Endogenous testicular D-aspartic acid regulates gonadal aromatase activity in boar testis.
D-aspartic acid (D-Asp), aromatase enzyme activity and the putative D-Asp involvement on aromatase induction have been studied in the testis of mature boars. The peroxidase-antiperoxidase and the indirect immunofluorescence methods, applied to cryostat and paraffin sections, were used to evaluate D-Asp and aromatase distributions. D-Asp level was dosed by an enzymatic method performed on boar testis extracts. Biochemical aromatase activity was determined by in vitro experiments carried out on testis extracts. D-Asp immunoreactivity was found in Leydig cells, and, to a lesser extent, in germ cells. Analogously, aromatase immunoreactivity was present in Leydig cells, but absent from seminiferous tubule elements. In vitro experiments showed that the addition of D-Asp to testicular tissue acetone powder induced a significant increase of aromatase activity, as assessed by testosterone conversion to 17beta-estradiol. Enzyme Km was not affected by D-Asp (about 25 nM in control and D-Asp added tests). These findings suggest that D-Asp could be involved in the local regulation of aromatase in boar Leydig cells and intervenes in this organ's production of estrogens
Resolving embarrassing medical conditions with online health information
Purpose: Reliance on online health information is proliferating and the Internet has the potential to revolutionize the provision of public health information. The anonymity of online health information may be particularly appealing to people seeking advice on 'embarrassing' health problems. The purpose of this study was to investigate (1) whether data generated by the embarrassingproblems.com health information site showed any temporal patterns in problem resolution, and (2) whether successful resolution of a medical problem using online information varied with the type of medical problem.
Methods: We analyzed the responses of visitors to the embarrassingproblems.com website on the resolution of their problems. The dataset comprised 100,561 responses to information provided on 77 different embarrassing problems grouped into 9 classes of medical problem over an 82-month period. Data were analyzed with a Bernoulli Generalized Linear Model using Bayesian inference.
Results: We detected a statistically important interaction between embarrassing problem type and the time period in which data were collected, with an improvement in problem resolution over time for all of the classes of medical problem on the website but with a lower rate of increase in resolution for urinary health problems and medical problems associated with the mouth and face. As far as we are aware, this is the first analysis of data of this nature.
Conclusions: Findings support the growing recognition that online health information can contribute to the resolution of embarrassing medical problems, but demonstrate that outcomes may vary with medical problem type. The results indicate that building data collection into online information provision can help to refine and focus health information for online users
Optical Monitoring of 3C 390.3 from 1995 to 2004 and Possible Periodicities in the Historical Light Curve
We report V, R, and I band CCD photometry of the radio galaxy 3C 390.3
obtained with the 1.56-m telescope of the Shanghai Astronomical Observatory
from March 1995 to August 2004. Combining these data with data from the
literature, we have constructed a historical light curve from 1894 to 2004 and
searched for periodicities using the CLEANest program. We find possible periods
of 8.30+-1.17, 5.37+-0.49, 3.51+-0.21, and 2.13+-0.08 years.Comment: Accepted by AJ, 34 pages, 11 figure
Hammering K-wires is Superior to Drilling with Irrigation
Cooling during drilling Kirschner wires is not always effective in preventing thermal related damage. In this study, we used a human in vitro model and compared temperature elevation, insertion time, and extraction force between three Kirschner wire insertion methods—drilling with and without irrigation and pneumatic hammering. Forty five Kirschner wires were inserted into 15 fresh human cadaver metacarpals. All three insertion methods were applied in each metacarpal. Drilling without irrigation resulted in a temperature elevation of 67.25 ± 5.4 ºC with significantly lower values for drilling with irrigation (4.15 ± 0.6 ºC) and pneumatic hammering (31.52 ± 3.4 ºC). The insertion time for pneumatic hammering (47.63 ± 8.8 s) was significantly lower compared to drilling without irrigation (263.16 ± 36.5 s) and drilling with irrigation (196.10 ± 28.5 s). Extraction forces after drilling without irrigation, drilling with irrigation, and pneumatic hammering were 39.85 ± 4.1 N, 57.81 ± 6.5 N, and 62.23 ± 6.7 N, respectively. Pneumatic hammering is superior to drilling without irrigation, especially when irrigation is not possible
Frying Doughnuts: What can the reprocessing of X-rays to IR tell us about the AGN environment?
