    Droplet Microfluidics for the ex Vivo Expansion of Human Primary Multiple Myeloma Cells.

    We previously reported a new approach for micromanipulation and encapsulation of human stem cells using a droplet-based microfluidic device We demonstrated the possibility of encapsulating and culturing difficult-to-preserve primary human hematopoietic stem cells using an engineered double layered bead composed by an inner layer of alginate and an outer layer of puramatrix constructed using a soft technology without the use of any external force. In this work, we use this micro manipulation technique to build a 3D scaffold as a biomimetic model to recapitulate the niche of patient-derived multiple myeloma cells (MM cell) using a multilayered 3D tissue scaffold constructed in a microfluidic device and cultured in 10% FBS culture medium. In the current study, we included the use of this biomimetic model comprising supporting human Mesenchymal stem cells to show the mid-term survival of MM cells in the proposed structures. We found that the generated microniches were suitable for the maintenance of MM cells with and without supporting cells. Additionally, cultured MM cells in droplets were exposed to both Bortezomib and Lenalidomide to test their toxicity in the cultured patient derived cells. Results indicate that the maintained MM cells were consistently responding to the applied medication, opening a wide field of possibilities to use the presented micro device as an ex vivo platform for drug screening.This research was funded by European Commission H2020 Marie Curie Research Grants Scheme MSCA-IF-GF (705163).S

    Stellar energy loss rates beyond the standard model

    It is known that the dipolemoments of the neutrino lead to important astrophysical and cosmological effects. In this regard, within the context of a U(1)B−L model, we develop and present novel analytical formulas to assess the effects of the anomalous magnetic moment and electric dipole moment of the neutrino on the stellar energy loss rates through some common physical process of pair-annihilation e+e− → (γ, Z, Z ) → ν¯ν. Our results show that the stellar energy loss rates strongly depend on the effective magnetic moment of the neutrino, but also on the parameters which characterize the adopted U(1)B−L mode

    Las Tecnologías de la Información y la Comunicación (TIC) enriquecen el proceso enseñanza-aprendizaje; el uso de los ambientes en Realidad Aumentada como apoyo a los procedimientos odontológicos, a través de la utilización de procesos educativos en la preclínica de ortodoncia, surge como alternativa para mejorar la práctica de los estudiantes en diferentes rutinas. El objetivo general de la investigación fue diseñar, implementar y evaluar una aplicación de la Realidad Aumentada como apoyo a los procedimientos odontológicos. La investigación se realizó en tres fases. Al aplicar la estrategia didáctica propuesta se buscaba determinar si esta es una herramienta más eficaz que el método de enseñanza tradicional para la enseñanza de los contenidos relacionados con doblaje de alambres en ortodoncia.     Self-efficacy for weight control and quality of life in Mexican patients with hemophilia

    El autor es alumno egresado del Doctorado en Ciencias de la Salud becario de CONACYTIntroduction: Perceived self-efficacy is closely related to an adequate quality of life en general well-being of people, but few studies have studied this relationship in people with hemophilia. Objective: To determine the degree of relationship between perceived self-efficacy for weight control and quality of life in people with and without hemophilia, comparing both variables in case-control groups. Methods: The sample made up, for convenience, of 40 participants, with two matched groups. The group of cases consisted of 20 males between 19 and 24 years of age (M = 19.50, SD = 1.47) with hemophilia type A. The control group consisted of 20 males who did not present hemophilia, matched in age and weight status, between the ages of 18 and 24 years of age (M= 19.59, SD= 1.44). Participants answered the “Self-efficacy for Weight Control” questionnaire and the World Health Organization Quality of Life Bref” questionnaire. Descriptive statistics where used to describe the factors of each instrument. Student's t-test was used to compare differences between the two groups. Results: Only the Daily physical activity dimension showed a statistical difference in the control group. According to the Pearson correlation, a positive correlation was found between the Scheduled physical activity, Daily physical activity and Physical health dimensions. Conclusions: It is concluded that physical exercise, weight control, nutritional counseling and psychological support are essential for the quality of life, especially for people with hemophilia, which complements medical treatmentEl autor es alumno egresado del Doctorado en Ciencias de la Salud becario de CONACY

