    Fortified architecture between preservation and reuse strategies: the twentieth century restoration projects of Castelfranco in Finale Ligure

    [EN] The town of Finale Ligure, situated on the western coast of Liguria, was the site of the Del Carretto Marquisate until the sixteenth century. After that, it was under the control of the Spanish Crown (seventeenth century) and it has been an independent territory of the Republic of Genoa for a long time. The three castles were built on the top of Finale hills and they were the symbol of its independence. Gavone castle, established on the top of the historical town, has been the site of the Marquisate since the twelfth century. S. Giovanni castle was built by the Spanish in order to improve the town defensive system in the second half of the seventeenth century. Castelfranco, built by the Genoese in the fourteenth century, was rehashed many times by the Spanish and in the nineteenth century by the Savoia family. The three castles still recall these historical events and are therefore witnesses of the Finale present and past history. They are the result of the different transformations occurred over the centuries. In recent times, Castelfranco has been opened to the public and today it houses art exhibitions and cultural events. The restoration of the castle is the last step of a long-lasting rehabilitation project history that has been developed since the 1900s, when the Municipality suggested to turn it into a hotel. The article analyses the restoration projects of Castelfranco that have been carried out in the first half of the twentieth century, which had different methodologies and approaches. Though this study the article highlights the perception that the town had about the castle, identifying the changes in the balance between reuse and conservation strategies after the first Italian preservation laws.Brusa, E.; Stanga, C.     Nitric oxide and P-glycoprotein modulate the phagocytosis of colon cancer cells

    The anticancer drug doxorubicin induces the synthesis of nitric oxide, a small molecule that enhances the drug cytotoxicity and reduces the drug efflux through the membrane pump P-glycoprotein (Pgp). Doxorubicin also induces the translocation on the plasma membrane of the protein calreticulin (CRT), which allows tumour cells to be phagocytized by dendritic cells. We have shown that doxorubicin elicits nitric oxide synthesis and CRT exposure only in drug-sensitive cells, not in drug-resistant ones, which are indeed chemo-immunoresistant. In this work, we investigate the mechanisms by which nitric oxide induces the translocation of CRT and the molecular basis of this chemo-immunoresistance. In the drug-sensitive colon cancer HT29 cells doxorubicin increased nitric oxide synthesis, CRT exposure and cells phagocytosis. Nitric oxide promoted the translocation of CRT in a guanosine monophosphate (cGMP) and actin cytoskeleton-dependent way. CRT translocation did not occur in drug-resistant HT29-dx cells, where the doxorubicin-induced nitric oxide synthesis was absent. By increasing nitric oxide with stimuli other than doxorubicin, the CRT exposure was obtained also in HT29-dx cells. Although in sensitive cells the CRT translocation was followed by the phagocytosis, in drug-resistant cells the phagocytosis did not occur despite the CRT exposure. In HT29-dx cells CRT was bound to Pgp and only by silencing the latter the CRT-operated phagocytosis was restored, suggesting that Pgp impairs the functional activity of CRT and the tumour cells phagocytosis. Our work suggests that the levels of nitric oxide and Pgp critically modulate the recognition of the tumour cells by dendritic cells, and proposes a new potential therapeutic approach against chemo-immunoresistant tumours

    Discovery of the first heavily obscured QSO candidate at z>6z>6 in a close galaxy pair

    While theoretical arguments predict that most of the early growth of supermassive black holes (SMBHs) happened during heavily obscured phases of accretion, current methods used for selecting z>6z>6 quasars (QSOs) are strongly biased against obscured QSOs, thus considerably limiting our understanding of accreting SMBHs during the first Gyr of the Universe from an observational point of view. We report the ChandraChandra discovery of the first heavily obscured QSO candidate in the early universe, hosted by a close (≈5\approx5 kpc) galaxy pair at z=6.515z=6.515. One of the members is an optically classified type 1 QSO, PSO167-13. The companion galaxy was first detected as a [C II] emitter by ALMA. An X-ray source is significantly (P=0.9996P=0.9996) detected by ChandraChandra in the 2-5 keV band, with <1.14<1.14 net counts in the 0.5-2 keV band, although the current positional uncertainty does not allow a conclusive association with either PSO167-13 or its companion galaxy. From X-ray photometry and hardness-ratio arguments, we estimated an obscuring column density of NH>2×1024 cm−2N_H>2\times10^{24}\,\mathrm{cm^{-2}} and NH>6×1023 cm−2N_H>6\times10^{23}\,\mathrm{cm^{-2}} at 68%68\% and 90%90\% confidence levels, respectively. Thus, regardless of which of the two galaxies is associated with the X-ray emission, this source is the first heavily obscured QSO candidate at z>6z>6.Comment: A&A Letters, accepte

    Chandra and Magellan/FIRE follow-up observations of PSO167-13: an X-ray weak QSO at z=6.515z=6.515

