    Cavernous hemangioma of the parotid gland in adults

    Hemangiomas account for 0.4-0.6% of all tumors of the parotid gland and most of them occur in children, never - theless in adults hemangiomas are very rare. We report the case of a 62 year old woman with a mass in the parotid right tail associated with fluctuating swelling episodes unrelated to meals and with a slowly progressive growth. The provisional diagnosis was a pleomorphic adenoma, so a right superficial parotidectomy was performed. During surgery, the macroscopic appearance makes suspect a vascular lesion. The histopathological result was a cavernous hemangioma. The classic clinical presentation of a parotid hemangioma is an intraglandular mass associated or not with skin lesions characterized by reddish macules and/or papules, and a vibration or pulsation when palpating the parotid region. In imaging tests, phleboliths could be observed which are very suggestive of a hemangioma or a vascular malformation. In the absence of these signs, the diagnosis could be difficult, particularly in an adult due to its low prevalence, with about 50 cases reported worldwide. However a hemangioma should be considered in the differential diagnosis of parotid tumors in adults

    Praderas de fanerógamas marinas en la bahía de Cádiz: conservación y gestión

    Comunicación técnicaLa bahía de Cádiz es un humedal costero protegido por figuras de protección nacionales (Parque Natural) e internacionales (LIC, Convención Ramsar) dada su importancia ecológica. La biodiversidad biológica incluye numerosas especies de macroalgas, invertebrados, peces y aves. Un hecho de singular importancia, y poco conocido por las administraciones públicas, es que en este humedal conviven 3 de las 4 especies de fanerógamas marinas, o más estrictamente angiospermas marinas, de Europa. Las praderas de Cymodocea nodosa y Zostera noltii junto con escasos rodales de Zostera marina, proporcionan numerosos servicios ecológicos que están siendo estudiados por el grupo de Estructura y Dinámica de Ecosistemas Acuáticos de la Universidad de Cádiz. Desde hace 15 años hemos desarrollado proyectos tanto nacionales como internacionales en la bahía con el fin de poner en práctica bases ecológicas para la gestión de estas praderas como parte integrante del ecosistema. Estas herramientas abarcan desde un nivel ecofisiológico muy reduccionista (tasas fotosintéticas, incorporación de nutrientes), hasta un nivel más holista (mapas de cobertura, efecto a gran escala de variables ambientales, utilización de información para la estimación de la calidad ecológica de la masa de agua). Las actuaciones desarrolladas incluyen la consolidación de una red de voluntariado (FAMAR) para la recogida y análisis de la información. El trabajo que se presenta resume la información disponible, con series de datos recogidos y analizados durante la última década, para la gestión de las praderas y el estado de conservación de las mismas.     ConBr lectin modulates MAPKs and Akt pathways and triggers autophagic glioma cell death by a mechanism dependent upon caspase-8 activation

    Glioblastoma multiforme is the most aggressive type of glioma, with limited treatment and poor prognosis. Despite some advances over the last decade, validation of novel and selective antiglioma agents remains a challenge in clinical pharmacology. Prior studies have shown that leguminous lectins may exert various biological effects, including antitumor properties. Accordingly, this study aimed to evaluate the mechanisms underlying the antiglioma activity of ConBr, a lectin extracted from the Canavalia brasiliensis seeds. ConBr at lower concentrations inhibited C6 glioma cell migration while higher levels promoted cell death dependent upon carbohydrate recognition domain (CRD) structure. ConBr increased p38MAPK and JNK and decreased ERK1/2 and Akt phosphorylation. Moreover, ConBr inhibited mTORC1 phosphorylation associated with accumulation of autophagic markers, such as acidic vacuoles and LC3 cleavage. Inhibition of early steps of autophagy with 3-methyl-adenine (3-MA) partially protected whereas the later autophagy inhibitor Chloroquine (CQ) had no protective effect upon ConBr cytotoxicity. ConBr also augmented caspase-3 activation without affecting mitochondrial function. Noteworthy, the caspase-8 inhibitor IETF-fmk attenuated ConBr induced autophagy and C6 glioma cell death. Finally, ConBr did not show cytotoxicity against primary astrocytes, suggesting a selective antiglioma activity. In summary, our results indicate that ConBr requires functional CRD lectin domain to exert antiglioma activity, and its cytotoxicity is associated with MAPKs and Akt pathways modulation and autophagy- and caspase-8- dependent cell death.Fil: Wolin, Ingrid A.     The morphometric acclimation to depth explains the long-term resilience of the seagrass Cymodocea nodosa in a shallow tidal lagoon

