    Culex quinquefasciatus vitellogenesis: morphological and biochemical aspects

    The vitellogenic process in Culex quinquefasciatus, which is triggered by a blood meal, involves the synthesis, distribution and storage of the nutrients necessary for embryo development. The fat body of an adult female Cx. quinquefasciatus revealed two cell types: large trophocytes and small, eosinophilic, "oenocyte-like" cells, which show no morphological changes throughout the gonotrophic cycle. Trophocytes, which only begin to synthesise vitellogenin (Vg) 12 h post-blood meal (PBM), undergo a series of morphological changes following engorgement. These changes include the expansion of the rough endoplasmic reticulum (RER) and Golgi complex, which are later destroyed by autophagosomes. At 84 h PBM, trophocytes return to their pre-engorgement morphology. The ovarian follicles of non-blood-fed Cx. quinquefasciatus contain a cluster of eight undifferentiated cells surrounded by follicular epithelium. After engorgement, the oocyte membrane facing the perioocytic space increases its absorptive surface by microvilli development; large amounts of Vg and lipids are stored between 24 and 48 h PBM. Along with yolk storage in the oocyte, follicular cells exhibit the development of RER cisternae and electron-dense granules begin to fill the perioocytic space, possibly giving rise to endochorion. Later in the gonotrophic cycle, electron-dense vesicles, which are possible exochorion precursors, fuse at the apical membrane of follicular cells.     Theoretical simulation of different 3D separator geometries for lithium-ion batteries

    The battery separator is an essential component of batteries and affects their cycling performance. In this work, the effect of different 3D geometries of the battery separator on battery performance was studied keeping the same volume. It was observed that the different geometries affect the cycling performance, the best geometry being the perforated one that cycled up to 90 C. The cycling performance is affected by parameters such as the separator thickness and the electrolyte volume.     Solid polymer electrolytes based on lithium bis(trifluoromethanesulfonyl)imide/poly(vinylidene fluoride -co-hexafluoropropylene) for safer rechargeable lithium-ion batteries

    The increasing use of electronic portable systems and the consequent energy demand, leads to the need to improve energy storage systems. According to that and due to safety issues, high-performance non-flammable electrolytes and solid polymer electrolytes (SPE) are needed.SPE containing different amounts of lithium bis(trifluoromethanesulfonyl) imide (LiTFSI) into a poly(vinylidene fluoride-co-hexafluoropropylene), PVDF-HFP, polymer matrix have been prepared by solvent casting. The addition of LiTFSI into PVDF-HFP allows to tailor thermal, mechanical and electrical properties of the composite.In particular, the ionic conductivity of the composites increases with LiTFSI content, the best ionic conductivities of 0.0011 mS/cmat 25 degrees C and 0.23 mS/cmat 90 degrees C were obtained for the PVDF-HFP/LiTFSI composites with 80 wt % of LiTFSI.This solid electrolyte allows the fabrication of Li metallic/SPE/C-LiFePO4 half-cells with a discharge capacity of 51.2 mAh/ g at C/20. Further, theoretical simulations show that the discharge capacity value depends on the lithium concentration and percentage of free ions and is independent of the solid polymer electrolyte thickness. On the other hand, the voltage plateau depends on the SPE thickness.     Evaluation of antifungal activity and potential application as fluorescent probes of indolenine and benzo[e]indole-based squarylium dyes

    The antifungal performance and the possible use as fluorescent probes of a series of squarylium dyes derived from indolenine and benzo[e]indole previously synthesized was evaluated. Some photophysical properties were performed in ethanol and phosphate buffer, and the type of aggregates form in phosphate buffer was analyzed. Using the 1,3-diphenylisobenzofuran assay, a qualitative assessment of the capacity of dyes to produce singlet oxygen after irradiation was performed. Regarding the antifungal activity, this was studied through a broth microdilution assay using Saccharomyces cerevisiae PYCC 4072 as a biological model. The effect of irradiation of the dyes, with an appropriate light emitting diode system, on the antifungal activity was also evaluated, and it was verified that some of the dyes improve their activity after irradiation. Using fluorescence microscopy techniques, the colocalization of dyes in S. cerevisae cells was investigated and it was possible to verify that some of the squarylium dyes with a barbituric moiety in the four-membered central ring stained and accumulated preferentially in the mitochondrial web and perinuclear membrane of the cells.     Perinatal factors associated with early deaths of preterm infants born in Brazilian Network on Neonatal Research centers

