    Quantum dot interactions with and toxicity to Shewanella oneidensis MR-1

    Combining abiotic photosensitisers such as quantum dots (QDs) with non-photosynthetic bacteria presents an intriguing concept into the design of artificial photosynthetic organisms and solar-driven fuel production. Shewanella oneidensis MR-1 (MR-1) is a versatile bacterium concerning respiration, metabolism and biocatalysis, and is a promising organism for artificial photosynthesis as the bacterium's synthetic and catalytic ability provides a potential system for bacterial biohydrogen production. MR-1's hydrogenases are present in the periplasmatic space. It follows that for photoenergised electrons to reach these enzymes, QDs will need to be able to enter the periplasm, or electrons need to enter the periplasm via the Mtr pathway that is responsible for MR-1's extracellular electron transfer ability. As a step towards this goal, various QDs were tested for their photo-reducing potential, nanotoxicology and further for their interaction with MR-1. CdTe/CdS/TGA, CdTe/CdS/Cysteamine, a commercial, negatively charged CdTe and CuInS2/ZnS/PMAL QDs were examined. The photoreduction potential of the QDs was confirmed by measuring their ability to photoreduce methyl viologen with different sacrificial electron donors. The commercial CdTe and CuInS2/ZnS/PMAL QDs showed no toxicity towards MR-1 as evaluated by a colony-forming units method and a fluorescence viability assay. Only the commercial negatively charged CdTe QDs showed good interaction with MR-1. With transmission electron microscopy, QDs were observed both in the cytoplasm and periplasm. These results inform on the possibilities and bottlenecks when developing bionanotechnological systems for the photosynthetic production of biohydrogen by MR-1

    EveTAR: Building a Large-Scale Multi-Task Test Collection over Arabic Tweets

    This article introduces a new language-independent approach for creating a large-scale high-quality test collection of tweets that supports multiple information retrieval (IR) tasks without running a shared-task campaign. The adopted approach (demonstrated over Arabic tweets) designs the collection around significant (i.e., popular) events, which enables the development of topics that represent frequent information needs of Twitter users for which rich content exists. That inherently facilitates the support of multiple tasks that generally revolve around events, namely event detection, ad-hoc search, timeline generation, and real-time summarization. The key highlights of the approach include diversifying the judgment pool via interactive search and multiple manually-crafted queries per topic, collecting high-quality annotations via crowd-workers for relevancy and in-house annotators for novelty, filtering out low-agreement topics and inaccessible tweets, and providing multiple subsets of the collection for better availability. Applying our methodology on Arabic tweets resulted in EveTAR , the first freely-available tweet test collection for multiple IR tasks. EveTAR includes a crawl of 355M Arabic tweets and covers 50 significant events for which about 62K tweets were judged with substantial average inter-annotator agreement (Kappa value of 0.71). We demonstrate the usability of EveTAR by evaluating existing algorithms in the respective tasks. Results indicate that the new collection can support reliable ranking of IR systems that is comparable to similar TREC collections, while providing strong baseline results for future studies over Arabic tweets

    Immunisation with Recombinant PfEMP1 Domains Elicits Functional Rosette-Inhibiting and Phagocytosis-Inducing Antibodies to Plasmodium falciparum

    BACKGROUND: Rosetting is a Plasmodium falciparum virulence factor implicated in the pathogenesis of life-threatening malaria. Rosetting occurs when parasite-derived P. falciparum Erythrocyte Membrane Protein One (PfEMP1) on the surface of infected erythrocytes binds to human receptors on uninfected erythrocytes. PfEMP1 is a possible target for a vaccine to induce antibodies to inhibit rosetting and prevent severe malaria. METHODOLOGY/FINDINGS: We examined the vaccine potential of the six extracellular domains of a rosette-mediating PfEMP1 variant (ITvar9/R29var1 from the R29 parasite strain) by immunizing rabbits with recombinant proteins expressed in E. coli. Antibodies raised to each domain were tested for surface fluorescence with live infected erythrocytes, rosette inhibition and phagocytosis-induction. Antibodies to all PfEMP1 domains recognized the surface of live infected erythrocytes down to low concentrations (0.02-1.56 µg/ml of total IgG). Antibodies to all PfEMP1 domains except for the second Duffy-Binding-Like region inhibited rosetting (50% inhibitory concentration 0.04-4 µg/ml) and were able to opsonize and induce phagocytosis of infected erythrocytes at low concentrations (1.56-6.25 µg/ml). Antibodies to the N-terminal region (NTS-DBL1α) were the most effective in all assays. All antibodies were specific for the R29 parasite strain, and showed no functional activity against five other rosetting strains. CONCLUSIONS/SIGNIFICANCE: These results are encouraging for vaccine development as they show that potent antibodies can be generated to recombinant PfEMP1 domains that will inhibit rosetting and induce phagocytosis of infected erythrocytes. However, further work is needed on rosetting mechanisms and cross-reactivity in field isolates to define a set of PfEMP1 variants that could induce functional antibodies against a broad range of P. falciparum rosetting parasites

