    The Betic Ophiolites and the Mesozoic Evolution of the Western Tethys

    The Betic Ophiolites consist of numerous tectonic slices, metric to kilometric in size, of eclogitized mafic and ultramafic rocks associated to oceanic metasediments, deriving from the Betic oceanic domain. The outcrop of these ophiolites is aligned along 250 km in the Mulhacen Complex of the Nevado-Filabride Domain, located at the center-eastern zone of the Betic Cordillera (SE Spain). According to petrological/geochemical inferences and SHRIMP (Sensitive High Resolution Ion Micro-Probe) dating of igneous zircons, the Betic oceanic lithosphere originated along an ultra-slow mid-ocean ridge, after rifting, thinning and breakup of the preexisting continental crust. The Betic oceanic sector, located at the westernmost end of the Tethys Ocean, developed from the Lower to Middle Jurassic (185-170 Ma), just at the beginning of the Pangaea break-up between the Iberia-European and the Africa-Adrian plates. Subsequently, the oceanic spreading migrated northeastward to form the Ligurian and Alpine Tethys oceans, from 165 to 140 Ma. Breakup and oceanization isolated continental remnants, known as the Mesomediterranean Terrane, which were deformed and affected by the Upper Cretaceous-Paleocene Eo-Alpine high-pressure metamorphic event, due to the intra-oceanic subduction of the Jurassic oceanic lithosphere and the related continental margins. This process was followed by the partial exhumation of the subducted oceanic rocks onto their continental margins, forming the Betic and Alpine Ophiolites. Subsequently, along the Upper Oligocene and Miocene, the deformed and metamorphosed Mesomediterranean Terrane was dismembered into different continental blocks collectively known as AlKaPeCa microplate (Alboran, Kabylian, Peloritan and Calabrian). In particular, the Alboran block was displaced toward the SW to occupy its current setting between the Iberian and African plates, due to the Neogene opening of the Algero-Provencal Basin. During this translation, the different domains of the Alboran microplate, forming the Internal Zones of the Betic and Rifean Cordilleras, collided with the External Zones representing the Iberian and African margins and, together with them, underwent the later alpine deformation and metamorphism, characterized by local differences of P-T (Pressure-Temperature) conditions. These Neogene metamorphic processes, known as Meso-Alpine and Neo-Alpine events, developed in the Nevado-Filabride Domain under Ab-Ep amphibolite and greenschists facies conditions, respectively, causing retrogradation and intensive deformation of the Eo-Alpine eclogites.This research was funded by Project CGL2009-12369 of the Spanish Ministry of Science and Innovation, co-financed with FEDER funds, and by Research Group RNM 333 of Junta de Andalucía (Spain)

    Bves Modulates Tight Junction Associated Signaling

    Blood vessel epicardial substance (Bves) is a transmembrane adhesion protein that regulates tight junction (TJ) formation in a variety of epithelia. The role of TJs within epithelium extends beyond the mechanical properties. They have been shown to play a direct role in regulation of RhoA and ZONAB/DbpA, a y-box transcription factor. We hypothesize that Bves can modulate RhoA activation and ZONAB/DbpA activity through its regulatory effect on TJ formation. Immortalized human corneal epithelial (HCE) cells were stably transfected with Flag-tagged full length chicken Bves (w-Bves) or C-terminus truncated Bves (t-Bves). We found that stably transfected w-Bves and t-Bves were interacting with endogenous human Bves. However, interaction with t-Bves appeared to disrupt cell membrane localization of endogenous Bves and interaction with ZO-1. w-Bves cells exhibited increased TJ function reflected by increased trans-epithelial electrical resistance, while t-Bves cells lost TJ protein immunolocalization at cell-cell contacts and exhibited decreased trans-epithelial electrical resistance. In parental HCE and w-Bves cells ZONAB/DbpA and GEF-H1 were seen at cell borders in the same pattern as ZO-1. However, expression of t-Bves led to decreased membrane localization of both ZONAB/DbpA and GEF-H1. t-Bves cells had increased RhoA activity, as indicated by a significant 30% increase in FRET activity compared to parental HCE cells. ZONAB/DbpA transcriptional activity, assessed using a luciferase reporter probe, was increased in t-Bves cells. These studies demonstrate that Bves expression and localization can regulate RhoA and ZONAB/DbpA activity

    Potent suppression of vascular smooth muscle cell migration and human neointimal hyperplasia by KV1.3 channel blockers

