    Non-adiabatic pressure loss boundary condition for modelling turbocharger turbine pulsating flow

    This paper presents a simplified methodology of pulse flow turbine modelling, as an alternative over the meanline integrated methodology outlined in previous work, in order to make its application to engine cycle simulation codes much more straight forward. This is enabled through the development of a bespoke non-adiabatic pressure loss boundary to represent the turbine rotor. In this paper, turbocharger turbine pulse flow performance predictions are presented along with a comparison of computation duration against the previously established integrated meanline method. Plots of prediction deviation indicate that the mass flow rate and actual power predictions from both methods are highly comparable and are reasonably close to experimental data. However, the new boundary condition required significantly lower computational time and rotor geometrical inputs. In addition, the pressure wave propagation in this simplified unsteady turbine model at different pulse frequencies has also been found to be in agreement with data from the literature, thereby supporting the confidence in its ability to simulate the wave action encountered in turbine pulse flow operation

    A Review of Heat Transfer in Turbochargers

    The conventional powertrain has seen a continuous wave of energy optimization, focusing heavily on boosting and engine downsizing. This trend is pushing OEMs to consider turbocharging as a premium solution for exhaust energy recovery. Turbocharger is an established, economically viable solution which recovers waste energy from the exhaust gasses, and in the process providing higher pressure and mass of air to the engine. However, a turbocharger has to be carefully matched to the engine. The process of matching a turbocharger to an engine is implemented in the early stages of design, through air system simulations. In these simulations, a turbocharger component is represented largely by performance maps and it serves as a boundary condition to the engine. The thermodynamic parameters of a turbocharger are calculated through the performance maps which are usually generated experimentally in gas test stands and used as look-up table in the engine models. Thus, the operational of the engine is dictated by the air flow thermodynamic parameters (pressure, temperature and mass flow) from the turbocharger compressor; this in turn will determine the thermodynamic parameters for the exhaust gas entering the turbocharger turbine. The importance and its sensitivity dictate that any heat transfer affecting the experiments to acquire the performance maps will cause errors in the characterization of a turbocharger. This will consequently lead to inaccurate predictions from the engine model if the heat transfer effects are not properly accounted for. The current paper provides a comprehensive review on how the industry and academics are addressing the heat transfer issue through advancing researches. The review begins by defining the main issues related with heat transfer in turbochargers and the state-of-the-art research looking into it. The paper also provides some inputs and recommendations on the research areas which should be further investigated in the years to come

    Radial turbine sound and noise characterisation with acoustic transfer matrices by means of fast one-dimensional models

A definitive version was subsequently published as https://doi.org/10.1177/1468087419889429.[EN] Estimating correctly the turbine acoustics can be valuable during the engine design stage; in fact, it can lead to a more optimised design of the silencer and aftertreatment, as well as to better prediction of the scavenging effects. However, obtaining the sound and noise emissions of radial turbocharger turbines with low computational costs can be challenging. To consider these effects in a time-efficient manner, the acoustic response of single-entry radial turbines can be characterised by means of acoustic transfer matrices that change with the operating conditions. Exploiting the different time-scales of the acoustic phenomena and the change in the operating point of the turbine, lookup tables of acoustic transfer matrices can be computed. Then, the obtained characterisation can be used in mean-value engine models. This article presents a method for generating these lookup tables by means of fast one-dimensional simulations of thoroughly validated fidelity, in terms of both acoustics and extrapolation capabilities.     Experimental validation of a one-dimensional twin-entry radial turbine model under non-linear pulse conditions

A definitive version was subsequently published as https://doi.org/10.1177/1468087419869157[EN] This article presents the experimental validation of a complete integrated one-dimensional twin-scroll turbine model able to be used in reciprocating internal combustion engine unsteady simulations. A passenger car with a twin-entry-type turbine has been tested under engine-like pulse conditions by means of a specifically built gas stand. To obtain high-resolution quality data, the turbine and turbine line pipes have been instrumented with mean and instantaneous pressure sensors as well as temperature and mass flow sensors, employing a uniquely designed rotating valve for the pulse generation. This experimental configuration enables to obtain the pressure decomposition in both inlets and outlets of the turbine. Using the experimental data obtained, the model is fully validated, with special focus on the reflected and transmitted components for analysing the performance of the model and its non-linear acoustics prediction capabilities.     Characterization of long-term effector-memory T-cell responses in patients with resected high-risk melanoma receiving a melanoma Peptide vaccine

