    Gene induction during differentiation of human monocytes into dendritic cells: an integrated study at the RNA and protein levels

    Changes in gene expression occurring during differentiation of human monocytes into dendritic cells were studied at the RNA and protein levels. These studies showed the induction of several gene classes corresponding to various biological functions. These functions encompass antigen processing and presentation, cytoskeleton, cell signalling and signal transduction, but also an increase in mitochondrial function and in the protein synthesis machinery, including some, but not all, chaperones. These changes put in perspective the events occurring during this differentiation process.     Performance of Five Serological Tests in the Diagnosis of Visceral and Cryptic Leishmaniasis: A Comparative Study

    Introduction: Leishmaniasis is a major health problem and its diagnosis still represents a challenge. Since consistent evidence on the comparison of serological methods is lacking, our work aims to compare five serological tests for the diagnosis of visceral and asymptomatic leishmaniasis in southern France, a region where leishmaniasis is endemic. Methodology: Serum samples from 75 patients living in Nice, France were retrospectively analyzed. They included patients affected by visceral leishmaniasis (VL; n = 25), asymptomatic carriers (AC; n = 25) and negative controls (n = 25). Each sample was tested using two immunochromatographic tests (ICT; IT LEISHÂź and TruQuick IgG/IgMÂź), an indirect fluorescent antibody test (IFAT) and two Western Blotting (WB; LDBio BIORADÂź and an in-house method). Results: Diagnosis of VL with IFAT and TruQuickÂź showed the highest diagnostic performance parameters. IFAT had 100% sensitivity and specificity, while TruQuick had 96% sensitivity and 100% specificity. Finally, the two tests showed high accuracy (100% for IFAT and 98% for TruQuick) for the AC group. WB LDBioÂź was the only method able to detect Leishmania latent infection, with a sensitivity of 92%, and a specificity of 100%, with a Negative Predictive Value (NPV) of 93%. This performance is reflected in the high accuracy of the test. Conclusions: The data obtained with TruQuickÂź supports its application in the rapid diagnosis of leishmaniasis in endemic areas, a feature not shown by IFAT despite its high diagnostic performance. Regarding the diagnosis of asymptomatic leishmaniasis, the best results were obtained with WB LDBioÂź, confirming previous studies

    Primary hepatic lymphoma presenting as fulminant hepatic failure with hyperferritinemia: A case report

    <p>Abstract</p> <p>Introduction</p> <p>Primary hepatic lymphoma is an unusual form of non-Hodgkin's lymphoma that usually presents with constitutional symptoms, hepatomegaly and signs of cholestatic jaundice. Diffuse hepatic infiltration is uncommon and presentation with acute hepatic failure even more rare. The presence of markedly elevated ferritin levels can complicate the evaluation process and suggest alternative diagnoses.</p> <p>We present the case of a middle-aged woman exhibiting pancytopenia, hyperferritinemia and rapidly deteriorating to develop acute hepatic failure. Her initial clinical picture led to a working diagnosis of adult onset Still's disease with probable hemophagocytic syndrome before her worsening liver function necessitated a percutaneous liver biopsy and establishment of the final diagnosis of primary hepatic lymphoma.</p> <p>Conclusion</p> <p>Primary hepatic lymphoma is an uncommon malignancy and its manifestation as progressive hepatitis or acute fulminant hepatic failure can be difficult to diagnose. The presence of constitutional symptoms, pancytopenia and high ferritin levels can complicate the evaluation process. A liver biopsy early in the course of liver dysfunction may establish the diagnosis without a higher risk of bleeding complications seen once liver failure sets in.</p

    Since the 2003 Iraq war, the Middle East and North Africa has entered into a New Regional Cold War, characterised by two competing logics: on the one hand, the politicisation of sectarianism opposing a Saudi-led Sunni bloc against an Iran-led Shia bloc and, on the other, an intra-Sunni cleavage around the mobilisation of political Islam, embodied by the Muslim Brotherhood and its supporters vs its opponents. Blending Buzan and Weaver’s regional security complex theory with Donnelly’s notion of ‘heterarchy’ and applying it to the cold wars the region has experienced, the similarities and differences between the Arab Cold War of the 1950s/60s and the New Regional Cold War reveal the increasing number of heterarchic features within the regional security complex: multiple and heterogeneous power centres, different power rankings, a more visible and relevant role of non-state and transnational actors, and the fragmentation of regional norms

    Impact of Genotyping of Mycobacterium tuberculosis on Public Health Practice in Massachusetts

    Massachusetts was one of seven sentinel surveillance sites in the National Tuberculosis Genotyping and Surveillance Network. From 1996 through 2000, isolates from new patients with tuberculosis (TB) underwent genotyping. We describe the impact that genotyping had on public health practice in Massachusetts and some limitations of the technique. Through genotyping, we explored the dynamics of TB outbreaks, investigated laboratory cross-contamination, and identified Mycobacterium tuberculosis strains, transmission sites, and accurate epidemiologic links. Genotyping should be used with epidemiologic follow-up to identify how resources can best be allocated to investigate genotypic findings

    Emergence of a unique group of necrotizing mycobacterial diseases.

