    Phloem loading in the sucrose-export-defective (SXD-1) mutant maize is limited by callose deposition at plasmodesmata in bundle sheath-vascular parenchyma interface

    Using Lucifer Yellow we have demonstrated that the phloem-loading pathway from the mesophyll to the bundle sheath-vascular parenchyma interface in Zea mays source leaves follows a symplasmic route in small and intermediate vascular bundles in control as well as in the green sections of mutant sucrose-export-defective (SXD-1) plants. In the anthocyanin-rich mutant leaf sections, Lucifer Yellow transport was prohibited along the same path, at the bundle sheath-vascular parenchyma interface in particular. Plasmodesmata at the latter interface in SXD-1 anthocyanin-rich leaf sections appear to be structurally altered through callose deposition at the plasmodesmal orifices. We suggest that a transport bottleneck at the bundle sheath-vascular parenchyma interface is thus orchestrated and regulated through callose formation, preventing symplasmic transport across this important loading interface

    Pulmonary Effects of Neonatal Hydrocortisone Treatment in Ventilator-Dependent Preterm Infants

    Background/Objective. Hydrocortisone, administered to ventilated preterm neonates to facilitate extubation, has no adverse long-term effects, but short-term pulmonary effects have not been described previously. In the present study, we analyzed effects of hydrocortisone on ventilator settings and FiO2 in ventilator-dependent preterm infants. Patients and Methods. Fifty-five preterm children were included in this retrospective cohort study. Hydrocortisone was administered at a postnatal age of > 7 days to treat chronic lung disease (CLD). Ventilator settings before and after hydrocortisone administration were recorded as well as FiO2 at 36 weeks' gestational age. Presence of cerebral palsy was assessed at a mean corrected age of 24.1 months. Results. Hydrocortisone administered at a median postnatal age of 14 days significantly reduced FiO2 from a median of 0.39 to 0.30, mean airway pressure (MAP) from a median of 10.0 cm H2O to 7.6 cm H2O, and PaCO2 from a median of 53.5 mmHg to 47 mmHg. Extubation was achieved in all patients. CLD at 36 weeks was present in 11 of the 52 patients (21.1%). None developed cerebral palsy. Conclusions. Hydrocortisone was effective in reducing the FiO2, MAP, and PaCO2 and facilitated extubation. Hydrocortisone was not associated with cerebral palsy

    We consider here a dilute hot plasma which is homogeneous and in equilibrium in the framework of classical relativistic statistical mechanics as formulated recently by the authors. No external fields are present. The two-particle distribution function of the plasma is calculated for any temperature. This distribution will be physically significant, as long as the creation of pairs can be neglected. Several limits which apply at low or high temperatures are discussed.     Biological modelling of the radiation dose escalation effect of regional hyperthermia in cervical cancer

    Background Locoregional hyperthermia combined with radiotherapy significantly improves locoregional control and overall survival for cervical tumors compared to radiotherapy alone. In this study biological modelling is applied to quantify the effect of radiosensitization for three cervical cancer patients to evaluate the improvement in equivalent dose for the combination treatment with radiotherapy and hyperthermia. Methods The Linear-Quadratic (LQ) model extended with temperature-dependent LQ-parameters α and β was used to model radiosensitization by hyperthermia and to calculate the conventional radiation dose that is equivalent in biological effect to the combined radiotherapy and hyperthermia treatment. External beam radiotherapy planning was performed based on a prescription dose of 46Gy in 23 fractions of 2Gy. Hyperthermia treatment using the AMC-4 system was simulated based on the actual optimized system settings used during treatment. Results The simulated hyperthermia treatments for the 3 patients yielded a T50 of 40.1 °C, 40.5 °C, 41.1 °C and a T90 of 39.2 °C, 39.7 °C, 40.4 °C, respectively. The combined radiotherapy and hyperthermia treatment resulted in a D95 of 52.5Gy, 55.5Gy, 56.9Gy in the GTV, a dose escalation of 7.3–11.9Gy compared to radiotherapy alone (D95 = 45.0–45.5Gy). Conclusions This study applied biological modelling to evaluate radiosensitization by hyperthermia as a radiation-dose escalation for cervical cancer patients. This model is very useful to compare the effectiveness of different treatment schedules for combined radiotherapy and hyperthermia treatments and to guide the design of clinical studies on dose escalation using hyperthermia in a multi-modality setting

    MAXI J1659-152: the shortest orbital period black-hole binary

    Following the detection of a bright new X-ray source, MAXI J1659-152, a series of observations was triggered with almost all currently flying high-energy missions. We report here on XMM-Newton, INTEGRAL and RXTE observations during the early phase of the X-ray outburst of this transient black-hole candidate. We confirm the dipping nature in the X-ray light curves. We find that the dips recur on a period of 2.4139+/-0.0005 hrs, and interpret this as the orbital period of the system. It is thus the shortest period black-hole X-ray binary known to date. Using the various observables, we derive the properties of the source. The inclination of the accretion disk with respect to the line of sight is estimated to be 60-75 degrees. The companion star to the black hole is possibly a M5 dwarf star, with a mass and radius of about 0.15 M_sun and 0.23 R_sun, respectively. The system is rather compact (orbital separation is about 1.35 R_sun) and is located at a distance of roughly 7 kpc. We present an agglomeration of stellar polarization catalogs with results for 9286 stars. We have endeavored to eliminate errors, provide accurate (arcsecond) positions, sensibly weight multiple observations of the same star, and provide reasonable distances.     Increased susceptibility to Chrysanthemum Yellows phytoplasma infection in Atcals7ko plants is accompanied by enhanced expression of carbohydrate transporters

