    Neutron Autoradiography Combined With UV-C Sensitization: Toward the Intracellular Localization of Boron

    Our group has reported the imprint formation of biological material on polycarbonate nuclear track detectors by UV-C exposure, which is used as an approach to simultaneously visualize cell imprints and nuclear tracks coming from the boron neutron capture reaction. Considering that the cell nucleus has a higher UV-C absorption than the cytoplasm and that hematoxylin preferentially stains the nucleus, we proposed to enhance the contrast between these two main cell structures by hematoxylin staining before UV-C sensitization. In this study, several experiments were performed in order to optimize UV-C exposure parameters and chemical etching conditions for cell imprint formation using the SK-BR-3 breast cancer cell line. The proposed method improves significantly the resolution of the cell imprints. It allows clear differentiation of the nucleus from the rest of the cell, together with nuclear tracks pits. Moreover, it reduces considerably the UV-C exposure time, an important experimental issue. The proposed methodology can be applied to study the boron distribution independently from the chosen cell line and/or boron compounds.Fil: Gadan, Mario Alberto.     The use of GIS and Multi-Criteria evaluation (MCE) to determine scenarios of potential floodings in fluvial basins : study case: cuenca Guanabo, Habana city

    El trabajo muestra la potencialidad que presentan los Sistemas de Información Geográfica (SIG) y la evaluación multicriterio (EMC), para determinar con precisión, la cartografía de las zonas susceptibles al desarrollo de inundaciones recurrentes en cuencas fluviales. La capacidad de análisis y manejo de la información espacial que poseen los SIG en la actualidad, conjuntamente con el procesamiento estadístico apoyados en el uso de la EMC, permiten cuantificar factores del medio natural y establecer grados de adecuación, reduciendo la subjetividad durante el procesamiento analítico. Las zonas susceptibles determinadas con el empleo de esta metodología se comparó con estudios anteriores y los resultados presentan una buena coincidencia espacial aunque ésta mejora en precisión cartográfica. Sin duda los resultados obtenidos constituyen valiosas herramientas de gestión, los cuales ayudan a la toma de decisiones en los territorios e incrementan el nivel de conocimiento para reducir los riesgos de desastres naturales.The work shows the potential that Geographic Information Systems and Multi-Criteria Evaluation (EMC) have, to determine accurately, the cartography of favorable areas to recurrent floods development in fluvial basins. The analysis and special information handling capacity that the SIGs have nowadays, together with statistic processing resting in the (EMC) use, make it possible to quantify natural resources factors and establish degrees of fitness, reducing subjectivity during analytic processing. The capable zones determined with the employment of this methodology were compared with previous studies and the result presents a good spatial coincidence though this one improves in cartographic precision.     Pituitary Neoplasm Nomenclature Workshop: Does Adenoma Stand the Test of Time?

    The WHO Classification of Endocrine Tumours designates pituitary neoplasms as adenomas. A proposed nomenclature change to pituitary neuroendocrine tumors (PitNETs) has been met with concern by some stakeholder groups. The Pituitary Society coordinated the Pituitary Neoplasm Nomenclature (PANOMEN) workshop to address the topic. Experts in pituitary developmental biology, pathology, neurosurgery, endocrinology, and oncology, including representatives nominated by the Endocrine Society, European Society of Endocrinology, European Neuroendocrine Association, Growth Hormone Research Society, and International Society of Pituitary Surgeons. Clinical epidemiology, disease phenotype, management, and prognosis of pituitary adenomas differ from that of most NETs. The vast majority of pituitary adenomas are benign and do not adversely impact life expectancy. A nomenclature change to PitNET does not address the main challenge of prognostic prediction, assigns an uncertain malignancy designation to benign pituitary adenomas, and may adversely affect patients. Due to pandemic restrictions, the workshop was conducted virtually, with audiovisual lectures and written précis on each topic provided to all participants. Feedback was collated and summarized by Content Chairs and discussed during a virtual writing meeting moderated by Session Chairs, which yielded an evidence-based draft document sent to all participants for review and approval. There is not yet a case for adopting the PitNET nomenclature. The PANOMEN Workshop recommends that the term adenoma be retained and that the topic be revisited as new evidence on pituitary neoplasm biology emerges

    The European Renal Association - European Dialysis and Transplant Association Registry Annual Report 2014 : a summary

