    Synergistic Antifungal Study of PEGylated Graphene Oxides and Copper Nanoparticles against Candida albicans

This article is an open access article distributed under the terms and conditions of the Creative Commons Attribution (CC BY) license (http://creativecommons.org/licenses/by/4.0/).The coupling reactions of polyethylene glycol (PEG) with two different nano-carbonaceous materials, graphene oxide (GO) and expanded graphene oxide (EGO), were achieved by amide bond formations. These reactions yielded PEGylated graphene oxides, GO-PEG and EGO-PEG. Whilst presence of the newly formed amide links (NH-CO) were confirmed by FTIR stretches observed at 1732 cm−1 and 1712 cm−1, the associated Raman D- and G-bands resonated at 1311/1318 cm−1 and 1584/1595 cm−1 had shown the carbonaceous structures in both PEGylated products remain unchanged. Whilst SEM images revealed the nano-sheet structures in all the GO derivatives (GO/EGO and GO-PEG/EGO-PEG), TEM images clearly showed the nano-structures of both GO-PEG and EGO-PEG had undergone significant morphological changes from their starting materials after the PEGylated processes. The successful PEGylations were also indicated by the change of pH values measured in the starting GO/EGO (pH 2.6–3.3) and the PEGylated GO-PEG/EGO-PEG (pH 6.6–6.9) products. Initial antifungal activities of selective metallic nanomaterials (ZnO and Cu) and the four GO derivatives were screened against Candida albicans using the in vitro cut-well method. Whilst the haemocytometer count indicated GO-PEG and copper nanoparticles (CuNPs) exhibited the best antifungal effects, the corresponding SEM images showed C. albicans had, respectively, undergone extensive shrinkage and porosity deformations. Synergistic antifungal effects all GO derivatives in various ratio of CuNPs combinations were determined by assessing C. albicans viabilities using broth dilution assays. The best synergistic effects were observed when a 30:70 ratio of GO/GO-PEG combined with CuNPs, where MIC50 185–225 μm/mL were recorded. Moreover, the decreased antifungal activities observed in EGO and EGO-PEG may be explained by their poor colloidal stability with increasing nanoparticle concentrations.Peer reviewe

    Detección de hembras en estro en una empresa pecuaria

    Data registered from January 1994 up to December 2008 were collected to evaluate estrual females detection performance at Triángulo 4 Livestock Center in Camagüey province. Seasonal and trend components from the reproduction control system were determined as well as their crossed correlations with detection of females at first estrus and total of estrus. The trend component showed a steady performance from 1994 to 2000, but it decreased from 2001 to 2008. Estrual female number increased from July to November, but dramatically diminished from December to May depending on pasture availability. Seasonal series revealed temporary dependence relations for several lags among reproductive categories, births, and estrual females detection. This evidenced the existence of aseasonal factors. Differences between temporary dependence relations and the expected ones for births and puerperal heifers related to detection of females at first estrus ranged from eight to five months. Non-pregnant and inseminated heifers were out of the expected range. This confirms organizational and control deficiencies concerning reproduction.Para evaluar el comportamiento de la recogida de hembras en estro, en la Empresa Pecuaria Triángulo 4 en la provincia de Camagüey, Cuba, se utilizaron los registros archivados por la brigada de reproducción (enero de 1994diciembre de 2008). Se determinó la tendencia y la estacionalidad de los componentes del sistema de control de la reproducción, y las correlaciones cruzadas entre estos y la recogida de hembras en primer y total de estros, apreciándose comportamiento estable en el período 1994-2000 y decreciente entre el 2001 y 2008 de la tendencia para la recogida de hembras en estro. La recogidas de hembras en estro mostró incremento en el período julio-noviembre y descenso marcado entre diciembre y mayo, asociados con la disponibilidad de los pastos. Para categorías reproductivas afines, reveló que fueron determinantes en el comportamiento reproductivo estacional evaluado. El empleo de series estacionarias permitió revelar la existencia de relaciones de dependencia temporal entre las categorías reproductivas, los nacimientos y la recogida de hembras en estro en varios retardos, lo que evidenció la presencia de otros factores independientes de la estación. Existieron diferencias de ocho y cinco meses entre las relaciones observadas y las esperadas para los nacimientos y las recentinas con relación a las hembras recogidas en el primer estro, las hembras vacías y las inseminadas estuvieron fuera del rango esperado, esto demuestra deficiencias en la organización y control de la reproducción

    Improving the Efficiency of the Bowden Cable Terminal Injection Process for the Automotive Industry

    Control cables transfer force between two separate locations by a flexible mean, and hence, they are important in the automotive industry and many others; their terminals interact with both moving and moved mechanisms, so they must be strong. Cable terminals are commonly made of ZAMAK and are created by injection molding. However, such a production method requires leaving extra material to allow the correct molding, also known as sprues, which are removed later in the process. In this case, the sprues were separating from the terminals in an uncontrolled way. In this work, the cause of sprues separating prematurely from the terminals in a production line is addressed. The whole process was analyzed, and each possible solution was evaluated using process improvement techniques and the Finite Element Method, leading to the best solutions. Molds, mold structures, and auxiliary equipment were improved, resulting in a minimally invasive intervention and remaining compatible with other equipment. Cost analyses were done, indicating an investment return in less than a year. The modification led to a reduction of 62.6% in the sprue mass, while porosity was reduced by 10.2% and 55.9%, corresponding to two terminal models.     Analysis of thermal effects in endoscopic nanocarriers-based photodynamic therapy applied to esophageal diseases

