Accumulation and elimination dynamics of the hydroxybenzoate saxitoxin analogues in mussels Mytilus galloprovincialis exposed to the toxic marine Dinoflagellate Gymnodinium catenatum
Paralytic shellfish poisoning (PSP) is a severe food-borne illness, caused by the ingestion of seafood containing paralytic shellfish toxins (PST), which are naturally produced by marine dinoflagellates and accumulate in shellfish during algae blooms. Novel PST, designated as hydroxybenzoate analogues (also known as GC toxins), was relatively recently discovered in Gymnodinium catenatum strains worldwide. However, to date, there have been no studies examining their accumulation in shellfish. In this study, mussels (Mytilus galloprovincialis) were exposed to G. catenatum for five days and then exposed to a non-toxic diet for 24 h, to investigate the toxin's accumulation/elimination dynamics. As determined by UHPLC-HILIC-MS/MS, the hydroxybenzoate analogues, GC1 to GC6, comprised 41% of the algae toxin profile and only 9% in mussels. Elimination of GC toxins after 24 h was not evident. This study highlights that a relevant fraction of PST in mussels are not routinely analysed in monitoring programs and that there is a need to better understand the toxicological potential of the hydroxybenzoate analogues, in order to properly address the risk of G. catenatum blooms.Centre for Environment, Fisheries and Aquaculture Science - contract code DP402;

The Increasing environmental concern in the modern society, which leads to even the states themselves set policy goals, not change the economic inequality of the same. This environmental-biased attitude implies that companies have to adjust to not only the legal framework but above all to improve the image and degree of confidence to the stakeholders, which may also be a way of improving business performance.
In order to meet a sustainable environmental policy it is necessary to characterize the situation or environmental quality (environmental assessment) because it serves as a basis for understanding and further analysis of the environmental situation of a company, assisting in the design of actions made in order to prevent, control and correct any environmental problems that eventually may occur.
In this study the methodology includes the following steps: defining the criteria for environmental assessment; collecting and organizing information about the canning industry in question; environmental assessment and the identification and assessment of impacts despite industry subject matter is not obliged to comply with the environmental impact assessment and environmental permitting, namely the provisions of paragraph i) of section 7.4 of Annex III of the Regional Legislative Decree 30/2010/A, from November 15th, since it does not have enough capacity to produce 75 tons of finished products per day. The methodology also included the verification of compliance with legal and regulatory requirements, as well as the identification of environmental aspects of higher relevance. This approach allowed the preparation of an environmental synthesis that encompasses proposals for mitigation measures. The survey identified in particular water consumption; production of waste water; consumption and energy production; emissions to water and air; transport, waste and noise as the main environmental concerns..
Noise and atmospheric emissions associated to steam generators were the environmental subjects with higher environmental significance considering their impacts, with areas where noise exceeded the exposure limit and the atmospheric emissions of nitrogen oxides is also above the threshold, as well as particles, nitrogen oxides and sulfur dioxide parameters, all exceeding the minimum mass threshold limit values as the mass flow rates defined in the Ordinance 80/2006, from 23th January
Perception of organizational politics and the effect of authenticity and affective commitment on deviance and task performance in contact centers
This study aimed to understand employees’ reactions to organizational politics in Contact Centers. Drawing from a sample of 187 supervisor-employee dyads, we studied the relationship between employees’ perceptions of organizational politics and supervisor-rated task performance and deviance, and mediation effects by authenticity at work and affective commitment. Results indicate that workers tend to react to workplace politics with deviant behavior and worse task performance. We found that the relationship between perceived politics and task performance was mediated by authenticity. The relationship between perceived politics and supervisor-rated deviance was mediated by affective commitment to the organization. Implications for management are discussed
Formation, structure and rheological properties of biopolymers systems
Palavras-chave: proteína, polissacarídeo, interação, gel, emulsão, biopolímero, solução, reologia, separação de fases, CLSM, ¿rheo-SALS¿Abstract: Biopolymers are widely used in dairy products, canned foods, bakery products, salad dressings, beverages, sauces, soups and other processed foodstuffs to improve textural characteristics, flavour and shelf life. The aim of the present Ph.D. thesis was to study the protein-polysaccharide interactions in aqueous systems (pH 7.0) or acidified gels. It was studied seven different biopolymers used in the food industry, being two proteins and five polysaccharides. Soy protein isolate (SPI), Na-caseinate and xanthan were studied in aqueous solutions and gels systems, while locust bean gum (LBG), gellan, Na-alginate and k-carrageenan were studied only in aqueous solutions. The protein-polysaccharide interactions were evaluated as following: 1) microstructures ¿ by using confocal (CLSM), atomic force and phase contrast microscopes and rheo-SALS (small angle light scattering); 2) macrostructures, evaluated trough small- and large-strain rheology under shear or compression; 3) chemical analysis - in order to determine the kind of interaction forces that maintained the gels structure. The results on the physical properties of pure biopolymers solutions revealed that the proteins showed Newtonian behaviour, while the polysaccharides showed either Newtonian or non-Newtonian behaviour depending on its type and solution concentration. The annealing temperature of xanthan solutions and the addition of sucrose caused a reduction on the solution