Perdiendo en los últimos minutos del encuentro, el equipo atacante incorpora a 6 o más jugadores al ataqueThe aim of this study was to examine the effects of the situational variable match status on corner kicks performance indicators in 95 matches played during the final stages of the 2012 UEFA European Championships and the 2010 FIFA World Cup. Video recordings of the matches were analyzed and coded post-event using notational analysis. Multiple interactions between the performance indicators and match status were analyzed using the Chi-squared automatic interaction detection (CHAID) decision-tree method. The results show that when a corner kick is taken during the last 30 minutes of the match, teams that are losing place 6 or more attackers in the shooting area, while teams that are drawing place 2-5 attackers in this area. In the same situation, teams that are drawing place 1-2 defenders at the goalposts while winning teams place non

In soccer, dead-ball moves are those in which the ball is returned to play from a stationary position following an interruption of play. The aim of this study was to analyse the effectiveness of one such dead-ball move, namely corner kicks, and to identify the key variables that determine the success of a shot or header following a corner, thereby enabling a model of successful corner kicks to be proposed. We recorded 554 corner kicks performed during the 2010 World Cup in South Africa and carried out a univariate, bivariate and multivariate analysis of the data. The results indicated that corners were of limited effectiveness in terms of the success of subsequent shots or headers. The analysis also revealed a series of variables that were significantly related to one another, and this enabled us to propose an explanatory model. Although this model had limited explanatory power, it nonetheless helps to understand the execution of corner kicks in practical terms. As acções de bola parada (ABP) no futebol são todas as situações de jogo que se iniciam depois de uma interrupção regulamentar do mesmo. The parasitic worm,Schistosoma mansoni, expresses unusual fucosylated glycans in a stage-dependent manner that can be recognized by the human innate immune receptor DC-SIGN, thereby shaping host immune responses. We have developed a synthetic approach for mono- and bis-fucosylated LacdiNAc (LDN-F and LDN-DF, respectively), which are epitopes expressed on glycolipids and glycoproteins ofS. mansoni. It is based on the use of monosaccharide building blocks having carefully selected amino-protecting groups, facilitating high yielding and stereoselective glycosylations. The molecular interaction between the synthetic glycans and DC-SIGN was studied by NMR and molecular modeling, which demonstrated that the alpha 1,3-fucoside of LDN-F can coordinate with the Ca2+-ion of the canonical binding site of DC-SIGN allowing for additional interactions with the underlying LDN backbone. The 1,2-fucoside of LDN-DF can be complexed in a similar manner, however, in this binding mode GlcNAc and GalNAc of the LDN backbone are placed away from the protein surface resulting in a substantially lower binding affinity. Glycan microarray binding studies showed that the avidity and selectivity of binding is greatly enhanced when the glycans are presented multivalently, and in this format Le(x)and LDN-F gave strong responsiveness, whereas no binding was detected for LDN-DF. The data indicates thatS. mansonihas developed a strategy to avoid detection by DC-SIGN in a stage-dependent manner by the addition of a fucoside to a number of its ligands. This research was supported by the Netherlands Organization for Scientific Research (NWO; TOP-PUNT grant 718.015.003 to G.-J.B.), the Human Frontier Science Program Organization (HFSP; grant LT000747/2018-C to L.U.), the European Research Council (ERC-2017-AdG, project number 788143-RECGLYC-ANMR to J.J.-B.), the Agencia Estatal Investigacion of Spain (AEI; grant RTI2018-094751-B-C21 to J.J.-B.) and the Severo Ochoa Excellence Accreditation (SEV-2016-0644 to J.J.-B.)

Molecular Recognition of Glycan-Bearing Glycomacromolecules Presented at Membrane Surfaces by Lectins: An NMR View

Lectin–glycan interactions are at the heart of a multitude of biological events. Glycans are usually presented in a multivalent manner on the cell surface as part of the so-called glycocalyx, where they interact with other entities. This multivalent presentation allows us to overcome the typical low affinities found for individual glycan–lectin interactions. Indeed, the presentation of glycans may drastically impact their binding by lectins, highly affecting the corresponding binding affinity and even selectivity. In this context, we herein present the study of the interaction of a variety of homo- and heteromultivalent lactose-functionalized glycomacromolecules and their lipid conjugates with two human galectins. We have employed as ligands the glycomacromolecules, as well as liposomes decorated with those structures, to evaluate their interactions in a cell-mimicking environment. Key details of the interaction have been unravelled by NMR experiments, both from the ligand and receptor perspectives, complemented by cryo-electron microscopy methods and molecular dynamics simulations.M.H. and L.H. thank the DFG for support through the ViroCarb research consortium (HA5950/5-2) and the CeMSA@HHU (Center for Molecular and Structural Analytics @ Heinrich-Heine University) for recording the mass spectrometric and the NMR-spectroscopic data for the structural conformation of the glycomacromolecules and their lipid conjugates. The different possible interacting epitopes have been deeply analysed and characterized, providing evidence of the presence of glycan structures not found in previous MS-based analyses. The interaction of the RBD 13C-labelled glycans with different human lectins, which are expressed in different organs and tissues that may be affected during the infection process, has also been evaluated by NMR. In particular, 15N-labelled galectins (galectins-3, -7 and -8 N-terminal), Siglecs (Siglec-8, Siglec-10), and C-type lectins (DC-SIGN, MGL) have been employed. Complementary experiments from the glycoprotein perspective or from the lectin's point of view have permitted to disentangle the specific interacting epitopes in each case. Based on these findings, 3D models of the interacting complexes have been proposed.publishersversionpublishe