    In this work we propose a predictive model that allows the study of thermal effects produced when the optical radiation interacts with an esophageal or stomach disease with gold nanoparticles embedded. The model takes into account light distribution in the tumor tissue by means of a Monte Carlo method. Mie theory is used to obtain the gold nanoparticles optical properties and the thermal model employed is based on the bio-heat equation.     The cold-sensing ion channel TRPM8 regulates central and peripheral clockwork and the circadian oscillations of body temperature

    [Abstract] Aim: Physiological functions in mammals show circadian oscillations, synchronized by daily cycles of light and temperature. Central and peripheral clocks participate in this regulation. Since the ion channel TRPM8 is a critical cold sensor, we investigated its role in circadian function. Methods: We used TRPM8 reporter mouse lines and TRPM8-deficient mice. mRNA levels were determined by in situ hybridization or RT-qPCR and protein levels by immunofluorescence. A telemetry system was used to measure core body temperature (Tc). Results: TRPM8 is expressed in the retina, specifically in cholinergic amacrine interneurons and in a subset of melanopsin-positive ganglion cells which project to the central pacemaker, the suprachiasmatic nucleus (SCN) of the hypothalamus. TRPM8-positive fibres were also found innervating choroid and ciliary body vasculature, with a putative function in intraocular temperature, as shown in TRPM8-deficient mice. Interestingly, Trpm8-/- animals displayed increased expression of the clock gene Per2 and vasopressin (AVP) in the SCN, suggesting a regulatory role of TRPM8 on the central oscillator. Since SCN AVP neurons control body temperature, we studied Tc in driven and free-running conditions. TRPM8-deficiency increased the amplitude of Tc oscillations and, under dim constant light, induced a greater phase delay and instability of Tc rhythmicity. Finally, TRPM8-positive fibres innervate peripheral organs, like liver and white adipose tissue. Notably, Trpm8-/- mice displayed a dysregulated expression of Per2 mRNA in these metabolic tissues.     Characterising a genetic stronghold amidst pervasive admixture: Morelet’s crocodiles (Crocodylus moreletii) in central Yucatan

    When backcrosses are fertile, interbreeding between endangered taxa can lead to the admixture of gene pools under threat. One such case pertains to the Mesoamerican crocodile Crocodylus moreletii, a species which shows strong signatures of both recent hybridisation and historic intogression with the American crocodile C. acutus across large parts of its range. In the present paper, we use RAD-seq derived SNPs (4980 nuclear and seven mtDNA loci) to demonstrate that C. moreletii populations inhabiting the region of Calakmul in central Yucatan (Mexico) are rather unaffected by hybridization, despite being surrounded by coastal areas where pervasive admixture has previously been documented. All (based on fastSTRU CTU RE) and 96% (based on NGSadmix) of 84 genotyped individuals from 18 sampled waterbodies (locally termed aguadas) were free from nuclear introgression of C. acutus DNA at at threshold of 0.95. Seven individuals (8%) possessed a C. acutus mtDNA haplotype, five of which were derived from two adjacent, rather peripheral aguadas. Spatial inferences based on a DAPC and fineRADstructure further showed that the region of Calakmul is inhabited by three genetic clusters spanning across a set of distinct aguadas each. Taken together, our findings reveal that central Yucatan contains the currently largest documented stronghold of C. moreletii populations only marginally affected by introgression, which has major implications for the conservation management of this important flagship species

    Episodic molecular outflow in the very young protostellar cluster Serpens South

    The loss of mass from protostars, in the form of a jet or outflow, is a necessary counterpart to protostellar mass accretion. Outflow ejection events probably vary in their velocity and/or in the rate of mass loss. Such `episodic´ ejection events have been observed during the Class 0 protostellar phase (the early accretion stage), and continue during the subsequent class I phase that marks the first one million years of star formation. Previously observed episodic-ejection sources were relatively isolated; however, the most common sites of star formation are clusters. Outflows link protostars with their environment and provide a viable source of turbulence that is necessary for regulating star formation in clusters, but it is not known how an accretion-driven jet or outflow in a clustered environment manifests itself in its earliest stage. This early stage is important in establishing the initial conditions for momentum and energy transfer to the environment as the protostar and cluster evolve. Here we report that an outflow from a very young class 0 protostar, at the hub of the very active and filamentary Serpens South protostellar cluster, shows unambiguous episodic events. The 12CO (J=2-1) emission from the protostar reveals 22 distinct features of outflow ejecta, the most recent having the highest velocity. The outflow forms bipolar lobes --- one of the first detectable signs of star formation --- which originate from the peak of 1-mm continuum emission. Emission from the surrounding C18O envelope shows kinematics consistent with rotation and an infall of material onto the protostar. The data suggest that episodic accretion-driven outflow begins in the earliest phase of protostellar evolution, and that the outflow remains intact in a very clustered environment, probably providing efficient momentum transfer for driving turbulence. Fil: Plunkett, Adele L. .