elasticity. It was observed that sucrose affected the rheological properties over all frequency range studied for initially anisotropic or biphasic xanthan solutions. It was proposed a new model based on BST (Blatz, Sharda, Tschoegl, 1974) equation to predict a great number of mechanical properties, which revealed a very good fit to the data. The slower the acidification by GDL (glucone-delta-lactone) it was observed a more interconnected and harder network in Na caseinate and SPI pure gels. The increase of the breaking stress or strain was obtained by increasing the SPI concentration in pure gels, by adding xanthan or by increasing the protein concentration in gels made with xanthan. In another hand, the addtion of xanthan let to weaker heat-induced SPI gels, which was attributed to the electrostatic interactions between xanthan and the b-7S subunit of SPI. In mixed protein-polysaccharide solutions, the results indicated that k-carrageenan was more compatible with both soy protein and Nacaseinate than alginate. However, the Na-caseinate ¿ Na-alginate mixture showed a greater capacity to form water-in-water emulsions than Na-caseinate - carrageenan system as observed by rheo-SALS and CLSM. In addition, the system with SPI and Na-alginate also formed emulsions but with a gelified protein dispersed phase. The interfacial tension of systems containing gellan was very low resulting in homogeneous mixtures. How much cultural events are "worth" for host communities: case of faz música Lisboa 2014
This paper studies the economic and social impact of Faz Música Lisboa on the segment
of society that feels from any direct or indirect consequence due to its existence. A
qualitative research based on surveys and interviews is made to retract a list of the
benefits and costs that each stakeholder perceives. Relying on the quantifiable variables, it
is performed a cost-benefit analysis to measure how much the event is “worth” for the
community. I conclude this is a viable project, as it brings a positive net benefit to the
society, value that could increase with a higher institutional support
Bivalves play a marked role in coastal marine ecosystems, impacting primary
productivity with a strong top-down control on phytoplankton communities. In
addition, they are an important food source for higher trophic levels, including
humans. However, bivalves can be impaired by several external factors, either
biotic or abiotic, natural or anthropogenic. Exposure to these stressors, coupled
with intensive farming, raised the need for adequate management of the wild
populations and farmed production. To improve the difficult balance between
ecological and socio-economic interests, it must be investigated how bivalves
cope with external stressors. For this reason, the effects caused by the
following stressors were studied in the present thesis: (i) harmful algal blooms
(HABs) and (ii) climate change drivers in the seawater, such as warming (W)
and acidification (A). HABs are increasing in frequency, intensity, persistence
and geographic distribution, but are ambiguous regarding their ecological
impacts and physiological effects on bivalves. A strong relationship is emerging
between HABs and climate change drivers, what may affect both shellfisheries
and wild populations. Hence, the objectives of this work are: 1) To determine
whether native and invasive species cope with exposure to HABs differently,
providing new insights on species-specificities and ecosystem functioning
fragilities in the presence of invasive species and HABs; 2) To evaluate how
commercially valuable bivalve species cope with simultaneous exposure to
several climate change drives and HABs, providing new insights on how
environmental changes affects toxicokinetics, physiological and genotoxic
bivalves responses, under HABs; 3) To assess, marine biotoxins
bioaccessibility, contributing for new information relevant for risk assessment.
Different approaches were used to achieve these goals. Complementary field
and laboratory works were performed to assess toxicokinetics patterns of
marine biotoxins in native vs. invasive bivalve species. Bivalves were exposed
to toxic blooming algae species under climate change scenarios in laboratory controlled conditions to assess toxicokinetics as well as physiological and
genotoxic responses. Naturally contaminated bivalves were used to investigate
their role as vectors of marine biotoxins to humans through in vitro digestion
methodology.
Relevant new data were obtained regarding the effects of several marine
biotoxins in bivalves. Assessing accumulation of regulated and non-regulated
biotoxins in native (Ruditapes decussatus) and invasive (R. philippinarum) clam
species from Aveiro Lagoon revealed that higher toxin content, particularly
regarding OA-group, the most abundant and frequent toxins in the Portuguese
coast, is reached by native species. Accumulation of lower toxin levels by
invasive clam may then favour farmers interest for their production. The kinetics
and genotoxicity study in native and invasive clams reinforced the existence of
species-specific behaviours. Exposure to the dinoflagellate Prorocentrum lima, an OA and dinophysistoxin-1 producer, increases the pressure over native
clams, with higher toxin accumulation and genetic damage, as well as early
and increased induction of DNA repair activity. Invasive clams, on the other
hand, are better adapted to cope with these challenges.
Investigation of the effects of combined exposure to multiple stressors revealed
that W, A and HABs alter the accumulation/elimination dynamics of Paralytic
Shellfish Poisoning (PSP) toxins in mussels (Mytilus galloprovincialis). Lower
accumulation levels and slower elimination rates were observed. The predicted
climate change scenarios and exposure to HABs may then lead to lower
contamination levels but to longer harvesting closures. Simultaneous exposure
to altered environmental conditions and HABs also had significant impacts in
the antioxidant system and DNA integrity, resulting in an organ-specific
modulation of the antioxidant response, increasing genetic damage and
preventing/retarding DNA damage repair. However, the DNA damage observed
seems to be non-oxidative. While the investigation of factors enhancing the
elimination of toxins was not a main objective of this study, it was shown that
acidification might promote PSP toxins elimination in mussels.
