    NvPrdm14d-expressing neural progenitor cells contribute to non-ectodermal neurogenesis in Nematostella vectensis

    Neurogenesis has been studied extensively in the ectoderm, from which most animals generate the majority of their neurons. Neurogenesis from non-ectodermal tissue is, in contrast, poorly understood. Here we use the cnidarian Nematostella vectensis as a model to provide new insights into the molecular regulation of non-ectodermal neurogenesis. We show that the transcription factor NvPrdm14d is expressed in a subpopulation of NvSoxB(2)-expressing endodermal progenitor cells and their NvPOU4-expressing progeny. Using a new transgenic reporter line, we show that NvPrdm14d-expressing cells give rise to neurons in the body wall and in close vicinity of the longitudinal retractor muscles. RNA-sequencing of NvPrdm14d::GFP-expressing cells and gene knockdown experiments provide candidate genes for the development and function of these neurons. Together, the identification of a population of endoderm-specific neural progenitor cells and of previously undescribed putative motoneurons in Nematostella provide new insights into the regulation of non-ectodermal neurogenesis.publishedVersio

    First evidence of a main channel generated by the Mediterranean Outflow Water after its exit from the Gibraltar Strait

    An extensive terrace comprising sandy sheeted drifts characterises the proximal sector (close to the Straits of Gibraltar) of the Contourite Depositional System (CDS) of the Gulf of Cadiz and the western continental margin of Portugal. A novel morphosedimentary study over this terrace has been executed based on new multibeam echosounder data, seismic profiles, and surficial sediments samples collected during the CONTOURIBER-1 Cruise (2010). Based on that study, an impressive large channel laterally connected with the central part of the Strait of Gibraltar (Camarinal Sill) has been identified. It is associated southwestward with a huge contourite levee (sand bank), which has been generated by overflow processes related to the Mediterranean Outflow Water (MOW). In the middle to the distal part of the terrace, there are other minor channels, erosional scour alignments and depositional features. The mapping of all these features coupled with CTD data allows to further understand the pathway and deceleration of the MOW, in addition to aid the conceptual identification of contourite terrace

    Going beyond body exposure therapy. Presenting an innovative Virtual Reality and Eye-Tracking body-related attentional bias task

    The present study provides preliminary findings of an innovative body-related attentional bias modification task (ABMT) using Virtual Reality (VR) and Eye-tracking (ET) technologies. Analyses were carried out on a sample composed of college women (n = 35) who were divided into groups of those with high body dissatisfaction (n = 16) and those with low body dissatisfaction (n = 19). All participants were exposed to an immersive virtual environment in which they were embodied in a virtual body that resembled the measurements of their real body. Subjects performed the body-related attentional bias modification task for 20 minutes. Eating Disorder (ED) symptomatology disturbances and body-related attentional bias (AB) were measured before and after a single session of the VR ABMT. Results showed a significant (p<.05) reduction of fear of gaining weight after the intervention among women with high body dissatisfaction. Our results indicated promising evidence in favor of using this ABMT, particularly among women with high body dissatisfaction. In addition, the current research provides a new application of VR and ET technologies that might open a wide range of possibilities for designing and developing new body-related interventions among patients with EDs and women with body image disturbances

    Buried volcanic structures in the Gulf of Naples (Southern Tyrrhenian Sea, Italy) resulting from high resolution magnetic survey and seismic profiling

    In this paper we present a correlation between volcanic structures and magnetic anomalies in the Gulf of Naples (Southern Tyrrhenian Sea) based on high resolution magnetic profiling. A densely spaced grid of magnetic profiles coupled with multichannel seismics (seismic source Watergun 15 cubic inch) was recorded in the Gulf of Naples, representing an active volcanic area during the Late Quaternary (volcanic centers of Somma-Vesuvius, Phlegraean Fields and Ischia and Procida islands). The dataset was collected during the oceanographic cruise GMS00-05 which took place during October-November 2000 in the South Tyrrhenian Sea onboard of the R/V Urania (National Research Council, Italy). Shallow volcanic structures in the subsurface of the gulf were recognized by seismo-stratigraphic analysis of high resolution profiles; the volcanic nature of some of these structures was inferred identifying the magnetic anomalies on a high resolution magnetic anomaly map of the gulf. Even if qualitative, the correlations between seismic and magnetic profiles allow us to better assess the geological structure of the Gulf of Naples

