Apolipoprotein B is regulated by gonadotropins and constitutes a predictive biomarker of IVF outcomes
International audienceAbstractBackgroundFollicular fluid (FF) is an important micro-environment influencing oocyte growth, its development competence, and embryo viability. The FF content analysis allows to identify new relevant biomarkers, which could be predictive of in vitro fertilization (IVF) outcomes. Inside ovarian follicle, the amount of FF components from granulosa cells (GC) secretion, could be regulated by gonadotropins, which play a major role in follicle development.MethodsThis prospective study included 61 female undergoing IVF or Intra-cytoplasmic sperm injection (ICSI) procedure. Apolipoprotein B (APOB) concentrations in follicular fluid and APOB gene and protein expression in granulosa cells from reproductively aged women undergoing an in vitro fertilization program were measured. The statistical analyses were performed according to a quartile model based on the amount of APOB level found in FF.ResultsAmounts of APOB were detected in human FF samples (mean ± SD: 244.6 ± 185.9 ng/ml). The odds of obtaining an oocyte in the follicle and a fertilized oocyte increased significantly when APOB level in FF was higher than 112 ng/ml [i.e., including in Quartile Q 2, Q3 and Q4] (p = 0.001; p < 0.001, respectively). The probabilities of obtaining an embryo and a top quality embryo on day 2, were significantly higher if APOB levels were within the ranges of 112 and 330 ng/ml (i.e. in Q2 and Q3) or 112 and 230 ng/ml (i.e. in Q2), respectively (p < 0.001; p = 0.047, respectively). In addition, our experiments in vitro indicated that APOB gene and protein expression, along with APOB content into culture were significantly under-expressed in GC upon stimulation with gonadotropins (follicular stimulating hormone: FSH and/or human chorionic gonadotropin: hCG).ConclusionWe are reporting a positive and statistically significant associations between APOB and oocyte retrieval, oocyte fertilization, and embryo quality. Using an experimental study component, the authors report significant reduced APOB expression and content for luteinized granulosa cells cultured in the presence of gonadotropins

Ageing is characterized by immunosenescence and the progressive decline in immunity in association with an increased frequency of infections and chronic disease. This complex process affects both the innate and adaptive immune systems with a progressive decline in most immune cell populations and defects in activation resulting in loss of function. Although host genetics and environmental factors, such as stress, exercise and diet can impact on the onset or course of immunosenescence, the mechanisms involved are largely unknown. This review focusses on identifying the most significant aspects of immunosenescence and on the evidence that nutritional intervention might delay this process, and consequently improve the quality of life of the elderly
Fish oil administration in older adults: is there potential for adverse events? A systematic review of the literature
ackground: Omega-3 (n-3) fatty acid supplementation is becoming increasingly popular. However given its
antithrombotic properties the potential for severe adverse events (SAE) such as bleeding has safety implications,
particularly in an older adult population. A systematic review of randomized control trials (RCT) was conducted to
explore the potential for SAE and non-severe adverse events (non-SAE) associated with n-3 supplementation in
older adults.
Methods: A comprehensive search strategy using Medline and a variety of other electronic sources was conducted.
Studies investigating the oral administration of n-3 fish oil containing eicosapentaenoic acid (EPA), docosahexaenoic
acid (DHA) or both against a placebo were sourced. The primary outcome of interest included reported SAE
associated with n-3 supplementation. Chi-square analyses were conducted on the pooled aggregate of AEs.
Results: Of the 398 citations initially retrieved, a total of 10 studies involving 994 older adults aged ≥60 years were
included in the review. Daily fish oil doses ranged from 0.03 g to 1.86 g EPA and/or DHA with study durations
ranging from 6 to 52 weeks. No SAE were reported and there were no significant differences in the total AE rate
between groups (n-3 intervention group: 53/540; 9.8%; placebo group: 28/454; 6.2%; p= 0.07). Non-SAE relating to
gastrointestinal (GI) disturbances were the most commonly reported however there was no significant increase in
the proportion of GI disturbances reported in participants randomized to the n-3 intervention (n-3 intervention
group: 42/540 (7.8%); placebo group: 24/454 (5.3%); p= 0.18).
