This emerging field, abetted by government funding and subsidies, needs the capacity to explore new ideas and expand on existing technologies through facilities that enable and facilitate research and production. With increased threats of global warming, imminent climate change, and rising oil prices, solar power will be a critical component of America's future, and will require significant investment in the near term. To facilitate the solar industry and emerging clean energy technologies, this thesis project proposes an incubator for start-up companies involved with silicon-based technologies, thin-film technologies, or organic PVs to occupy and invest in. The premise of the program is to provide a series of shared spaces - wet labs, clean rooms, conference spaces, reception areas, and other communal resources - along with private spaces, including offices, research facilities, and meeting rooms. The project is both a R&D facility as well as a demonstration laboratory for new technologies. Located 12 miles north of Boston at the Industr-Plex Superfund site in Woburn, Massachusetts, the design aims to revitalize and environmentally reinvigorate the local area.by Alice Rosenberg.M.Arch

    Rapid profiling of DNA replication dynamics using mass spectrometry-based analysis of nascent DNA

    The primary method for probing DNA replication dynamics is DNA fiber analysis, which utilizes thymidine analog incorporation into nascent DNA, followed by immunofluorescent microscopy of DNA fibers. Besides being time-consuming and prone to experimenter bias, it is not suitable for studying DNA replication dynamics in mitochondria or bacteria, nor is it adaptable for higher-throughput analysis. Here, we present mass spectrometry-based analysis of nascent DNA (MS-BAND) as a rapid, unbiased, quantitative alternative to DNA fiber analysis. In this method, incorporation of thymidine analogs is quantified from DNA using triple quadrupole tandem mass spectrometry. MS-BAND accurately detects DNA replication alterations in both the nucleus and mitochondria of human cells, as well as bacteria. The high-throughput capability of MS-BAND captured replication alterations in an E. coli DNA damage-inducing gene library. Therefore, MS-BAND may serve as an alternative to the DNA fiber technique, with potential for high-throughput analysis of replication dynamics in diverse model systems

    Pre-existing virus-specific CD8+ T-cells provide protection against pneumovirus-induced disease in mice

    Pneumoviruses such as pneumonia virus of mice (PVM), bovine respiratory syncytial virus (bRSV) or human (h)RSV are closely related pneumoviruses that cause severe respiratory disease in their respective hosts. It is well-known that T-cell responses are essential in pneumovirus clearance, but pneumovirus-specific T-cell responses also are important mediators of severe immunopathology. In this study we determined whether memory- or pre-existing, transferred virus-specific CD8 + T-cells provide protection against PVM-induced disease. We show that during infection with a sublethal dose of PVM, both natural killer (NK) cells and CD8 + T-cells expand relatively late. Induction of CD8 + T-cell memory against a single CD8 + T-cell epitope, by dendritic cell (DC)-peptide immunization, leads to partial protection against PVM challenge and prevents Th2 differentiation of PVM-induced CD4 T-cells. In addition, adoptively transferred PVM-specific CD8 + T-cells, covering the entire PVM-specific CD8 + T-cell repertoire, provide partial protection from PVM-induced disease. From these data we infer that antigen-specific memory CD8 + T-cells offer significant protection to PVM-induced disease. Thus, CD8 + T-cells, despite being a major cause of PVM-associated pathology during primary infection, may offer promising targets of a protective pneumovirus vaccine

    HIV and early hospital readmission: evaluation of a tertiary medical facility in Lilongwe, Malawi

    Abstract Background Delivery of quality healthcare in resource-limited settings is an important, understudied public health priority. Thirty-day (early) hospital readmission is often avoidable and an important indicator of healthcare quality. Methods We investigated the prevalence of all-cause early readmission and its associated factors using age and sex adjusted risk ratios (RR) and 95% confidence intervals (CI). A retrospective review of the medical ward database at Kamuzu Central Hospital in Lilongwe, Malawi was conducted between February and December 2013. Results There were 3547 patients with an index admission of which 2776 (74.4%) survived and were eligible for readmission. Among these patients: 49.7% were male, mean age was 39.7 years, 36.1% were HIV-positive, 34.6% were HIV-negative, and 29.3% were HIV-unknown. The prevalence of early hospital readmission was 5.5%. Diagnoses associated with 30-day readmission were HIV-positive status (RR = 2.41; 95% CI: 1.64–3.53) and malaria (RR = 0.45; 95% CI: 0.22–0.91). Other factors associated with readmission were multiple diagnoses (excluding HIV) (RR = 1.52; 95% CI: 1.11–2.06), and prolonged length of stay (≥ 16 days) at the index hospitalization (RR = 3.63; 95% CI: 1.72–7.67). Conclusion Targeting HIV-infected inpatients with multiple diagnoses and longer index hospitalizations may prevent early readmission and improve quality of care

    The topography of transmembrane segment six is altered during the catalytic cycle of P-glycoprotein