    Background: This article summarizes the European Renal Association - European Dialysis and Transplant Association Registry's 2014 annual report. It describes the epidemiology of renal replacement therapy (RRT) for end-stage renal disease (ESRD) in 2014 within 35 countries. Methods: In 2016, the ERA-EDTA Registry received data on patients who in 2014 where undergoing RRT for ESRD, from 51 national or regional renal registries. Thirty-two registries provided individual patient level data and 19 provided aggregated patient level data. The incidence, prevalence and survival probabilities of these patients were determined. Results: In 2014, 70 953 individuals commenced RRT for ESRD, equating to an overall unadjusted incidence rate of 133 per million population (pmp). The incidence ranged by 10-fold; from 23 pmp in the Ukraine to 237 pmp in Portugal. Of the patients commencing RRT, almost two-thirds were men, over half were aged >= 65 years and a quarter had diabetes mellitus as their primary renal diagnosis. By day 91 of commencing RRT, 81% of patients were receiving haemodialysis. On 31 December 2014, 490 743 individuals were receiving RRT for ESRD, equating to an unadjusted prevalence of 924 pmp. This ranged throughout Europe by more than 10-fold, from 157 pmp in the Ukraine to 1794 pmp in Portugal. In 2014, 19 406 kidney transplantations were performed, equating to an overall unadjusted transplant rate of 36 pmp. Again this varied considerably throughout Europe. For patients commencing RRT during 2005-09, the 5-year-adjusted patient survival probabilities on all RRT modalities was 63.3% (95% confidence interval 63.0-63.6). The expected remaining lifetime of a 20-to 24-year-old patient with ESRD receiving dialysis or living with a kidney transplant was 21.9 and 44.0 years, respectively. This was substantially lower than the 61.8 years of expected remaining lifetime of a 20-year-old patient without ESRD.Peer reviewe

    Clonal chromosomal mosaicism and loss of chromosome Y in elderly men increase vulnerability for SARS-CoV-2

    The pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2, COVID-19) had an estimated overall case fatality ratio of 1.38% (pre-vaccination), being 53% higher in males and increasing exponentially with age. Among 9578 individuals diagnosed with COVID-19 in the SCOURGE study, we found 133 cases (1.42%) with detectable clonal mosaicism for chromosome alterations (mCA) and 226 males (5.08%) with acquired loss of chromosome Y (LOY). Individuals with clonal mosaic events (mCA and/or LOY) showed a 54% increase in the risk of COVID-19 lethality. LOY is associated with transcriptomic biomarkers of immune dysfunction, pro-coagulation activity and cardiovascular risk. Interferon-induced genes involved in the initial immune response to SARS-CoV-2 are also down-regulated in LOY. Thus, mCA and LOY underlie at least part of the sex-biased severity and mortality of COVID-19 in aging patients. Given its potential therapeutic and prognostic relevance, evaluation of clonal mosaicism should be implemented as biomarker of COVID-19 severity in elderly people.     Inter-comparison of boron concentration measurements at INFN-University of Pavia (Italy) and CNEA (Argentina)

    An inter-comparison of three boron determination techniques was carried out between laboratories from INFN-University of Pavia (Italy) and CNEA (Argentina): alpha spectrometry (alpha-spect), neutron capture radiography (NCR) and quantitative autoradiography (QTA). Samples of different nature were analysed: liquid standards, liver homogenates and tissue samples from different treatment protocols. The techniques showed a good agreement in a concentration range of interest in BNCT (1-100ppm), thus demonstrating their applicability as precise methods to quantify boron and determine its distribution in tissues

    Production of IVF transgene-expressing bovine embryos using a novel strategy based on cell cycle inhibitors