Active galactic nuclei (AGN) produce vast amounts of high energy radiation
deep in their central engines. X-rays either escape the AGN or are absorbed and
re-emitted mostly as IR. By studying the dispersion in the ratio of observed
mid-IR luminosity to observed 2-10keV X-ray luminosity (R_{ir/x}) in AGN we can
investigate the reprocessing material (possibly a torus or donut of dust) in
the AGN central engine, independent of model assumptions. We studied the ratio
of observed mid-IR and 2-10keV X-ray luminosities in a heterogeneous sample of
245 AGN from the literature. We found that when we removed AGN with prominent
jets, ~90% of Type I AGN lay within a very tight dispersion in luminosity ratio
(1<R_{ir/x}<30). This implies that the AGN central engine is extremely uniform
and models of the physical AGN environment (e.g. cloud cover, turbulent disk,
opening angle of absorbing structures such as dusty tori) must span a very
narrow range of parameters. We also found that the far-IR(100um) to mid-IR
(12um) observed luminosity ratio is an effective descriminator between heavily
obscured AGN and relatively unobscured AGN.Comment: 12 pages, MNRAS accepte
Cryo-EM structural studies of the agonist complexed human TRPV4 ion-channel reveals novel structural rearrangements resulting in an open-conformation
The human transient receptor potential vanilloid 4 (hTRPV4) ion channel plays a critical role in a variety of biological processes. Whilst the activation of hTRPV4 gating properties has been reported for a broad spectrum of stimuli, including synthetic 4α-phorbols, the molecular basis of the activation is poorly understood. Here we report the novel cryo-EM structure of the hTRPV4 determined in the presence of the archetypical phorbol acid agonist, 4α-PDD. Complementary mutagenesis experiments support the EM-identified binding site as well as allowing rationalization of disruptive mutants located outside of the 4α-PDD binding site. This work represents the first structural information of hTRPV4 in a ligand-induced open conformation. Together, our data reveal the underlying molecular mechanisms resulting in the opening of the central pore and ion-channel activation and provide a structural template for designing inhibitors targeting the open-state conformation of hTRPV4
On the cosmic evolution of the scaling relations between black holes and their host galaxies: Broad Line AGN in the zCOSMOS survey
(Abriged) We report on the measurement of the rest frame K-band luminosity
and total stellar mass of the hosts of 89 broad line Active Galactic Nuclei
detected in the zCOSMOS survey in the redshift range 1<z<2.2. The unprecedented
multiwavelength coverage of the survey field allows us to disentangle the
emission of the host galaxy from that of the nuclear black hole in their
Spectral Energy Distributions. We derive an estimate of black hole masses
through the analysis of the broad Mg II emission lines observed in the
medium-resolution spectra taken with VIMOS/VLT as part of the zCOSMOS project.
We found that, as compared to the local value, the average black hole to host
galaxy mass ratio appears to evolve positively with redshift, with a best fit
evolution of the form (1+z)^{0.68 \pm0.12 +0.6 -0.3}, where the large
asymmetric systematic errors stem from the uncertainties in the choice of IMF,
in the calibration of the virial relation used to estimate BH masses and in the
mean QSO SED adopted. A thorough analysis of observational biases induced by
intrinsic scatter in the scaling relations reinforces the conclusion that an
evolution of the MBH-M* relation must ensue for actively growing black holes at
early times: either its overall normalization, or its intrinsic scatter (or
both) appear to increase with redshift. This can be interpreted as signature of
either a more rapid growth of supermassive black holes at high redshift, a
change of structural properties of AGN hosts at earlier times, or a significant
mismatch between the typical growth times of nuclear black holes and host
