    Time to stop blaming parents: A case study of parents’ and children’s experiences of family life and in marginalised communities with implications for parenting interventions, educational welfare policy and practice

    The inclusion of the voices of parents and children from marginalised communities is quite a departure from traditional rigid assumptions in parenting research. This study sought to explore parents' experience of family life and participation in the universal roll-out of Parents Plus Children's Programme (PPCP), as well as children's perspectives on how they navigate the different settings of home, school and community in a Dublin urban area of low socio-economic status (SES). Applying a community psychology perspective and the lens of Bronfenbrenner’s (1979) bio-ecological model, the primary research question posed was how can parents’ and children’s perspectives and experiences of family life and PPCP in marginalised communities inform formal educational welfare policy and practice through Delivering Equality of Opportunity in Schools (DEIS). Employing a case study approach, qualitative research methods were used to gather data from seven parents (via semi-structured interviews) and eight children (via mosaic-arts approach). This study found that their ability to parent effectively was either supported, hindered or disrupted by people, community influences (e.g. crime) and situations (e.g. adequate housing), often outside of their control. While PPCP was a support to the majority of parents, where isolation and marginalisation was felt most profoundly in the community, PPCP couldn’t address the larger social issues impacting on parenting practices. For children, how their families were perceived in the school, especially Traveller families, influenced their school experiences. Compared to their settled peers, Traveller children stated that they did not like school, nor the school their parents, and that it would be one of the first places they would change in their community. The findings of this study has implications for how parenting interventions, educational welfare policy and practice, as well as all-of-government policy, can better support families in marginalised communities

    Investigating epidermogenesis in a human skin equivalent model

    Skin is the largest, and arguably, the most important organ of the body. It is a complex and multi-dimensional tissue, thus making it essentially impossible to fully model in vitro in conventional 2-dimensional culture systems. In view of this, rodents or pigs are utilised to study wound healing therapeutics or to investigate the biological effects of treatments on skin. However, there are many differences between the wound healing processes in rodents compared to humans (contraction vs. re-epithelialisation) and there are also ethical issues associated with animal testing for scientific research. Therefore, the development of skin equivalent (HSE) models from surgical discard human skin has become an important area of research. The studies in this thesis compare, for the first time, native human skin and the epidermogenesis process in a HSE model. The HSE was reported to be a comparable model for human skin in terms of expression and localisation of key epidermal cell markers. This validated HSE model was utilised to study the potential wound healing therapeutic, hyperbaric oxygen (HBO) therapy. There is a significant body of evidence suggesting that lack of cutaneous oxygen results in and potentiates the chronic, non-healing wound environment. Although the evidence is anecdotal, HBO therapy has displayed positive effects on re-oxygenation of chronic wounds and the clinical outcomes suggest that HBO treatment may be beneficial. Therefore, the HSE was subjected to a daily clinical HBO regime and assessed in terms of keratinocyte migration, proliferation, differentiation and epidermal thickening. HBO treatment was observed to increase epidermal thickness, in particular stratum corneum thickening, but it did not alter the expression or localisation of standard epidermal cell markers. In order to elucidate the mechanistic changes occurring in response to HBO treatment in the HSE model, gene microarrays were performed, followed by qRT-PCR of select genes which were differentially regulated in response to HBO treatment. The biological diversity of the HSEs created from individual skin donors, however, overrode the differences in gene expression between treatment groups. Network analysis of functional changes in the HSE model revealed general trends consistent with normal skin growth and maturation. As a more robust and longer term study of these molecular changes, protein localisation and expression was investigated in sections from the HSEs undergoing epidermogenesis in response to HBO treatment. These proteins were CDCP1, Metallothionein, Kallikrein (KLK) 1 and KLK7 and early growth response 1. While the protein expression within the HSE models exposed to HBO treatment were not consistent in all HSEs derived from all skin donors, this is the first study to detect and compare both KLK1 and CDCP1 protein expression in both a HSE model and native human skin. Furthermore, this is the first study to provide such an in depth analysis of the effect of HBO treatment on a HSE model. The data presented in this thesis, demonstrates high levels of variation between individuals and their response to HBO treatment, consistent with the clinical variation that is currently observed