    OBJETIVO: Avaliar os fatores perinatais associados ao óbito neonatal precoce em prematuros com peso ao nascer entre 400 e 1.500 g. MÉTODOS: Coorte prospectiva e multicêntrica dos nascidos vivos com idade gestacional de 23 a 33 semanas e peso de 400-1.500 g, sem malformações em oito maternidades públicas terciárias universitárias entre junho de 2004 e maio de 2005. As características maternas e neonatais e a morbidade nas primeiras 72 horas de vida foram comparadas entre os prematuros que morreram ou sobreviveram até o sexto dia de vida. As variáveis perinatais associadas ao óbito neonatal precoce foram determinadas por regressão logística. RESULTADOS: No período, 579 recém-nascidos preencheram os critérios de inclusão. O óbito precoce ocorreu em 92 (16%) neonatos, variando entre as unidades de 5 a 31%, e tal diferença persistiu controlando-se por um escore de gravidade clínica (SNAPPE-II). A análise multivariada para o desfecho óbito neonatal intra-hospitalar precoce mostrou associação com: idade gestacional de 23-27 semanas (odds ratio - OR = 5,0; IC95% 2,7-9,4), ausência de hipertensão materna (OR = 1,9; IC95% 1,0-3,7), Apgar 0-6 no 5º minuto (OR = 2,8; IC95% 1,4-5,4), presença de síndrome do desconforto respiratório (OR = 3,1; IC95% 1,4-6,6) e centro em que o paciente nasceu. CONCLUSÃO: Importantes fatores associados ao óbito neonatal precoce em prematuros de muito baixo peso são passíveis de intervenção, como a melhora da vitalidade fetal ao nascer e a diminuição da incidência e gravidade da síndrome do desconforto respiratório. As diferenças de mortalidade encontradas entre os centros apontam para a necessidade de identificar as melhores práticas e adotá-las de maneira uniforme em nosso meio.OBJECTIVE:To evaluate perinatal factors associated with early neonatal death in preterm infants with birth weights (BW) of 400-1,500 g. METHODS: A multicenter prospective cohort study of all infants with BW of 400-1,500 g and 23-33 weeks of gestational age (GA), without malformations, who were born alive at eight public university tertiary hospitals in Brazil between June of 2004 and May of 2005. Infants who died within their first 6 days of life were compared with those who did not regarding maternal and neonatal characteristics and morbidity during the first 72 hours of life. Variables associated with the early deaths were identified by stepwise logistic regression. RESULTS: A total of 579 live births met the inclusion criteria. Early deaths occurred in 92 (16%) cases, varying between centers from 5 to 31%, and these differences persisted after controlling for newborn illness severity and mortality risk score (SNAPPE-II). According to the multivariate analysis, the following factors were associated with early intrahospital neonatal deaths: gestational age of 23-27 weeks (odds ratio - OR = 5.0; 95%CI 2.7-9.4), absence of maternal hypertension (OR = 1.9; 95%CI 1.0-3.7), 5th minute Apgar 0-6 (OR = 2.8; 95%CI 1.4-5.4), presence of respiratory distress syndrome (OR = 3.1; 95%CI 1.4-6.6), and network center of birth. CONCLUSION: Important perinatal factors that are associated with early neonatal deaths in very low birth weight preterm infants can be modified by interventions such as improving fetal vitality at birth and reducing the incidence and severity of respiratory distress syndrome. The heterogeneity of early neonatal rates across the different centers studied indicates that best clinical practices should be identified and disseminated throughout the country.Ministério da Saúd

    Marine and coastal environmental education in the context of global climate changes - synthesis and subsidies for ReBentos (Coastal Benthic Habitats Monitoring Network)

