    Influence of bacterial physiology on processing of selenite, biogenesis of nanomaterials and their thermodynamic stability

    We explored how Ochrobactrum sp. MPV1 can convert up to 2.5 mM selenite within 120 h, surviving the challenge posed by high oxyanion concentrations. The data show that thiol-based biotic chemical reaction(s) occur upon bacterial exposure to low selenite concentrations, whereas enzymatic systems account for oxyanion removal when 2 mM oxyanion is exceeded. The selenite bioprocessing produces selenium nanomaterials, whose size and morphology depend on the bacterial physiology. Selenium nanoparticles were always produced by MPV1 cells, featuring an average diameter ranging between 90 and 140 nm, which we conclude constitutes the thermodynamic stability range for these nanostructures. Alternatively, selenium nanorods were observed for bacterial cells exposed to high selenite concentration or under controlled metabolism. Biogenic nanomaterials were enclosed by an organic material in part composed of amphiphilic biomolecules, which could form nanosized structures independently. Bacterial physiology influences the surface charge characterizing the organic material, suggesting its diverse biomolecular composition and its involvement in the tuning of the nanomaterial morphology. Finally, the organic material is in thermodynamic equilibrium with nanomaterials and responsible for their electrosteric stabilization, as changes in the temperature slightly influence the stability of biogenic compared to chemogenic nanomaterials

    A study of the core of the Shapley Concentration: VI. Spectral properties of galaxies

    We present the results of a study of the spectral properties of galaxies in the central part of the Shapley Concentration, covering an extremely wide range of densities, from the rich cluster cores to the underlying supercluster environment. Our sample is homogeneous, in a well defined magnitude range (17=<b_J=<18.8) and contains ~1300 spectra of galaxies at the same distance, covering an area of ~26 sq.deg. These characteristics allowed an accurate spectral classification, that we performed using a Principal Components Analysis technique. This spectral classification, together with the [OII] equivalent widths and the star formation rates, has been used to study the properties of galaxies at different densities: cluster, intercluster (i.e. galaxies in the supercluster but outside clusters) and field environment. No significant differences are present between samples at low density regimes (i.e. intercluster and field galaxies). Cluster galaxies, instead, not only have values significantly different from the field ones, but also show a dependence on the local density. Moreover, a well defined morphology-density relation is present in the cluster complexes, although these structures are known to be involved in major merging events. Also the mean equivalent width of [OII] shows a trend with the local environment, decreasing at increasing densities, even if it is probably induced by the morphology-density relation. Finally we analyzed the mean star formation rate as a function of the density, finding again a decreasing trend (at ~ 3sigma significance level).     PO-332 Genomic landscapes, neoantigen profiles and biological impact of MLH1 inactivation in cancer cells

    Introduction Alterations in DNA repair pathways are thought to fuel tumour progression. Mismatch Repair (MMR) deficient cancers show peculiar biological features such as an indolent progression and a resolute therapeutic response to checkpoint inhibitors. The genomic and biological bases of the peculiar clinical features are poorly understood. Further progress in this area is limited by the paucity of models to study the impact of MMR genes inactivation at the genomic and biological levels. To address this issue we developed a bioinformatic workflow to monitor the neoantigen repertoire induced by inactivation of the Mlh1 gene (a key player of the MMR machinery), in murine cell lines. Material and methods We inactivated Mlh1 throughout the CRISPR-Cas9 technology in CT26 (colon cancer), PDAC (pancreatic cancer) and TSA (breast cancer) murine cell lines. We performed whole exome sequencing (WES) at different time points and then we quantified the amount of mutations (SNVs and indels). We generated a pipeline that characterises the neoantigen repertoire, starting from annotated alterations and the HLA of specific murine strain. In parallel, we inoculated MMR-proficient and -deficient cells in immuno-compromised and -competent mice and monitored their growth. Results and discussions In all pre-clinical models analysed we found a massive increment in the number of non-synonymous alterations (up to 100% increase respect to basal population) after Mlh1 inactivation. Notably, analysis of MMR deficient mouse cells at different time points showed a renewal of mutational profile and consequently an accumulation of predicted neoantigens. We further characterised the SNVs and frameshifts acquired by Mlh1-knockout cells. In agreement with data in human tumours, the fraction of predicted neoantigens derived from frameshifts was higher than the SNV-derived neoantigens. When injected in immuno-compromised mice the Mlh1-knockout cells and their wild type counterpart showed comparable growth. On the contrary, MMR-deficient cells but not their control counterpart grew poorly in immuno-competent mice and responded promptly to treatment with checkpoint inhibitors. Conclusion We find that Mlh1 gene inactivation drives dynamic neoantigen profiles, which can be monitored with an ad hoc bioinformatic pipeline. These analyses provide mechanistic support to understand why MMR deficient cells engage the immune system of the host, foster immune surveillance and tumour control

    Detection of Invasive Borrelia burgdorferi Strains in North-Eastern Piedmont, Italy.