Methods: Cross-sectional study of prevalence, carried out on 838 members of the human talent in health (professionals, technicians, technologists, and auxiliaries) of Colombian health institutions in the study period with the application of the EASE instrument. Univariate analysis using descriptive statistical techniques, chi-square and Student’s t-test, and bivariate analysis with a Poisson regression model using the institucional SPSS v. 26. Results: The prevalence of adverse events in the last 5 years was 33.8%, presenting levels of acute stress qualifying as Medium-high emotional overload at 21.91%, while extreme acute stress was at 3.53%. The prevalence of risk for presenting acute stress after being involved in an adverse event was PR: 1.30 (CI: 1.24–1.36). Conclusion: Acute stress in human talent after adverse events is limiting health and care capacity and must be efficiently addressed by health institutions. Psychosocial risk is linked within the framework of the patient safety program and the institutional occupational health and safety management systems.publishersversionpublishe

    Case report: Cytokine hemoadsorption in a case of hemophagocytic lymphohistiocytosis secondary to extranodal NK/T-cell lymphoma

A full microbiological panel was carried out, including screening for imported disease, standard serologies and cultures for bacterial and fungal infection. A liver biopsy and bone marrow aspirate were performed, confirming the diagnosis. The patients fulfilled the HLH-2004 diagnostic criteria, and according to the 2018 Consensus Statements by the HLH Steering Committee of the Histiocyte Society, dexamethasone and etoposide were started. There was an associated hypercytokinemia and, due to refractory distributive shock, rescue therapy with cytokine hemoadsorption was performed during 24 h (within day 2 and 3 from ICU admission). After starting this procedure, rapid hemodynamic control was achieved with a significant reduction in vasopressor support requirements. This case report highlights that cytokine hemoadsorption can be an effective since rapid decrease in IL-10 levels and a significant hemodynamic improvement was achieved

    Synthesis and in vitro evaluation of an antiangiogenic cancerspecific dual-targeting 177Lu-Au-nanoradiopharmaceutical

    The aim of this research was to synthesize and chemically characterize a cancer-specific 177Lu-Aunanoradiopharmaceutical based on gold nanoparticles (NPs), the nuclear localization sequence (NLS)-Arg-Gly- Asp peptide and an aptamer (HS-pentyl-pegaptanib) to target both the a(v)b(3) integrin and the vascular endothelial growth factor (VEGF) overexpressed in the tumor neovasculature, as well as to evaluate by the tube formation assay, the nanosystem capability to inhibit angiogenesis. 177Lu-NP-RGD-NLS-Aptamer was obtained with a radiochemical purity of 99 ± 1%. Complete inhibition of tube formation (angiogenesis) was demonstrated when endothelial cells (EA.hy926), cultured in a 3D-extracellular matrix support, were treated with the developed nanosystem.Mexican National Council of Science and Technology (CONACYT-SEP-CB-2014-01-242443)

    A clinical and molecular portrait of non-metastatic anal squamous cell carcinoma

    Anal squamous cell carcinoma (ASCC) is a rare gastrointestinal malignancy associated with high-risk Human papillomavirus (HPV) infection. Despite improved outcomes in non-metastatic ASCC, definitive chemoradiotherapy constitutes the standard treatment for localized disease. Evidences for predictive and prognostic biomarkers are limited. Here, we performed a viral, immune, and mutational characterization of 79 non-metastatic ASCC patients with complete definitive chemoradiotherapy. HPV-16 was detected in 91% of positive cases in single infections (78%) or in coinfections with multiple genotypes (22%). Fifty-four percent of non-metastatic ASCC cases displayed mutations affecting cancer driver genes such as PIK3CA (21% of cases), TP53 (15%), FBXW7 (9%), and APC (6%). PD-L1 expression was detected in 57% of non-metastatic ASCC. Increased PD-L1 positive cases (67%) were detected in patients with complete response compared with non-complete response to treatment (37%) (p = 0.021). Furthermore, patients with PD-L1 positive tumors were significantly associated with better disease-free survival (DFS) and overall survival (OS) compared with patients with PD-L1 negative tumors (p = 0.006 and p = 0.002, respectively). PD-L1 expression strongly impacts CR rate and survival of non-metastatic ASCC patients after standard definitive chemoradiotherapy.