    Evaluation of the ECOSSE model for simulating soil organic carbon under Miscanthus and short rotation coppice-willow crops in Britain

    In this paper, we focus on the impact on soil organic carbon (SOC) of two dedicated energy crops: perennial grass Miscanthus x Giganteus (Miscanthus) and short rotation coppice (SRC)-willow. The amount of SOC sequestered in the soil is a function of site-specific factors including soil texture, management practices, initial SOC levels and climate; for these reasons, both losses and gains in SOC were observed in previous Miscanthus and SRC-willow studies. The ECOSSE model was developed to simulate soil C dynamics and greenhouse gas emissions in mineral and organic soils. The performance of ECOSSE has already been tested at site level to simulate the impacts of land-use change to short rotation forestry (SRF) on SOC. However, it has not been extensively evaluated under other bioenergy plantations, such as Miscanthus and SRC-willow. Twenty-nine locations in the United Kingdom, comprising 19 paired transitions to SRC-willow and 20 paired transitions to Miscanthus, were selected to evaluate the performance of ECOSSE in predicting SOC and SOC change from conventional systems (arable and grassland) to these selected bioenergy crops. The results of the present work revealed a strong correlation between modelled and measured SOC and SOC change after transition to Miscanthus and SRC-willow plantations, at two soil depths (0–30 and 0–100 cm), as well as the absence of significant bias in the model. Moreover, model error was within (i.e. not significantly larger than) the measurement error. The high degrees of association and coincidence with measured SOC under Miscanthus and SRC-willow plantations in the United Kingdom, provide confidence in using this process-based model for quantitatively predicting the impacts of future land use on SOC, at site level as well as at national level

    Harm minimisation for self-harm: a cross-sectional survey of British clinicians’ perspectives and practises

    ObjectiveHarm minimisation for self-harm is an alternative to preventive strategies and focuses on maximising safety when self-harming. We explored the views of clinicians on harm minimisation for self-harm to describe reported use and acceptability in clinical practice.DesignA cross-sectional study using an online survey consisting of fixed-choice and open-ended questions.SettingPrimary and secondary care practices in England, Scotland and Wales.ParticipantsSnowball sampling of UK-based clinicians (n=90; 67% female) working with people who self-harm and who have or have not previously recommended harm minimisation methods to patients.ResultsOf the 90 clinicians sampled, 76 (84%) reported having recommended harm minimisation techniques to people in their care who self-harm. Commonly recommended techniques were snapping rubber bands on one’s wrist and squeezing ice. Other techniques, such as teaching use of clean instruments when self-harming, were less likely to be recommended. Perceived client benefits included harm reduction and promotion of the therapeutic relationship. Perceived potential limitations of a harm minimisation approach for self-harm were (a) potential worsening of self-harm outcomes; (b) ethical reservations; (c) doubts about its effectiveness and appropriateness; and (d) lack of training and clear policies within the workplace.ConclusionsIn our sample of UK-based clinicians in various settings, harm minimisation for self-harm was broadly recommended for clients who self-harm due to perceived client benefits. However, future policies on harm minimisation must address clinicians’ perceived needs for training, well-defined guidelines, and clear evidence of effectiveness and safety to mitigate some clinician concerns about the potential for further harm.</jats:sec

    Health worker performance in the management of paediatric fevers following in-service training and exposure to job aids in Kenya

    BACKGROUND: Improving the way artemether-lumefantrine (AL) is provided to patients attending clinics is critical to maximize the benefit of this new medicine. In 2007, a new initiative was launched in one part of Kenya to improve malaria case-management through enhanced in-service training and provision of job aids. METHODS: An evaluation of the intervention using pre- and post-intervention cross sectional health facility surveys was conducted in Bondo district. The surveys included: audit of government health facilities, health worker structured interviews and exit interviews with caretakers of sick children below five years of age. The outcome indicators were the proportions of febrile children who had AL prescribed, AL dispensed, and four different dispensing and counseling tasks performed. RESULTS: At baseline 33 government health facilities, 48 health workers and 386 febrile child consultations were evaluated. At follow-up the same health facilities were surveyed and 36 health workers and 390 febrile child consultations evaluated. The findings show: 1) no health facility or health worker was exposed to all components of the intervention; 2) the proportion of health workers who received the enhanced in-service training was 67%; 3) the proportion of febrile children with uncomplicated malaria treated with the first-line anti-malarial drug, artemether-lumefantrine (AL), at health facilities where AL was in stock increased from 76.9% (95%CI: 69.4, 83.1) to 87.6% (95% CI: 82.5, 91.5); 4) there were modest but non-significant improvements in dispensing and counseling practices; and 5) when the analyses were restricted to health workers who received the enhanced in-service training and/or had received new guidelines and job aids, no significant improvements in reported case-management tasks were observed compared to baseline. CONCLUSION: In-service training and provision of job aids alone may not be adequate to improve the prescribing, dispensing and counseling tasks necessary to change malaria case-management practices and the inclusion of supervision and post-training follow-up should be considered in future clinical practice change initiatives