    Behavioral Voluntary and Social Bioassays Enabling Identification of Complex and Sex-Dependent Pain-(-Related) Phenotypes in Rats with Bone Cancer

    Simple Summary Bone metastases are one of the most common complications in patients with advanced cancer that result in pain, which is usually severe, thereby significantly reducing the patient's quality of life. Although preclinical pain research in rodents is improving, the pain phenotyping methods currently used have been criticized. This study aimed to identify in detail pain phenotypes of cancer-induced bone pain (CIBP) in both sexes of rats. CIBP in the splint bone on one side results in a distinct CIBP-related phenotype characterized by mechanical hypersensitivity, resting pain, and antalgic gait in both sexes. Progression of tumor growth leads to the establishment of the CIBP phenotype that appears earlier in male than in female rats and affects rat-specific social behaviors in both sexes. We demonstrate social transfer of pain in a bone cancer model in both sexes, resulting in mechanical and, in females, also heat hypervigilance in non-tumor bearing control rats. Cancer-induced bone pain (CIBP) is a common and devastating symptom with limited treatment options in patients, significantly affecting their quality of life. The use of rodent models is the most common approach to uncovering the mechanisms underlying CIBP; however, the translation of results to the clinic may be hindered because the assessment of pain-related behavior is often based exclusively on reflexive-based methods, which are only partially indicative of relevant pain in patients. To improve the accuracy and strength of the preclinical, experimental model of CIBP in rodents, we used a battery of multimodal behavioral tests that were also aimed at identifying rodent-specific behavioral components by using a home-cage monitoring assay (HCM). Rats of all sexes received an injection with either heat-deactivated (sham-group) or potent mammary gland carcinoma Walker 256 cells into the tibia. By integrating multimodal datasets, we assessed pain-related behavioral trajectories of the CIBP-phenotype, including evoked and non-evoked based assays and HCM. Using principal component analysis (PCA), we discovered sex-specific differences in establishing the CIBP-phenotype, which occurred earlier (and differently) in males. Additionally, HCM phenotyping revealed the occurrence of sensory-affective states manifested by mechanical hypersensitivity in sham when housed with a tumor-bearing cagemate (CIBP) of the same sex. This multimodal battery allows for an in-depth characterization of the CIBP-phenotype under social aspects in rats. The detailed, sex-specific, and rat-specific social phenotyping of CIBP enabled by PCA provides the basis for mechanism-driven studies to ensure robustness and generalizability of results and provide information for targeted drug development in the future

    Pulmonary Safety Profile of Esc Peptides and Esc-Peptide-Loaded Poly(lactide-co-glycolide) Nanoparticles: A Promising Therapeutic Approach for Local Treatment of Lung Infectious Diseases

    In recent years, we have discovered Esc(1-21) and its diastereomer (Esc peptides) as valuable candidates for the treatment of Pseudomonas lung infection, especially in patients with cystic fibrosis (CF). Furthermore, engineered poly(lactide-co-glycolide) (PLGA) nanoparticles (NPs) were revealed to be a promising pulmonary delivery system of antimicrobial peptides. However, the "ad hoc" development of novel therapeutics requires consideration of their stability, tolerability, and safety. Hence, by means of electrophysiology experiments and preclinical studies on healthy mice, we demonstrated that neither Esc peptides or Esc-peptide-loaded PLGA NPs significantly affect the integrity of the lung epithelium, nor change the global gene expression profile of lungs of treated animals compared to those of vehicle-treated animals. Noteworthy, the Esc diastereomer endowed with the highest antimicrobial activity did not provoke any pulmonary pro-inflammatory response, even at a concentration 15-fold higher than the efficacy dosage 24 h after administration in the free or encapsulated form. The therapeutic index was ≥70, and the peptide was found to remain available in the bronchoalveolar lavage of mice, after two days of incubation. Overall, these studies should open an avenue for a new up-and-coming pharmacological approach, likely based on inhalable peptide-loaded NPs, to address CF lung disease

