    Micropropagação e Germinação das Espécies Endémicas da Ilha da Berlenga

    O decréscimo do número de exemplares de algumas das espécies vegetais endémicas da ilha da Berlenga é resultado do impacto exercido pelo Homem, sendo um cenário preocupante. A conservação destas espécies ainda é possível a partir de técnicas de cultura in vitro. O objetivo principal deste trabalho foi o estabelecimento de protocolos que permitam a multiplicação por micropropagação ou por germinação de sementes, de três espécies endémicas da ilha da Berlenga, Pulicaria microcephala, Armeria berlengensis e Herniaria berlengiana, que se encontram categorizadas como espécies em risco de extinção ou criticamente ameaçadas. No estabelecimento de novos cultivos, foi determinada a taxa de sucesso na desinfeção das três espécies, sendo que os melhores resultados obtidos foram de 24,4% para P. microcephala, 15,79% para A. berlengensis e 18,2% para H. berlengiana, com diferentes processos de desinfeção. Avaliou-se igualmente, a taxa de sucesso na germinação de P. microcephala, que variou entre 6,19 e 73,02%. Em A. berlengensis obtiveram-se valores inferiores de germinação (20%), facto que, em conjunto com uma taxa de fecundidade de 3,5%, corrobora a necessidade da micropropagação como estratégia para a recuperação desta espécie. Foram realizados ainda ensaios de aclimatização com P. microcephala e A. berlengensis e estudadas as taxas de sucesso dos mesmos. Em P. microcephala a taxa de sucesso chegou aos 90,90%, quando realizada com uma luminosidade de 5790 lux, no entanto, e apesar da taxa de sucesso é apenas de 63,64% em plantas que foram aclimatizadas no exterior com luminosidade, humidade e temperatura não controladas, natural, estas apresentaram maior quantidade de flores. As plantas de A. berlengensis de menores dimensões registaram os melhores resultados de aclimatização, com 80% de sucesso. Foi ainda estudada a adição ao meio de cultura para o desenvolvimento de P. microcephala, de hormonas de crescimento e NaCl, bem como testados diferentes valores de pH. Este estudo demonstra que a micropropagação, seguida de aclimatização, é a melhor solução até agora encontrada para contribuir na conservação destas três espécies

    Star-Shaped Gold Nanoparticles as Friendly Interfaces for Protein Electrochemistry: the Case Study of Cytochrome c

Direct electron transfer between cytochrome c and nanostar-coated pyrolytic graphite electrodes was observed with the protein in solution. The electrochemical response was improved at nanostars functionalized with a 1 : 1 mixture of 11-mercaptoundecanoic acid and 4-mercaptobenzoic acid in comparison with gold nanospheres coated with a similar functionalization. Further immobilization of cytochrome c on pyrolytic graphite while conjugated with the same nanostars guaranteed the maintenance of the protein's native properties, whereas direct adsorption on the bare or nanostar-modified electrodes resulted in an altered conformational state.     Nursing Professional Self-Concept: A Scoping Review Protocol

