    Synthesis of Oligosaccharides Derived from Lactulose (OsLu) Using Soluble and Immobilized Aspergillus oryzae β-Galactosidase

    β-Galactosidase from Aspergillus oryzae offers a high yield for the synthesis of oligosaccharides derived from lactulose (OsLu) by transgalactosylation. Oligosaccharides with degree of polymerization (DP) ≥ 3 have shown to possess higher in vitro bifidogenic effect than di- and tetrasaccharides. Thus, in this work, an optimization of reaction conditions affecting the specific selectivity of A. oryzae β-galactosidase for synthesis of OsLu has been carried out to enhance OsLu with DP ≥ 3 production. Assays with β-galactosidase immobilized onto a glutaraldehyde–agarose support were also carried out with the aim of making the process cost-effective and industrially viable. Optimal conditions with both soluble and immobilized enzyme for the synthesis of OsLu with DP ≥ 3 were 50 °C, pH 6.5, 450 g/L of lactulose, and 8 U/mL of enzyme, reaching yields of ca. 50% (w/v) of total OsLu and ca. 20% (w/v) of OsLu with DP 3, being 6′-galactosyl-lactulose the major one, after a short reaction time. Selective formation of disaccharides, however, was favored at 60 °C, pH 4.5, 450 g/L of lactulose and 8 U/mL of enzyme. Immobilization increased the enzymatic stability to temperature changes and allowed to reuse the enzyme.     Characterization and improvement of rheological properties of sodium caseinate glycated with galactose, lactose and dextran

    10 páginas, 7 figuras, 2 tablas.-- El pdf del artículo es la versión de autor.The aim of this work was to investigate the effect of non-enzymatic glycosylation with galactose, lactose, and 10 kDa dextran on the rheological properties of sodium caseinate. To promote the formation of covalent complexes, the reaction was done in solid state (aw = 0.67), pH 7.0 (0.1 M sodium phosphate buffer), and temperature set at 50 and 60 °C. The progress of Maillard reaction was indirectly traced by measuring the formation of the Amadori compound, through furosine (2-furoylmethyl-lysine) analysis, and brown polymers, and the resulting glycoconjugates were characterized by LC/ESI-MS and SEC. Results showed a higher reactivity of galactose than lactose and dextran to form the glycoconjugates, due to its smaller molecular weight. Glycation with galactose and lactose increased the viscosity of caseinate and also altered its flow characteristics from Newtonian to shear-thinning. Oscillatory testing showed a higher elastic modulus (G′) in glycoconjugates when compared to non-glycated caseinate, especially with galactose, where a gel-like behaviour was observed after long incubation times. Glycation with dextran did not produce substantial improvements in the rheological properties of caseinate, probably due to the limited extent of the reaction.     First Report of Soybean (Glycine max) Disease Caused by Pseudomonas aeruginosa in Cuba

    Soybean (Glycine max L.) has become one of the most widely consumed foods, however diseases caused by microorganisms can affect yields and seed quality. In March 2015, during routine survey in a soybean growing area in Pinar del Rio province, Cuba; disease symptoms were observed in some leaves. These included water-soaked necrotic spots with surrounded chlorotic halos, especially on the margins of the leaves. To identify the possible pathogens involved, leaves were disinfected with tap water, 70% ethanol and were rinsed with sterile distilled water. Small segments from diseased tissue were macerated in sterile 0.85% NaCl solution, decimal dilutions were performed and 20 µL aliquots were streaked onto King´s B medium (KB). After 24 hours of incubation at 28 oC, a fluorescent pseudomonad was isolated. Colonies were round, smooth and produced yellowish-green diffusible pigments on KB.     Determinantes del financiamiento externo de las PyMEs del MERCOSUR

    El presente trabajo analiza un conjunto de variables como posibles determinantes en el acceso al financiamiento externo de las PyMEs pertenecientes a cuatro países miembros del MERCOSUR: Argentina, Brasil, Uruguay y Paraguay. Se utiliza la base de datos Enterprise Survey del Banco Mundial y se aplican modelos Logit a fin de probar las hipótesis planteadas. Los resultados encontrados indican que el financiamiento con proveedores y el tamaño de la empresa son variables significativas para el acceso al financiamiento de las PyMEs en la mayoría de los países, mientras que las variables que miden las exportaciones, el género del gerente y el sector al que pertenecen las PyMEs alteran su significatividad de acuerdo al país analizado. Las variables que miden la antigüedad de la empresa y su forma jurídica resultaron ser no significativas en todos los países de la muestra.This paper analyzes a set of variables as possible determinants in access to external financing of SMEs belonging to four-member countries of MERCOSUR: Argentina, Brazil, Uruguay and Paraguay. The World Bank Enterprise Survey database is used and Logit models are applied in order to test the hypotheses proposed. The results found indicate that the financing with suppliers and the size of the company are significant variables for the access to financing of SMEs in most of the countries, while the variables that measure exports, the gender of the manager and the sector which belong to SMEs alter their significance according to the country analyzed. The variables that measure the age of the company and its legal form were not significant in all the countries in the sample

