    Double Trouble: Commingled Effects of Fast Food and Sugar-Sweetened Beverage Consumption and the Intervening Role of Physical Activity on Childhood Obesity

    Children are exposed to a great deal of food and beverage promotion. This is particularly concerning given that the prevalence of childhood obesity, a critical public health challenge, may be partially due to the increased consumption of fast food and sugar-sweetened beverages. However, there are lingering questions about the complex relationships between fast food and sugar-sweetened beverage consumption, physical activity levels, and childhood obesity. To address these complex relationships, this research examines the interaction of fast food and sugar-sweetened beverage consumption, along with the frequency of physical activity, in leading to the likelihood of a child being overweight or obese. A primary concern is the compensatory role of physical activity in offsetting the direct and indirect effects of fast food and sugar-sweetened beverage consumption on childhood obesity. For a sample of more than 4000 children between five and eleven years of age, results show that physical activity can partially counter the direct relationship between (1) fast food consumption and obesity and (2) sugar-sweetened beverage consumption and obesity. However, this intervening role of physical activity indicates that activity level is unable to compensate for the combined effect of high fast food and high sugar-sweetened drink consumption on obesity likelihood. Conclusions for the public health community and possible implications for policy makers interested in the combined roles of fast food and sugar-sweetened drink consumption on childhood health and obesity are offered

    UX007 for the treatment of long chain-fatty acid oxidation disorders: Safety and efficacy in children and adults following 24weeks of treatment.

    BACKGROUND: Long-chain fatty acid oxidation disorders (LC-FAOD) lead to accumulation of high concentrations of potentially toxic fatty acid intermediates. Newborn screening and early intervention have reduced mortality, but most patients continue to experience frequent hospitalizations and significant morbidity despite treatment. The deficient energy state can cause serious liver, muscle, and heart disease, and may be associated with an increased risk of sudden death. Triheptanoin is a medium odd-chain fatty acid. Anaplerotic metabolites of triheptanoin have the potential to replace deficient tricarboxylic acid (TCA) cycle intermediates, resulting in net glucose production as a novel energy source for the treatment of LC-FAOD. STUDY DESIGN: A single-arm, open-label, multicenter Phase 2 safety and efficacy study evaluated patients with severe LC-FAOD evidenced by ongoing related musculoskeletal, cardiac, and/or hepatic events despite treatment. After a four-week run-in on current regimen, investigational triheptanoin (UX007) was titrated to a target dose of 25-35% of total daily caloric intake. Patients were evaluated on several age/condition-eligible endpoints, including submaximal exercise tests to assess muscle function/endurance (12-minute walk test; 12MWT) and exercise tolerance (cycle ergometry), and health related quality of life (HR-QoL). Results through 24weeks of treatment are presented; total study duration is 78weeks. RESULTS: Twenty-nine patients (0.8 to 58years) were enrolled; most qualified based on severe musculoskeletal disease. Twenty-five patients (86%) completed the 24-week treatment period. At Week 18, eligible patients (n=8) demonstrated a 28% increase (LS mean=+181.9 meters; p=0.087) from baseline (673.4meters) in 12MWT distance. At Week 24, eligible patients (n=7) showed a 60% increase in watts generated (LS mean=+409.3W; p=0.149) over baseline (744.6W) for the exercise tolerance test. Improvements in exercise tests were supported by significant improvements from baseline in the adult (n=5) self-reported SF-12v2 physical component summary score (LS mean=+8.9; p CONCLUSIONS: In patients with severe LC-FAOD, UX007 interim study results demonstrated improved exercise endurance and tolerance, and were associated with positive changes in self-reported HR-QoL

    The emission line near 1319 A in solar and stellar spectra

    An emission line near 1319 A is one of the strongest unidentified lines in the ultraviolet spectra of cool dwarf stars. In most line lists it is identified as a transition in N I, although its intensity would then be anomalous and the observed wavelength does not fit precisely that expected for N I. The line is also observed in cool giant stars. The measured wavelength of the line in stellar spectra is 1318.94 (+,- 0.01) A. Observations of giant stars provide further evidence that this line is not due to N I. It is proposed that this line is a decay from a previously unknown level in S I, which lies above the first ionization limit. This is identified with the 3d singlet D (odd parity) term. The previous tentative assignment of this term to the S I line at 1309.3 A then needs to be revised. The 1309.3 A line has been identified here for the first time in an astrophysical source. The singlet D (odd parity) level could, in principle, be populated by collisions from nearby autoionizing levels that have large number-densities, through population by di-electronic capture. Spin-orbit interaction with the autoionizing triplet D (odd parity) term might also lead to di-electronic capture into the singlet D (odd parity) level. A line at 1309.87 A observed in cool giant stars is identified as a transition in P II, pumped by the O I resonance lines.Comment: 9 pages, 3 figures, to be published in Monthly Notices of the Royal Astronomical Societ