    Nurses are considered one of the largest professional groups in healthcare, generating positive health outcomes for people at various stages of life. A significant impact on the construction of the professional self—or professional self-concept—is considered to exist through the educational process, influenced by factors such as the family and societal expectations often presented by teachers, tutors, and peers. Improving professional self-concept in nursing can offer specific gains in personal, relational, social, and interpersonal communication skills, favoring evolution in the academic and clinical path. This scoping review aims to map the literature related to the state of knowledge regarding professional self-concept in nursing. This scoping review will follow JBI recommendations with the PCC mnemonic and report its findings through PRISMA-ScR using a specific instrument made by the researchers. Providing healthcare complying with high scientific standards requires the professional to have enough self-confidence in his work and skills. Surface-enhanced Raman scattering (SERS)-based immunosensors are especially suited for this purpose. Here, we present a highly sensitive and selective SERS immunoassay, demonstrating for example the detection of horseradish peroxidase (HRP), in a sandwich format. The strength of our biosensor lies in merging: (i) SERS-immunotags based on gold nanostars, allowing exceptional intense SERS from attached Raman probes, covalent attachment of anti-HRP antibodies by a simple chemical method providing exceptional antigen binding activity; (ii) the ease of preparation of the capture platform from a regenerated cellulose-based hydrogel, a transparent material, ideal for microfluidics applications, with low background fluorescence and Raman signal, particularly suited for preserving high activity of the covalently bound anti-HRP antibodies. The sandwich complexes formed were characterised by atomic force microscopy, and by scanning electron microscopy coupled with electron diffraction spectroscopy; and (iii) the robustness of the simple Classical Least Squares method for SERS data analysis, resulting in superior discrimination of SERS signals from the background and much better data fitting, compared to the commonly used peak integral method. Our SERS immunoassay greatly improves the detection limits of traditional enzyme-linked immunosorbent assay approaches, and its performance is better or comparable to those of existing SERS-based immunosensors. Our approach successfully overcomes the main challenges of application at point-of-care, including increasing reproducibility, sensitivity, and specificity, associated with an environmentally friendly and robust design. Also, the proposed design withstands several cycles of regeneration, a feature absent in paper-SERS immunoassays and this opens the way for sensitive multiplexing applications on a microfluidic platform.preprintpublishe

    Avaliação da qualidade da atenção básica em 37 municípios do centro-oeste paulista: características da organização da assistência

    The State of São Paulo, Brazil, has a broad network of primary health care services (PHCS) whose organizational profile is heterogeneous and not well known. This study analyzes the organization of PHCS in 37 municipalities located in the central-west region of São Paulo, as the first part of a broader project that evaluates the quality of these services. This is a cross-sectional study conducted through a structured questionnaire answered by the local managers and health teams. The questions addressed institutional characteristics and the organization and management of the work process. The questionnaires were sent to 131 UBS (Primary Health Care Units) distributed across 37 municipalities. There were responses from 113 units (87%) located in 32 municipalities (86.4%). Of the overall number of units, 57 (50%) are traditional UBS, 26 (22.8%) are family health units, and 31 (27.2%) are mixed. Most services (62%, 70/113) do not work with catchment areas delimited in a planned way. Services are divided between those who perform 70% to 100% of scheduled medical consultations (37.6%, 41/109) and those who perform 70% to 100% of non-scheduled consultations (39.4%, 43/109). Sixty-five units (63.7%, 65/102) do not have community-based committees. Based on the collected data, it is possible to discuss the main features of the health programs, procedures and activities carried out by PHCS. The predominant organizational profiles reveal deficiencies of structure and process in relation to the guidelines issued by SUS (Brazil's National Health System). The development of self-assessment instruments allows the teams to appropriate their work in a critical way and to carry out new technological arrangements to improve the quality of the care provided.No Estado de São Paulo, existe uma rede extensiva de serviços de Atenção Básica (AB) com perfil organizacional heterogêneo e pouco conhecido. Este estudo objetiva caracterizar a organização dos serviços de AB em 37 municípios do Centro-oeste paulista, como primeira etapa de um projeto de avaliação da qualidade desses serviços. Trata-se de um estudo transversal conduzido mediante questionário estruturado, autorrespondido pelos gerentes e equipes locais, com questões que abordam características institucionais e de organização e gerência do trabalho. Esses questionários foram enviados para 131 UBS, distribuídas em 37 municípios. Obteve-se resposta de 113 unidades (87%) localizadas em 32 municípios (86,4%). Do total de unidades, 57 (50%) são UBS tradicionais, 26 (22,8%), Unidades de Saúde da Família, e 31, (27,2%) organizadas segundo formas mistas. A maioria dos serviços (62%, 70/113) não trabalha com área de abrangência delimitada de modo planejado. Os serviços se polarizam entre aqueles que realizam entre 70 e 100% de consultas médicas agendadas (37,6%, 41/109), e aqueles que realizam entre 70 e 100% de não agendadas (39,4%, 43/109). Não possuem conselhos locais organizados 65 unidades (63,7%, 65/102). Os dados coletados permitem discutir as características dos principais programas, procedimentos e ações realizados pelos serviços. Os perfis organizacionais predominantes apontam a presença de deficiências de estrutura e processo em relação às diretrizes do SUS. O desenvolvimento de instrumentos de autoavaliação permite que as equipes se apropriem, de forma crítica, de seu trabalho, e possam elaborar novos arranjos tecnológicos para melhoria da qualidade