    Although most diseases due to pathogenic mycobacteria are caused by Mycobacterium tuberculosis, several other mycobacterial diseases-caused by M. ulcerans (Buruli ulcer), M. marinum, and M. haemophilum-have begun to emerge. We review the emergence of diseases caused by these three pathogens in the United States and around the world in the last decade. We examine the pathophysiologic similarities of the diseases (all three cause necrotizing skin lesions) and common reservoirs of infection (stagnant or slow-flowing water). Examination of the histologic and pathogenic characteristics of these mycobacteria suggests differences in the modes of transmission and pathogenesis, though no singular mechanism for either characteristic has been definitively described for any of these mycobacteria

    Birbeck granule-like "organized smooth endoplasmic reticulum" resulting from the expression of a cytoplasmic YFP-tagged langerin

    Langerin is required for the biogenesis of Birbeck granules (BGs), the characteristic organelles of Langerhans cells. We previously used a Langerin-YFP fusion protein having a C-terminal luminal YFP tag to dynamically decipher the molecular and cellular processes which accompany the traffic of Langerin. In order to elucidate the interactions of Langerin with its trafficking effectors and their structural impact on the biogenesis of BGs, we generated a YFP-Langerin chimera with an N-terminal, cytosolic YFP tag. This latter fusion protein induced the formation of YFP-positive large puncta. Live cell imaging coupled to a fluorescence recovery after photobleaching approach showed that this coalescence of proteins in newly formed compartments was static. In contrast, the YFP-positive structures present in the pericentriolar region of cells expressing Langerin-YFP chimera, displayed fluorescent recovery characteristics compatible with active membrane exchanges. Using correlative light-electron microscopy we showed that the coalescent structures represented highly organized stacks of membranes with a pentalaminar architecture typical of BGs. Continuities between these organelles and the rough endoplasmic reticulum allowed us to identify the stacks of membranes as a form of "Organized Smooth Endoplasmic Reticulum" (OSER), with distinct molecular and physiological properties. The involvement of homotypic interactions between cytoplasmic YFP molecules was demonstrated using an A206K variant of YFP, which restored most of the Langerin traffic and BG characteristics observed in Langerhans cells. Mutation of the carbohydrate recognition domain also blocked the formation of OSER. Hence, a "double-lock" mechanism governs the behavior of YFP-Langerin, where asymmetric homodimerization of the YFP tag and homotypic interactions between the lectin domains of Langerin molecules participate in its retention and the subsequent formation of BG-like OSER. These observations confirm that BG-like structures appear wherever Langerin accumulates and confirm that membrane trafficking effectors dictate their physiology and, illustrate the importance of molecular interactions in the architecture of intracellular membranes

    An ecological study of grassland field margins in Northern Ireland

    The plant diversity of agricultural grassland field margins in Northern Ireland is assessed to provide an ecological basis for agri-environmental management. Approximately 70 per cent of the land area of Northern Ireland (NI) is used for agriculture, of which grassland, typically with hedged boundaries, comprises most. Improved grasslands are predominant, with less semi- improved grassland and a much smaller area of species-rich semi-natural grassland. The thesis aims were to model: a) the regional ecological influence of agricultural management factors on the plant species composition of grassland field margins and b) vegetation composition along the field boundary, field margin, managed grass field spatial gradient. The methodology was through statistically structured quadrat sampling. Multivariate ordination was carried out by detrended correspondence analysis and canonical correspondence analysis to assess the main regional and spatial explanatory environmental gradients influencing plant species composition. A hierarchical typology of Neutral grassland field margins was created with two-way indicator species analysis, to facilitate the implementation of agri-environment scheme management. A soil fertility gradient inferred from weighted vegetation indicators for nitrogen and pH was the main regional environment gradient influencing the species composition of field margins. This gradient was related to grassland management intensity and field boundary structure. A spatial composition gradient between the field margin, field boundary and grass field was also related to field boundary structure and grassland management intensity. This highlights the spatial and temporal variability of the field margin zone as a habitat to be targeted by agri-environment schemes.EThOS - Electronic Theses Online ServiceGBUnited Kingdo