    Aim - The aim of the study was to determine the potential for KV1 potassium channel blockers as inhibitors of human neoinitimal hyperplasia. Methods and results - Blood vessels were obtained from patients or mice and studied in culture. Reverse transcriptasepolymerase chain reaction and immunocytochemistry were used to detect gene expression. Whole-cell patch-clamp, intracellular calcium measurement, cell migration assays, and organ culture were used to assess channel function.  KV1.3 was unique among the  KV1 channels in showing preserved and up-regulated expression when the vascular smooth muscle cells switched to the proliferating phenotype. There was strong expression in neointimal formations. Voltage-dependent potassium current in proliferating cells was sensitive to three different blockers of  KV1.3 channels. Calcium entry was also inhibited. All three blockers reduced vascular smooth muscle cell migration and the effects were non-additive. One of the blockers (margatoxin) was highly potent, suppressing cell migration with an IC of 85 pM. Two of the blockers were tested in organ-cultured human vein samples and both inhibited neointimal hyperplasia. Conclusion - KV1.3 potassium channels are functional in proliferating mouse and human vascular smooth muscle cells and have positive effects on cell migration. Blockers of the channels may be useful as inhibitors of neointimal hyperplasia and other unwanted vascular remodelling events

    A systematic review of tests for lymph node status in primary endometrial cancer

    <p>Abstract</p> <p>Background</p> <p>The lymph node status of a patient is a key determinate in staging, prognosis and adjuvant treatment of endometrial cancer. Despite this, the potential additional morbidity associated with lymphadenectomy makes its role controversial. This study systematically reviews the accuracy literature on sentinel node biopsy; ultra sound scanning, magnetic resonance imaging (MRI) and computer tomography (CT) for determining lymph node status in endometrial cancer.</p> <p>Methods</p> <p>Relevant articles were identified form MEDLINE (1966–2006), EMBASE (1980–2006), MEDION, the Cochrane library, hand searching of reference lists from primary articles and reviews, conference abstracts and contact with experts in the field. The review included 18 relevant primary studies (693 women). Data was extracted for study characteristics and quality. Bivariate random-effect model meta-analysis was used to estimate diagnostic accuracy of the various index tests.</p> <p>Results</p> <p>MRI (pooled positive LR 26.7, 95% CI 10.6 – 67.6 and negative LR 0.29 95% CI 0.17 – 0.49) and successful sentinel node biopsy (pooled positive LR 18.9 95% CI 6.7 – 53.2 and negative LR 0.22, 95% CI 0.1 – 0.48) were the most accurate tests. CT was not as accurate a test (pooled positive LR 3.8, 95% CI 2.0 – 7.3 and negative LR of 0.62, 95% CI 0.45 – 0.86. There was only one study that reported the use of ultrasound scanning.</p> <p>Conclusion</p> <p>MRI and sentinel node biopsy have shown similar diagnostic accuracy in confirming lymph node status among women with primary endometrial cancer than CT scanning, although the comparisons made are indirect and hence subject to bias. MRI should be used in preference, in light of the ASTEC trial, because of its non invasive nature.</p

    Effectiveness of cricoid pressure in preventing gastric aspiration during rapid sequence intubation in the emergency department: study protocol for a randomised controlled trial

    <p>Abstract</p> <p>Background</p> <p>Cricoid pressure is considered to be the gold standard means of preventing aspiration of gastric content during Rapid Sequence Intubation (RSI). Its effectiveness has only been demonstrated in cadaveric studies and case reports. No randomised controlled trials comparing the incidence of gastric aspiration following emergent RSI, with or without cricoid pressure, have been performed. If improperly applied, cricoid pressure increases risk to the patient. The clinical significance of aspiration in the emergency department is unknown. This randomised controlled trial aims to; 1. Compare the application of the 'ideal" amount of force (30 - 40 newtons) to standard, unmeasured cricoid pressure and 2. Determine the incidence of clinically defined aspiration syndromes following RSI using a fibrinogen degradation assay previously described.</p> <p>Methods/design</p> <p>212 patients requiring emergency intubation will be randomly allocated to either control (unmeasured cricoid pressure) or intervention groups (30 - 40 newtons cricoid pressure). The primary outcome is the rate of aspiration of gastric contents (determined by pepsin detection in the oropharyngeal/tracheal aspirates or treatment for aspiration pneumonitis up to 28 days post-intubation). Secondary outcomes are; correlation between aspiration and lowest pre-intubation Glasgow Coma Score, the relationship between detection of pepsin in trachea and development of aspiration syndromes, complications associated with intubation and grade of the view on direct largyngoscopy.</p> <p>Discussion</p> <p>The benefits and risks of cricoid pressure application will be scrutinised by comparison of the incidence of aspiration and difficult or failed intubations in each group.     GTC Follow-up Observations of Very Metal-Poor Star Candidates from DESI

    The observations from the Dark Energy Spectroscopic Instrument (DESI) will significantly increase the numbers of known extremely metal-poor stars by a factor of ~ 10, improving the sample statistics to study the early chemical evolution of the Milky Way and the nature of the first stars. In this paper we report high signal-to-noise follow-up observations of 9 metal-poor stars identified during the DESI commissioning with the Optical System for Imaging and low-Intermediate-Resolution Integrated Spectroscopy (OSIRIS) instrument on the 10.4m Gran Telescopio Canarias (GTC). The analysis of the data using a well-vetted methodology confirms the quality of the DESI spectra and the performance of the pipelines developed for the data reduction and analysis of DESI data.Comment: 13 pages, 4 figures, to be submitted to ApJ, data available from https://doi.org/10.5281/zenodo.802084
