellular pathways through their ability to promote the binding of GTP to heterotrimeric G proteins. Regulator of G protein signaling (RGS) proteins increase the intrinsic GTPase activity of G-subunits and are widely regarded as negative regulators of G protein signaling. Using yeast we demonstrate that GTP hydrolysis is not only required for desensitization, but is essential for achieving a high maximal (saturated level) response. Thus RGS-mediated GTP hydrolysis acts as both a negative (low stimulation) and positive (high stimulation) regulator of signaling. To account for this we generated a new kinetic model of the G protein cycle where GGTP enters an inactive GTP-bound state following effector activation. Furthermore, in vivo and in silico experimentation demonstrates that maximum signaling output first increases and then decreases with RGS concentration. This unimodal, non-monotone dependence on RGS concentration is novel. Analysis of the kinetic model has revealed a dynamic network motif that shows precisely how inclusion of the inactive GTP-bound state for the G produces this unimodal relationship

    Phase relationships for molecular salts

    The pharmaceutical industry has for many years been interested in the formation of salts as a means of controlling the chemical/physical properties of API (active pharmaceutical ingredient) molecules. While current interest in cocrystals has led to study of their formation and phase behaviour, comparatively little data is available for salts. This work aims to come some way to redress that balance. Initial studies focussed on a single base manually ground with a number of acids (leading on from previous solution work). Salt formation was noted in a significant number of cases. The work was followed by two more detailed and contrasting case studies. These case studies are ephedrine with pimelic acid and benzoic acid. Both mono- and di-basic salts of ephedrine and pimelic acid were formed both by grinding and solution methods. Binary and aqueous ternary phase diagrams were measured to allow assessment of pure phase regions. The ideality of the systems was assessed and, while the binary phase diagram showed some relatively ideal regions, the ternary system appeared highly non-ideal. Effects on the shape and size of phase regions have been considered, especially the role ionisation plays in the ternary non-ideality. Using fundamental equations solubility has been determined as a function of pH and compared to experimental data. The pH value has also been calculated using concentrations of acid and base along the experimental liquidus of the ternary phase diagram and comparisons made with measured pH values. From these comparisons it was determined that the non-ideality and shape of the liquidus in the ephedrine/pimelic acid/water ternary phase diagram is not solely due to ionisation of the components. Some major issues that may arise in attempted salt formation studies are discussed in relation to the ephedrine and benzoic acid system. Aqueous salt formation was not successful; therefore reasons for this failure are discussed. It is concluded that while the ternary phase behaviour of a salt should be considered, it does not mean the ionisation behaviour (and hence the speciation diagram) should be ignored.     Ribosomal DNA sequence heterogeneity reflects intraspecies phylogenies and predicts genome structure in two contrasting yeast species

    The ribosomal RNA encapsulates a wealth of evolutionary information, including genetic variation that can be used to discriminate between organisms at a wide range of taxonomic levels. For example, the prokaryotic 16S rDNA sequence is very widely used both in phylogenetic studies and as a marker in metagenomic surveys and the internal transcribed spacer region, frequently used in plant phylogenetics, is now recognized as a fungal DNA barcode. However, this widespread use does not escape criticism, principally due to issues such as difficulties in classification of paralogous versus orthologous rDNA units and intragenomic variation, both of which may be significant barriers to accurate phylogenetic inference. We recently analyzed data sets from the Saccharomyces Genome Resequencing Project, characterizing rDNA sequence variation within multiple strains of the baker's yeast Saccharomyces cerevisiae and its nearest wild relative Saccharomyces paradoxus in unprecedented detail. Notably, both species possess single locus rDNA systems. Here, we use these new variation datasets to assess whether a more detailed characterization of the rDNA locus can alleviate the second of these phylogenetic issues, sequence heterogeneity, while controlling for the first. We demonstrate that a strong phylogenetic signal exists within both datasets and illustrate how they can be used, with existing methodology, to estimate intraspecies phylogenies of yeast strains consistent with those derived from whole-genome approaches. We also describe the use of partial Single Nucleotide Polymorphisms, a type of sequence variation found only in repetitive genomic regions, in identifying key evolutionary features such as genome hybridization events and show their consistency with whole-genome Structure analyses. We conclude that our approach can transform rDNA sequence heterogeneity from a problem to a useful source of evolutionary information, enabling the estimation of highly accurate phylogenies of closely related organisms, and discuss how it could be extended to future studies of multilocus rDNA systems.