    New evidences of the interplay between a turbidite canyon (Guadiaro canyon, NW Alboran Sea) and current-driven along slope processes

The work is based on data acquired during the FAUCES surveys, comprising high-resolution bathymetry and seismic profiles, in combination with a database comprising seismic profiles acquired since the 70s. The interaction between downslope and alongslope processes occurs on both margins of the Alboran Sea, but this interaction is especially complex in the Spanish margin, where the turbidite canyons cut the continuity of terraced plastered drifts. The interaction may vary between the dominance of downslope processes over the alongslope processes (i.e., Almeria turbidite system), the alternation between downslope and alongslope deposits (which is the case of the most recent lobe of the Guadiaro fan) and the influence of alongslope processes over downslope processes. The Guadiaro canyon provides an example of the last case: After a first phase in which the canyon was incised, during a second phase (Pliocene) downslope processes were dominant, as indicated by the chaotic facies infilling the canyon incision. A last phase (Quaternary) characterized by stratified discontinuous facies indicate a greater influence of contourite alongslope deposits. During the stage two and mostly during stage three, the lateral accretion of packages (LAPs) of stratified facies with NE progradation trend have been recognized on its SW side (right margin), progressively invading of the Guadiaro Palaeocanyon and affecting its current location and shape (Fig. 1). These findings have been confirmed by isochore maps, in which a depocentre of Quaternary age runs parallel to the canyon on its SW side. The oceanography of the study area is marked by the closeness to the Strait of Gibraltar, located to the west. The Mediterranean intermediate waters, comprising Western Intermediate Waters (WIW), Levantine Intermediate Waters (LIW) and the upper portion of Tyrrhenian Dense Waters (TDWi), flow along the Spanish margin towards the Strait. On the opposite, the Atlantic Waters enter forming a strong jet into the Mediterranean. These two groups of water masses are separated by a pycnocline, along which internal waves formed in the Camarinal Sill travel eastwards. The NE progradation of the LAPs affecting the Guadiaro canyon points to the much stronger effect of the Atlantic Jet and the eastwardtravelling internal waves over the intermediate MWs directed towards the Strait of Gibraltar. Ultimately, these findings may shed light on one of the reasons behind the sudden abandonment and obliteration of the Estepona Palaeocanyon after the BQD, possibly located where the Atlantic Jet and the internal waves cease their sediment transport towards the NE and where the weaker SW transport by the intermediate Mediterranean Waters become the only alongslope transport mechanismContribution from Project FAUCES - CTM2015-65461-C2-R (MINECO/FEDER

    1st International Experts' Meeting on Agitation: Conclusions Regarding the Current and Ideal Management Paradigm of Agitation.

    Agitation is a heterogeneous concept without a uniformly accepted definition, however, it is generally considered as a state of cognitive and motor hyperactivity characterized by excessive or inappropriate motor or verbal activity with marked emotional arousal. Not only the definition but also other aspects of agitated patients' care are still unsolved and need consensus and improvement. To help the discussion about agitation among experts and improve the identification, management, and treatment of agitation, the 1st International Experts' Meeting on Agitation was held in October 2016 in Madrid. It was attended by 20 experts from Europe and Latin America with broad experience in the clinical management of agitated patients. The present document summarizes the key conclusions of this meeting and highlights the need for an updated protocol of agitation management and treatment, the promotion of education and training among healthcare professionals to improve the care of these patients and the necessity to generate clinical data of agitated episodes

This ebook presents contributions addressing these issues from different disciplinary points of view. The proposed interpretations allow useful comparisons and may constitute the beginning of a broader and more articulated discussion
