    Patterns of long‐term vegetation change vary between different types of semi‐natural grasslands in Western and Central Europe

    Questions: Has plant species richness in semi‐natural grasslands changed over recent decades? Do the temporal trends of habitat specialists differ from those of habitat generalists? Has there been a homogenization of the grassland vegetation? Location: Different regions in Germany and the UK. Methods: We conducted a formal meta‐analysis of re‐survey vegetation studies of semi‐natural grasslands. In total, 23 data sets were compiled, spanning up to 75 years between the surveys, including 13 data sets from wet grasslands, six from dry grasslands and four from other grassland types. Edaphic conditions were assessed using mean Ellenberg indicator values for soil moisture, nitrogen and pH. Changes in species richness and environmental variables were evaluated using response ratios. Results: In most wet grasslands, total species richness declined over time, while habitat specialists almost completely vanished. The number of species losses increased with increasing time between the surveys and were associated with a strong decrease in soil moisture and higher soil nutrient contents. Wet grasslands in nature reserves showed no such changes or even opposite trends. In dry grasslands and other grassland types, total species richness did not consistently change, but the number or proportions of habitat specialists declined. There were also considerable changes in species composition, especially in wet grasslands that often have been converted into intensively managed, highly productive meadows or pastures. We did not find a general homogenization of the vegetation in any of the grassland types. Conclusions: The results document the widespread deterioration of semi‐natural grasslands, especially of those types that can easily be transformed to high production grasslands. The main causes for the loss of grassland specialists are changed management in combination with increased fertilization and nitrogen deposition. Dry grasslands are most resistant to change, but also show a long‐term trend towards an increase in more mesotrophic species

    The Effect of Bifidobacterium on Reducing Symptomatic Abdominal Pain in Patients with Irritable Bowel Syndrome: A Systematic Review

    Probiotics, specifically Bifidobacteria, may improve abdominal pain in patients with irritable bowel syndrome (IBS); however, results from randomised controlled trials (RCTs) are conflicting. Here, we systematically reviewed the efficacy of Bifidobacteria on abdominal pain in IBS. We searched MEDLINE, EMBASE and the Cochrane Controlled Trials Register from inception to 20 May 2019, without language or date restrictions. The search strategy comprised of the combination of three concepts: supplementation, abdominal pain, and IBS. Inclusion criteria included double-blind placebo-controlled RCTs featuring Bifidobacteria supplementation in Rome-diagnosed IBS patients. A total of 8 RCTs involving a total of 1045 patients with Rome diagnosed IBS were included. The dose of total Bifidobacteria ranged from 106 to > 1011 cfu (colony-forming unit) and duration of supplementation ranged between 2 and 8 weeks. Bifidobacteria was delivered through either intake of fermented milk products, encapsulation or via a malted milk beverage, with all studies assessing abdominal pain via a visual analogue Likert scale. From the studies included, 50% (n = 4) of studies found a statistically significant improvement in abdominal pain following Bifidobacteria supplementation compared to placebo, 38% (n = 3) of studies found non-significant improvements and 12% (n = 1) showed a statistically significant dose-response effect of improvement. The evidence shows a heterogeneity of effect for Bifidobacteria dependent upon strain, dosage and delivery method. While not all studies demonstrate significant improvements in abdominal pain, none of the selected studies reported an increase in pain or other adverse effects

    Phylogeography and Molecular Evolution of Potato virus Y

    Potato virus Y (PVY) is an important plant pathogen, whose host range includes economically important crops such as potato, tobacco, tomato, and pepper. PVY presents three main strains (PVYO, PVYN and PVYC) and several recombinant forms. PVY has a worldwide distribution, yet the mechanisms that promote and maintain its population structure and genetic diversity are still unclear. In this study, we used a pool of 77 complete PVY genomes from isolates collected worldwide. After removing the effect of recombination in our data set, we used Bayesian techniques to study the influence of geography and host species in both PVY population structure and dynamics. We have also performed selection and covariation analyses to identify evolutionarily relevant amino acid residues. Our results show that both geographic and host-driven adaptations explain PVY diversification. Furthermore, purifying selection is the main force driving PVY evolution, although some indications of positive selection accounted for the diversification of the different strains. Interestingly, the analysis of P3N-PIPO, a recently described gene in potyviruses, seems to show a variable length among the isolates analyzed, and this variability is explained, in part, by host-driven adaptation