    A multiproxy study distinguishes environmental change from diagenetic alteration in the recent sedimentary record of the inner Cadiz Bay (SW Spain)

    In this study, we reconstruct the recent environmental evolution of the inner Cadiz Bay using sedimentary records reaching back as far as AD 1700. We report lithological descriptions of the sediments and extensive mineralogical and geochemical analyses. An extraction technique that identifies different Fe phases provides an assessment of diagenetic alteration, which allows an estimation of the original organic matter inputs to the inner Cadiz Bay. Downcore variations in Corg/N ratios, δ13Corg and δ15N are related to changes in organic matter sources and the trophic state of the water column. The downcore records of selected trace metals (e.g. Pb, Zn and Cu) are interpreted to reflect changes in heavy metal pollution in the bay, while records of other elements (e.g. Mn and P) are likely overprinted by diagenetic alteration. Major environmental shifts took place during the 20th century, when the population around Cadiz Bay increased exponentially. Increases in sediment accumulation rates, organic matter inputs and heavy metal contents, in parallel with increases in δ13Corg and δ15N over this period, are interpreted as direct effects of the increasing anthropogenic influence in the area. The results of this study suggest that multiproxy approaches and detailed consideration of diagenetic overprinting are required to reconstruct past environmental conditions from coastal sediments

    Quasiexperimental intervention study protocol to optimise the use of new antibiotics in Spain: the NEW_SAFE project

    Introduction Ceftaroline, tedizolid, dalbavancin, ceftazidime-avibactam and ceftolozane-tazobactam are novel antibiotics used to treat infections caused by multidrug-resistant pathogens (MDR). Their use should be supervised and monitored as part of an antimicrobial stewardship programme (ASP). Appropriate use of the new antibiotics will be improved by including consensual indications for their use in local antibiotic guidelines, together with educational interventions providing advice to prescribers to ensure that the recommendations are clearly understood. Methods and analysis This study will be implemented in two phases. First, a preliminary historical cohort (2017-2019) of patients from 13 Andalusian hospitals treated with novel antibiotics will be analysed. Second, a quasiexperimental intervention study will be developed with an interrupted time-series analysis (2020-2021). The intervention will consist of an educational interview between prescribers and ASP leaders at each hospital to reinforce the proper use of novel antibiotics. The educational intervention will be based on a consensus guideline designed and disseminated by leaders after the retrospective cohort data have been analysed. The outcomes will be acceptance of the intervention and appropriateness of prescription. Incidence of infection and colonisation with MDR organisms as well as incidence ofClostridioides difficileinfection will also be analysed. Changes in prescription quality between periods and the safety profile of the antibiotics in terms of mortality rate and readmissions will also be measured. Ethics and dissemination Ethical approval will be obtained from the Andalusian Coordinating Institutional Review Board. The study is being conducted in compliance with the protocol and regulatory requirements consistent with International Council of Harmonisation E6 Good Clinical Practice and the ethical principles of the latest version of the Declaration of Helsinki. The results will be published in peer-reviewed journals and disseminated at national and international conferences

    Epidemiology of Invasive Fungal Infections in Latin America

    The pathogenic role of invasive fungal infections (IFIs) has increased during the past two decades in Latin America and worldwide, and the number of patients at risk has risen dramatically. Working habits and leisure activities have also been a focus of attention by public health officials, as endemic mycoses have provoked a number of outbreaks. An extensive search of medical literature from Latin America suggests that the incidence of IFIs from both endemic and opportunistic fungi has increased. The increase in endemic mycoses is probably related to population changes (migration, tourism, and increased population growth), whereas the increase in opportunistic mycoses may be associated with the greater number of people at risk. In both cases, the early and appropriate use of diagnostic procedures has improved diagnosis and outcome
