    Evaluation of the agronomic performance of 'Syrah' and 'Tempranillo' when grafted on 12 rootstocks

    Beyond pest resistance, rootstocks significantly influence the performance of grapevine varieties. However, the effect of the rootstock is strongly affected by its interaction with the environment, and it is therefore necessary to evaluate their influence in a particular terroir. With the aim of evaluating the influence of 12 rootstocks on the agronomic performance of 'Syrah' and 'Tempranillo', a trial was established in 2011 and 2012 in Miranda de Arga (Navarra, Spain), under the typical environmental conditions of the Ebro Valley. Growth and yield, as well as industrial and phenolic maturity parameters were analysed during four consecutive seasons (2015-2018). Most rootstocks showed a similar performance with both varieties, not always following the trends reported in bibliography, which highlights the relevance of studying rootstocks in different conditions. 3309 C was the rootstock conferring the highest vigour, whereas the lowest were provided by 420 A MGt and 'Fercal'. The implications on grape composition were much more diverse, and were partially conditioned by yield. Results were obtained during the four first harvests of the vineyard, and could therefore change to some extent as the vineyard reaches stability

    Intrinsically disordered chromatin protein NUPR1 binds to the enzyme PADI4

    The nuclear protein 1 (NUPR1) is an intrinsically disordered protein involved in stress-mediated cellular conditions. Its paralogue nuclear protein 1-like (NUPR1L) is p53-regulated, and its expression down-regulates that of the NUPR1 gene. Peptidyl-arginine deiminase 4 (PADI4) is an isoform of a family of enzymes catalyzing arginine to citrulline conversion; it is also involved in stress-mediated cellular conditions. We characterized the interaction between NUPR1 and PADI4 in vitro, in silico, and in cellulo. The interaction of NUPR1 and PADI4 occurred with a dissociation constant of 18 ± 6 μM. The binding region of NUPR1, mapped by NMR, was a hydrophobic polypeptide patch surrounding the key residue Ala33, as pinpointed by: (i) computational results; and, (ii) site-directed mutagenesis of residues of NUPR1. The association between PADI4 and wild-type NUPR1 was also assessed in cellulo by using proximity ligation assays (PLAs) and immunofluorescence (IF), and it occurred mainly in the nucleus. Moreover, binding between NUPR1L and PADI4 also occurred in vitro with an affinity similar to that of NUPR1. Molecular modelling provided information on the binding hot spot for PADI4. This is an example of a disordered partner of PADI4, whereas its other known interacting proteins are well-folded. Altogether, our results suggest that the NUPR1/PADI4 complex could have crucial functions in modulating DNA-repair, favoring metastasis, or facilitating citrullination of other proteins

    Objetivo: Analizar las características poblacionales y los métodos diagnósticos de patología cervical para la prevención del cáncer de cérvix de tres hospitales españoles para mejorar y unificar los programas de cribado y prevención. Material y métodos: Estudio retrospectivo de las características demográficas y clínicas de 408 mujeres con patología cervical uterina diagnosticadas en 3 hospitales españoles. Se comparan los factores de riesgo, el proceso diagnóstico y la indicación de tratamiento de dos grupos: las que requirieron conización cervical (n=222) y las que no precisaron tratamiento quirúrgico (n=186). También se analizan las recomendaciones vacunales y su grado de cumplimiento. Resultados: Las mujeres conizadas usaron más anticoncepción hormonal y tienen un mayor hábito tabáquico mientras que el número de compañeros sexuales es mayor en pacientes no conizadas. Más del 50% de pacientes con biopsia cervical positiva presentaron un resultado igual o más grave en la anatomía patológica de la pieza quirúrgica. Existen diferencias significativas en sensibilidad y valor predictivo positivo de la citología y de la determinación de HPV entre hospitales. La recomendación de vacunación en ambos grupos fue similar, el porcentaje de mujeres que no la cumplieron fue elevado y significativamente mayor entre pacientes conizadas. Conclusión: En nuestro medio las mujeres conizadas tienen características clínicas y epidemiológicas diferentes a las no conizadas, existen diferencias entre las técnicas diagnósticas de distintos hospitales y sin embargo la concordancia entre biopsia y resultado del cono es elevada. Sigue siendo necesaria una correcta educación sanitaria en relación con la vacunación en mujeres con patología cervical. Background: To analyse the characteristics of the population and diagnostic methods related to the cervical cancer prevention program in three different-level hospitals of a Spanish region in order to improve and unify the screening program. Methods: We retrospectively studied demographic and clinical characteristics of 408 women with cervical lesions diagnosed in three hospitals in Aragon (Spain). Correlation between risk factors, diagnosis process and conisation indication was analysed divided in two groups: conisation required (n=222) or non-conisation (n=186). We also assessed the number of vaccine recommendations made to the patients and the degree of compliance. Results: Conisaited women more frequently used a combined hormonal contraceptive method and are more smokers, while the sexual partners are more in women without conisation. More than 50% of women con positive biopsy was confirmed after surgical treatment. There are significant differences between sensibility and positive predictive value of pap-smear and HPV determination in different hospitals. The recombinant vaccine was recommended to both groups at a similar rate. The percentage of women who were recommended to receive the vaccine but chose not to do it, was high in both groups but significantly higher in the Conisation group. Conclusion: In our environment conisaited women have different clinical and epidemiological profiles, there are differences between diagnosis techniques in different hospitals, however, the concordance between biopsy and definitive result is high. A good sanitary education is necessary in relation with the vaccination of women with cervical pathology

    A transcriptomic snapshot of early molecular communication between Pasteuria penetrans and Meloidogyne incognita