Finally, the bioaccessibility studies revealed a significant reduction in the OA group content available after in vitro digestion, suggesting an overestimation of
exposure to these biotoxins and an over-conservative approach in safety levels
definition and risk assessment studies. This was the first study assessing
bioaccessibility of OA-group toxins, pointing to in vitro digestion as a promising
tool to obtain accurate data regarding toxin ingestion from bivalves to the
consumer. Overall, the complexity of the relation between HABs, bivalves and
the environmental factors to consider in marine biotoxins management is
increasing, and only continuous and extensive monitoring of environmental,
biological and anthropogenic conditions may allow for a healthier balance
between environmental and socio-economic interests.
simultânea a alterações climáticas e HABs, focando o efeito destas
dano no ADN.
prolongados. A exposição simultânea a condições ambientais alteradas e
O dano encontrado no ADN parece, no
acessibilidade destes compostos no organismo humano.
The Influence of the Structural Pillars of Digital Academic Entrepreneurship on University Students’ Entrepreneurial Intention
In the dynamic, complex and emerging environment which characterises the phenomenon of digital academic entrepreneurship, accelerating in the post Covid-19 period, digital technology has become essential in educational processes. Setting out from the general lines of the Theory of Planned Behaviour and Sociotechnical Theory, this research aimed to analyse the influence of digital academic entrepreneurship on university students’ entrepreneurial intention. Specifically, four studies were carried out from the following research question: What are the influences of the pillars of digital academic entrepreneurship on university students’ entrepreneurial intention? The four specific studies aimed to: (1) Map studies on digital entrepreneurship, digital technology and digital transformation, and propose a structural model of analysis for digital academic entrepreneurship; (2) Propose a conceptual model of analysis to allow assessment of the Soft Skills dimension; (3) Propose a conceptual model of analysis to allow assessment of the Hard Skills dimension, and (4) Analyse the influence of the pillars (Hard Skills and Soft Skills-.) of digital academic entrepreneurship on university students’ entrepreneurial intention and how that relation can be mediated by the theory of planned behaviour.
The methodological strategy adopted here was based on an exploratory study, as this type of study aims to explain the reasons, ideas and motivations underlying the phenomenon of digital academic entrepreneurship and its influence on university students’ entrepreneurial intention. To validate the structural model proposed in this research, a quantitative approach was also adopted, using a final sample of 761 Portuguese (51%) and Brazilian (49%) university students. The systematic literature review (SLR) method was also used to understand the extent and depth of the existing body of work.
Data analysis was carried out through exploratory and confirmatory factor analysis using structural equation models. The results allow the conclusion that the pillars of digital academic entrepreneurship (Hard and Soft Skills) have a direct and indirect positive influence on university students’ entrepreneurial intention. The empirical evidence also suggests that the direct effects of Hard Skills (Management tools, Digital processes and Digital products) and Soft Skills (Individual characteristics; Cultural characteristics and Knowledge sharing) are slightly less than the total indirect effects between these pillars and entrepreneurial intention, when considering the mediations of Entrepreneurial Attitude and Perceived Behavioural Control, and even when inserting Subjective Norms as a mediator with these dimensions. These results raise a number of pertinent questions that warrant deeper reflection, not just in the scientific and academic community, but also among political and government entities. This research also provides important evidence to expand digital entrepreneurial education in higher education institutions (HEI), as it contributes to greater understanding of intention and the factors leading to formation of that intention among higher education students. Other implications for theory and practice are also presented.
The clinical gut: Examining the cognitive processes underlying therapist’s intuitive judgments in the psychotherapy session
Understanding the underlying cognitive processes of clinical judgments may be critical to optimizing clinical care and focusing future research on clinical psychology and psychotherapy. The present dissertation brings insights from decision science to the clinical psychology realm to achieve the overarching goal of understanding clinical intuitions about those expressing psychological suffering and symptoms of mental illness. Specifically, this research studies clinical intuitions and the tendency to confirm hypotheses in different clinical and person perception domains. In three empirical chapters, we present three research lines based on experimental studies. By investigating the cognitive processes underlying the generation and testing of psychological disorder diagnoses, the adjustment of trait inferences and personality impressions according to contextual information, and the judgment of competence and the predictions of future suffering in conditions of unbearable suffering and euthanasia decisions, this research demonstrated how task characteristics favored confirmatory strategies and explored under which conditions this tendency was reduced or eliminated. This dissertation further explores the implications of using confirmatory and disconfirmatory processing strategies in the context of psychotherapy and proposes new directions for further investigation. This research provided empirical evidence we hope will stimulate the area of clinical decision making and to bridge the gap between research in clinical psychology and psychotherapy practice and training.