    Main conclusion: Loss of CALS7 appears to confer increased susceptibility to phytoplasma infection in Arabidopsis, altering expression of genes involved in sugar metabolism and membrane transport. Abstract: Callose deposition around sieve pores, under control of callose synthase 7 (CALS7), has been interpreted as a mechanical response to limit pathogen spread in phytoplasma-infected plants. Wild-type and Atcals7ko mutants were, therefore, employed to unveil the mode of involvement of CALS7 in the plant’s response to phytoplasma infection. The fresh weights of healthy and CY-(Chrysanthemum Yellows) phytoplasma-infected Arabidopsis wild type and mutant plants indicated two superimposed effects of the absence of CALS7: a partial impairment of photo-assimilate transport and a stimulated phytoplasma proliferation as illustrated by a significantly increased phytoplasma titre in Atcal7ko mutants. Further studies solely dealt with the effects of CALS7 absence on phytoplasma growth. Phytoplasma infection affected sieve-element substructure to a larger extent in mutants than in wild-type plants, which was also true for the levels of some free carbohydrates. Moreover, infection induced a similar upregulation of gene expression of enzymes involved in sucrose cleavage (AtSUS5, AtSUS6) and transmembrane transport (AtSWEET11) in mutants and wild-type plants, but an increased gene expression of carbohydrate transmembrane transporters (AtSWEET12, AtSTP13, AtSUC3) in infected mutants only. It remains still unclear how the absence of AtCALS7 leads to gene upregulation and how an increased intercellular mobility of carbohydrates and possibly effectors contributes to a higher susceptibility. It is also unclear if modified sieve-pore structures in mutants allow a better spread of phytoplasmas giving rise to higher titre

    The sieve-element endoplasmic reticulum: A focal point of phytoplasma-host plant interaction?

    The rough endoplasmic reticulum (r-ER) is of paramount importance for adaptive responses to biotic stresses due to an increased demand for de novo synthesis of immunity-related proteins and signaling components. In nucleate cells, disturbance of r-ER integrity and functionality leads to the “unfolded protein response” (UPR), which is an important component of innate plant immune signalling. In contrast to an abundance of reports on r-ER responses to biotic challenges, sieve-element endoplasmic reticulum (SE-ER) responses to phytoplasma infection have not been investigated. We found that morphological SE-ER changes, associated with phytoplasma infection, are accompanied by differential expression of genes encoding proteins involved in shaping and anchoring the reticulum. Phytoplasma infection also triggers an increased release of bZIP signals from the (SE-ER)/r-ER and consequent differential expression of UPR-related genes. The modified expression patterns seem to reflect a trade-off between survival of host cells, needed for the phytoplasmic biotrophic lifestyle, and phytoplasmas. Specialized plasmodesmata between sieve element and companion cell may provide a corridor for transfer of phytoplasma effectors inducing UPR-related gene expression in companion cells

    Detailed Radio to Soft Gamma-ray Studies of the 2005 Outburst of the New X-ray Transient XTE J1818-245

    XTE J1818-245 is an X-ray nova that experienced an outburst in 2005, first seen by the RXTE satellite. The source was observed simultaneously at various wavelengths up to soft gamma-rays with the INTEGRAL satellite, from 2005 February to September. X-ray novae are extreme systems that often harbor a black hole, and are known to emit throughout the electromagnetic spectrum when in outburst. We analyzed radio, (N)IR, optical, X-ray and soft gamma-ray observations and constructed simultaneous broad-band X-ray spectra. Analyzing both the light curves in various energy ranges and the hardness-intensity diagram enabled us to study the long-term behavior of the source. Spectral parameters were typical of the Soft Intermediate States and the High Soft States of a black hole candidate. The source showed relatively small spectral variations in X-rays with considerable flux variation in radio. Spectral studies showed that the accretion disc cooled down from 0.64 to 0.27 keV in 100 days and that the total flux decreased while the relative flux of the hot medium increased. Radio emission was detected several times, and, interestingly, five days after entering the HSS. Modeling the spectral energy distribution from the radio to the soft gamma-rays reveals that the radio flares arise from several ejection events. XTE J1818-245 is likely a black hole candidate transient source that might be closer than the Galactic Bulge. The results from the data analysis trace the physical changes that took place in the system at a maximum bolometric luminosity of (0.4-0.9)e38 erg/s (assuming a distance between 2.8-4.3 kpc) and they are discussed within the context of disc and jet models.Comment: Accepted for publication in Astronomy and Astrophysics. 11 Figures, 3 Table

    Effects of Antenatal Glucocorticoid Therapy on Hippocampal Histology of Preterm Infants

    Objective: To investigate if antenatal glucocorticoid treatment has an effect on hippocampal histology of the human preterm newborn. Patients and Methods: Included were consecutive neonates with a gestational age between 24 and 32 weeks, who were born between 1991 to 2009, who had died within 4 days after delivery and underwent brain autopsy. Excluded were neonates with congenital malformations and neonates treated postnatally with glucocorticoids. The brains were routinely fixed, samples of the hippocampus were stained with haematoxylin and eosin and sections were examined for presence or absence of large and small neurons in regions of the hippocampus. Additional staining with GFAP, neurofilament and vimentin was performed to evaluate gliosis and myelination. The proliferation marker Ki67 was used to evaluate neuronal proliferation. Staining with acid fuchsin-thionin was performed to evaluate ischemic damage. Results: The hippocampi of ten neonates who had been treated with antenatal glucocorticoids showed a lower density of large neurons (p = 0.01) and neurons irrespective of size (p = 0.02) as compared to eleven neonates who had not been treated with glucocorticoids. No difference was found in density of small neurons, in myelination, gliosis, proliferation or ischemic damage.