Conclusions: The potential for AEs appear mild-moderate at worst and are unlikely to be of clinical significance. The
use of n-3 fatty acids and the potential for SAE should however be further researched to investigate whether this
evidence is consistent at higher doses and in other populations. These results also highlight that well-documented data
outlining the potential for SAE following n-3 supplementation are limited nor adequately reported to draw definitive
conclusions concerning the safety associated with n-3 supplementation. A more rigorous and systematic approach for
monitoring and recording AE data in clinical settings that involve n-3 supplementation is required.The authors would like to acknowledge funding

Male infertility: an obstacle to sexuality?
International audienceInteractions between infertility and sexuality are numerous and complex. Infertile men may suffer from sexual dysfunction (SD) when undergoing an assisted reproductive technology programme. We undertook a review both in French and English of the available data on male SD when being diagnosed with a fertility problem with a specific focus on azoospermic men. The review was performed over a 30-year time period using PubMed/Medline. The sexual concerns and needs of infertile/sterile men for whom potential parenting can be compromised were evaluated. When diagnosed with infertility, men usually go through a crisis that can have a deleterious effect on their sexuality with sometimes a feeling of sexual inadequacy. Infertile men will feel stigmatized because they are perceived as being deficient in a specific component of their masculinity. Hence, subsequent SD may occur that can impact the couple sexuality and the infertility management. However, little is known on how the announcement of azoospermia may affect male on a sexual and psychological point of view. The present review suggests that a global management through a healthcare network (biologist, andrologist, sexologist and psychologist) is required which will allow to consider infertility and its subsequent sexual disorders as a whole and not as dichotomized issues
Effects of pulsed near infrared light (NIR) on Bacillus subtilis spores
International audienceIn this study, we develop a characterization of bacterial spore resistance to NIR pulsed light under modalities traditionally used in multiphoton microscopy. Energy dose and laser power are both key parameters in spore and bacterial cell inactivation. Surprisingly, spores and vegetative cells seem to show a similar sensitivity to pulsed NIR, spores being only 2-fold more resistant than their vegetative counterparts. This work enables us to eliminate certain hypotheses concerning the main driver of spore inactivation processes. Our findings suggest that damage leading to inactivation is mainly caused by photochemical reactions characterized by multiple possible pathways, including DNA damage or oxidation processes
Analysis of multivariate images in fluorescence microscopy
International audienceA multivariate image is an image stack in which each pixel contains several variables. Such images are common in many fields (medicine, imaging microscopy, satellite imaging...) and their analysis requires adapted multivariate statistical methods. In fluorescence imaging microscopy, different probes or different measurements such as intensity, fluorescence lifetime or spectral information can be observed from one view. However, this is not yet analysed as multivariate images. Here, we are presenting a full approach of multivariate analysis of fluorescence microscopy images and we are proposing a free R package (multifluo) to conduct it
Similarly increased congenital anomaly rates after intrauterine insemination and IVF technologies: a retrospective cohort study
International audienceBACKGROUND: While intrauterine insemination (IUI), a simple, inexpensive and non-invasive technique, is the most used assisted reproduction technology (ART) worldwide, the risk of major birth defects following IUI is paradoxically not well documented. METHODS: Retrospective cohort study performed in Burgundy, France, over a 9-year period which consisted of the cross analysis of two prospective databases, the Burgundy perinatal network database and the database of the assisted conception units in Burgundy. A total of 1348 ART singletons [in vitro fertilization technologies (IVFT): n= 903; IUI: n= 445] matched with 4044 infants conceived naturally, 552 ART twins (IVFT: n= 362; IUI: n= 190) matched with 1656 twins who were conceived naturally. The major birth defects were categorized according to the European Surveillance of Congenital Anomalies classification EUROCAT. RESULTS: Compared with naturally conceived singletons, singletons born after IUI and IVFT had a higher prevalence of major congenital malformations, with adjusted odd ratios (AOR) of 2.0 [95% confidence interval (CI) 1.0-3.8] and 2.0 (CI 1.3-3.1); 3.6 and 4.2% of infants born, respectively. All twins and unlike-sex twins born after IVFT but not IUI, have an increased prevalence of major birth defects compared with naturally conceived twins; AOR of 3.0 (CI 1.6-5.6) and 3.7 (CI 1.1-16.9), respectively. When comparing IUI with IVFT, no differences were observed for singletons (AOR 1.0; CI 0.4-2.2), all twins (AOR 0.4; CI 0.1-1.2) and unlike-sex twins (AOR 0.3; CI 0.1-4.5). CONCLUSIONS: The risk of major birth defects in singletons conceived through IUI was increased over naturally conceived singletons. This risk was no different from that observed after IVFT
