    Forecasting demand at the individual stock-keeping-unit (SKU) level often necessitates the use of statistical methods, such as exponential smoothing. In some organizations, however, statistical forecasts will be subject to judgemental adjustments by managers. Although a number of empirical and ‘laboratory’ studies have been performed in this area, no formal OR modelling has been conducted to offer insights into the impact such adjustments may have on supply chain performance and the potential development of mitigation mechanisms. This is because of the associated dynamic complexity and the situation-specific nature of the problem at hand. In conjunction with appropriate stock control rules, demand forecasts help decide how much to order. It is a common practice that replenishment orders may also be subject to judgemental intervention, adding further to the dynamic system complexity and interdependence. The system dynamics (SD) modelling method can help advance knowledge in this area, where mathematical modelling cannot accommodate the associated complexity. This study, which constitutes part of a UK government funded (EPSRC) project, uses SD models to evaluate the effects of forecasting and ordering adjustments for a wide set of scenarios involving: three different inventory policies; seven different (combinations of) points of intervention; and four different (combinations of) types of judgmental intervention (optimistic and pessimistic). The results enable insights to be gained into the performance of the entire supply chain. An agenda for further research concludes the paper

    Leaf water δ 18/O, δ2H and d-excess isoscapes for Australia using region-specific plant parameters and non-equilibrium vapour

    Oxygen (δ18O) and hydrogen (δ2H) isotope ratios, and their relationship to one another (d-excess) are altered as water travels from the atmosphere to the land surface, into soils and plants and back to the atmosphere. Plants return water to the atmosphere through transpiration (evaporation through the stomata), which causes isotopic fractionation concentrating the heavier isotopes (18O and 2H) in the water that remains behind in the leaves. The degree of isotopic fractionation during transpiration is controlled largely by climate, and as a result can be predicted using process-based models and climate data. The modelled transpirational isotopic fractionation can be applied to plant source water isotopic values to predict leaf water isotope ratios and generate maps of isotopic composition, or isoscapes. This approach of mechanistic modelling has been well demonstrated in the first generation of global leaf water isoscapes (PLoS One, 3(6), e2447, 2008). However, use of leaf water isoscapes in fields such as hydrology, ecology, and forensics requires a new generation of updated region-specific isoscapes. Here, we generate leaf water isoscapes of δ18O, δ2H and d-excess for Australia, the driest vegetated continent on Earth, where leaf water represents a critical water resource for ecosystems. These isoscapes represent an improvement over previous global isoscapes due to their higher resolution, region-specific, empirically derived plant parameters, and non-equilibrium corrections for water vapour isotopic composition. The new isoscapes for leaf water are evaluated relative to observed isotope ratios of leaf cellulose and cherry juice. The model predictions for annual average leaf water isotope ratios showed strong correlations with these plant tissues that integrate over time. Moreover, inclusion of region-specific leaf temperature estimates and non-equilibirum vapour corrections improved prediction accuracy. Regionally based isoscapes provide improved characterisations of average leaf water isotope ratios needed to support research in hydrology, plant ecophysiology, atmospheric science, ecology, and geographic provenancing of biological materials

    The eventization of leisure and the strange death of alternative Leeds

    The communicative potential of city spaces as leisure spaces is a central assumption of political activism and the creation of alternative, counter-cultural and subcultural scenes. However, such potential for city spaces is limited by the gentrification, privatization and eventization of city centres in the wake of wider societal and cultural struggles over leisure, work and identity formation. In this paper, we present research on alternative scenes in the city of Leeds to argue that the eventization of the city centre has led to a marginalization and of alternative scenes on the fringes of the city. Such marginalization has not caused the death of alternative Leeds or political activism associated with those scenes—but it has changed the leisure spaces (physical, political and social) in which alternative scenes contest the mainstream

    A method for purification, identification and validation of DNMT1 mRNA binding proteins

    DNA methyltransferase 1 (DNMT1) is the enzyme responsible for the maintenance of DNA methylation patterns during cell division. DNMT1 expression is tightly regulated within the cell cycle. Our previous study showed that the binding of a protein with an apparent size of ~40 kDa on DNMT1 3’-UTR triggered the destabilization of DNMT1 mRNA transcript during Go/G1 phase. Using RNA affinity capture with the 3’-UTR of DNMT1 mRNA and matrix-assisted laser desorption-time of flight tandem mass spectrometry (MALDI-TOF-MS-MS) analysis, we isolated and identified AUF 1 (AU-rich element ARE:poly-(U)-binding/degradation factor) as the binding protein. We then validated the role of this protein in the destabilization of DNMT1 mRNA.     End of the Kiaman Superchron in the Permian of SW England: Magnetostratigraphy of the Aylesbeare Mudstone and Exeter groups