    As changes in coastal and marine environments are expected to negatively affect Brazilian ecosystems, the importance of Marine Environmental Education (MEE) comes to the fore. However, so far only 32 contributions related to this issue have been published in Brazil. The MEE workgroup of ReBentos aims at promoting EE and the communication of marine ecological research to the scholastic public as a whole, as well as to groups which exert an influence on general perception, such as the media, politicians, and scientists. This paper presents an overview of the initiatives of MEE in Brazil, with emphasis on the ReBentos projects and guidelines. The conceptual background of action is based on the Rio'92 Treaty on Environmental Education, thereby implying an MEE with Transdisciplinar, emancipatory and reflexive characteristics, directed to changes in values, principles and attitudes. During the period 2011 to 2015, 10 projects were developed from Alagoas to Santa Catarina States, involving the development, implementation and testing through scientific research of 16 MEE activity-models. The didactic material subsequently produced comprised three books and 21 book-chapters. A public of around 6,500 Conservation Unit visitors, 250 public school teachers and 800 high school students have been impacted to date. To act as monitors and multipliers, 250 undergraduate students and professionals were trained. Research project evaluation generated the publication of nine papers. As a further step, the need for protocol elaboration for each model is placed in evidence, in order to direct and facilitate future initiatives.A importância da educação ambiental marinha (EAM) vem tomando relevância à medida que aumenta a expectativa de impactos nos ecossistemas brasileiros ocasionados por mudanças nos ambientes costeiros. Entretanto, apenas 32 contribuições sobre esse assunto foram publicadas no Brasil. O grupo de trabalho em EAM da ReBentos objetiva promover a comunicação da pesquisa ecológica marinha para o público escolar como um todo, bem como a grupos com influência na percepção comum, como a mídia, políticos e cientistas. Este trabalho apresenta uma visão das iniciativas em EAM no Brasil, com ênfase nos projetos e diretrizes da ReBentos. A base conceitual de ação é o Tratado de Educação Ambiental da Rio 92, implicando em um ensino com características transdisciplinares, reflexivas e emancipatórias, dirigidas a mudanças em valores, princípios e atitudes. Durante o período de 2011 a 2015, 10 modelos de atividade foram desenvolvidos, de Alagoas a Santa Catarina, envolvendo sua concepção, implementação e teste por meio de pesquisa científica. O material didático produzido compreendeu três livros e 21 capítulos de livros. Um público total ao redor de 5500 visitantes de UCs, 250 professores de escolas públicas e 800 estudantes foi impactado. Como monitores e multiplicadores, foram treinados 250 estudantes de graduação e profissionais. Projetos de avaliação de pesquisa geraram nove trabalhos científicos. Como uma próxima iniciativa, é salientada a necessidade de elaboração de protocolos para cada modelo, a fim de direcionar e facilitar futuras iniciativas

    Molecular epidemiology and antimicrobial susceptibility of enterococci recovered from Brazilian intensive care units

    ABSTRACT: We studied the antimicrobial resistance and the molecular epidemiology of 99 enterococcal surveillance isolates from two hospitals of Brasília, Brazil. Conventional biochemical tests were used to identify the enterococcal species and the disk diffusion method was used to determine their resistance profiles. Enterococcus faecalis (76%) and E. faecium (9%) were the most prevalent species. No enterococci showed the vanA or vanB vancomycin resistance phenotypes or genotypes. Only the intrinsically resistant species E. gallinarum (n=2) and E. casseliflavus (n=3) harbored the vancomycin-resistance genes vanC1 and vanC2/3, respectively. We found E. faecalis isolates with high-level resistance to gentamicin (22%) and streptomycin (8%) and both E. faecalis and E. faecium isolates with resistance to more than two antimicrobials (84% and 67%, respectively). Nine E. faecalis isolates (12%) were resistant to ampicillin; the minimal inhibitory concentration (MIC) values were 16µg/mL (n=6) and 32µg/mL (n=3). Among these ampicillin-resistant E. faecalis, seven were also resistant to gentamicin, ciprofloxacin, rifampin, penicillin, chloramphenicol, tetracycline and erythromycin. Pulsed-field gel electrophoresis classified those isolates in three different genotypes, suggesting dissemination of genetically related ampicillin-resistant E. faecalis strains among different patients