    Understanding Youth Worker Job Stress

    Work in out-of-school learning programs can be stressful, and job stress may have cascading effects for the children and youth that attend. Fortunately, workplace supports can help decrease this stress. In this study, we aimed to understand how youth workers’ personal and work-related demands as well as supports predict on-the-job stress. We used multilevel modeling to investigate the demands and supports of a sample of 111 youth workers nested in 25 programs. Results suggested that job stress systematically varies at the program level. We found that stress at home and a negative staffing climate is associated with higher stress and the presence of supervisor support is associated with lower staff stress. Supervisor support, in particular, likely can play a key role in decreasing youth worker stress. We discuss implications for training supervisors and structuring programs to support staff and to ultimately foster more positive out-of-school program experiences for the children and youth that attend

    Three decades of change in the Skagerrak coastal ecosystem, shaped by eutrophication and coastal darkening

    Global coastal ecosystems are under accelerating pressure from human activities and climate change. In this study we explore a long-term time series (mostly 1990–2016) from major Norwegian rivers, together with coastal time series from the Norwegian Skagerrak coast. The aims are to: 1) analyse long-term trends in riverine loadings to Skagerrak, changes in coastal water quality and pelagic and benthic species composition, and 2) to describe the relationships between human drivers (eutrophication and coastal darkening) and community structure of benthic communities. Riverine discharge and transport increased by 23–85% over the time period, corresponding to a 40–78% increase in concentrations of suspended particulate material in coastal waters and reduced surface salinity, connected to the reported coastal darkening of coastal waters. There was a worsening in ecological status for hard-bottom benthic communities (0–30 m) and a reduction in the lower growth depth limit of several macroalgae species. A structural shift in the community composition from macroalgae towards increased abundance of animals was found to be related to coastal darkening and reduced eutrophication. The concentration of coastal inorganic nutrients (DIN, PO4) declined by 27–36%, in response to management efforts to reduce eutrophication in European regional seas. Results indicate that reduced eutrophication was related to increased ecological status of the deep soft-bottom community (350 m), with a reduction in opportunistic polychaetes and an increase in filter feeding bivalves. This work highlights how climate change and other human-induced changes in a boreal ecosystem intensifies land-ocean interactions, and calls for more adaptive monitoring, where traditional water quality programs and policies need to evolve iteratively as new information emerges and the major drivers of the systems change.publishedVersio

    Identifying Causal Genes at the Multiple Sclerosis Associated Region 6q23 Using Capture Hi-C.

    BACKGROUND: The chromosomal region 6q23 has been found to be associated with multiple sclerosis (MS) predisposition through genome wide association studies (GWAS). There are four independent single nucleotide polymorphisms (SNPs) associated with MS in this region, which spans around 2.5 Mb. Most GWAS variants associated with complex traits, including these four MS associated SNPs, are non-coding and their function is currently unknown. However, GWAS variants have been found to be enriched in enhancers and there is evidence that they may be involved in transcriptional regulation of their distant target genes through long range chromatin looping. AIM: The aim of this work is to identify causal disease genes in the 6q23 locus by studying long range chromatin interactions, using the recently developed Capture Hi-C method in human T and B-cell lines. Interactions involving four independent associations unique to MS, tagged by rs11154801, rs17066096, rs7769192 and rs67297943 were analysed using Capture Hi-C Analysis of Genomic Organisation (CHiCAGO). RESULTS: We found that the pattern of chromatin looping interactions in the MS 6q23 associated region is complex. Interactions cluster in two regions, the first involving the rs11154801 region and a second containing the rs17066096, rs7769192 and rs67297943 SNPs. Firstly, SNPs located within the AHI1 gene, tagged by rs11154801, are correlated with expression of AHI1 and interact with its promoter. These SNPs also interact with other potential candidate genes such as SGK1 and BCLAF1. Secondly, the rs17066096, rs7769192 and rs67297943 SNPs interact with each other and with immune-related genes such as IL20RA, IL22RA2, IFNGR1 and TNFAIP3. Finally, the above-mentioned regions interact with each other and therefore, may co-regulate these target genes. CONCLUSION: These results suggest that the four 6q23 variants, independently associated with MS, are involved in the regulation of several genes, including immune genes. These findings could help understand mechanisms of disease and suggest potential novel therapeutic targets