    The objective was to evaluate the effects of cell cycle inhibitors (6-dimethylaminopurine [DMAP], and dehydroleukodine [DhL]) on transgene expression efficiency and on mosaic expression patterns of IVF bovine zygotes cytoplasmically injected with oolema vesicles coincubated with transgene. The DNA damage induced by the transgene or cell cycle inhibitors was measured by detection of phosphorylated histone H2AX foci presence (marker of DNA double-stranded breaks). Cloning of egfp blastomeres was included to determine continuity of expression after additional rounds of cellular division. The pCX-EGFP [enhanced green fluorescent protein gene (EGFP) under the chimeric cytomegalovirus IE-chicken-β-actin enhancer promoter control] gene plasmid (50 ng/μL) was injected alone (linear or circular exogenous DNA, leDNA and ceDNA, respectively) or associated with ooplasmic vesicles (leDNA-v or ceDNA-v). The effects of 2 mm DMAP or 1 μm DhL for 6 h (from 15 to 21 h post IVF) was evaluated for groups injected with vesicles. The DMAP increased (P < 0.05) egfp homogenous expression relative to transgene alone (21%, 18%, and 11% for leDNA-v + DMAP, leDNA-v, and leDNA, respectively) and also increased (P < 0.05) the phosphorylated histone H2AX foci area. Expression of egfp was higher (P < 0.05) for linear than for circular pCX-EGFP, and egfp blastocyst rates were higher (P < 0.05) for groups injected with linear transgene coincubated with vesicles than for linear transgene alone (95%, 77%, 84%, and 52% for leDNA-v + DMAP, leDNA-v + DhL, leDNA-v, and leDNA, respectively). Moreover, DMAP tended to improve egfp blastocysts rates for both circular and linear transgenes. Based on fluorescent in situ hybridization (FISH) analysis, there was evidence of integration in egfp embryos. Finally, clones derived from leDNA-v + DMAP had the highest egfp expression rates (96%, 65%, and 65% for leDNA-v + DMAP, leDNA-v, and leDNA, respectively).     Bases metodológicas y resultados de la implementación de la Encuesta Nacional de Niños, Niñas y Mujeres en México 2015

    Objetivo. Describir la metodología y los resultados del levantamiento de la Encuesta Nacional de Niños, Niñas y Mujeres en México (ENIM 2015). Material y métodos. La ENIM 2015 es una encuesta probabilística con muestreo polietápico, estratificado y por conglomerados, con representatividad regional, por estratos rural y urbano, y para la población indígena. Se aplicaron cuestionarios para obtener información sobre el hogar, mujeres de 15 a 49 años, niños y niñas menores de cinco años y niños y niñas y adolescentes de 5 a 17 años. Resultados. La tasa de respuesta en hogares y en mujeres fue de 94%; se obtuvo información de 10 760 hogares y 12 110 mujeres; para los niños y niñas y adolescentes y niños y niñas menores de cinco años, estos valores fueron de 98%, 11 607 y 8 066, respectivamente. Conclusión. El diseño probabilístico de la ENIM 2015 permite generar indicadores que se pueden desagregar en cinco regiones, para los estratos rural y urbano y para la población indígena, así como una línea base para 15 indicadores de los ODS

    Everything you must know about Azospirillum and its impact on agriculture and beyond

    Azospirillum is one of the most studied plant growth-promoting bacteria (PGPB); it represents a common model for plant-bacterial interactions. While Azospirillum brasilense is the species that is most widely known, at least 22 species, including 17 firmly validated species, have been identified, isolated from agricultural soils as well as habitats as diverse as contaminated soils, fermented products, sulfide springs, and microbial fuel cells. Over the last 40 years, studies on Azospirillum-plant interactions have introduced a wide array of mechanisms to demonstrate the beneficial impacts of this bacterium on plant growth. Multiple phytohormones, plant regulators, nitrogen fixation, phosphate solubilization, a variety of small-sized molecules and enzymes, enhanced membrane activity, proliferation of the root system, enhanced water and mineral uptake, mitigation of environmental stressors, and competition against pathogens have been studied, leading to the concept of the Multiple Mechanisms Hypothesis. This hypothesis is based on the assumption that no single mechanism is involved in the promotion of plant growth; it posits that each case of inoculation entails a combination of a few or many mechanisms. Looking specifically at the vast amount of information about the stimulatory effect of phytohormones on root development and biological nitrogen fixation, the Efficient Nutrients Acquisition Hypothesis model is proposed. Due to the existence of extensive agriculture that covers an area of more than 60 million hectares of crops, such as soybeans, corn, and wheat, for which the bacterium has proven to have some agronomic efficiency, the commercial use of Azospirillum is widespread in South America, with over 100 products already in the market in Argentina, Brazil, and Uruguay. Studies on Azospirillum inoculation in several crops have shown positive and variable results, due in part to crop management practices and environmental conditions. The combined inoculation of legumes with rhizobia and Azospirillum (co-inoculation) has become an emerging agriculture practice in the last several years, mainly for soybeans, showing high reproducibility and efficiency under field conditions. This review also addresses the use of Azospirillum for purposes other than agriculture, such as the recovery of eroded soils or the bioremediation of contaminated soils. Furthermore, the synthetic mutualistic interaction of Azospirillum with green microalgae has been developed as a new and promising biotechnological application, extending its use beyond agriculture.Fil: Cassan, Fabricio Dario.
