    Concepts and Benefits of Lunar Core Drilling

    Understanding lunar material at depth is critical to nearly every aspect of NASA s Vision and Strategic Plan. As we consider sending human s back to the Moon for brief and extended periods, we will need to utilize lunar materials in construction, for resource extraction, and for radiation shielding and protection. In each case, we will be working with materials at some depth beneath the surface. Understanding the properties of that material is critical, thus the need for Lunar core drilling capability. Of course, the science benefit from returning core samples and operating down-hole autonomous experiments is a key element of Lunar missions as defined by NASA s Exploration Systems Architecture Study.     Non-Raft AC2 Defines a cAMP Signaling Compartment That Selectively Regulates IL-6 Expression in Airway Smooth Muscle Cells

    Adenylyl cyclase (AC) isoforms differ in their tissue distribution, cellular localization, regulation, and protein interactions. Most cell types express multiple AC isoforms. We hypothesized that cAMP produced by different AC isoforms regulates unique cellular responses in human bronchial smooth muscle cells (BSMC). Overexpression of AC2, AC3, or AC6 had distinct effects on forskolin (Fsk)-induced expression of a number of known cAMP-responsive genes. These data show that different AC isoforms can differentially regulate gene expression. Most notable, overexpression and activation of AC2 enhanced interleukin 6 (IL-6) expression, but overexpression of AC3 or AC6 had no effect. IL-6 production by BSMC was induced by Fsk and select G protein-coupled receptor (GPCR) agonists, though IL-6 levels did not directly correlate with global cAMP levels. Treatment with PKA selective 6-Bnz-cAMP or Epac selective 8-CPT-2Me-cAMP cAMP analogs revealed a predominant role for PKA in cAMP-mediated induction of IL-6. IL-6 promoter mutations demonstrated that AP-1 and CRE transcription sites were required for Fsk to stimulate IL-6 expression. Our present study defines an AC2 cAMP signaling compartment that specifically regulates IL-6 expression in BSMC via Epac and PKA and demonstrates that other AC isoforms are excluded from this pool

    Assessing the rural food environment for advancing sustainable healthy diets: Insights from India

    World agricultural production has seen significant growth in the past four decades, yet malnutrition remains a persistent problem, particularly in the global south and more so in the rural areas. Need for a holistic approach to food systems is becoming crucial in designing policies that support the transition to sustainable and healthy diets. The present study is aimed to understand the rural food environment in the Telangana state in southern India by analyzing the combination of external and personal factors affecting food choices, attitudes, and consumption behavior. We developed a scoring-based methodology to assess the external and personal domains and dimensions to understand the food environment. The results showed that rural households favored carbohydrate-rich food groups obtained mostly from their own production or subsidized sources. On the other hand, protein and micronutrient-rich food groups were neglected due to affordability and preference for taste, cultural factors, and the limitations of external food environment. The findings of this study provide a deeper understanding of the food environment in low and middle-income countries (LMICs) conext. By highlighting the interplay between agriculture, food environments, and nutrition outcomes, this study contributes to the ongoing effort to address the global malnutrition crisis and support the development of healthier and more sustainable food systems. These findings can be useful to guide policy actions towards achieving food security and nutrition in the rural regions where food environments are under rapid transitions in the LMICs

    Cost–utility analysis of imatinib mesilate for the treatment of advanced stage chronic myeloid leukaemia

    Imatinib mesilate (Glivec®, Novartis Pharmaceuticals) is a novel therapy for the treatment of chronic myeloid leukaemia (CML). We evaluated the cost-effectiveness of imatinib (600 mg daily) when used for the treatment of patients in advanced stages of CML (accelerated phase and blast crisis) against conventional therapies of combination chemotherapy (DAT) and palliative care in hospital or at home. A Markov model simulated the transitions of hypothetical patient cohorts and outcomes were modelled for 5 years from the start of treatment. Costs were estimated from the perspective of the UK National Health Service. Over 5 years, a patient in accelerated phase will, on average, accrue an additional 2.09 QALYs with imatinib compared to conventional therapies, while patients in blast crisis will accrue an additional 0.58 quality-adjusted life-years (QALYs) with imatinib compared to conventional therapies. The costs per additional QALY gained from treatment with imatinib compared with conventional therapies were £29 344 (accelerated phase) and £42 239 (blast crisis). The results were particularly sensitive to the price of imatinib, improvements in quality of life, and the duration of haematological responses. We conclude that treatment of CML with imatinib confers considerably greater survival and quality of life than conventional treatments but at a cost

    Status of the light ion source developments at CEA/Saclay

    ACC NIMInternational audienceSILHI (High Intensity Light Ion Source) is an ECR ion source producing high intensity proton ordeuteron beams at 95 keV. It is now installed in the IPHI site building, on the CEA/Saclay center. IPHI is a frontend demonstrator of high power accelerator. The source regularly delivers more than 130 mA protons in CWmode and already produced more than 170 mA deuterons in pulsed mode at nominal energy. The last beamcharacterisations, including emittance measurements, space charge compensation analysis and diagnosticimprovements, will be reported. Taking into account the SILHI experience, new developments are in progress tobuild and test a 5 mA deuteron source working in CW mode. This new source will also operate at 2.45 GHz andpermanent magnets will provide the magnetic configuration. This source, of which the design will be discussed,will have to fit in with the SPIRAL 2 accelerator developed at GANIL to produce Radioactive Ion Beams.     Oxygen isotopic constraints on the origin and parent bodies of eucrites, diogenites, and howardites