    A epidemia do HIV no Brasil é concentrada em populações-chave. Organizações não governamentais (ONGs) que atuam em defesa dos direitos da população LGBT e de pessoas vivendo com HIV/aids podem contribuir para o desenvolvimento de políticas de prevenção. O objetivo deste estudo é avaliar o envolvimento em ONGs e analisar sua associação com a participação individual em ações de educação em saúde, testagem e prevenção às infecções sexualmente transmissíveis (IST) e ao HIV, como parte de um estudo nacional de vigilância biológica e comportamental entre homens que fazem sexo com homens (HSH). Trata-se de estudo transversal utilizando respondent driven sampling (RDS) em 12 cidades brasileiras. A magnitude da associação do envolvimento em ONGs com cada ação foi avaliada pela estimativa de odds ratio por meio de regressão logística por amostragem complexa, considerando cada cidade como um estrato e ponderando pelo estimador de Gile. Foi estimada a proporção relativa atribuída ao envolvimento em ONGs para cada evento avaliado. Dentre 4.176 participantes, a maioria tinha menos de 25 anos (56,5%) e baixo nível econômico (56,7%). Um quarto dos HSH referiu se envolver em ONGs, que foi significativamente associado com as ações avaliadas: receber preservativo e gel lubrificante, participar de palestra, receber material educativo e aconselhamento em IST, conhecimento de profilaxia pós-exposição (PEP) e profilaxia pré-exposição (PrEP), testagem para sífilis e HIV, ter aceitação do autoteste e saber onde realizar teste para HIV. ONGs têm um papel histórico na resposta à epidemia de HIV no Brasil e, apesar de terem sofrido significativa redução de recursos nos últimos anos, mantêm relevante atuação nas ações de saúde pública.The HIV epidemic in Brazil is concentrated in key populations. Nongovernmental organizations (NGOs) working in defense of LGBT rights and the rights of persons living with HIV/AIDS can contribute to the development of prevention policies. The current study’s objectives are to assess involvement in NGOs and analyze the association with individual participation in health education, testing, and STI and HIV prevention activities as part of a national study on biological and behavioral surveillance in men who have sex with men (MSM). This is a cross-sectional study using respondent-driven sampling in 12 Brazilian cities. The magnitude of the association between involvement in NGOs and each activity was assessed with estimation of odds ratios via logistic regression with complex sampling, considering each city as a stratum and weighting by the Gile estimator. The relative proportion attributed to involvement in NGOs was estimated for each event. Among 4,176 participants, the majority were under 25 years of age (56.5%) and with low socioeconomic status (56.7%). One-fourth of MSM reported being involved in NGOs, which was associated significantly with the target activities: receiving free condoms and lubricant gel, participating in talks on STIs, receiving educational materials and counseling on STIs, prior knowledge of postexposure prophylaxis (PEP) and pre-exposure prophylaxis (PrEP), testing for syphilis and HIV, acceptance of HIV self-testing, and knowing where to get an HIV test. NGOs play a historical role in the response to the HIV epidemic in Brazil, and despite a significant cutback in resources in recent years, they have maintained relevant work in public health activities.La epidemia de VIH en Brasil se concentra en poblaciones clave.     Evidence for Reductive Genome Evolution and Lateral Acquisition of Virulence Functions in Two Corynebacterium pseudotuberculosis Strains

