    Dual Effect of Secondary Solutes on Binding Equilibria: Contributions from Solute–Reactant Interactions and Solute–Water Interactions

    This study examines the role of water in binding equilibria with a special focus on secondary solutes (cosolutes) that influence the equilibrium but are not constituents of the final product. Using a thermodynamic framework that includes an explicit term for the release of water molecules upon binding, this investigation reveals how solutes may alter equilibria by changing the activity of the reactants, reflected in ΔG°(obs), and by changing the chemical potential of the solvent, reflected in ΔGS. The framework is applied to four experimental binding systems that differ in the degree of electrostatic contributions. The model systems include the chelation of Ca2+ by EDTA and three host–guest reactions; the pairings of p-sulfonatocalix[4]arene with tetramethylammonium ion, cucurbit[7]uril with N-acetyl-phenylalanine-amide, and β-cyclodextrin with adamantane carboxylate are tested. Each reaction pair is examined by isothermal titration calorimetry at 25 °C in the presence of a common osmolyte, sucrose, and a common chaotrope, urea. Molar solutions of trehalose and phosphate were also tested with selected models. In general, cosolutes that enhance binding tend to reduce the solvation free energy penalty and cosolutes that weaken binding tend to increase the solvation free energy penalty. Notably, the nonpolar–nonpolar interaction between adamantane carboxylate and β-cyclodextrin is characterized by a ΔGS value near zero. The results with β-cyclodextrin, in particular, prompt further discussions of the hydrophobic effect and the biocompatible properties of trehalose. Other investigators are encouraged to test and refine the approach taken here to further our understanding of solvent effects on molecular recognition

    The dairy sector in the Azores Islands: possibilities and main constraints towards increased added value

    Technical NoteThe Azores archipelago is the most suitable region for dairy production in Portugal, representing 30% of the overall Portuguese dairy production. It has a production system characterized by an average milk yield of 6216 kg/cow/year, and the predominance of pasture-based feeding and cows that have longer productive lives and lower incidence of metabolic/production diseases, such as acidosis or mastitis. The biggest problem with the Azores Islands dairy sector is the cost of transport, as the main markets are located in continental Portugal, over 1500 km away, and local dairy products have to compete with dairy products produced in mainland Portugal and in the rest of the European Union. Herein, the evolution of the dairy sector in the Azores Islands from 2007 to 2017 is presented. A SWOT (Strength, Weakness, Opportunity, and Threat) analysis was performed to find potential solutions to increase the value of the Azorean dairy sector. The most relevant solution considered was the valorization of the dairy production through three major aspects: higher milk quality, namely, better organoleptic properties; lower carbon footprint (not considering transport costs); and higher levels of animal welfare. Three examples are shown of such valorization: protected denomination of origin (PDO) cheeses, the “happy cows” program, and the production of an organic milk, from the Terceira Island.     Mean-field limit of systems with multiplicative noise

    A detailed study of the mean-field solution of Langevin equations with multiplicative noise is presented. Three different regimes depending on noise-intensity (weak, intermediate, and strong-noise) are identified by performing a self-consistent calculation on a fully connected lattice. The most interesting, strong-noise, regime is shown to be intrinsically unstable with respect to the inclusion of fluctuations, as a Ginzburg criterion shows. On the other hand, the self-consistent approach is shown to be valid only in the thermodynamic limit, while for finite systems the critical behavior is found to be different. In this last case, the self-consistent field itself is broadly distributed rather than taking a well defined mean value; its fluctuations, described by an effective zero-dimensional multiplicative noise equation, govern the critical properties. These findings are obtained analytically for a fully connected graph, and verified numerically both on fully connected graphs and on random regular networks. The results presented here shed some doubt on what is the validity and meaning of a standard mean-field approach in systems with multiplicative noise in finite dimensions, where each site does not see an infinite number of neighbors, but a finite one. The implications of all this on the existence of a finite upper critical dimension for multiplicative noise and Kardar-Parisi-Zhang problems are briefly discussed.Comment: 9 Pages, 8 Figure

    Tactical Voting in Plurality Elections

    How often will elections end in landslides? What is the probability for a head-to-head race? Analyzing ballot results from several large countries rather anomalous and yet unexplained distributions have been observed. We identify tactical voting as the driving ingredient for the anomalies and introduce a model to study its effect on plurality elections, characterized by the relative strength of the feedback from polls and the pairwise interaction between individuals in the society. With this model it becomes possible to explain the polarization of votes between two candidates, understand the small margin of victories frequently observed for different elections, and analyze the polls' impact in American, Canadian, and Brazilian ballots. Moreover, the model reproduces, quantitatively, the distribution of votes obtained in the Brazilian mayor elections with two, three, and four candidates.Comment: 7 pages, 4 figure

