    Microsphere-Based Scaffolds Encapsulating Tricalcium Phosphate And Hydroxyapatite For Bone Regeneration

    Bioceramic mixtures of tricalcium phosphate (TCP) and hydroxyapatite (HAp) are widely used for bone regeneration because of their excellent cytocompatibility, osteoconduction, and osteoinduction. Therefore, we hypothesized that incorporation of a mixture of TCP and HAp in microsphere-based scaffolds would enhance osteogenesis of rat bone marrow stromal cells (rBMSCs) compared to a positive control of scaffolds with encapsulated bone-morphogenic protein-2 (BMP-2). Poly(D,L-lactic-co-glycolic acid) (PLGA) microsphere-based scaffolds encapsulating TCP and HAp mixtures in two different ratios (7:3 and 1:1) were fabricated with the same net ceramic content (30 wt%) to evaluate how incorporation of these ceramic mixtures would affect the osteogenesis in rBMSCs. Encapsulation of TCP/HAp mixtures impacted microsphere morphologies and the compressive moduli of the scaffolds. Additionally, TCP/HAp mixtures enhanced the end-point secretion of extracellular matrix (ECM) components relevant to bone tissue compared to the “blank” (PLGA-only) microsphere-based scaffolds as evidenced by the biochemical, gene expression, histology, and immunohistochemical characterization. Moreover, the TCP/HAp mixture groups even surpassed the BMP-2 positive control group in some instances in terms of matrix synthesis and gene expression. Lastly, gene expression data suggested that the rBMSCs responded differently to different TCP/HAp ratios presented to them. Altogether, it can be concluded that TCP/HAp mixtures stimulated the differentiation of rBMSCs toward an osteoblastic phenotype, and therefore may be beneficial in gradient microsphere-based scaffolds for osteochondral regeneration

    This paper describes a computational tool developed to aid the analysis of chemical, isotopic and production data of geothermal wells. ANAGEOT is an interactive and user-friendly software that allows quickly and efficient manner, modification, retrieval and visualization of the wide amounts of data involved in the study of reservoirs. ANAGEOT optimizes time and resources, due to high quantity of historical data of most of the wells of various petroleum and geothermal fields which involve information sometimes of very long time periods. The system has the flexibility to work with different fields, considering a database for each one A very important feature of this tool is that the reports, charts and graphs are generated in a way which can be edited at any time. Similarly these can be used as source files for other applications, because they are generated in wide commercial use applications such as Microsoft Word, Excel and Golden Grapher

    Psychedelics Promote Structural and Functional Neural Plasticity.

    Atrophy of neurons in the prefrontal cortex (PFC) plays a key role in the pathophysiology of depression and related disorders. The ability to promote both structural and functional plasticity in the PFC has been hypothesized to underlie the fast-acting antidepressant properties of the dissociative anesthetic ketamine. Here, we report that, like ketamine, serotonergic psychedelics are capable of robustly increasing neuritogenesis and/or spinogenesis both in vitro and in vivo. These changes in neuronal structure are accompanied by increased synapse number and function, as measured by fluorescence microscopy and electrophysiology. The structural changes induced by psychedelics appear to result from stimulation of the TrkB, mTOR, and 5-HT2A signaling pathways and could possibly explain the clinical effectiveness of these compounds. Our results underscore the therapeutic potential of psychedelics and, importantly, identify several lead scaffolds for medicinal chemistry efforts focused on developing plasticity-promoting compounds as safe, effective, and fast-acting treatments for depression and related disorders

    MicroRNAs and Drinking: Association between the Pre-miR-27a rs895819 Polymorphism and Alcohol Consumption in a Mediterranean Population

    Recently, microRNAs (miRNA) have been proposed as regulators in the different processes involved in alcohol intake, and differences have been found in the miRNA expression profile in alcoholics. However, no study has focused on analyzing polymorphisms in genes encoding miRNAs and daily alcohol consumption at the population level. Our aim was to investigate the association between a functional polymorphism in the pre-miR-27a (rs895819 A>G) gene and alcohol consumption in an elderly population. We undertook a cross-sectional study of PREvencion con DIeta MEDiterranea (PREDIMED)-Valencia participants (n = 1007, including men and women aged 67 +/- 7 years) and measured their alcohol consumption (total and alcoholic beverages) through a validated questionnaire. We found a strong association between the pre-miR-27a polymorphism and total alcohol intake, this being higher in GG subjects (5.2 +/- 0.4 in AA, 5.9 +/- 0.5 in AG and 9.1 +/- 1.8 g/day in GG; p(adjusted) = 0.019). We also found a statistically-significant association of the pre-miR-27a polymorphism with the risk of having a high alcohol intake (> 2 drinks/day in men and > 1 in women): 5.9\% in AA versus 17.5\% in GG; p(adjusted) < 0.001. In the sensitivity analysis, this association was homogeneous for sex, obesity and Mediterranean diet adherence.     Microsphere-Based Osteochondral Scaffolds Carrying Opposing Gradients Of Decellularized Cartilage And Demineralized Bone Matrix