    Expanding the role of young people in research: Towards a better understanding of their lives

    The participation of young people in the research process can be empowering for the participants and valuable for the research outcomes. This paper presents the methods used and outcomes of involving youth in the development of priorities for the Health Behaviour in School-aged Children study in Ireland.Two participative workshops were conducted with young people: the first focused on identifiying what is important for people to understand about young people’s lives; the second served to expand and prioritise the themes identified, for the purpose of developing questionnaire items. Following idea generation, discussion and voting four themes emerged as priorities. These were; ‘Diversity and Individuality’, ‘Independence’, ‘Mental Health’ and ‘Bullying’.The process enabled young people to prioritise dimensions of their lives that deserved further quantitative research attention. The findings of the study identify the potential of such a methodology to transform the path of any research project concerning young people

    Multicentre evaluation of the Boehringer Mannheim/Hitachi 917 analysis system

    The new selective access analysis system BM/Hitachi 917 was evaluated in an international multicentre study, mainly according to the ECCLS protocol for the evaluation of analysers in clinical chemistry. Forty-three different analytes, covering 56 different methods enzymes, substrates, electrolytes, specific proteins, drugs and urine applications were tested in seven European clinical chemistry laboratories. Additionally, the practicability of the BM/ Hitachi 917 was tested according to a standardized questionnaire. Within-run CVs (median of 3 days) for enzymes, substrates and electrolytes were <2% except for creatine-kinase MB isoform and lipase at low concentration. For proteins, drugs and urine analytes the within-run CVs were < 4% except for digoxin and albumin in urine. Between-day median CVs were generally < 3% for enzymes, substrates and electrolytes, and < 6% for proteins, drugs and urine analytes, except for lipase, creatine kinase and MB isoform, D-dimer, glycosylated haemoglobin, rheumatoid factors, digoxin, digitoxin, theophylline and albumin in urine in some materials. Linearity was found according to the test specifications or better and there were no relevant effects seen in drift and carry-over testing. The interference results clearly show that also for the BM/Hitachi 917 interference exists sometimes, as could be expected because of the chemistries applied. It is a situation that can be found in equivalent analysers as well. The accuracy is acceptable regarding a 95–105% recovery in standard reference material, with the exception of the creatinine Jaffé method. Most of the 160 method comparisons showed acceptable agreement according to our criteria: enzymes, substrates, urine analytes deviation of slope ± 5%, electrolytes ± 3%, and proteins and drugs ± 10%. The assessment of practicability for 14 groups of attributes resulted in a grading of one–three scores better for the BM/Hitachi 917 than the present laboratory situation. In conclusion, the results of the study showed good analytical performance and confirmed the usefulness of the system as a consolidated workstation in medium-sized to large clinical chemistry laboratories

    Discoveries from a Near-infrared Proper Motion Survey using Multi-epoch 2MASS Data

    We have conducted a 4030-square-deg near-infrared proper motion survey using multi-epoch data from the Two Micron All-Sky Survey (2MASS). We find 2778 proper motion candidates, 647 of which are not listed in SIMBAD. After comparison to DSS images, we find that 107 of our proper motion candidates lack counterparts at B-, R-, and I-bands and are thus 2MASS-only detections. We present results of spectroscopic follow-up of 188 targets that include the infrared-only sources along with selected optical-counterpart sources with faint reduced proper motions or interesting colors. We also establish a set of near-infrared spectroscopic standards with which to anchor near-infrared classifications for our objects. Among the discoveries are six young field brown dwarfs, five "red L" dwarfs, three L-type subdwarfs, twelve M-type subdwarfs, eight "blue L" dwarfs, and several T dwarfs. We further refine the definitions of these exotic classes to aid future identification of similar objects. We examine their kinematics and find that both the "blue L" and "red L" dwarfs appear to be drawn from a relatively old population. This survey provides a glimpse of the kinds of research that will be possible through time-domain infrared projects such as the UKIDSS Large Area Survey, various VISTA surveys, and WISE, and also through z- or y-band enabled, multi-epoch surveys such as Pan-STARRS and LSST.Comment: To appear in the September 2010 issue of The Astrophysical Journal, Supplement Serie