PLoS ONE. 2011;6(4): e18551.Background: Corynebacterium pseudotuberculosis, a Gram-positive, facultative intracellular pathogen, is the etiologic agent of the disease known as caseous lymphadenitis (CL). CL mainly affects small ruminants, such as goats and sheep; it also causes infections in humans, though rarely. This species is distributed worldwide, but it has the most serious economic impact in Oceania, Africa and South America. Although C. pseudotuberculosis causes major health and productivity problems for livestock, little is known about the molecular basis of its pathogenicity. Methodology and Findings: We characterized two C. pseudotuberculosis genomes (Cp1002, isolated from goats; and CpC231, isolated from sheep). Analysis of the predicted genomes showed high similarity in genomic architecture, gene content and genetic order. When C. pseudotuberculosis was compared with other Corynebacterium species, it became evident that this pathogenic species has lost numerous genes, resulting in one of the smallest genomes in the genus. Other differences that could be part of the adaptation to pathogenicity include a lower GC content, of about 52%, and a reduced gene repertoire. The C. pseudotuberculosis genome also includes seven putative pathogenicity islands, which contain several classical virulence factors, including genes for fimbrial subunits, adhesion factors, iron uptake and secreted toxins. Additionally, all of the virulence factors in the islands have characteristics that indicate horizontal transfer. Conclusions: These particular genome characteristics of C. pseudotuberculosis, as well as its acquired virulence factors in pathogenicity islands, provide evidence of its lifestyle and of the pathogenicity pathways used by this pathogen in the infection process.     MAMMALS IN PORTUGAL : A data set of terrestrial, volant, and marine mammal occurrences in P ortugal

    Mammals are threatened worldwide, with 26% of all species being includedin the IUCN threatened categories. This overall pattern is primarily associatedwith habitat loss or degradation, and human persecution for terrestrial mam-mals, and pollution, open net fishing, climate change, and prey depletion formarine mammals. Mammals play a key role in maintaining ecosystems func-tionality and resilience, and therefore information on their distribution is cru-cial to delineate and support conservation actions. MAMMALS INPORTUGAL is a publicly available data set compiling unpublishedgeoreferenced occurrence records of 92 terrestrial, volant, and marine mam-mals in mainland Portugal and archipelagos of the Azores and Madeira thatincludes 105,026 data entries between 1873 and 2021 (72% of the data occur-ring in 2000 and 2021). The methods used to collect the data were: live obser-vations/captures (43%), sign surveys (35%), camera trapping (16%),bioacoustics surveys (4%) and radiotracking, and inquiries that represent lessthan 1% of the records. The data set includes 13 types of records: (1) burrowsjsoil moundsjtunnel, (2) capture, (3) colony, (4) dead animaljhairjskullsjjaws, (5) genetic confirmation, (6) inquiries, (7) observation of live animal (8),observation in shelters, (9) photo trappingjvideo, (10) predators dietjpelletsjpine cones/nuts, (11) scatjtrackjditch, (12) telemetry and (13) vocalizationjecholocation. The spatial uncertainty of most records ranges between 0 and100 m (76%). Rodentia (n=31,573) has the highest number of records followedby Chiroptera (n=18,857), Carnivora (n=18,594), Lagomorpha (n=17,496),Cetartiodactyla (n=11,568) and Eulipotyphla (n=7008). The data setincludes records of species classified by the IUCN as threatened(e.g.,Oryctolagus cuniculus[n=12,159],Monachus monachus[n=1,512],andLynx pardinus[n=197]). We believe that this data set may stimulate thepublication of other European countries data sets that would certainly contrib-ute to ecology and conservation-related research, and therefore assisting onthe development of more accurate and tailored conservation managementstrategies for each species.     Pervasive gaps in Amazonian ecological research

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by 2050.
