Biodegradable Prussian blue nanoparticles for photothermal immunotherapy of advanced cancers

Multifunctional nanoparticles represent a class of materials with diverse therapy and imaging properties that can be exploited for the treatment of cancers that have significantly progressed or advanced, which are associated with a poor patient prognosis. Here, we describe the use of biodegradable Prussian blue nanoparticles (PBNPs) in combination with anti-CTLA-4 checkpoint blockade immunotherapy for the treatment of advanced cancers. Our nanoparticle synthesis scheme yields PBNPs that possess pH-dependent intratumoral stability and photothermal therapy (PTT) properties, and degrade under mildly alkaline conditions mimicking the blood and lymph. Studies using PBNPs for PTT in a mouse model of neuroblastoma, a hard-to-treat cancer, demonstrate that PTT causes rapid reduction of tumor burden and growth rates, but results in incomplete responses to therapy and tumor relapse. Studies to elucidate the underlying immunological responses demonstrate that PTT causes increased tumor infiltration of lymphocytes and T cells and a systemic activation of T cells against re-exposed tumor cells in a subset of treated mice. PBNP-based PTT in combination with anti-CTLA-4 immunotherapy results in complete tumor regression and long-term survival in 55.5% of neuroblastoma tumor-bearing mice compared to only 12.5% survival in mice treated with anti-CTLA-4 alone and 0% survival both in mice treated with PTT alone, or remaining untreated. Further, all of the combination therapy-treated mice exhibit protection against tumor rechallenge indicating the development of antitumor immunity as a consequence of therapy. Our studies indicate the immense potential of our combination photothermal immunotherapy in improving the prognosis and outlook for patients with advanced cancers

Plan estratégico para incrementar la recaudación tributaria 2021 en la Municipalidad Distrital de San José – La Libertad

Se tuvo como fin proponer un plan estratégico (PE) para incrementar la recaudación tributaria, se realizó una investigación descriptiva, pues se procedió a narrar la problemática de ambas variables, así mismo no experimental y transversal, puesto que las variables no fueron estimuladas y los instrumentos se usaron en un solo momento en una fecha pactada. La población intervenida fueron el jefe de Administración Tributaria y los contribuyentes que están formalmente inscritos en el padrón los cuales son 12,185. Como técnicas se suministró la encuesta. El resultado más relevante es que el 75,1% está TD que en la comuna se realicen actividades y operaciones para mejorar la recaudación de impuestos, el 6,3% es indiferente y el 18,8% está totalmente de acuerdo. Por lo tanto, no se está cumpliendo la misión. Finalmente concluye que la propuesta debe estar en base a la teoría de Ferrel, Hartline y Lucas, (2002), donde indica que todo plan estratégico debe contener las dimensiones: Misión, Visión. La imagen y la narrativa como herramientas para el abordaje psicosocial en escenarios de violencia de los Municipios de San Martin Cesar, Astrea Cesar, plato, Magdalena.El enfoque narrativo ofrece un clima más relajado, invitando a los participantes a innovar y a ser creativos durante el abordaje y afrontamiento de sus problemas, a través de la exteriorización donde puedan liberarse y deconstruir sobre sus experiencias de violencia en oportunidades de transformación y de esperanza. Sin duda alguna este enfoque posestructuralista permite al psicólogo afianzar en términos de comunicación y cercanía la relación existente con los grupos víctimas de violencia, convirtiendo al proceso de intervención una acción significativa y con garantías de mejoramiento psicosocial. Por medio del presente trabajo se abordarán los análisis de dos casos narrados en el contexto de la violencia con el fin de darles la trazabilidad psicosocial para la atención colectiva y estratégica de afrontamiento postraumático.The narrative approach offers a more relaxed climate, inviting participants to innovate and be creative while addressing and addressing their problems, through externalization where they can free themselves and deconstruct their experiences of violence in opportunities for transformation and hope. Self-reported glaucoma prevalence and related factors, contribution to reported visual impairment, and functional burden in a cross-sectional study in Colombia