To access the final edited and published work see https://doi.org/10.1021/acsbiomaterials.6b00071.Extracellular matrix (ECM) “raw materials” such as demineralized bone matrix (DBM) and cartilage matrix have emerged as leading scaffolding materials for osteochondral regeneration owing to their capacity to facilitate progenitor/resident cell recruitment, infiltration, and differentiation without adding growth factors. Scaffolds comprising synthetic polymers are sturdy yet generally lack cues for guiding cell differentiation. We hypothesized that opposing gradients of decellularized cartilage (DCC) and DBM in polymeric microsphere-based scaffolds would provide superior regeneration compared to polymer-only scaffolds in vivo. Poly(D,L-lactic-co-glycolic acid) (PLGA) microsphere-based scaffolds were fabricated, either with opposing gradients of DCC and DBM encapsulated (GRADIENT) or without DCC and DBM (BLANK control), and implanted into rabbit osteochondral defects in medial femoral condyles. After 12 weeks, gross morphological evaluation showed that the repair tissue in about 30% of the implants was either slightly or significantly depressed, hinting toward rapid polymer degradation in scaffolds from both of the groups. Additionally, no differences were observed in gross morphology of the repair tissue between the BLANK and GRADIENT groups. Mechanical testing revealed no significant differences in model parameter values between the two groups. Histological observations demonstrated that the repair tissue in both of the groups was fibrous in nature with the cells demonstrating notable proliferation and matrix deposition activity. No adverse inflammatory response was observed in any of the implants from the two groups. Overall, the results emphasize the need to improve the technology in terms of altering the DBM and DCC concentrations, and tailoring the polymer degradation to these concentrations.R01 AR056347Kansas Bioscience Authority Rising Star Awar

    CR1 Knops blood group alleles are not associated with severe malaria in the Gambia

    The Knops blood group antigen erythrocyte polymorphisms have been associated with reduced falciparum malaria-based in vitro rosette formation (putative malaria virulence factor). Having previously identified single-nucleotide polymorphisms (SNPs) in the human complement receptor 1 (CR1/CD35) gene underlying the Knops antithetical antigens Sl1/Sl2 and McC(a)/McC(b), we have now performed genotype comparisons to test associations between these two molecular variants and severe malaria in West African children living in the Gambia. While SNPs associated with Sl:2 and McC(b+) were equally distributed among malaria-infected children with severe malaria and control children not infected with malaria parasites, high allele frequencies for Sl 2 (0.800, 1,365/1,706) and McC(b) (0.385, 658/1706) were observed. Further, when compared to the Sl 1/McC(a) allele observed in all populations, the African Sl 2/McC(b) allele appears to have evolved as a result of positive selection (modified Nei-Gojobori test Ka-Ks/s.e.=1.77, P-value &lt;0.05). Given the role of CR1 in host defense, our findings suggest that Sl 2 and McC(b) have arisen to confer a selective advantage against infectious disease that, in view of these case-control study data, was not solely Plasmodium falciparum malaria. Factors underlying the lack of association between Sl 2 and McC(b) with severe malaria may involve variation in CR1 expression levels

    Was evaluated the growth of the common carp (Cyprinus carpio), lettuce (Lactuca sativa), spinach (Spinacia oleracea) during 12 weeks, incorporating the supply of three alimentary diets. Were used three tanks, in which they have deposited 28 fishes, 20 plants of lettuce, and 20 plants of spinach, for each tank and additionally were planted a control group of these plants. The diets were handled with concentrated food and duckweed (Lemna minor), in concentrations of 100% concentrated, 70% concentrated and 30% duckweed, and 50% concentrated and 50% duckweed. Weekly it was measured the longitude, width, and weight of the fishes, in the plants the heights, number of leaves, and in the water the quantity of ammonium, nitrite, nitrate, pH, and temperature. At the end of the experimental project was evident, that there were no considerable differences in the growth of the fishes, nevertheless the fishes of three tank they had a longitude of 0.24 cm on average with tank 1. The one tank allowed that the number of leaves and height of the lettuce to have a significant difference in relation with the control group and the plants of the spinach were better developed in the control group than in the plants of an aquaponic system.&nbsp;Se evaluó el crecimiento de la carpa común (Cyprinus carpio), lechuga (Lactuca sativa) y espinaca (Spinacia oleracea) durante 12 semanas, incorporando el suministro de tres dietas alimenticias. Se usaron tres tanques, en los cuales se depositaron 28 peces, 20 plantas de lechuga, 20 de espinaca, para cada tanque y adicionalmente se sembraron testigos de las plantas para tener un grupo control. Las dietas se manejaron con concentrado y lenteja de agua (Lemna minor), en concentraciones de 100% concentrado, 70% concentrado y 30% lenteja, y 50% concentrado y 50% lenteja. Cada semana se midió la longitud, ancho y peso de los peces, en las plantas la altura y el número de hojas y en el agua la cantidad de amonio, nitritos, nitratos, pH y temperatura. Al final del proyecto experimental se evidenció, que no hubo diferencias considerables en el crecimiento de los peces, sin embargo, los peces del tanque 3 tuvieron una longitud de 0.24 cm en promedio de diferencia con el tanque 1. El tanque 1 permitió que el número de hojas y altura de la lechuga tuviera unas diferencias significativas en relación con el grupo control y las plantas de espinaca se desarrollaron mejor en el grupo control que en las plantas del sistema acuapónico