    Structural evidence has demonstrated that P-glycoprotein (P-gp) undergoes considerable conformational changes during catalysis, and these alterations are important in drug interaction. Knowledge of which regions in P-gp undergo conformational alterations will provide vital information to elucidate the locations of drug binding sites and the mechanism of coupling. A number of investigations have implicated transmembrane segment six (TM6) in drug-P-gp interactions, and a cysteine-scanning mutagenesis approach was directed to this segment. Introduction of cysteine residues into TM6 did not disturb basal or drug-stimulated ATPase activity per se. Under basal conditions the hydrophobic probe coumarin maleimide readily labeled all introduced cysteine residues, whereas the hydrophilic fluorescein maleimide only labeled residue Cys-343. The amphiphilic BODIPY-maleimide displayed a more complex labeling profile. The extent of labeling with coumarin maleimide did not vary during the catalytic cycle, whereas fluorescein maleimide labeling of F343C was lost after nucleotide binding or hydrolysis. BODIPY-maleimide labeling was markedly altered during the catalytic cycle and indicated that the adenosine 5'-(beta,gamma-imino)triphosphate-bound and ADP/vanadate-trapped intermediates were conformationally distinct. Our data are reconciled with a recent atomic scale model of P-gp and are consistent with a tilting of TM6 in response to nucleotide binding and ATP hydrolysis

    Evaluation of status of commercial fish stocks in European marine subareas using mean trophic levels of fish landings and spawning stock biomass

    Most of the fish stocks in the world, including European fish stocks, are threatened by overfishing and/or degraded environmental conditions. Although the Common Fisheries Policy (CFP) is the main policy instrument managing fish stocks in Europe, there is continued concern as to whether commercial fish stocks will achieve Good Environmental Status (GEnS) in 2020 in accordance with the Marine Strategy Framework Directive (MSFD). In this context, the evaluation of the status of fish stocks in the subareas of FAO fishing area 27 was carried out using mean trophic levels (MTL) in fish landings and spawning stock biomass (SSB). Comparisons were made before and after 2008 to establish whether the trend is positive or negative. The main data sources for landings and SSB were the International Council for the Exploration of the Sea (ICES) advisory reports. MTLs in landing and SSB were determined for each subarea and the subareas were categorized into four groups, according to MTLs after 2008. The first group (subareas I + II, V) had higher MTL in landings and higher MTL in SSB after 2008. Therefore, fisheries in these subareas appear sustainable. The second group was subareas VIII + IX, for which the fish stocks have higher MTL in landings but low MTL in SSB, indicating that SSB was being overfished. The third was subarea (VI), where fish stocks have lower MTL in landings than those in SSB after 2008, which may indicate that fish stocks are recovering. Fish stocks in the fourth group (subareas III, IV and VII) had low MIL in landings and the MTL in SSB was lower than that of landings before 2008. This may be due to heavy fishing. In addition, we estimated the harvest rate (HR) of the fish stocks before and after 2008. The results showed that most of the fish stocks have lower HR after 2008, indicating that the status has improved, perhaps due to improvements in the implementation of CFP. However, some fish stocks showed high HR even after 2008, so that new management options are still needed. Other factors such as eutrophication, seafloor disturbances, marine pollution, invasive species etc., influence SSB ecosystem health options and should also be incorporated in the management criteria. Most of these environmental pressures are of high priority in the MSFD, and therefore the findings of this study will be useful for both CFP and MSFD. (C) 2016 The Authors.     Challenges in the Evaluation of Interventions to Improve Engagement Along the HIV Care Continuum in the United States: A Systematic Review.

    In the United States (US), there are high levels of disengagement along the HIV care continuum. We sought to characterize the heterogeneity in research studies and interventions to improve care engagement among people living with diagnosed HIV infection. We performed a systematic literature search for interventions to improve HIV linkage to care, retention in care, reengagement in care and adherence to antiretroviral therapy (ART) in the US published from 2007-mid 2015. Study designs and outcomes were allowed to vary in included studies. We grouped interventions into categories, target populations, and whether results were significantly improved. We identified 152 studies, 7 (5%) linkage studies, 33 (22%) retention studies, 4 (3%) reengagement studies, and 117 (77%) adherence studies. 'Linkage' studies utilized 11 different outcome definitions, while 'retention' studies utilized 39, with very little consistency in effect measurements. The majority (59%) of studies reported significantly improved outcomes, but this proportion and corresponding effect sizes varied substantially across study categories. This review highlights a paucity of assessments of linkage and reengagement interventions; limited generalizability of results; and substantial heterogeneity in intervention types, outcome definitions, and effect measures. In order to make strides against the HIV epidemic in the US, care continuum research must be improved and benchmarked against an integrated, comprehensive framework

    Public awareness and healthcare professional advice for obesity as a risk factor for cancer in the UK:a cross-sectional survey

    BACKGROUND: Overweight and obesity is the second biggest preventable cause of cancer after smoking, causing ~3.4 million deaths worldwide. This study provides current UK data on awareness of the link between obesity and cancer by socio-demographic factors, including BMI, and explores to what degree healthcare professionals provide weight management advice to patients. METHODS: Cross-sectional survey of 3293 adults completed an online survey in February/March 2016, weighted to be representative of the UK population aged 18+. RESULTS: Public awareness of the link between obesity and cancer is low (25.4% unprompted and 57.5% prompted). Higher levels of awareness existed for least deprived groups (P < 0.001), compared to more deprived groups. Most respondents had seen a healthcare practitioner in the past 12 months (91.6%) and 17.4% had received advice about their weight, although 48.4% of the sample were overweight/obese. CONCLUSION: Cancer is not at the forefront of people’s minds when considering health conditions associated with overweight or obesity. Socio-economic disparities exist in health knowledge across the UK population, with adults from more affluent groups being most aware. Healthcare professionals are uniquely positioned to provide advice about weight, but opportunities for intervention are currently under-utilized in healthcare settings