    Weyl's law and quantum ergodicity for maps with divided phase space

    For a general class of unitary quantum maps, whose underlying classical phase space is divided into several invariant domains of positive measure, we establish analogues of Weyl's law for the distribution of eigenphases. If the map has one ergodic component, and is periodic on the remaining domains, we prove the Schnirelman-Zelditch-Colin de Verdiere Theorem on the equidistribution of eigenfunctions with respect to the ergodic component of the classical map (quantum ergodicity). We apply our main theorems to quantised linked twist maps on the torus. In the Appendix, S. Zelditch connects these studies to some earlier results on `pimpled spheres' in the setting of Riemannian manifolds.     Antigenic diversity is generated by distinct evolutionary mechanisms in African trypanosome species

    Antigenic variation enables pathogens to avoid the host immune response by continual switching of surface proteins. The protozoan blood parasite Trypanosoma brucei causes human African trypanosomiasis ("sleeping sickness") across sub-Saharan Africa and is a model system for antigenic variation, surviving by periodically replacing a monolayer of variant surface glycoproteins (VSG) that covers its cell surface. We compared the genome of Trypanosoma brucei with two closely related parasites Trypanosoma congolense and Trypanosoma vivax, to reveal how the variant antigen repertoire has evolved and how it might affect contemporary antigenic diversity. We reconstruct VSG diversification showing that Trypanosoma congolense uses variant antigens derived from multiple ancestral VSG lineages, whereas in Trypanosoma brucei VSG have recent origins, and ancestral gene lineages have been repeatedly co-opted to novel functions. These historical differences are reflected in fundamental differences between species in the scale and mechanism of recombination. Using phylogenetic incompatibility as a metric for genetic exchange, we show that the frequency of recombination is comparable between Trypanosoma congolense and Trypanosoma brucei but is much lower in Trypanosoma vivax. Furthermore, in showing that the C-terminal domain of Trypanosoma brucei VSG plays a crucial role in facilitating exchange, we reveal substantial species differences in the mechanism of VSG diversification. Our results demonstrate how past VSG evolution indirectly determines the ability of contemporary parasites to generate novel variant antigens through recombination and suggest that the current model for antigenic variation in Trypanosoma brucei is only one means by which these parasites maintain chronic infections

    221 Newborn-Screened Neonates with Medium-Chain Acyl-Coenzyme A Dehydrogenase Deficiency: Findings from the Inborn Errors of Metabolism Collaborative

    Introduction: There is limited understanding of relationships between genotype, phenotype and other conditions contributing to health in neonates with medium-chain acyl-coenzyme A dehydrogenase deficiency (MCADD) identified through newborn screening. Methods: Retrospective analysis of comprehensive data from a cohort of 221 newborn-screened subjects identified as affected with MCADD in the Inborn Errors of Metabolism-Information System (IBEM-IS), a long term follow-up database of the Inborn Errors of Metabolism Collaborative, was performed. Results: The average age at notification of first newborn screen results to primary care or metabolic providers was 7.45 days. The average octanoylcamitine (C8) value on first newborn screen was 11.2 mu mol/L (median 8.6, range 036-43.91). A higher C8 level correlated with an earlier first subspecialty visit. Subjects with low birth weight had significantly lower C8 values. Significantly higher C8 values were found in symptomatic newborns, in newborns with abnormal lab testing in addition to newborn screening and/or diagnostic tests, and in subjects homozygous for the c.985A\u3eG ACADM gene mutation or compound heterozygous for the c.985A\u3eG mutation and deletions or other known highly deleterious mutations. Subjects with neonatal symptoms, or neonatal abnormal labs, or neonatal triggers were more likely to have at least one copy of the severe c.985A\u3eG ACADM gene mutation. C8 and genotype category were significant predictors of the likelihood of having neonatal symptoms. Neonates with select triggers were more likely to have symptoms and laboratory abnormalities. Conclusions: This collaborative study is the first in the United States to describe health associations of a large cohort of newborn-screened neonates identified as affected with MCADD. The IBEM-IS has utility as a platform to better understand the characteristics of individuals with newborn-screened conditions and their follow-up interactions with the health system.     The Impact of Gymnastics on Children’s Physical Self-Concept and Movement Skill Development in Primary Schools

    This study evaluated the effectiveness of an 8-week gymnastics curriculum on children’s movement competence and their physical self-concept. There were 113 children (46% girls, 49% intervention) with a mean age of 9.4 years (SD = 1.8) that participated. Intervention children underwent 8 weeks of gymnastics and the comparison group continued with their standard curriculum. Age was a significant co-variate, a separate analysis was conducted on the lower (grades 2 and 4) and upper (grade 6) groups. The lower age group showed significant improvement in favor of the gymnastic group in fundamental movement skills. The upper age group showed a significant improvement for the control group in general body coordination and fundamental movement skills. For all grades, the physical self-concept showed a significant main effect in favor of the gymnastics group. The gymnastics intervention was found to be of particular benefit for developing children’s movement competence and physical self-concept in younger children