    The chronology of Permian strata in SW England is fragmentary and largely based on radiometric dating of associated volcanic units. Magnetostratigraphy from the c. 2 km of sediments in the Exeter and Aylesbeare Mudstone groups was undertaken to define a detailed chronology, using the end of the Kiaman Superchron and the overlying reverse and normal polarity in the Middle and Upper Permian as age constraints. The palaeomagnetic directions are consistent with other European Permian palaeopoles, with data passing fold and reversal tests. The end of the Kiaman Superchron (in the Wordian) occurs in the uppermost part of the Exeter Group. The overlying Aylesbeare Mudstone Group is early Capitanian to latest Wuchiapingian in age. The Changhsingian and most of the Lower Triassic sequence is absent. Magnetostratigraphic comparison with the Southern Permian Basin shows that the Exeter and Aylesbeare Mudstone groups are closely comparable in age with the Havel and Elbe Subgroups of the Rotliegend II succession. The Altmark unconformities in these successions appear similar in age to the sequence boundaries in SW England, indicating that both may be climate controlled. Clasts in the Exeter Group, from unroofing of the Dartmoor Granite, first occurred at a minimum of c. 8 myr after formation of the granite

    What Qualities Are Most Important to Making a Point of Care Test Desirable for Clinicians and Others Offering Sexually Transmitted Infection Testing?

    To investigate the possible effects of different levels of attributes of a point-of-care test (POCT) on sexually transmitted infection (STI) professionals' decisions regarding an ideal POCT for STI(s).An online survey was designed based on a large-scale in-depth focus discussion study among STI experts and professionals. The last section of the survey "build your own POCT" was designed by employing the discrete choice experiment approach. Practicing clinicians from two venues, STI-related international conference attendees and U.S. STD clinic clinicians were invited to participate in the survey. Conditional logistical regression modeling was used for data analysis.Overall, 256 subjects took the online survey with 218 (85%) completing it. Most of the participants were STD clinic clinicians who already used some POCTs in their practice. "The time frame required" was identified as a major barrier that currently made it difficult to use STI POCTs. Chlamydia trachomatis was the organism chosen as the top priority for a new POCT, followed by a test that would diagnose early seroconversion for HIV, and a syphilis POCT. Without regard to organism type selected, sensitivity of 90-99% was always the most important attribute to be considered, followed by a cost of $20. However, when the test platform was prioritized for early HIV seroconversion or syphilis, sensitivity was still ranked as most important, but specificity was rated second most important.STI professionals preferred C. trachomatis as the top priority for a new POCT with sensitivity over 90%, low cost, and a very short completion time

    Assessment of the centre of pressure pattern and moments about S2 in scoliotic subjects during normal walking

    Background Context: Research employing gait measurements indicate asymmetries in ground reaction forces and suggest relationships between these asymmetries, neurological dysfunction and spinal deformity. Although, studies have documented the use of centre of pressure (CoP) and net joint moments in gait assessment and have assessed centre of mass (CoM)-CoP distance relationships in clinical conditions, there is a paucity of information relating to the moments about CoM. It is commonly considered that CoM is situated around S2 vertebra in normal upright posture and hence this study uses S2 vertebral prominence as reference point relative to CoM. Purpose: To assess and establish asymmetry in the CoP pattern and moments about S2 vertebral prominence during level walking and its relationship to spinal deformity in adolescents with scoliosis. Patient sample: Nine Adolescent Idiopathic Scoliosis subjects (8 females and 1 male with varying curve magnitudes and laterality) scheduled for surgery within 2-3 days after data collection, took part in this study. Outcome measures: Kinetic and Kinematic Gait assessment was performed with an aim to estimate the CoP displacement and the moments generated by the ground reaction force about the S2 vertebral prominence during left and right stance during normal walking. Methods: The study employed a strain gauge force platform to estimate the medio-lateral and anterior-posterior displacement of COP and a six camera motion analysis system to track the reflective markers to assess the kinematics. The data were recorded simultaneously. Results: Results indicate wide variations in the medio lateral direction CoP, which could be related to the laterality of both the main and compensation curves. This variation is not evident in the anterior-posterior direction. Similar results were recorded for moments about S2 vertebral prominence. Subjects with higher left compensation curve had greater displacement to the left. Conclusion: Although further longitudinal studies are needed, results indicate that the variables identified in this study are applicable to initial screening and surgical evaluation of scoliosis. © 2008 Chockalingam et al; licensee BioMed Central Ltd