    Cadiz Bay is a shallow mesotidal lagoon with extensive populations of the seagrass Cymodocea nodosa at intertidal and shallow subtidal elevations. This work aims to understand the mechanisms behind the resilience of this species to gradual sea level rise by studying its acclimation capacity to depth along the shallow littoral, and therefore, to gradual variations in the light environment. To address this objective, these populations have been monitored seasonally over a 10 year period, representing the longest seasonal database available in the literature for this species. The monitoring included populations at 0.4, -0.08 and -0.5 m LAT. The results show that C. nodosa has a strong seasonality for demographic and shoot dynamic properties - with longer shoots and larger growth in summer (high temperature) than in winter (low temperature), but also some losses. Moreover, shoots have different leaf morphometry depending on depth, with small and dense shoots in the intertidal areas (0.4 m) and sparse large shoots in the subtidal ones (-0.08 and 0.5 m). These differences in morphometry and shoot dynamic properties, combined with the differences in shoot density, explain the lack of differences in meadow production balance (i.e. meadow growth - meadow losses) between the intertidal (0.4 m) and the deepest population (-0.5 m), supporting the long term resilience of Cymodocea nodosa in Cadiz Bay. This study contributes to the understanding of the mechanisms behind seagrass stability and resilience, which is particularly important towards predicting the effects of climate change on these key coastal ecosystems, and also highlights the value of continuous long-term monitoring efforts to evaluate seagrass trajectories

    Detection of Taura Syndrome Virus (TSV) in Litopenaeus vannamei in the Philippines

    Farming of Litopenaeus vannamei in the Philippines was seen as a measure to increase shrimp production in the country. Taura Syndrome Virus (TSV) is a viral pathogen reported to be associated with Litopenaeus vannamei and other shrimp species. TSV outbreak impedes the production of Litopenaeus vannamei population and other species susceptible to the virus. Detection of TSV in L. vannamei in the Philippines calls for proper mitigation and appropriate actions for aquaculture and environmental management towards sustainable development. This study embarked on the detection of TSV using a purposive sampling method. Samples showing morphological symptoms of TSV such as redness of the carapace and pleopods and necrotic gill tissue collected from sites in Luzon and Visayas in the Philippines were processed. Viral RNA was extracted from the gills, the central area of infiltration and replication site of the virus. Complementary DNA (cDNA) was synthesized from the RNA templates and were subjected to RT-PCR under specific conditions using TSV specific primers against a positive control. Selected sites in the Philippines were proven to be positive of infection as they showed morphological symptoms and RT-PCR result in having a band at around 200 base pairs (bp). Percentage of prevalence results were obtained: Bulacan with 5 out of 15 samples (33%), Batangas with 7 out of 15 samples (47%), Bohol with 1 out of 15 samples (7%) and Cebu with 2 out of 15 samples (13%). The Batangas samples showed the highest prevalence of 47% (7 out of 15 samples) that were positive of the infection. This study reported the presence of TSV in L. vannamei in the country using morphological and molecular techniques

    O Semiárido brasileiro por muito tempo foi visto pelo prisma do combate à seca, o que culminou em políticas ineficientes para resolução de problemas advindos da pouca oferta de água. Por muito tempo o desenvolvimento de técnicas e o gerenciamento dessas ficou a cargo dos chefes da família, onde as mulheres ficavam de fora do processo. O presente trabalho buscou realizar uma reflexão sobre as tecnologias sociais de convivência com o semiárido, a agroecologia e a questão de gênero. A tecnologia aqui discutida, desenvolvida a partir da parceria de diferentes setores (universidades e movimentos sociais), é a de reuso de água, que busca aumentar a oferta de água para produção agrícola dos moradores do assentamento Hipólito, no município de Mossoró (RN). O reuso de água apresenta benefícios sociais, ambientais e econômicos, que se pretende concretizá-los a partir da implementação dessa prática no referido assentamento. O reuso aqui retratado não se reduz ao aumento da oferta de água, mas e refere a uma proposta de resiliência às atividades dos agricultores e agricultoras familiares e, principalmente, ao envolvimento das mulheres no processo de implementação dessa tecnologia, contribuindo para a emancipação social dessas

    Constitutive gene expression profile segregates toxicity in locally advanced breast cancer patients treated with high-dose hyperfractionated radical radiotherapy