    Abstract: The authors determined whether long-term memory T cells could be detected in patients who received a multipeptide vaccine for high-risk resected melanoma. Five HLA-A*0201 patients received a vaccine that included the gp100 209-217 (210M) peptide with Montanide ISA 51. Peripheral blood mononuclear cells were obtained before therapy, after 6 months of vaccinations, and from 18 months to 36 months later. The presence of gp100 antigen-specific cytolytic T cells was measured by ELISPOT, tetramer and chromium release assays. Tetramer-positive CD8 + cells were phenotyped by flow cytometry for markers including CD44, CD45RA, and CCR7. T-cell avidity and its evolution over time were examined in selected patients. Epitope spreading was analyzed by assessment of gp100 [280][281][282][283][284][285][286][287][288] (288V) T cells. All patients exhibited a significant increase in tetramer-positive gp100-specific CD8 + T cells that decayed at different rates over 18 to 36 months after vaccinations. Cells from all patients exhibited an effector-memory phenotype and were generally CD45 RA low/CCR7 negative and CD44 positive. Tetramer-positive cells declined over time in four of the five patients, but the proportion of tetramer-positive CD8 + cells that secreted gamma-interferon rose, suggesting enrichment for effector cells. Epitope spreading for the gp100 280-288 (288V) epitope was detected. One patient maintained a population of 2.5% circulating gp100 tetramer-positive cells over 36 months. Avidity analysis showed no changes over time after induction of antigen-specific T cells. Vaccination with a heteroclitic melanoma antigen peptide with Montanide ISA 51 generated populations of circulating functional effector-memory T cells that were specific for gp100 and long-lived in the circulation for periods of 18 to 36 months after vaccination

    Laparoscopy in management of appendicitis in high-, middle-, and low-income countries: a multicenter, prospective, cohort study.

    BACKGROUND: Appendicitis is the most common abdominal surgical emergency worldwide. Differences between high- and low-income settings in the availability of laparoscopic appendectomy, alternative management choices, and outcomes are poorly described. The aim was to identify variation in surgical management and outcomes of appendicitis within low-, middle-, and high-Human Development Index (HDI) countries worldwide. METHODS: This is a multicenter, international prospective cohort study. Consecutive sampling of patients undergoing emergency appendectomy over 6 months was conducted. Follow-up lasted 30 days. RESULTS: 4546 patients from 52 countries underwent appendectomy (2499 high-, 1540 middle-, and 507 low-HDI groups). Surgical site infection (SSI) rates were higher in low-HDI (OR 2.57, 95% CI 1.33-4.99, p = 0.005) but not middle-HDI countries (OR 1.38, 95% CI 0.76-2.52, p = 0.291), compared with high-HDI countries after adjustment. A laparoscopic approach was common in high-HDI countries (1693/2499, 67.7%), but infrequent in low-HDI (41/507, 8.1%) and middle-HDI (132/1540, 8.6%) groups. After accounting for case-mix, laparoscopy was still associated with fewer overall complications (OR 0.55, 95% CI 0.42-0.71, p < 0.001) and SSIs (OR 0.22, 95% CI 0.14-0.33, p < 0.001). In propensity-score matched groups within low-/middle-HDI countries, laparoscopy was still associated with fewer overall complications (OR 0.23 95% CI 0.11-0.44) and SSI (OR 0.21 95% CI 0.09-0.45). CONCLUSION: A laparoscopic approach is associated with better outcomes and availability appears to differ by country HDI. Despite the profound clinical, operational, and financial barriers to its widespread introduction, laparoscopy could significantly improve outcomes for patients in low-resource environments.     Pooled analysis of WHO Surgical Safety Checklist use and mortality after emergency laparotomy

    Background The World Health Organization (WHO) Surgical Safety Checklist has fostered safe practice for 10 years, yet its place in emergency surgery has not been assessed on a global scale. The aim of this study was to evaluate reported checklist use in emergency settings and examine the relationship with perioperative mortality in patients who had emergency laparotomy. Methods In two multinational cohort studies, adults undergoing emergency laparotomy were compared with those having elective gastrointestinal surgery. Relationships between reported checklist use and mortality were determined using multivariable logistic regression and bootstrapped simulation. Results Of 12 296 patients included from 76 countries, 4843 underwent emergency laparotomy. After adjusting for patient and disease factors, checklist use before emergency laparotomy was more common in countries with a high Human Development Index (HDI) (2455 of 2741, 89.6 per cent) compared with that in countries with a middle (753 of 1242, 60.6 per cent; odds ratio (OR) 0.17, 95 per cent c.i. 0.14 to 0.21, P <0001) or low (363 of 860, 422 per cent; OR 008, 007 to 010, P <0.001) HDI. Checklist use was less common in elective surgery than for emergency laparotomy in high-HDI countries (risk difference -94 (95 per cent c.i. -11.9 to -6.9) per cent; P <0001), but the relationship was reversed in low-HDI countries (+121 (+7.0 to +173) per cent; P <0001). In multivariable models, checklist use was associated with a lower 30-day perioperative mortality (OR 0.60, 0.50 to 073; P <0.001). The greatest absolute benefit was seen for emergency surgery in low- and middle-HDI countries. Conclusion Checklist use in emergency laparotomy was associated with a significantly lower perioperative mortality rate.     Guidelines for the use and interpretation of assays for monitoring autophagy (4th edition)1.

    In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide.
