    Sound and noise in high school gymnasiums

    The goal of this research was to characterize the interior acoustics of high schools sports facilities using objective parameters. In situ measurements were done in 68 school gymnasiums in Portugal (Volume from 450 to 16,190 m3 with a median of 4,720 m3) regarding LAeqBN (background noise without gym classes), LAeqPE (ongoing Physical Education classes), RT, and RASTI. The results for LAeqBN were from 30 to 59 dB with a median of 42 dB. For the LAeqPE were found values from 68 to 90 dB with a median of 80 dB. For the RT(500/1k/2k), room values from 2.5 to 8.1 s with a median of 4.8 s, were measured. The room average RASTI values were from 0.26 to 0.54 with a median of 0.34. These sports rooms proved to be highly reverberant, almost without sound absorbing materials, which might be harmful, especially for the PE teachers. The subjective perception of the PE teachers was analyzed through questionnaires where it was verified that they feel most discomfort when it comes for noise conditions. This was supported by the objective results obtained. A multicriteria method to assess the overall acoustic quality of school gymnasiums is presented. Ideal values for those acoustic parameters are presented

    New insights on the effects of formulation type and compositional mixtures over the antioxidant and cytotoxic activities of dietary supplements based-on hepatoprotective plants

    Artichoke (A), borututu (B) and milk thistle (M) are included in several supplements to provide beneficial effects. Different formulations (infusions, pills and syrups), as also different proportions of A, B and M (1:1:1, 2:1:1, 1:2:1, 1:1:2) within each formulation were assayed to optimize the desired benefits. The antioxidant activity, anti-hepatocellular carcinoma activity, hepatotoxicity and bioactive compounds contents were evaluated. Syrups tended to be the formulation with highest antioxidant activity and total phenolics and flavonoids content; otherwise, pills were the worst formulation. In what concerns A:B:M ratios, the results did not reveal so pronounced differences. None of the assayed mixtures resulted to be toxic (up to the maximum assayed dose) for liver primary cells (PLP2), but some samples, especially infusions, showed toxicity for the hepatocellular carcinoma cell line (HepG2).     Galectin-3 negatively regulates the frequency and function of CD4+CD25+Foxp3+ regulatory T cells and influences the course of Leishmania major infection

    Galectin-3, an endogenous glycan-binding protein, plays essential roles during microbial infection by modulating innate and adaptive immunity. However, the role of galectin-3 within the CD4+CD25+Foxp3+ T regulatory (TREG) cell compartment has not yet been explored. Here, we found, in a model of Leishmania major infection, that galectin-3 deficiency increases the frequency of peripheral TREG cells both in draining lymph nodes (LNs) and sites of infection. These observations correlated with an increased severity of the disease, as shown by increased footpad swelling and parasite burden. Galectin-3-deficient (Lgals3−/−) TREG cells displayed higher CD103 expression, showed greater suppressive capacity, and synthesized higher amounts of IL-10 compared with their wild-type (WT) counterpart. Furthermore, both TREG cells and T effector (TEFF) cells from Lgals3−/− mice showed higher expression of Notch1 and the Notch target gene Hes-1. Interestingly, Notch signaling components were also altered in both TREG and TEFF cells from uninfected Lgals3−/− mice. Thus, endogenous galectin-3 regulates the frequency and function of CD4+CD25+Foxp3+ TREG cells and alters the course of L. major infection.Fil: Fermino, Marise L..     Spray-dried Spirulina platensis as an effective ingredient to improve yogurt formulations: testing different encapsulating solutions

    The consumption of foods functionalized with spirulina might have positive health effects. However, spirulinabased food products are usually associated with unpleasant flavor and odor, and can present non-homogeneous appearance, impairing consumers’ acceptance. Moreover, it is important to assure bioactivity maintenance. To develop a novel food ingredient, spirulina was chemically characterized, and spray-dried using two encapsulating materials: i) maltodextrin and ii) maltodextrin crosslinked with citric acid. Thereafter, free and encapsulated spirulina were evaluated for their bioactive properties. Microencapsulated spirulina presented higher thermal stability than the base materials, while showing better anti-inflammatory activity without exerting cytotoxicity. Free and encapsulated spirulina were further added to yogurts to validate their suitability as functionalizing agents. Yogurts added with encapsulated spirulina presented a more homogeneous appearance, and the best solution was spirulina encapsulated in maltodextrin crosslinked with citric acid, considering the nutritional profile, attractive color, and improved antioxidant activity throughout storage time.The authors are grateful to the Foundation for Science and Technology (FCT, Portugal) and FEDER under Program PT2020 for financial support to CIMO (UID/AGR/00690/2019); L.     The lectin-specific activity of Toxoplasma gondii microneme proteins 1 and 4 binds Toll-like receptor 2 and 4 N-glycans to regulate innate immune priming.