    SummaryFollowing reports of human cases of Lyme borreliosis from the Ossola Valley, a mountainous area of Piemonte, north‐western Italy, the abundance and altitudinal distribution of ticks, and infection of these vectors with Borrelia burgdorferi sensu lato were evaluated. A total of 1662 host‐seeking Ixodes ricinus were collected by dragging from April to September 2011 at locations between 400 and 1450 m above sea level. Additional 104 I. ricinus were collected from 35 hunted wild animals (4 chamois, 8 roe deer, 23 red deer). Tick density, expressed as the number of ticks per 100 m2, resulted highly variable among different areas, ranging from 0 to 105 larvae and from 0 to 22 nymphs. A sample of 352 ticks (327 from dragging and 25 from wild animals) was screened by a PCR assay targeting a fragment of the 16S rRNA gene of B. burgdorferi s.l. Positive samples were confirmed with a PCR assay specific for the 5S‐23S rRNA intergenic spacer region and sequenced. Four genospecies were found: B. afzelii (prevalence 4.0%), B. lusitaniae (4.0%), B. garinii (1.5%) and B. valaisiana (0.3%). Phylogenetic analysis based on the ospC gene showed that most of the Borrelia strains from pathogenic genospecies had the potential for human infection and for invasion of secondary body sites

    Fiasco: a multidetector optimized for semiperipheral heavy ion collisions at Fermi energies

    The Fiasco multidetector is a low-threshold apparatus, optimized for the investigation of peripheral to semi-central collisions in heavy ion reactions at Fermi energies. It consists of three types of detectors. The first detector layer is a shell of 24 position-sensitive Parallel Plate Avalanche Detectors (PPADs), covering about 70% of the forward hemisphere, which measure the velocity vectors of the heavy ðZ\10Þ reaction products. Below and around the grazing angle, behind the most forward PPADs, there are 96 DE–E silicon telescopes (with thickness of 200 and 500 mm; respectively); they are mainly used to measure the energy of the projectile-like fragment and to identify its charge and, via the time-of-flight of the PPADs, also its mass. Finally, behind most of the PPADs there are 158 (or 182, depending on the configuration) scintillation detectors, mostly of the phoswich type, which cover 25–30% of the forward hemisphere; they identify both light charged particles ðZ ¼ 1; 2Þ and intermediate mass fragments ð3pZt20Þ; measuring also their time-of-flight. r 2003 Elsevier B.V.     Ecosistemi per la ricerca Atti Convegno ACNP/NILDE Trieste, 22-23 maggio 2014

    Il secondo convegno congiunto ACNP / NILDE: ecosistemi per la ricerca è stato ospitato dal 22 al 23 maggio 2014 dall’Università di Trieste. Sotto gli auspici della stessa Università di Trieste e degli altri enti di ricerca del Friuli Venezia Giulia1 sono stati affrontati in un’ottica internazionale i temi del rapporto tra cataloghi collettivi e servizi interbibliotecari, e il più generale ambito dei servizi bibliografici per la ricerca scientifica. ACNP e NILDE possono essere considerati un vero e proprio ecosistema. Le biblioteche e i bibliotecari collaborano tra di loro in maniera reciproca e secondo modalità interconnesse, offrendo agli utenti servizi sempre più evoluti e dinamici. Questo ecosistema, essendo aperto, mette i propri servizi a disposizione della ricerca scientifica in senso generale. Il convegno di Trieste ha offerto l’occasione di investigare e proporre soluzioni innovative, interconnessioni e relazioni nuove e più proficue. Il convegno ha presentato alcune rilevanti esperienze internazionali in tema di servizi interbibliotecari e cataloghi collettivi e la prosecuzione di attività che erano state proposte come spunti di ispirazione nel convegno precedente2 inoltre si sono condotte delle riflessioni sulle nuove esigenze dell’utenza. Una ultima parte è dedicata alla illustrazione degli sviluppi tecnici e le prospettive future di ACNP e di NILDE. Hanno partecipato all’evento oltre 200 colleghi italiani e stranieri ed i relatori dei 18 contributi provenivano da Italia, Germania, Austria, Grecia, Slovenia e Stati Uniti. Il carattere di confronto e di condivisione delle esperienze tipico della realtà partecipativa di ACNP e NILDE, è emerso anche in questa occasione nella tavola rotonda - di cui viene riportato un resoconto dettagliato - che ha affrontato il tema della valutazione della ricerca dal punto di vista delle biblioteche. Inoltre, la molto partecipata sessione poster ha ospitato 13 lavori- anch’essi riportati nel volume - incentrati oltre che su ACNP e NILDE anche sul tema ricerca e sistema biblioteca, dando ottimi spunti di partecipazione, dialogo e confronto sulle diverse realtà in cui operiamo

    VIMOS Ultra-Deep Survey (VUDS): Witnessing the assembly of a massive cluster at z ~ 3.3

    International audienceUsing new spectroscopic observations obtained as part of the VIMOS Ultra-Deep Survey (VUDS), we performed a systematic search for overdense environments in the early universe (z> 2) and report here on the discovery of Cl J0227-0421, a massive protocluster at z = 3.29. This protocluster is characterized by both the large overdensity of spectroscopically confirmed members, δgal = 10.5 ± 2.8, and a significant overdensity in photometric redshift members. The halo mass of this protocluster is estimated by a variety of methods to be ~3 × 1014ℳ⊙ at z ~ 3.3, which, evolved to z = 0 results in a halo mass rivaling or exceeding that of the Coma cluster. The properties of 19 spectroscopically confirmed member galaxies are compared with a large sample of VUDS/VVDS galaxies in lower density field environments at similar redshifts. We find tentative evidence for an excess of redder, brighter, and more massive galaxies within the confines of the protocluster relative to the field population, which suggests that we may be observing the beginning ofenvironmentally induced quenching. The properties of these galaxies are investigated, including a discussion of the brightest protocluster galaxy, which appears to be undergoing vigorous coeval nuclear and starburst activity. The remaining member galaxies appear to have characteristics that are largely similar to the field population. Though we find weaker evidence of the suppression of the median star formation rates among and differences in the stacked spectra of member galaxies with respect to the field, we defer any conclusions about these trends to future work with the ensemble of protostructures that are found in the full VUDS sample