    Using incomplete floristic monitoring data from habitat mapping programmes to detect species trends

    Aim: The loss of biodiversity has raised serious concerns about the entailing losses of ecosystem services. Here, we explore the potential of repeated habitat mapping data to identify floristic changes over time. Using one German federal state as a case study, we assessed floristic changes between the 1980s and 2010s. These habitat data have great potential for analysis because of their high spatial coverage while also posing methodological challenges such as incomplete observation data. We developed a modelling approach that accounts for incomplete observations and explored the ability to detect temporal trends. Location: The Federal State of Schleswig‐Holstein (Germany) Methods: We compiled plant species lists from the earliest (1980s) and most recent (2010s) habitat mapping survey and aligned differing habitat definitions across mapping campaigns. A total of 5,503 mapped polygons, each with a list of species records, intersected the two surveys. We accounted for underrecorded species by assigning occurrence probabilities, based on species co‐occurrence information across all surveys, using Beals' index and tested the robustness of this approach by simulation experiments. For those species with significant increases and decreases in occurrence probability, we linked these trends to the species' functional characteristics. Results: We found a systematic loss of species that are moderately threatened. Species that indicate low nitrogen supply and high soil moisture declined, suggesting a shift towards a more eutrophic and drier landscape. Importantly, assessing specific plant traits associated with losses, we also detected a decrease in species with reddish and blueish flowers and species providing nectar, pointing to a decrease of insect‐pollinated taxa. Main conclusions: The identified changes raise concerns that plant biodiversity has fundamentally changed over the last three decades, with concomitant consequences for ecosystem services, especially pollination. Given the general lack of historical standardized data, our approach for trend analyses using incomplete observation data may be widely applicable to assess long‐term biodiversity change

    Resistance Breaking Tomato Spotted Wilt Virus Isolates on Resistant Tomato Cultivars in Italy

    In summer 2012, typical symptoms of Tomato spotted wilt disease occurred on tomato plants of different resistant cultivars (carrying the Sw5 gene), in different fields in Foggia province (Apulia region, southern Italy). The percentage of symptomatic plants was 20-30% and, in particular cases of advanced stage of cultivation, it reached up to 50% at the end of cycle. All TSWV isolates induced similar, very serious, systemic symptoms of bronzing in all resistant, infected tomato hybrids. Leaf extracts from these samples were tested by ELISA for the detection of TSWV, CMV, TMV, ToMV, INSV, PVY, AMV, PepMV, PVX, TYLCV, TYLCSV, PZSV and by molecular hybridization for the detection of PSTVd and CLVd. TSWV was detected in all the tested samples. The correspondent virus isolate was inoculated mechanically and by Frankliniella occidentalis on to a set of different tomato hybrids, in order to investigate for its ability to overcome the resistance gene Sw5, beyond that on some herbaceous test plants. The TSWV isolate, after three single lesion passages in Nicotiana glutinosa, induced symptoms typical of TSWV in mechanically inoculated seedlings grown from the same seed lots as the field-grown tomato plants. The virus isolate, designated TSWV-RB-TI, inoculated to plants of five commercial tomato hybrids carrying the Sw5 gene (that had previously shown resistance to different TSWV isolates), and five pepper hybrids carrying the Tsw gene, was later detected by ELISA in most inoculated tomato plants of each hybrid: systemic infection occurred a few days before local lesions appeared in inoculated leaves as a consequence of the typical hypersensitivity reaction caused by the Sw5 gene. By contrast, pepper hybrids carrying the Tsw gene were uninfected by TSWV-RB-TI isolate. The RB isolate did not differ noticeably in transmission efficiency when it was tested with the thrips F. occidentalis. This is not the first report of a TSWV strain infecting tomato plants carrying the Sw5 gene. Our observations demonstrate that new evolved strains of TSWV have emerged in southern Italy in 2012, able to overcome the Sw5 resistance gene in tomato hybrids previously known to be resistant to the virus