    Infection of host cells by Toxoplasma gondii is an active process, which is regulated by secretion of microneme (MICs) and rhoptry proteins (ROPs and RONs) from specialized organelles in the apical pole of the parasite. MIC1, MIC4 and MIC6 assemble into an adhesin complex secreted on the parasite surface that functions to promote infection competency. MIC1 and MIC4 are known to bind terminal sialic acid residues and galactose residues, respectively and to induce IL-12 production from splenocytes. Here we show that rMIC1- and rMIC4-stimulated dendritic cells and macrophages produce proinflammatory cytokines, and they do so by engaging TLR2 and TLR4. This process depends on sugar recognition, since point mutations in the carbohydrate-recognition domains (CRD) of rMIC1 and rMIC4 inhibit innate immune cells activation. HEK cells transfected with TLR2 glycomutants were selectively unresponsive to MICs. Following in vitro infection, parasites lacking MIC1 or MIC4, as well as expressing MIC proteins with point mutations in their CRD, failed to induce wild-type (WT) levels of IL-12 secretion by innate immune cells. However, only MIC1 was shown to impact systemic levels of IL-12 and IFN-γ in vivo. Together, our data show that MIC1 and MIC4 interact physically with TLR2 and TLR4 N-glycans to trigger IL-12 responses, and MIC1 is playing a significant role in vivo by altering T. gondii infection competency and murine pathogenesis

    Imago & mirabilia : les formes del prodigi a la Mediterrània medieval

    Hem organitzat el present volum a partir de tres àmbits que, sense cap pretensió totalitzadora ni teòrica, mostrin la vivacitat del prodigi medieval i la riquesa de la seva exploració. El primer àmbit és el titulat Geografies. El sagrat i el profà tracen una topografia del meravellós que omple la geografia d'indrets singulars, escenaris de tota mena de prodigis, alguns de remots o d'altres encara avinents. La tradicional història sagrada i l'heterogeni santoral, la literatura i els tractats pseudo-científics de l'antiguitat, les cròniques de viatgers i pelegrins o la mateixa cartografia, tot contribueix a fixar els indrets de la meravella com a lieux de mémoire. En efecte, el meravellós també respon a una topografia, a uns espais concrets on es troba, es reconeix i es difon; una memòria precisa on tothora són recordades les mirabilia. El segon àmbit del volum és Objectes. A l'edat mitjana, relíquies, objectes valuosos, rars o exòtics van ser buscats amb afany i els van creure portadors de virtuts prodigioses o reminiscències de criatures tan llunyanes com fabuloses. Presents a les corts, als tresors d'església o a les llars benestants, van esdevenir formes tangibles del sobrenatural. Alguns de procedents de l'Orient cristià van assolir una remarcable dimensió llegendària. I molts d'ells van desvetllar una arkhaiología, una «història de les coses antigues», amb la qual es va anar acompanyant l'objecte prodigiós d'una llegenda que reverberava la seva condició meravellosa i remota. Per últim, el tercer àmbit és Relats. Les narracions escrites i també els relats visuals van ser vehicles fonamentals per a la formació i la difusió del prodigi a l'edat mitjana. Les diverses formes de la meravella troben en la literatura profana o en l'hagiogràfica uns potents mitjans d'expressió i de la seva concreció. Cròniques, novel·les i relats hagiogràfics són portaveus de successos extraordinaris, i la seva enunciació o visualització és causa d'estupor. Miracle, llegenda i aventura esdevenen presències constants en la literatura i en les arts figuratives medievals

    A search for ultra-high-energy photons at the Pierre Auger Observatory exploiting air-shower universality

    The Pierre Auger Observatory is the most sensitive detector to primary photons with energies above ∼0.2 EeV. It measures extensive air showers using a hybrid technique that combines a fluorescence detector (FD) with a ground array of particle detectors (SD). The signatures of a photon-induced air shower are a larger atmospheric depth at the shower maximum (Xmax_{max}) and a steeper lateral distribution function, along with a lower number of muons with respect to the bulk of hadron-induced background. Using observables measured by the FD and SD, three photon searches in different energy bands are performed. In particular, between threshold energies of 1-10 EeV, a new analysis technique has been developed by combining the FD-based measurement of Xmax_{max} with the SD signal through a parameter related to its muon content, derived from the universality of the air showers. This technique has led to a better photon/hadron separation and, consequently, to a higher search sensitivity, resulting in a tighter upper limit than before. The outcome of this new analysis is presented here, along with previous results in the energy ranges below 1 EeV and above 10 EeV. From the data collected by the Pierre Auger Observatory in about 15 years of operation, the most stringent constraints on the fraction of photons in the cosmic flux are set over almost three decades in energy

    Study on multi-ELVES in the Pierre Auger Observatory

    Since 2013, the four sites of the Fluorescence Detector (FD) of the Pierre Auger Observatory record ELVES with a dedicated trigger. These UV light emissions are correlated to distant lightning strikes. The length of recorded traces has been increased from 100 μs (2013), to 300 μs (2014-16), to 900 μs (2017-present), to progressively extend the observation of the light emission towards the vertical of the causative lightning and beyond. A large fraction of the observed events shows double ELVES within the time window, and, in some cases, even more complex structures are observed. The nature of the multi-ELVES is not completely understood but may be related to the different types of lightning in which they are originated. For example, it is known that Narrow Bipolar Events can produce double ELVES, and Energetic In-cloud Pulses, occurring between the main negative and upper positive charge layer of clouds, can induce double and even quadruple ELVES in the ionosphere. This report shows the seasonal and daily dependence of the time gap, amplitude ratio, and correlation between the pulse widths of the peaks in a sample of 1000+ multi-ELVES events recorded during the period 2014-20. The events have been compared with data from other satellite and ground-based sensing devices to study the correlation of their properties with lightning observables such as altitude and polarity