    Breast cancer patients show a wide variation in normal tissue reactions after radiotherapy. The individual sensitivity to x-rays limits the efficiency of the therapy. Prediction of individual sensitivity to radiotherapy could help to select the radiation protocol and to improve treatment results. The aim of this study was to assess the relationship between gene expression profiles of ex vivo un-irradiated and irradiated lymphocytes and the development of toxicity due to high-dose hyperfractionated radiotherapy in patients with locally advanced breast cancer. Raw data from microarray experiments were uploaded to the Gene Expression Omnibus Database (GEO accession GSE15341). We obtained a small group of 81 genes significantly regulated by radiotherapy, lumped in 50 relevant pathways. Using ANOVA and t-test statistical tools we found 20 and 26 constitutive genes (0 Gy) that segregate patients with and without acute and late toxicity, respectively. Non-supervised hierarchical clustering was used for the visualization of results. Six and 9 pathways were significantly regulated respectively. Concerning to irradiated lymphocytes (2 Gy), we founded 29 genes that separate patients with acute toxicity and without it. Those genes were gathered in 4 significant pathways. We could not identify a set of genes that segregates patients with and without late toxicity. In conclusion, we have found an association between the constitutive gene expression profile of peripheral blood lymphocytes and the development of acute and late toxicity in consecutive, unselected patients. These observations suggest the possibility of predicting normal tissue response to irradiation in high-dose non-conventional radiation therapy regimens. Prospective studies with higher number of patients are needed to validate these preliminary results

    Epidemiological and clinical characteristics of Streptococcus tigurinus endocarditis

    Background: Streptococcus tigurinus was recently described as a new streptococcal species within the viridans group streptococci (VGS). The objectives of the present work were to analyse the clinical and microbiological characteristics of S. tigurinus isolated from patients with bacteraemias, to determine the prevalence of S. tigurinus among VGS endocarditis in Spain, and to compare the clinical characteristics and outcomes of endocarditis caused by S. tigurinus and other VGS. Methods: Retrospective nationwide study, performed between 2008 and 2016 in 9 Spanish hospitals from 7 different provinces comprising 237 cases of infective endocarditis. Streptococcal isolates were identified by sequencing fragments of their 16S rRNA, sodA and groEL genes. Clinical data of patients with streptococcal endocarditis were prospectively collected according to a pre-established protocol. Results: Patients with endocarditis represented 7/9 (77.8%) and 26/86 (30.2%) of the bacteraemias caused by S. tigurinus and other VGS, respectively (p < 0.001), in two of the hospital participants. Among patients with streptococcal endocarditis, 12 different Streptococcus species were recognized being S. oralis, S. tigurinus and S. mitis the three more common. No relevant statistical differences were observed in the clinical characteristics and outcomes of endocarditis caused by the different VGS species. Conclusions: In this multicenter study performed in Spain, S. tigurinus showed a higher predilection for the endocardial endothelium as compared to other VGS.     Combined low initial DNA damage and high radiation-induced apoptosis confers clinical resistance to long-term toxicity in breast cancer patients treated with high-dose radiotherapy

Either higher levels of initial DNA damage or lower levels of radiation-induced apoptosis in peripheral blood lymphocytes have been associated to increased risk for develop late radiation-induced toxicity. It has been recently published that these two predictive tests are inversely related. The aim of the present study was to investigate the combined role of both tests in relation to clinical radiation-induced toxicity in a set of breast cancer patients treated with high dose hyperfractionated radical radiotherapy. METHODS. Peripheral blood lymphocytes were taken from 26 consecutive patients with locally advanced breast carcinoma treated with high-dose hyperfractioned radical radiotherapy. Acute and late cutaneous and subcutaneous toxicity was evaluated using the Radiation Therapy Oncology Group morbidity scoring schema. The mean follow-up of survivors (n = 13) was 197.23 months. Radiosensitivity of lymphocytes was quantified as the initial number of DNA double-strand breaks induced per Gy and per DNA unit (200 Mbp). Radiation-induced apoptosis (RIA) at 1, 2 and 8 Gy was measured by flow cytometry using annexin V/propidium iodide. RESULTS. Mean DSB/Gy/DNA unit obtained was 1.70 ± 0.83 (range 0.63-4.08; median, 1.46). Radiation-induced apoptosis increased with radiation dose (median 12.36, 17.79 and 24.83 for 1, 2, and 8 Gy respectively). We observed that those "expected resistant patients" (DSB values lower than 1.78 DSB/Gy per 200 Mbp and RIA values over 9.58, 14.40 or 24.83 for 1, 2 and 8 Gy respectively) were at low risk of suffer severe subcutaneous late toxicity (HR 0.223, 95%CI 0.073-0.678, P = 0.008; HR 0.206, 95%CI 0.063-0.677, P = 0.009; HR 0.239, 95%CI 0.062-0.929, P = 0.039, for RIA at 1, 2 and 8 Gy respectively) in multivariate analysis. CONCLUSIONS. A radiation-resistant profile is proposed, where those patients who presented lower levels of initial DNA damage and higher levels of radiation induced apoptosis were at low risk of suffer severe subcutaneous late toxicity after clinical treatment at high radiation doses in our series.