    Life cycle assessment of European anchovy (Engraulis encrasicolus) landed by purse seine vessels in northern Spain

    Purpose: The main purpose of this article is to assess the environmental impacts associated with the fishing operations related to European anchovy fishing in Cantabria (northern Spain) under a life cycle approach. Methods: The life cycle assessment (LCA) methodology was applied for this case study including construction, maintenance, use, and end of life of the vessels. The functional unit used was 1 kg of landed round anchovy at port. Inventory data were collected for the main inputs and outputs of 32 vessels, representing a majority of vessels in the fleet. Results and discussion: Results indicated, in a similar line to what is reported in the literature, that the production, transportation, and use of diesel were the main environmental hot spots in conventional impact categories. Moreover, in this case, the production and transportation of seine nets was also relevant. Impacts linked to greenhouse gas (GHG) emissions suggest that emissions were in the upper range for fishing species captured with seine nets and the value of global warming potential (GWP) was 1.44 kg CO2 eq per functional unit. The ecotoxicity impacts were mainly due to the emissions of antifouling substances to the ocean. Regarding fishery-specific categories, many were discarded given the lack of detailed stock assessments for this fishery. Hence, only the biotic resource use category was computed, demonstrating that the ecosystems' effort to sustain the fishery is relatively low. Conclusions: The use of the LCA methodology allowed identifying the main environmental hot spots of the purse seining fleet targeting European anchovy in Cantabria. Individualized results per port or per vessel suggested that there are significant differences in GHG emissions between groups. In addition, fuel use is high when compared to similar fisheries.     Deceptive body movements reverse spatial cueing in soccer

    This article has been made available through the Brunel Open Access Publishing Fund.The purpose of the experiments was to analyse the spatial cueing effects of the movements of soccer players executing normal and deceptive (step-over) turns with the ball. Stimuli comprised normal resolution or point-light video clips of soccer players dribbling a football towards the observer then turning right or left with the ball. Clips were curtailed before or on the turn (-160, -80, 0 or +80 ms) to examine the time course of direction prediction and spatial cueing effects. Participants were divided into higher-skilled (HS) and lower-skilled (LS) groups according to soccer experience. In experiment 1, accuracy on full video clips was higher than on point-light but results followed the same overall pattern. Both HS and LS groups correctly identified direction on normal moves at all occlusion levels. For deceptive moves, LS participants were significantly worse than chance and HS participants were somewhat more accurate but nevertheless substantially impaired. In experiment 2, point-light clips were used to cue a lateral target. HS and LS groups showed faster reaction times to targets that were congruent with the direction of normal turns, and to targets incongruent with the direction of deceptive turns. The reversed cueing by deceptive moves coincided with earlier kinematic events than cueing by normal moves. It is concluded that the body kinematics of soccer players generate spatial cueing effects when viewed from an opponent's perspective. This could create a reaction time advantage when anticipating the direction of a normal move. A deceptive move is designed to turn this cueing advantage into a disadvantage. Acting on the basis of advance information, the presence of deceptive moves primes responses in the wrong direction, which may be only partly mitigated by delaying a response until veridical cues emerge

    Self-Affirmation Improves Problem-Solving under Stress

    High levels of acute and chronic stress are known to impair problem-solving and creativity on a broad range of tasks. Despite this evidence, we know little about protective factors for mitigating the deleterious effects of stress on problem-solving. Building on previous research showing that self-affirmation can buffer stress, we tested whether an experimental manipulation of self-affirmation improves problem-solving performance in chronically stressed participants. Eighty undergraduates indicated their perceived chronic stress over the previous month and were randomly assigned to either a self-affirmation or control condition. They then completed 30 difficult remote associate problem-solving items under time pressure in front of an evaluator. Results showed that self-affirmation improved problem-solving performance in underperforming chronically stressed individuals. This research suggests a novel means for boosting problem-solving under stress and may have important implications for understanding how self-affirmation boosts academic achievement in school settings. © 2013 Creswell et al