    Limits on the LyC signal from z~3 sources with secure redshift and HST coverage in the E-CDFS field

    Aim: We aim to measure the LyC signal from a sample of sources in the Chandra deep field south. We collect star-forming galaxies (SFGs) and active galactic nuclei (AGN) with accurate spectroscopic redshifts, for which Hubble Space Telescope (HST) coverage and multi-wavelength photometry are available. Method: We selected a sample of about 200 sources at z~3. Taking advantage of HST resolution, we applied a careful cleaning procedure and rejected sources showing nearby clumps with different colours, which could be lower-z interlopers. Our clean sample consisted of 86 SFGs (including 19 narrow-band selected Lya emitters) and 8 AGN (including 6 detected in X-rays). We measured the LyC flux from aperture photometry in four narrow-band filters covering wavelengths below a 912 A rest frame (3.11<z<3.53). We estimated the ratio between ionizing (LyC flux) and 1400 A non-ionizing emissions for AGN and galaxies. Results: By running population synthesis models, we assume an average intrinsic L(1400 A)/L(900 A) ratio of 5 as the representative value for our sample. With this value and an average treatment of the lines of sight of the inter-galactic medium, we estimate the LyC escape fraction relative to the intrinsic value (fesc_rel(LyC)). We do not directly detect ionizing radiation from any individual SFG, but we are able to set a 1(2)sigma upper limit of fesc_rel(LyC)<12(24)%. This result is consistent with other non-detections published in the literature. No meaningful limits can be calculated for the sub-sample of Lya emitters. We obtain one significant direct detection for an AGN at z=3.46, with fesc_rel(LyC) = (72+/-18)%. Conclusions: Our upper limit on fescrel(LyC) implies that the SFGs studied here do not present either the physical properties or the geometric conditions suitable for efficient LyC-photon escape.Comment: Accepted for publication in A&A on Jan 5th, 201

    Residence Time (RT), a new parameter to predict neurosteroidogenic efficacy of Translocator Protein (TSPO) ligands: N,N-dialkyl-2-arylindol-3-ylglyoxylamides, a case study

    Targeting neuroactive steroid biosynthetic pathway by specific 18 kDa Translocator Protein (TSPO) ligands may represent a therapeutic approach in a variety of neurodegenerative and neuropsychiatric diseases. However, the lack of correlation between the binding affinity and the in vitro steroidogenic efficacy has limited the identification of lead compounds by a traditional affinity-based drug discovery strategy. Our recent researches indicate that the key factor for robust steroidogenic TSPO ligand efficacy is not the binding affinity per se, but rather the time the compound spends into the target, namely its Residence Time (RT). The assessment of this kinetic parameter during the in vitro characterization of compounds appears mandatory in order to obtain structure-efficacy relationships suitable for the future development of novel molecules with promising pharmacological properties

    Discovery of a rich proto-cluster at z=2.9 and associated diffuse cold gas in the VIMOS Ultra-Deep Survey (VUDS)

    [Abridged] We characterise a massive proto-cluster at z=2.895 that we found in the COSMOS field using the spectroscopic sample of the VIMOS Ultra-Deep Survey (VUDS). This is one of the rare structures at z~3 not identified around AGNs or radio galaxies, so it is an ideal laboratory to study galaxy formation in dense environments. The structure comprises 12 galaxies with secure spectroscopic redshift in an area of 7'x8', in a z bin of Dz=0.016. The measured galaxy number overdensity is delta_g=12+/-2. This overdensity has total mass of M~8.1x10^(14)M_sun in a volume of 13x15x17 Mpc^3. Simulations indicate that such an overdensity at z~2.9 is a proto-cluster that will collapse in a cluster of total mass M~2.5x10^(15)M_sun at z=0. We compare the properties of the galaxies within the overdensity with a control sample at the same z but outside the overdensity. We did not find any statistically significant difference between the properties (stellar mass, SFR, sSFR, NUV-r, r-K) of the galaxies inside and outside the overdensity. The stacked spectrum of galaxies in the overdensity background shows a significant absorption feature at the wavelength of Lya redshifted at z=2.895 (lambda=4736 A), with a rest frame EW = 4+/- 1.4 A. Stacking only background galaxies without intervening sources at z~2.9 along their line of sight, we find that this absorption feature has a rest frame EW of 10.8+/-3.7 A, with a detection S/N of ~4. These EW values imply a high column density (N(HI)~3-20x10^(19)cm^(-2)), consistent with a scenario where such absorption is due to intervening cold gas streams, falling into the halo potential wells of the proto-cluster galaxies. However, we cannot exclude the hypothesis that this absorption is due to the diffuse gas within the overdensity.Comment: 15 pages, 9 figures, accepted for publication in A&A (revised version after referee's comments and language editing