    The discovery of hundreds of QSOs in the first Gyr of the Universe powered by already grown SMBHs challenges our knowledge of SMBH formation. In particular, investigations of z>6z>6 QSOs presenting notable properties can provide unique information on the physics of fast SMBH growth in the early universe. We present the results of follow-up observations of the z=6.515z=6.515 radio-quiet QSO PSO167-13, which is interacting with a close companion galaxy. The PSO167-13 system has been recently proposed to host the first heavily obscured X-ray source at high redshift. We observed PSO167-13 with Chandra/ACIS-S (177 ks), and obtained new spectroscopic observations (7.2 h) with Magellan/FIRE. No significant X-ray emission is detected from the PSO167-13 system, suggesting that the obscured X-ray source previously tentatively detected was either due to a strong background fluctuation or is highly variable. The upper limit (90% confidence level) on the X-ray emission of PSO167-13 (L2−10 keV<8.3×1043 ergs−1L_{2-10\,\mathrm{keV}}<8.3\times10^{43}\,\mathrm{erg s^{-1}}) is the lowest available for a z>6z>6 QSO. The ratio between the X-ray and UV luminosity of αox<−1.95\alpha_{ox}<-1.95 makes PSO167-13 a strong outlier from the αox−LUV\alpha_{ox}-L_{UV} and LX−LbolL_X-L_{\mathrm{bol}} relations. In particular, its X-ray emission is >6>6 times weaker than the expectation based on its UV luminosity. The new Magellan/FIRE spectrum of PSO167-13 is strongly affected by the unfavorable sky conditions, but the tentatively detected C IV and Mg II emission lines appear strongly blueshifted. The most plausible explanations for the X-ray weakness of PSO167-13 are intrinsic weakness or small-scale absorption by Compton-thick material. The possible strong blueshift of its emission lines hints at the presence of nuclear winds, which could be related to its X-ray weakness.Comment: Accepted for publication on A&

    Gastrointestinal presentation of kawasaki disease: A red flag for severe disease?

    Background Kawasaki disease (KD) is a febrile systemic vasculitis of unknown etiology and the main cause of acquired heart disease among children in the developed world. To date, abdominal involvement at presentation is not recognized as a risk factor for a more severe form of the disease. Objective To evaluate whether presenting abdominal manifestations identify a group at major risk for Intravenous immunoglobulin (IVIG)-resistance and coronary lesions. Methods Retrospective study of KD patients diagnosed between 2000 and 2015 in 13 pediatric units in Italy. Patients were divided into 2 groups according to the presence or absence of abdominal manifestations at onset. We compared their demographic and clinical data, IVIG-responsiveness, coronary ectasia/aneurysms, laboratory findings from the acute and subacute phases. Results 302 patients (181 boys) were enrolled: 106 patients with, and 196 patients without presenting abdominal features. Seasonality was different between the groups (p = 0.034). Patients with abdominal manifestations were younger (p = 0.006) and more frequently underwent delayed treatment (p = 0.014). In the acute phase, patients with abdominal presentation had higher platelet counts (PLT) (p = 0.042) and lower albuminemia (p = 0.009), while, in the subacute phase, they had higher white blood cell counts (WBC) and PLT (p = 0.002 and p < 0.005, respectively) and lower red blood cell counts (RBC) and hemoglobin (Hb) (p = 0.031 and p 0.009). Moreover, the above mentioned group was more likely to be IVIG-resistant (p < 0.005) and have coronary aneurysms (p = 0.007). In the multivariate analysis, presenting abdominal manifestations, age younger than 6 months, IVIG- resistance, delayed treatment and albumin concentration in the acute phase were independent risk factors for coronary aneurysms (respectively p<0.005, <0.005, = 0.005 and 0.009). Conclusions This is the first multicenter report demonstrating that presenting gastrointestinal features in KD identify patients at higher risk for IVIG-resistance and for the development of coronary aneurysms in a predominantly Caucasian population

    Clinical correlates of “pure” essential tremor: the TITAN study

    BackgroundTo date, there are no large studies delineating the clinical correlates of “pure” essential tremor (ET) according to its new definition.MethodsFrom the ITAlian tremor Network (TITAN) database, we extracted data from patients with a diagnosis of “pure” ET and excluded those with other tremor classifications, including ET-plus, focal, and task-specific tremor, which were formerly considered parts of the ET spectrum.ResultsOut of 653 subjects recruited in the TITAN study by January 2022, the data of 208 (31.8%) “pure” ET patients (86M/122F) were analyzed. The distribution of age at onset was found to be bimodal. The proportion of familial cases by the age-at-onset class of 20 years showed significant differences, with sporadic cases representing the large majority of the class with an age at onset above 60 years. Patients with a positive family history of tremor had a younger onset and were more likely to have leg involvement than sporadic patients despite a similar disease duration. Early-onset and late-onset cases were different in terms of tremor distribution at onset and tremor severity, likely as a function of longer disease duration, yet without differences in terms of quality of life, which suggests a relatively benign progression. Treatment patterns and outcomes revealed that up to 40% of the sample was unsatisfied with the current pharmacological options.DiscussionThe findings reported in the study provide new insights, especially with regard to a possible inversed sex distribution, and to the genetic backgrounds of “pure” ET, given that familial cases were evenly distributed across age-at-onset classes of 20 years. Deep clinical profiling of “pure” ET, for instance, according to age at onset, might increase the clinical value of this syndrome in identifying pathogenetic hypotheses and therapeutic strategies

    Plant–environment interactions through a functional traits perspective: a review of Italian studies

    Italy is among the European countries with the greatest plant diversity due to both a great environmental heterogeneity and a long history of man–environment interactions. Trait-based approaches to ecological studies have developed greatly over recent decades worldwide, although several issues concerning the relationships between plant functional traits and the environment still lack sufficient empirical evaluation. To draw insights on the association between plant functional traits and direct and indirect human and natural pressures on the environmental drivers, this article summarizes the existing knowledge on this topic by reviewing the results of studies performed in Italy adopting a functional trait approach on vascular plants, bryophytes and lichens. Although we recorded trait measurements for 1418 taxa, our review highlighted some major gaps in plant traits knowledge: Mediterranean ecosystems are poorly represented; traits related to belowground organs are still overlooked; traits measurements for bryophytes and lichens are lacking. Finally, intraspecific variation has been little studied at community level so far. We conclude by highlighting the need for approaches evaluating trait–environment relationship at large spatial and temporal scales and the need of a more effective contribution to online databases to tie more firmly Italian researchers to international scientific networks on plant traits
