    Determination of Gonyautoxin-4 in Echinoderms and Gastropod Matrices by Conversion to Neosaxitoxin Using 2-Mercaptoethanol and Post-Column Oxidation Liquid Chromatography with Fluorescence Detection

    Paralytic Shellfish Toxin blooms are common worldwide, which makes their monitoring crucial in the prevention of poisoning incidents. These toxins can be monitored by a variety of techniques, including mouse bioassay, receptor binding assay, and liquid chromatography with either mass spectrometric or pre- or post-column fluorescence detection. The post-column oxidation liquid chromatography with fluorescence detection method, used routinely in our laboratory, has been shown to be a reliable method for monitoring paralytic shellfish toxins in mussel, scallop, oyster and clam species. However, due to its high sensitivity to naturally fluorescent matrix interferences, when working with unconventional matrices, there may be problems in identifying toxins because of naturally fluorescent interferences that co-elute with the toxin peaks. This can lead to erroneous identification. In this study, in order to overcome this challenge in echinoderm and gastropod matrices, we optimized the conversion of Gonyautoxins 1 and 4 to Neosaxitoxin with 2-mercaptoethanol.     Influence of Different Shellfish Matrices on the Separation of PSP Toxins Using a Postcolumn Oxidation Liquid Chromatography Method

    The separation of PSP toxins using liquid chromatography with a post-column oxidation fluorescence detection method was performed with different matrices. The separation of PSP toxins depends on several factors, and it is crucial to take into account the presence of interfering matrix peaks to produce a good separation. The matrix peaks are not always the same, which is a significant issue when it comes to producing good, reliable results regarding resolution and toxicity information. Different real shellfish matrices (mussel, scallop, clam and oyster) were studied, and it was seen that the interference is not the same for each individual matrix. It also depends on the species, sampling location and the date of collection. It was proposed that separation should be accomplished taking into account the type of matrix, as well as the concentration of heptane sulfonate in both solvents, since the mobile phase varies regarding the matrix. Scallop and oyster matrices needed a decrease in the concentration of heptane sulfonate to separate GTX4 from matrix peaks, as well as dcGTX3 for oysters, with a concentration of 6.5 mM for solvent A and 6.25 mM for solvent B. For mussel and clam matrices, interfering peaks are not as large as they are in the other group, and the heptane sulfonate concentration was 8.25 mM for both solvents.     Orientación al mercado y renovación de espacios urbanos en destinos culturales: el caso de Sevilla

    La pérdida de competitividad de algunos destinos culturales consolidados les ha obligado a plantear el reforzamiento de los atractivos de su oferta de un modo integral, donde lo determinante no sólo es renovar el conjunto de productos del destino, sino realzar de un modo global las condiciones del escenario territorial en que el visitante disfruta su experiencia, y los atributos ambientales y paisajísticos que conforman el espacio turístico. El presente trabajo describe las acciones emprendidas por los responsables turísticos y urbanísticos de la ciudad de Sevilla durante la primera década del siglo XXI para conseguir la renovación de un destino ya maduro, estructuradas en torno al principio de orientación al mercado, autenticidad y sostenibilidad, como elementos clave en la búsqueda de fórmulas que incorporen valor a su oferta turística.On purpose to solve the loss of competitiveness of some consolidated cultural tourism destinations, it ́s necessary thinking in a depth strategic way with the aim to help in their progressive repositioning making-process. Improving competitiveness implies acting and understanding on the reinforcement of the main tourism ́s appeals as a hole, not only in the renewal of the set of tourism destination ́s product or services, but enhance the global conditions of territorial stage in which visitor enjoys his experience. The paper shows the policy developed by touristic and town-planning policymaker in Seville during the first decade of the 21st century.     Spanish researchers’ perceived difficulty writing research articles for English-medium journals: the impact of proficiency in English versus publication experience

    Previous quantitative studies suggest that the burden researchers who use English as an additional language perceive when writing research articles (RAs) for publication in English (as L2) is 24% greater than the burden they perceive when they write RAs for publication in their L1. It remains unclear precisely which aspects of research article (RA) writing in English present these writers with the greatest challenge and just why they perceive this increase in difficulty. A structured questionnaire comprising thirty-seven questions about researchers’ publication experiences in scientific journals in English and in Spanish was designed and sent out to all (n = 8,794) Spanish postdoctoral researchers at one research-only institution and four universities in Spain, yielding responses from 1,717 researchers. Our first results show that the discussion is the section that is perceived as more difficult to write for English-medium journals, across the four broad knowledge areas in a way that cannot be fully explained by their lower level of proficiency in English (as L2). This article proposes the rhetorical transfer hypothesis as a possible explanation for their additional difficulty. Our results also reveal that their increased perceived difficulty writing RA discussions in English (as L2) does not decrease noticeably until Spanish researchers report high or very high levels of proficiency in English (as L2) for academic or general purposes or have published on average at least 37 RAs as corresponding author in English-medium journals over the last ten years. Implications for English for Academic Purposes (EAP) research and pedagogy are discussed

    Comparative Analysis of Muscle Transcriptome between Pig Genotypes Identifies Genes and Regulatory Mechanisms Associated to Growth, Fatness and Metabolism.