    A few eucrites have anomalous oxygen isotopic compositions. To help understand their origin and identify additional samples, we have analyzed the oxygen isotopic compositions of 18 eucrites and four diogenites. Except for five eucrites, these meteorites have ?17O values that lie within 2 of their mean value viz., -0.242±0.016', consistent with igneous isotopic homogenization of Vesta. The five exceptional eucrites–NWA 1240, Pasamonte (both clast and matrix samples), PCA 91007, A-881394, and Ibitira–have ?17O values that lie respectively 4?, 5?, 5?, 15?, and 21 away from this mean value. NWA 1240 has a ?18O value that is 5? below the mean eucrite value. Four of the five outliers are unbrecciated and unshocked basaltic eucrites, like NWA 011, the first eucrite found to have an anomalous oxygen isotopic composition. The fifth outlier, Pasamonte, is composed almost entirely of unequilibrated basaltic clasts. Published chemical data for the six eucrites with anomalous oxygen isotopic compositions (including NWA 011) exclude contamination by chondritic projectiles as a source of the oxygen anomalies. Only NWA 011 has an anomalous Fe/Mn ratio, but several anomalous eucrites have exceptional Na, Ti, or Cr concentrations. We infer that the six anomalous eucrites are probably derived from five distinct Vesta-like parent bodies (Pasamonte and PCA 91007 could come from one body). These anomalous eucrites, like many unbrecciated eucrites from Vesta, are probably deficient in brecciation and shock effects because they were sequestered in small asteroids (~10 km diameter) during the Late Heavy Bombardment following ejection from Vesta-like bodies. The preservation of Vesta's crust and the lack of deeply buried samples from the hypothesized Vesta-like bodies are consistent with the removal of these bodies from the asteroid belt by gravitational perturbations from planets and protoplanets, rather than by collisonal grinding

    Terrorism, governmentality and the simulated city: the Boston Marathon bombing and the search for suspect two

    This article examines the online circulation of a photograph of the immediate aftermath of the 2013 Boston Marathon bombings taken by David Green, a Boston Marathon runner. The photograph fortuitously captured an image of Dzhokhar Tsarnaev, the younger Tsarnaev brother, running from the scene. Initially, Tsarnaev went unnoticed by online message board users and FBI investigators – he was just another face in the urban crowd. However, after he was identified as suspect two, he emerged in the photograph as the figure of terror, the condensed embodiment of the spectacular attack, and thus as a spectre, a figure whose appearance in the archive of the past haunts the future. The author examines how this spectral emergence simultaneously reveals the attack terrorism launches against everyday mediations and how everyday mediations respond to terrorist spectacle. He argues that photography sustains vernacular practices that also support the practices of urban governmentality. Understanding urban governmentality thus requires attending to the urban archive of visual mediation in which the relationships between past and present, image and reality, and surveillance and spectacle are always contingent and open to revision

    Light infrastructures and intimate publics in the vertical city

    This paper explores how the uneven distribution of light and darkness in the vertical city conditions residents’ capacities to form meaningful attachments to the places in which they dwell. Drawing on ethnographic material collected on a recent high-rise development in London, the paper explores how residents illuminate their homes in improvisatory engagement with the basic infrastructures that support their domestic lives and with the wider urban context. Four biographic vignettes reveal how people alter, adapt to or overturn the inadequacies of domestic infrastructures to carve out intimate spaces of inhabitation. The paper advances the idea of “light infrastructure” as a conceptual proposition for attending to the affective, esthetic and performative compositions of infrastructures in the night, and as an analytical proposition for developing a more hopeful and inclusive outlook on the ways people come to dwell, inhabit and feel at home in the vertical city at night

    Rab11 Is Required for Epithelial Cell Viability, Terminal Differentiation, and Suppression of Tumor-Like Growth in the Drosophila Egg Chamber

    The Drosophila egg chamber provides an excellent system in which to study the specification and differentiation of epithelial cell fates because all of the steps, starting with the division of the corresponding stem cells, called follicle stem cells, have been well described and occur many times over in a single ovary.Here we investigate the role of the small Rab11 GTPase in follicle stem cells (FSCs) and in their differentiating daughters, which include main body epithelial cells, stalk cells and polar cells. We show that rab11-null FSCs maintain their ability to self renew, even though previous studies have shown that FSC self renewal is dependent on maintenance of E-cadherin-based intercellular junctions, which in many cell types, including Drosophila germline stem cells, requires Rab11. We also show that rab11-null FSCs give rise to normal numbers of cells that enter polar, stalk, and epithelial cell differentiation pathways, but that none of the cells complete their differentiation programs and that the epithelial cells undergo premature programmed cell death. Finally we show, through the induction of rab11-null clones at later points in the differentiation program, that Rab11 suppresses tumor-like growth of epithelial cells. Thus, rab11-null epithelial cells arrest differentiation early, assume an aberrant cell morphology, delaminate from the epithelium, and invade the neighboring germline cyst. These phenotypes are associated with defects in E-cadherin localization and a general loss of cell polarity.While previous studies have revealed tumor suppressor or tumor suppressor-like activity for regulators of endocytosis, our study is the first to identify such activity for regulators of endocytic recycling. Our studies also support the recently emerging view that distinct mechanisms regulate junction stability and plasticity in different tissues