    © The Author(s). 2018Background: Southern root-knot nematode Meloidogyne incognita (Kofoid and White, 1919), Chitwood, 1949 is a key pest of agricultural crops. Pasteuria penetrans is a hyperparasitic bacterium capable of suppressing the nematode reproduction, and represents a typical coevolved pathogen-hyperparasite system. Attachment of Pasteuria endospores to the cuticle of second-stage nematode juveniles is the first and pivotal step in the bacterial infection. RNA-Seq was used to understand the early transcriptional response of the root-knot nematode at 8 h post Pasteuria endospore attachment. Results: A total of 52,485 transcripts were assembled from the high quality (HQ) reads, out of which 582 transcripts were found differentially expressed in the Pasteuria endospore encumbered J2 s, of which 229 were up-regulated and 353 were down-regulated. Pasteuria infection caused a suppression of the protein synthesis machinery of the nematode. Several of the differentially expressed transcripts were putatively involved in nematode innate immunity, signaling, stress responses, endospore attachment process and post-attachment behavioral modification of the juveniles. The expression profiles of fifteen selected transcripts were validated to be true by the qRT PCR. RNAi based silencing of transcripts coding for fructose bisphosphate aldolase and glucosyl transferase caused a reduction in endospore attachment as compared to the controls, whereas, silencing of aspartic protease and ubiquitin coding transcripts resulted in higher incidence of endospore attachment on the nematode cuticle. Conclusions: Here we provide evidence of an early transcriptional response by the nematode upon infection by Pasteuria prior to root invasion. We found that adhesion of Pasteuria endospores to the cuticle induced a down-regulated protein response in the nematode. In addition, we show that fructose bisphosphate aldolase, glucosyl transferase, aspartic protease and ubiquitin coding transcripts are involved in modulating the endospore attachment on the nematode cuticle. Our results add new and significant information to the existing knowledge on early molecular interaction between M. incognita and P. penetrans.Peer reviewedFinal Published versio

    Estudios globales sobre el cadmio en relación con Theobroma cacao: Un análisis bibliométrico desde Scopus (1996 -2020)

    The maximum cadmium tolerance standards were established by the European Union in 2014, for the importation of cocoa-based products, causing concern in the countries. Global studies on Theobroma cacao research related to cadmium activity in the atmosphere were analyzed. Bibliometric analyses in R and VOSviewer programs were used to examine 64 documents published in the Scopus database according to keywords. We identified 811 keywords in the co-occurrence of terms, 5 thematic groups in the bibliographic coupling, 20 institutions as most important affiliations, 20 countries of origin of corresponding authors, 112 institutions in co-authorship network of which 5 are in primary documents, and two groups in thematic similarity in co-citation of documents. The United States leads the scientific production with 11 papers, followed by Colombia (8) and Ecuador (7). In 1996, the first scientific article was registered for the network, with increases of up to 11 publications by 2020. In conclusion, the need to strengthen and create more research networks between countries, institutions, authors, and co-authors is evident. It is hoped that the results will allow a comprehensive unraveling of the cadmium-cocoa research trajectory, while at the same time yielding new prospective research.En el año 2014, la Unión Europea impuso estándares máximos de tolerancia en cadmio, para la importación de productos a base de cacao, causando preocupación en los países. Se analizó estudios globales referidos a investigaciones en Theobroma cacao, relacionados con la actividad del cadmio en la atmósfera. Se utilizaron análisis bibliométricos en los programas R y VOSviewer, para examinar 64 documentos publicados en la base de datos Scopus según palabras clave. Se identificaron 811 palabras clave en coocurrencias de términos, 5 grupos temáticos en acoplamiento bibliográfico, 20 instituciones como afiliaciones más importantes, 20 países de procedencia de autores corresponsales, 112 instituciones en red de coautoría de los cuales 5 están en documentos primarios, y dos grupos en similaridad temática en co-citación de documentos. Estados Unidos lidera la producción científica con 11 documentos, seguido de Colombia (8) y Ecuador (7). En 1996 se registró el primer artículo científico para la red, con incrementos de hasta 11 publicaciones al 2020. En conclusión, se evidencia la necesidad de fortalecer y crear más redes de investigaciones entre países, instituciones, autores y coautores. Se espera que los resultados permitan desentrañar de manera integral la trayectoria de investigaciones cadmio-cacao, al tiempo que arrojen nuevas investigaciones prospectivas

    Alcohol Drinking and Health in Ageing: A Global Scale Analysis of Older Individual Data through the Harmonised Dataset of ATHLOS

    We investigated the relation between alcohol drinking and healthy ageing by means of a validated health status metric, using individual data from the Ageing Trajectories of Health: Longitudinal Opportunities and Synergies (ATHLOS) project. For the purposes of this study, the ATHLOS harmonised dataset, which includes information from individuals aged 65+ in 38 countries, was analysed (n = 135,440). Alcohol drinking was reflected by means of three harmonised variables: alcohol drinking frequency, current and past alcohol drinker. A set of 41 self-reported health items and measured tests were used to generate a specific health metric. In the harmonised dataset, the prevalence of current drinking was 47.5% while of past drinking was 26.5%. In the pooled sample, current alcohol drinking was positively associated with better health status among older adults ((b-coef (95% CI): 1.32(0.45 to 2.19)) and past alcohol drinking was inversely related (b-coef (95% CI): -0.83 (-1.51 to -0.16)) with health status. Often alcohol consumption appeared to be beneficial only for females in all super-regions except Africa, both age group categories (65-80 years old and 80+), both age group categories, as well as among all the financial status categories (all p < 0.05). Regional analysis pictured diverse patterns in the association for current and past alcohol drinkers. Our results report the need for specific alcohol intake recommendations among older adults that will help them maintain a better health status throughout the ageing process