    Iberian ham production includes both purebred (IB) and Duroc-crossbred (IBxDU) Iberian pigs, which show important differences in meat quality and production traits, such as muscle growth and fatness. This experiment was conducted to investigate gene expression differences, transcriptional regulation and genetic polymorphisms that could be associated with the observed phenotypic differences between IB and IBxDU pigs. Nine IB and 10 IBxDU pigs were slaughtered at birth. Morphometric measures and blood samples were obtained and samples from Biceps femoris muscle were employed for compositional and transcriptome analysis by RNA-Seq technology. Phenotypic differences were evident at this early age, including greater body size and weight in IBxDU and greater Biceps femoris intramuscular fat and plasma cholesterol content in IB newborns. We detected 149 differentially expressed genes between IB and IBxDU neonates (p < 0.01 and Fold-Change > 1. 5). Several were related to adipose and muscle tissues development (DLK1, FGF21 or UBC). The functional interpretation of the transcriptomic differences revealed enrichment of functions and pathways related to lipid metabolism in IB and to cellular and muscle growth in IBxDU pigs. Protein catabolism, cholesterol biosynthesis and immune system were functions enriched in both genotypes. We identified transcription factors potentially affecting the observed gene expression differences. Some of them have known functions on adipogenesis (CEBPA, EGRs), lipid metabolism (PPARGC1B) and myogenesis (FOXOs, MEF2D, MYOD1), which suggest a key role in the meat quality differences existing between IB and IBxDU hams. We also identified several polymorphisms showing differential segregation between IB and IBxDU pigs. Among them, non-synonymous variants were detected in several transcription factors as PPARGC1B and TRIM63 genes, which could be associated to altered gene function. Taken together, these results provide information about candidate genes, metabolic pathways and genetic polymorphisms potentially involved in phenotypic differences between IB and IBxDU pigs associated to meat quality and production traits

    Effect of parameter calculation in direct estimation of the Lyapunov exponent in short time series

    The literature about non-linear dynamics offers a few recommendations, which sometimes are divergent, about the criteria to be used in order to select the optimal calculus parameters in the estimation of Lyapunov exponents by direct methods. These few recommendations are circumscribed to the analysis of chaotic systems. We have found no recommendation for the estimation of λ starting from the time series of classic systems. The reason for this is the interest in distinguishing variability due to a chaotic behavior of determinist dynamic systems of variability caused by white noise or linear stochastic processes, and less in the identification of non-linear terms from the analysis of time series. In this study we have centered in the dependence of the Lyapunov exponent, obtained by means of direct estimation, of the initial distance and the time evolution. We have used generated series of chaotic systems and generated series of classic systems with varying complexity. To generate the series we have used the logistic map

    La intensa competencia a la que se enfrentan los destinos culturales provoca que éstos se centren en la búsqueda de fórmulas que sirvan para generarles una ventaja competitiva, habiéndose señalado la autenticidad como uno de los elementos que en mayor medida inciden sobre la satisfacción y lealtad del visitante. Si bien son múltiples los elementos que pueden afectar a la autenticidad percibida, la gestión del patrimonio histórico que poseen las urbes cobra especial relevancia tanto desde el ámbito académico como de la gestión pública. Que los destinos turísticos empleen una estrategia de desarrollo sostenible permitiría poner en valor su patrimonio histórico y activar el triple componente sectorial que componen los Paisajes Urbanos (turismo, ocio y comercio), y que se constituye en la principal fuente de actividad económica de gran parte de los municipios. Es por ello, que el presente trabajo se dirige a analizar el impacto que presente la conservación y mantenimiento de los Pasajes Urbanos sobre la autenticidad percibida por el visitante y de ésta sobre las actividades que desarrolla una vez que se encuentra en el destino.The intense competition faced by cultural destinations causes them to focus on the search for formulas that will generate a competitive advantage, having noted the authenticity as one of the elements that have the greatest impact on visitor satisfaction and loyalty. While there are multiple elements that can affect the perceived authenticity, heritage management that the cities have special relevance from both academic and public management. Destinations that employ a sustainable development strategy would develop its historical heritage and enable triple sectoral component comprising the Urban Landscapes (tourism, leisure and commerce), and has become the main source of economic activity for much municipalities.     Identifying Spanish researchers’ needs for training in English for Research Publication Purposes: Methodological aspects of a large-scale online survey

    This paper mainly aims to report on the survey method employed in Phase 1 of the ENEIDA project to fulfil the following aims: a) locating those researchers at the five institutions participating in the project who might be interested in receiving ERPP training and in collaborating in subsequent phases of the project; b) identifying their specific needs vis-à-vis ERPP; and c) providing a context for future studies of Spanish-English intercultural rhetoric for research publication purposes. The paper also offers an overall characterisation of the informants to our survey, discusses some of the results to assess the relevance and viability of further phases of the project, and evaluates the database thus created. 24 researchers were first interviewed in-depth from one research-only institution and two universities. Interview responses were then used to design a structured questionnaire comprising thirty-seven questions related to both English and Spanish for research publication purposes. The questionnaire was piloted with 200 researchers selected from the total population of staff with doctorates (8,794) at the three institutions mentioned above, plus another two universities. The questionnaire was then sent out to the total population, yielding responses from 1717 researchers, which are kept in the ENEIDA Database. The findings suggest high levels of interest in ERPP amongst participants in that not only were 64% of respondents interested in future ERPP training, but also in that 96% of them were willing to receive information about how to participate in subsequent phases of the project. It is hoped that the information contained in the ENEIDA Database will allow us to: a) carry out precise needs analyses of specific groups of informants (e.g. according to specific disciplines); b) carry out in-depth studies of how relevant factors affect writing for research publication purposes of Spanish researchers, and c) design multiple case studies of their difficulties writing for research publication purposes grounded in sound researc