Objetivo Describir la prevalencia autorreportada de glaucoma en adultos mayores colombianos, enfatizando los factores de riesgo más importantes y las alteraciones funcionales de la vida diaria asociadas. Métodos Este es un análisis secundario de la encuesta de Salud, Bienestar y Envejecimiento realizada en el año 2015. El diagnóstico de glaucoma se obtuvo del autoinforme. Las variables funcionales se evaluaron mediante cuestionarios de actividades de la vida diaria. Se realizó un análisis descriptivo seguido de modelos de regresión bivariados y multivariados ajustando por variables de confusión. Resultados La prevalencia autoinformada de glaucoma fue de 5,67%, con mayor tasa en mujeres, OR 1,22 (1,13–1,40) p = ,003, mayor edad OR 1,02 (1,01–1,02) p < ,001, y con educación superior OR 1,38 (1,28 –1.50) p < .001. El glaucoma se asoció de forma independiente con diabetes OR 1,37 (1,18–1,61) p  < ,001 e hipertensión 1,26 (1,08–1,46) p = ,003. También mostró correlaciones estadísticamente significativas con mala SSR OR 1.15 (1.02–1.32) p < .001, discapacidad visual autoinformada 1.73 (1.50–2.01) p < .001 y deterioro en el manejo del dinero OR 1.59 (1.16–2.08) p = .002, compras de comestibles OR 1.57 (1.26–1.96) p < .001 y preparación de comidas OR 1.31 (1.06–1.63) p  = .013 y haber tenido caídas en el último año OR 1.14 (1.01–1.31) p = 0.041. Conclusión Nuestros hallazgos sugieren que la prevalencia autoinformada de glaucoma en adultos mayores en Colombia es más alta que los datos informados. El glaucoma y la discapacidad visual en adultos mayores representan un problema de salud pública, ya que el glaucoma se asoció con resultados adversos como pérdida funcional y riesgo de caídas, afectando la calidad de vida y su participación en la sociedad.Q2Q2Purpose Describe the self-reported prevalence of glaucoma in Colombian older adults, emphasizing the most important risk factors and associated daily-life functional alterations. Conclusion Our findings suggest the self-reported prevalence of glaucoma in older adults in Colombia to be higher than reported data. Polyphenic trait promotes liver cancer in a model of epigenetic instability in mice.

Hepatocellular carcinoma (HCC) represents the fifth-most common form of cancer worldwide and carries a high mortality rate attributed to lack of effective treatment. Males are 8 times more likely to develop HCC than females, an effect largely driven by sex hormones, albeit through still poorly understood mechanisms. We previously identified TRIM28 (tripartite protein 28), a scaffold protein capable of recruiting a number of chromatin modifiers, as a crucial mediator of sexual dimorphism in the liver. Trim28(hep-/-) mice display sex-specific transcriptional deregulation of a wide range of bile and steroid metabolism genes and development of liver adenomas in males. We now demonstrate that obesity and aging precipitate alterations of TRIM28-dependent transcriptional dynamics, leading to a metabolic infection state responsible for highly penetrant male-restricted hepatic carcinogenesis. Molecular analyses implicate aberrant androgen receptor stimulation, biliary acid disturbances, and altered responses to gut microbiota in the pathogenesis of Trim28(hep-/-) -associated HCC. Correspondingly, androgen deprivation markedly attenuates the frequency and severity of tumors, and raising animals under axenic conditions completely abrogates their abnormal phenotype, even upon high-fat diet challenge. This work underpins how discrete polyphenic traits in epigenetically metastable conditions can contribute to a cancer-prone state and more broadly provides new evidence linking hormonal imbalances, metabolic disturbances, gut microbiota, and cancer. Photothermal therapy generates a thermal window of immunogenic cell death in neuroblastoma

Nanoparticle-based photothermal therapy (PTT) has been widely investigated in cancer therapy as a rapid and minimally invasive tumor ablation technique. Over the past two decades, several reports utilizing nanoparticles for PTT of diverse tumor types in vitro and in vivo have been described. An emerging area of interest is the effect of PTT on the immune system during tumor therapy, since PTT not only causes tumor cell death, but can also release tumor antigens and endogenous adjuvants (e.g. heat shock proteins, damage-associated molecular patterns (DAMPs)) under certain conditions. These effects have the potential to increase tumor immunogenicity, which can trigger improved therapeutic responses. Engaging the immune system during PTT is important as it offers the potential for persistent treatment responses and immunological memory. Here, we describe a thermal “window” of immunogenic cell death (ICD) elicited by nanoparticle-based photothermal therapy (PTT) in an animal model of neuroblastoma.
